目的

观察艾灸对类风湿关节炎（RA）模型大鼠膝关节滑膜组织环鸟苷酸-腺苷酸合成酶（cGAS）/干扰素基因刺激因子（STING）信号通路的影响，探讨艾灸减轻RA模型大鼠滑膜炎性损伤的作用机制。
方法

SD大鼠随机分为空白组、模型组、艾灸组及雷公藤总甙组，每组10只。采用右后足趾注射弗氏完全佐剂结合风、寒、湿环境建立RA大鼠模型。艾灸组每日艾灸“足三里”或“肾俞”20 min；雷公藤总甙组每日给予雷公藤总甙混悬液（8 mg/kg）灌胃。以上两组干预均每日1次，持续15 d。排水法测量各组大鼠右后足趾关节肿胀度；HE染色法观察各组大鼠膝关节滑膜形态变化；透射电镜观察滑膜细胞线粒体结构变化；Western blot法检测各组大鼠膝关节滑膜组织cGAS、STING、TANK结合激酶1（TBK1）、磷酸化干扰素调节因子３（p-IRF3）和磷酸化核转录因子-κB p65（p-NF-κB p65）蛋白表达水平；ELISA法测定各组大鼠血清白细胞介素6（IL-6）、细胞间黏附分子-1（ICAM-1）、CXC趋化因子配体10（CXCL10）、干扰素β（IFN-β）的含量。
结果

与空白组比较，模型组大鼠关节肿胀度增大（

0.01），滑膜内衬细胞分布不规则、增生明显，可见大量炎性细胞浸润；滑膜组织中cGAS、STING、TBK1、p-IRF3、p-NF-κB p65蛋白表达升高（

<
0.01），血清中IL-6、ICAM-1、CXCL10、IFN-β含量升高（
P
<
0.01）；与模型组比较，艾灸组和雷公藤总甙组大鼠关节肿胀度减小（
P
<
0.01），滑膜内衬细胞分布较为规则、增生改善明显、出现少量的炎性细胞；滑膜组织中cGAS、STING、TBK1、p-IRF3、p-NF-κB p65蛋白表达降低（
P
<
0.01），血清中IL-6、ICAM-1、CXCL10、IFN-β含量降低（
P
<
0.01）。艾灸组与雷公藤总甙组比较，各项指标差异无统计学意义。
结论

艾灸能够减轻RA模型大鼠关节肿胀，降低炎性细胞因子的释放，改善滑膜炎性损伤，可能与其抑制cGAS/STING信号通路有关。
Abstract
Objective

To investigate the effect of moxibustion on the cyclic GMP-AMP synthase （cGAS）/stimulator of interferon genes （STING） signaling pathway in the synovial tissue of knee joints in rheumatoid arthritis （RA） model rats， and to explore the mechanism by which moxibustion alleviates synovial inflammatory injury.
Methods

Forty male SD rats were randomly divided into 4 groups （10 rats in each group）： blank， model， moxibustion， and triptolide groups. The RA model was induced by injecting complete Freund’s adjuvant into the right hind toe combined with exposure to wind， cold， and damp environments. The moxibustion group received daily 20 min treatment at “Zusanli” （ST36） or “Shenshu” （BL23） for 15 d； the triptolide group was administered triptolide suspension （8 mg/kg） via gavage for 15 d. The joint swelling degree of right hind toe was measured by water displacement； the synovial morphology was assessed via HE staining； the mitochondrial ultrastructure was observed by transmission electron microscopy； the synovial protein expression levels of cGAS， STING， TANK-binding kinase 1 （TBK1）， phosphorylated interferon regulatory factor 3 （p-IRF3） and phosphorylated nuclear factor-kappa B p65（p-NF-κB p65） were detected by Western blot； serum contents of interleukin-6 （IL-6）， intercellular adhesion molecule-1 （ICAM-1）， C-X-C motif chemokine ligand 10 （CXCL10）， and interferon-β （IFN-β） were measured by ELISA.
Results

Compared with the blank group， the model group showed increased joint swelling degree （

0.01）， irregular synovial lining cell distribution， marked hyperplasia， and inflammatory cell infiltration； synovial cGAS， STING， TBK1， p-IRF3， and p-NF-κB p65 protein expression levels and serum IL-6， ICAM-1， CXCL10， and IFN-β contents were significantly elevated （

0.01）. Compared with the model group， both moxibustion and triptolide interventions reduced joint swelling degree （
P
<
0.01）， improved synovial histopathology， and down regulated the aforementioned proteins and cytokines （
P
<
0.01）. No significant differences were observed between the moxibustion and triptolide groups.
Conclusion

Moxibustion attenuates joint swelling， inflammatory cytokine release， and synovial injury in RA model rats， potentially via suppression of the cGAS/STING pathway.
