      摘要:Objective: To observe the effect of intracerebroventricular(icv) injection of Sincalide(CCK-8) on the electroacupuncture(EA) analgesia by using discharges of neurons of Caudate nucleus(Cd) and radiant heat irradiation induced tail-flick reflex as the indexes.Methods: Eighty-six Wistar rats anesthetized by 20% urethane((1.0 g/kg)) were randomized into control,EA and EA+CCK-8 groups.Extracellular discharges of pain-excited neurons(PEN) and pain-inhibitory neurons(PIN) in Cd were recorded by using glass micropipettes(0.5-(1.0 μm) in diameter,10-(30 mΩ) in resistance).Radiant heat irradiation induced tail-flick latency (TFL) was used as the pain threshold index.EA((6 V),(15 Hz) and continuous electric pulses) was applied to bilateral "Zusanli"(ST 36) for(15 min).Fifteen minutes after EA stimulation,CCK-8((10 ng)/(10 μL)) was injected slowly into the lateral cerebral ventricle(A:(-0.2 mm);R:(1.5 mm);H:(3.0 mm)) in a duration of(2 min).Results: 1) A total of 105 neurons including 63 PEN and 42 PIN were recorded in Cd.The radiant heat induced TFL simultaneously caused an increase in the discharge of PEN or a reduction in that of PIN,showing a painful effect of radiant heat on both the rat's behavior and central neuronal activity.2) EA at bilateral "Zusanli"(ST 36) resulted in an inhibition of the electrical activity of PEN as well as a potentiation of the electrical activity of PIN and a prolongation of TFL,exhibiting an analgesic effect of EA.3) The analgesic effect of EA on PEN or PIN and TFL were simultaneously antagonized by icv injection of CCK-8((10 ng)/(10 μL)).Conclusion: The antagonism of CCK-8 to analgesic effect of EA shows the coordinative and consistent actions on the levels of discharges of central pain-related neurons and the whole behavior reflex.It suggests that reducing concentration of CCK-8 in the brain can potentiate the analgesic effect of clinical acupuncture,and the electrical activities of PEN and PIN being as the indexes of pain and analgesia are indeed feasible.

      摘要:Objective: To study the effect of electroacupuncture(EA) of "Dazhui"(GV 14) and "Mingmen"(GV 4) on hypothalamic corticotrophin releasing hormone(CRH),plasma adrenocorticotropic hormone(ACTH),serum cortisol(Cort) and tumor necrosis factor(TNF)-α contents in adjuvant arthritis rats.Methods: Thirty-six male Wistar rats were evenly randomized into normal control,model,EA-GV14 and EA-GV4 groups.Arthritis model was established by subcutaneous injection of Freund's complete adjuvant(FCA,(0.1 mL)) into the right hind-paw.EA((15 Hz),continuous pulses,(1 mA)) was applied to "Dazhui"(GV 14) and "Mingmen"(GV 4) for(20 min),once every other day and continuously for 8 times.After the rats were killed,hypothalamic tissue and blood sample were taken for detecting hypothalamic CRH,plasma ACTH,serum Cort and TNF-α contents with radioimmunoassay.Results: Compared with normal control group,the swelling rate of the right hindpaw(24 h) after injection of FCA and 16 days after EA increased significantly(P<0.01);while in comparison with model group,the swelling rates of EA-GV14 and EA-GV4 groups were markedly lower(P<0.01).Compared with normal control group,hypothalamic CRH,plasma ACTH,serum Cort and TNF-α contents of model group increased significantly(P<0.05);while in comparison with model group,hypothalamic CRH serum Cort and TNF-α contents in EA-GV14 group,and serum Cort level in EA-GV4 group decreased significantly(P<0.05).No significant differences were found between EA-GV14 and EA-GV4 groups(P>0.05) in these 4 indexes.Conclusion: EA has a marked anti-inflammatory effect in adjuvant arthritis rats and can relieve arthritis stress induced increase of hypothalamic CRH,plasma ACTH,serum Cort and TNF-α levels.

      摘要:Objective: To observe the effect of electroacupuncture(EA) on thymocyte proliferation and apoptosis in surgical trauma-stress rats.Methods: Thirty-six SD rats were evenly randomized into control,model and EA groups.Surgical trauma-stress model was established by making two longitudinal incisions((6 cm) and(5 cm) long) along the median back and abdominal lines and performing visceral exploration manipulation.EA((2 Hz) and(60 Hz) in the frequency,(2.85 s)/(1.05 s) in the duration of pulse-trains,and(1 mA)) was applied to unilateral "Zusanli"(ST 36) and "Lanwei"(EX-LE 7) for(30 min).After killing the rats,the thymus tissue was sampled to be processed into cell suspension for determining the thymocyte proliferation which was expressed by using stimulus index [optical density(OD) of the cellular culture fluid of EA group/OD of control group],and thymocyte apoptosis was assayed by TdT-mediated dUTP nick end labeling(TUNEL) technique.Results: Results showed that the rates of the apoptotic thymocytes in normal control,model and EA groups were 3.56±3.18%,15.32±4.67%,and 7.42±4.19% respectively;while the SI values of the aforementioned 3 groups were 1.96±0.09,1.40±0.10 and 1.65±0.10 separately.It indicated that after trauma-stress,the apoptosis of the thymus increased markedly and the cellular proliferation lowered significantly(P<0.01,0.001);following EA,the aforementioned effects of surgical trauma-stress were suppressed apparently(P<0.05).Conclusion: EA can improve trauma-stress induced immunodysfunction via inhibiting the apoptosis and facilitating the proliferation of thymocytes.

      摘要:Objective: To study the effect of electroacupuncture(EA) on the activity of nitric oxide synthase(NOS) in the colon,dorsal root ganglia and spinal cord in rats with pelvic pain for studying the role of NO in pain transmission.Methods: Twenty-six SD rats were randomly divided into control group(n=6),model group(n=10) and EA group(n=10).EA(4-(6 Hz),dense-sparse waves,0.5-(3 V)) was applied to bilateral "Zusanli"(ST 36) for(60 min).Pelvic pain model was established by local injection of 5% formalin((40 μL)) to the colonic wall from the anus.Behavioral reactions were observed.After transcardiac perfusion of saline and paraformaldehyde,the rat's colon near the anus,sacral 2-4 ganglia and the corresponding spinal cord segments were removed to be cut into sections((25 μm)).NOS activity of the colon,dorsal root ganglia and spinal cord tissues was determined by using nicotinamide-adenine dinucleotide phosphate-diaphorase(NADPH-d) histochemistry.Results: In comparison with model group,the cumulative score of pain behavior in EA group was significantly lower(P<0.01).While the integral OD values of NOS of the colonic wall,DRG and spinal cord in model group were significant higher than those in control group(P<0.01).Comparison between model and EA groups showed that the integral OD values of NOS of the 3 types of tissues in EA group were markedly lower than those in model group(P<0.01),indicating that EA could suppress pelvic pain induced increase of NOS activity.Conclusion: EA can relieve formalin induced pelvic pain and suppress pain produced increase of NOS activity,suggesting NO may be involved in the process of EA analgesia.

      摘要:Objective: To observe the influence of sectioning fat-strip along the Stomach Meridian on acupuncture analgesia.Methods: Sixteen SD rats were randomized into control group(n=6) and treatment group(n=10).After anesthesia with 3% pentobarbital((40 mg/kg)),a 6 mm long fat strip [6-(8 mm) proximal to "Zusanli"(ST 36)] along the Stomach Meridian of each rat in treatment group was excised,while for rats of control group,a same length of muscle strip beside the fat strip was removed.In treatment group,ST-36 was punctured with the acupuncture needle manipulated with uniform reinforcing-reducing method for(15 min).Pain threshold(tail flick latency responding to warm water stimulation) was detected before acupuncture,(0 min),(10 min),(20 min),(30 min) and(60 min) after acupuncture.Results: In control group,the pain threshold values between pre-and post-acupuncture before and after operation on the operation side,and between pre-and post-acupuncture on the non-operation side had no significant differences(P>0.05).There was a significant difference between control group and treatment group in the pain threshold on the operation side after acupuncture(P<0.001).In treatment group,the pain threshold values between pre-and post-acupuncture on the operation side had no significant difference(P>0.05);while those between pre-and post-acupuncture before operation,and between pre-and post-acupuncture on the non-operation side,and between operation side and non-operation side had significant differences(P<0.001),suggesting that after removal of the fat-strip along the Stomach Meridian,acupuncture induced increase of the pain threshold was abolished.Conclusion: The fat-strip structure along the Stomach Meridian is closely related to acupuncture of "Zusanli"(ST 36) induced analgesia.

      摘要:Objective: To observe the cardioprotective effects of repetitive pretreatment of electroacupuncture(EA) at higher intensity prior to the myocardial ischemia and reperfusion(MI/R) and to explore the involvement of beta-adrenergic receptor.Methods: Male SD rats were evenly randomized into normal control(NC),model,EA and EA+ propranolol groups.MI/R model was produced by occlusion of the anterior descending branch of the left coronary artery((30 min)) and reperfusion((10 min)) in the rats.ECG ST-segments and the ratio of infarct size/risk zone were detected,and the content of cardiac beta_1-adrenoceptor was assayed with immunoblot technique.EA((20 V),(20 Hz)) was applied to bilateral "Neiguan"(PC 6) for(30 min),once daily and continuously for 3 days.Propranolol((10 mg/kg),i.p) was given to the animals(15 min) before EA every time.Results: The elevated ST-segments of ECG in IR group recorded 30 min after ischemia and 10 min after reperfusion were significantly higher than those of NC group(P<0.01 and P<0.01 respectively);in EA group the elevated ST-segments were markedly reduced(P<0.01 and P<0.01 respectively vs IR group);while the elevated ST-segments in EAP group were almost the same as IR group,much higher than EA group(P<0.01 and P<0.01 respectively).The similar results were also achieved in the ratios of infarct size/risk zone in the 4 groups.Moreover,the content of cardiac beta_1-adrenoceptor(CCA) in EA group was significantly reduced as compared with IR group(P<0.01) in which CCA was significantly higher than NC group(P<0.01).In EAP group the CCA was almost the same as that in IR group(P>0.05) and much higher than EA group(P<0.05).Conclusion: The results of the present study indicate that by mimicking the ischemic preconditioning,pretreatment of EA can protect the heart from injury of MI/R in rats.The protective effect of EA is mediated by cardiac beta_1-adrenoceptor.

      摘要:Objective: To investigate the effect of electroacupuncture(EA) on the natural reward behavior and the related mechanism in depression rats.Methods: Thirty male SD rats were evenly randomized into control,model and EA groups.Depression model was established by using chronic stress stimulation including forced cool water swim,tail-clamping,fasting,electric shock,etc.,once daily,(21 days) altogether.One hour before stress stimulation in EA group,EA((2 Hz),(2 mA)) was applied to "Baihui"(GV 20) and "Yintang"(EX-HN 3) for(20 min),once daily,continuously for 21 days.The consumption of the rats' sucrose intake was detected before and after establishment of depression model.The rat's sexual behavior was recorded by calculating the times of mounting the female rat and sniffing female rat's pudendum in 15 minutes.Dopamine(DA) contents of frontal cortex,striate body and hypothalamus tissues were assayed by using high performance liquid chromatography electro-chemical detection technique.Results: Compared with control group,the male rats' sucrose intake amount,pudendum-sniffing times,DA contents of frontal cortex,striate body and hypothalamus tissues in model group decreased significantly(P<0.05,0.01);while compared with model group,the rats' sucrose intake amount,mounting and sniffing times and DA contents of the aforementioned 3 cerebral areas in EA group increased significantly(P<0.05).No significant differences were found between control and EA groups in sucrose intake amount,pudendum-sniffing times and DA contents of the 3 cerebral areas(P>0.05).Conclusion: EA can promote recovery of the rats' food rewarding and sexual reward and adjust the secretion of DA in depression rats.

    • Effect of Electroacupuncture on Behavioral Activities of Depression Rats

      摘要:Objective: To observe the effect of electroacupuncture(EA) combined with Clomipramine(Clo) on behavioral activities of depression rats.Methods: Forty-eight SD rats were randomized into normal control,model,EA,Clo(2.5 mg),Clo(5 mg) and EA+Clo(2.5 mg) groups,with 8 cases in each group.Chronic unpredictable stress(CUS) model was established by forcing the rat to have fasting,cool-water swim,water-deprivation,high temperature environment,etc.in 28 days.EA((4 Hz) and (60 Hz),duration of the pulse train:(2.5 s) and(5 s),≤(1 mA)) was applied to "Baihui"(GV 20) and right "Yanglingquan"(GB 34) for(30 min), once every other day,7 times altogether.CHP was given to the animals by intraperitoneal injection.The open-field test was used to observe the rats' rearing and crossing activities,and the forced swim test was employed to determine the rats' duration of immovability(DI) during(5 min) in a water pool.Results: In comparison with normal control group,the rearing and crossing movement times of rats in model group decreased significantly(P<0.05);while compared with model group,the rearing times of rats in EA,Clo(2.5 mg),Clo(5 mg) and EA+Clo(2.5 mg) groups,and crossing movement times of rats in EA,Clo(5 mg) and EA+Clo(2.5 mg) groups increased remarkably(P<0.05),and the effect of EA+Clo(2.5 mg) was significantly superior to those of EA and Clo(2.5 mg) groups in the rearing times and to that of EA group in the crossing times(P<0.05).In comparison with normal control group,the rats' DI in the water pool in model group prolonged significantly(P<0.05);while compared with model group,the DI of EA,Clo(5 mg) and EA+Clo(2.5 mg) shortened significantly(P<0.05),and the effect of EA+Clo(2.5 mg) group was significantly superior to that of EA group on the 28(th) day(P<0.05).Conclusion: EA works well in relieving

      摘要:Objective: To observe the time-effect of needling twelve Jing(well)-points on TNF-α levels in the serum and cerebral tissue in rats with cerebral ischemia(CI).Methods: A total of 96 male Wistar rats were randomized into normal control(n=8),sham-operation(n=8),CI(n=40) and acupuncture(n=40) groups.CI model was established by adopting photochemical method.The twelve Jing-points(bilateral LU 11,LI 1,PC 9,TE 1,HT 9,SI 1) were pricked with a filiform needle(gauge-30)(1 h),(3 h),(6 h),(12 h) and(24 h) after the establishment of CI model.The rats were killed respectively after six days of treatment for taking blood and brain tissue samples.Serum and cerebral TNF-α contents were assayed with radioimmunoassay according to the instructions of reagent kits.Results: Compared with normal control group,cerebral and serum TNF-α contents in CI group at(1 h),(3 h),(6 h),(12 h) and (24 h) after CI increased significantly(P<0.05);while comparison between acupuncture and CI groups showed that cerebral TNF-α contents of acupuncture group and serum TNF-α levels from(1 h) to(24 h) after CI were significantly lower than those of CI group at the same time courses(P<0.05,0.01). No significant differences were found between normal control and sham-operation groups,and between normal control and acupuncture groups in cerebral and serum TNF-α levels(P>0.05).Conclusion: Acupuncture of twelve Jing-points has a definite effect in reducing CI-induced increase of both cerebral and serum TNF-α contents,which may contribute to its action in improving cerebral ischemia in clinic.

      摘要:Objective: To observe the effect of electroacupuncture(EA) on the expression of brain-derived neurotrophic factor(BDNF) in substantia nigra compacta(SNC) of Parkinson's disease(PD) mice so as to study the synaptic plasticity.Methods: Twenty-four male mice(C57BL/6J) were averagely and randomly divided into normal control,EA,model and model +EA groups.PD model was induced by injection(i.p.) of 1-methyl,4-phenyl-1,2,3,6 tetrahydropyridine((30 mg/kg)).EA((2-100 Hz),2-(4 V),dense-sparse waves) was applied to "Hegu"(LI 4) and "Taichong"(LR 3) for(20 min),once daily.After 21 treatments,the mice were killed for sampling SNC tissue which was then cut into sections for detecting the expression of BDNF and BDNF mRNA by immunohistochemical and in situ hybridization techniques respectively.Results: Both immunohistochemical and in situ hybridization assays showed that compared with normal control group,the number of BDNF and BDNF mRNA positive cells and integral optical density(OD) values of model group were significantly lower(P<0.05,0.01);compared with model group,the number of BDNF and BDNF mRNA positive cells and integral OD values in model +EA group were significantly higher(P<0.05,0.01).No significant differences were found among normal control,EA and model +EA groups in these indexes.Conclusion: EA can inhibit the down-regulation of expression of BDNF and BDNF mRNA of SNC in PD mice which maybe contribute to its effect in raising the synaptic plasticity of the degenerative dopaminergic neurons.

      摘要:Objective: To observe the effect of acupuncture of "Zusanli"(ST 36) on blood flow of the stomach in the rat in order to evaluate the applied value of laser Doppler perfusion imager(LDPI) in acupuncture study.Methods: Thirty-eight Wistar rats were randomized into control group(n=18) and acupuncture group(n=20).Under anesthesia by 2% pentobarbital((2.5 mg/kg)),the rat's stomach body was exposed and put on a test pad.Then the rat was put in a constant temperature-box by controlling the temperature to be within 30-(35 ℃) and the change of temperature in an experiment to be ±(1 ℃).After inserting the acupuncture needle into "Zusanli"(ST 36),it was manipulated for(1 min),then kept in place for(10 min).The probe of LDPI was set to aim at the stomach with the distance between the probe and the stomach being about(20 cm).The images were analyzed by using the instrument-installed LDPI 2.5 software.Results: Under natural state,blood flow shown by the images presented a flabellate distribution from the lesser curvature to the greater curvature of the stomach,and decreased gradually during the experiment in the control group.After acupuncture of "Zusanli"(ST 36),the results showed that there were distinct differences in the perfusion images flow(increase of blood flow,particularly at the 10(th) min after acupuncture) on the middle part of the stomach and the region near the cardia in comparison with control group.Conclution: Acupuncture of "Zusanli"(ST 36) can increase the blood flow in the rat.The present finding demonstrates that LDPI can be used to provide an objective and specific assessment on the effects of acupuncture.

      摘要:Objective: To study the effect of electroacupuncture(EA) of "Hegu"(LI 4) and "Sibai"(ST 2) on the electrical activities of stomach-related neurons in the nucleus of solitary tract(NTS) of the rat.Methods: Experiments were performed on 62 Wistar rats anesthetized with mixture solution of urethane((420 mg/kg)) and chloralose((50 mg/kg),i.p.).The 4(th) ventricle of the brain was exposed for inserting glass micropipettes((1 μm) in diameter,10-(30 mΩ) in electrical resistance,and filled with(1 mol) KCl+2% pontamine sky blue)into NTS under the microscopic observation to record electrical activities of neurons.After opening the abdominal cavity and making an incision at the junction between the pylorus and duodenum,a plastic balloon was inserted into the stomach for dilating it.EA(2-(4 V),(20 Hz) and duration of(2 min)) was applied to the left "Hegu"(LI 4) and "Sibai"(ST 2).If the firing rate of the neuron decreased or increased by 25% or more after EA or gastric dilation stimulation,it was considered to be inhibitory or excitatory reaction.Results: 1) A total of 278 NTS neurons were recorded in the present study.Among them,132 neurons showed apparent excitatory or inhibitory responses to gastric distention via the implanted balloon and were called as stomach-related neurons.Among the 132 neurons,70 were excited,and 62 were inhibited,including 36/62 being inhibited completely.2) Of the 132 stomach-related neurons(17 inhibitory and 24 excitatory neurons),14/17 and 8/24 presented inhibitory response,and 3/17 and 16/24 excitatory response to EA of "Hegu"(LI 4);while 11/17 and 7/24 presented inhibitory response,6/17 and 17/24 excitatory response to EA of "Sibai"(ST 2) separately,indicating that signals of EA of LI-4and ST-2 and those from gastric dilation could converge to the same NTS neuron.Conclusion: The results indicate that information of EA of "Sibai"(ST 2) and "Hegu"(LI 4) and gastric distention may converge to the same individual NTS neuron.NTS may play an important role in the integration and convergence of the somatic(facial area and LI-4) message and visceral(stomach) message.The present research provides new evidence for the theory of meridian about the specific connection between Hegu Point and the face-mouth region.

      摘要:Objective: The spinal segmental distribution of neural innervations related to "Guanyuan"(CV 4) in rats has been investigated by horseradish peroxidase conjugated cholera toxin B subunit(CB-HRP) tracing technique.Methods: Twelve SD female rats were divided randomly into electroacupuncture group(EA) and control group(C).CB-HRP solution(0.03%(20 μL)) was injected into "Guanyuan" acupoint of the rats. The HRP labeled neurons were observed morphologically and counted in the related spinal dorsal root ganglia(DRG).Results: The sensory nerve afferents of "Guanyuan" acupoint mainly projected to the neurons of T_(11) to L_3 DRG.The labeled neurons were observed to be large,medium and small sized cells,of which the small size was in The majority.The number of labeled neurons in EA group was increased significantly compared with control group((P<0.05)).The nerve fibers projected to L_2 were activated by the EA treatment and the number of HRP labeled cells in L_2 DRG was increased significantly.Conclusion: EA at "Guanyuan" acupoint may affect the local innervation activity,resulting in the projection of the local primary sensory afferents focusing on L_2 DRG.So this study provides a new morphological evidence for transduction of acupuncture signals.

      摘要:Objective: To compare the effects of acupuncture of Baihui(GV 20),Shuigou(GV 26) and Shenmen(HT 7) on the cognitive ability in vascular dementia(VD) patients.Methods: Fifty cases of VD patients were randomly and evenly divided into body-acupoint,GV-20,GV-26,HT-7 and GV-20+GV-26+HT-7 groups.Body-acupoints were Jianyu(LI 15),Quchi(LI 11),Zusanli(ST 36),etc and were used in all the patients of the 5 groups.The acupuncture treatment was given once daily(except weekend days),4 weeks altogether.The therapeutic effects were assessed by using "Scale for Evaluating the Cognition Function"(SECF) before and after the treatment. Results: After the treatment,the total scores of SECF in GV-20,GV-26,HT-7 and GV-20+GV-26+HT-7 groups were significantly higher than those of their respective pre-treatment(P<0.05).Adding Baihui(GV 20) could significantly raise the patients' scores of orientation,short and long-term memory,animal naming,calculating and classification(P<0.05,0.01);adding Shuigou(GV 26) could raise the scores of orientation,calculating and classification(P<0.05);adding Shenmen(HT 7) could raise the scores of digital range,animal naming and classification(P<0.05).And adding GV-20, GV-26 and HT-7 could raise the scores of orientation,digital range,short-term memory,animal naming,calculation,classification and painting abilities(P<0.05,0.01).The effects of GV-20+GV-26+HT-7 group were significantly better than those of body-acupoint group in improving the total score,orientation,digital range,animal naming,calculation and painting(P<0.01);than that of GV-20 group in calculation(P<0.05);than those of GV-26 in digital range,animal naming,calculation and painting(P<0.05,0.01);and than that of HT-7 group in calculation(P<0.01).Conclusion: Acupuncture of the added Baihui(GV 20),Shuigou(GV 26) and Shenmen(HT 7) works better in improving VD patients' cognitive ability.

    • Study on the Homologous Phenomenon of Meridian and Orientation

      摘要:In the present paper,the authors analyze "the homologous phenomenon of the meridian of traditional Chinese medicine(TCM) and the orientation" and "the relationship between the Zang-or Fu-organ and the meridian" from the classical theory of "corresponding correlation between the Zang-or Fu-organ and the meridian" and the basic clinical principles about "selecting acupoints along the meridian".Results reveal that there exist many doubtful points in the doctrine of "corresponding correlation between the Zang-or Fu-organ and the meridian" including "non-specific correspondence" and "non-correspondence".For example,puncturing acupoints of the Pericardium Meridian can treat disorders of both the heart and the stomach.However,in the doctrine of "correlation between the Zang-or Fu-organ and the meridian",there are some reasonable elements which possess the phenomenon of "correspondence of orientation" while the doubtful points don't.The "correspondence of orientation" is quite complicated probably due to the organism's "diversity" and "multiple directions(meridians' distribution)" but may help us sort out a main thread from the classical meridian-collateral theory and clinical experience,favoring simplifying the researched objects.
