      摘要:Objective:To study the mechanism of acupuncture of "Baihui"(GV 20) and "Taiyang"(EX-HN 5) in improving cerebral ischemia(CI) injury and vascular endothelial cellular function.Methods: Eighty old male Wistar rats were randomized into control(n=8),model(n=32),acupuncture(n=32),sham-operation(n=8) groups.Model and acupuncture groups were further evenly and respectively divided into(1 d),(3 d),(5 d) and(10 d) subgroups.CI model was established by using middle cerebral artery occlusion(MCAO) method."Baihui"(GV 20) and "Taiyang"(EX-HN 5) were punctured with filiform needles which were twirled rapidly at a frequency of about 200 revolutions/min for(1 min)(1 time/10 min),and retained for 30 min.The treatment was given once daily,continuously for 1,3,5 and 10 days respectively in different subgroups.Endothelin(ET)-1,intercellular adhesion molecule (ICAM)-1,and factor Ⅷ related antigen(FⅧR-Ag) of CA_3 area tissue of the hippocampus were assayed with immunohistochemical method(SABC).Results: The CI rats' behavioral symptoms of(5 d) and(10 d) groups were improved significantly in comparison with the corresponding subgroups of model group (P<0.05,0.01).Compared with(1 d),(3 d),(5 d) and(10 d) subgroups of control group,the total area and integral optical density(IOD) values of ICAM-1,ET1 and FⅧR-Ag expression in CA_3 of hippocampus all increased significantly in the 4 subgroups of model group(P<0.01).Compared with 4 subgroups of model group,the total area values of acupuncture group,ICAM-1 expression in the 4 subgroups,ET-1 in(5 d) and(10 d) subgroups decreased significantly,and FⅧR-Ag expression in(3 d),(5 d) and(10) d subgroups increased;IOD values of ICAM-1 in(3 d),(5 d) and(10 d) subgroups,ET-1 in(5 d) and(10 d) subgroups decreased markedly,and FⅧR-Ag in(3 d),(5 d) and(10 d) subgroupsall increased considerably(P<0.05,0.01).Conclusion: Acupuncture of "Baihui"(GV 20) and "Taiyang"(EX-HN 5) can improve CI rats' behavior and regulate the expression of vascular endothelial ICAM-1,ET-1 and FⅧR-Ag in CA_3 of hippocampus and has accumulative effects.

      摘要:Objective: To explore the effect of accelerated aging and dementia on cerebral proteome and the regulative effect of acupuncture interference.Methods: Forty eight-month-old senescence-accelerated mice prone10(SAMP10) and their homologue,normal aging mice(SAMR1) of the same age were randomly and evenly assigned to P10-acupuncture,P10-sham-acupuncture,P10-control and R1-control groups with 10 cases in each group.P10-acupuncture group was treated by acupuncture of "Shanzhong"(CV 17),"Zhongwan"(CV 12),"Qihai"(CV 6),"Xuehai"(SP 10) and "Zusanli"(ST 36),once daily and continuously for 15 days except the 7(th) day.The sham-acupuncture group was treated by acupuncture of non-acupoint at the hypochondrium.The acupuncture needle was twirled rapidly with reinforcing method about(30 s) every time for each acupoint.At the end of the experiments,the mice under deep anesthesia were killed for sampling the brain tissue.After a series of process of the brain tissue,the expressive profiles of cerebral proteins were displayed by applying bi-dimensional electrophoresis technique.Results: With the accelerated senescence and dementia,the expression of 1 protein spot((100 kD/pI 5.5)) appeared and 2 proteins disappeared simultaneously in SAMP10 of P10-acupuncture,P10-sham-acupuncture and P10-control groups,but appeared in SAMR1.In addition,in SAMP10 group,6 proteins displayed abnormal expression including 2 proteins being down-regulated and 4 proteins up-regulated in the expression level which were reversed in P10-acupuncture group,suggesting acupuncture may alleviate the effect of aging and dementia.In P10-sham-acupuncture group,of the 4 proteins being up-regulated in the expression in SAMP10 group, 2 proteins showed up-regulation and the other 2 down-regulation.Conclusion: Some proteins of the cerebrum in SAMP10 show aberrant expression with the accelerated senescence,which can be improved by acupuncture.

      摘要:Objective: To observe the effects of electroacupuncture(EA) on hyperalgesia,inflammatory tissue repair and the expression of spinal COX-2 mRNA and protein in rats with chronic inflammatory pain.Methods: Chronic inflammatory pain model was established by injection of complete Freund's adjuvant(CFA) into the SD rat's left ankle-joint cavity of the hind limb.EA((2 Hz),(1 mA),continuous waves) was applied to "Yanglingquan"(GB 34) and "Kunlun"(BL 60) for(20 min),once daily,continuously for(1 d),(4 d),(6 d),(8 d) and(10 d) respectively.For observing changes of tissue repair of the focus,HE staining technique was used;and for determining the expression of spinal COX-2 mRNA and COX-2 protein in the dorsal horns of the lumbar spinal cord,immunohistochemical and RT-PCR techniques were carried out.Results: After 6 days' EA treatment,the hyperalgesic score of EA group(n=6) was significantly lower than that of model group(n=6);and after 8 and 10 days' treatment,the decreased hyperalgesic score of EA group was similar to that of control group(n=6,P>0.05).Compared with model 1((4 d) after injection of CFA,n=3) and 2((14 d) after injection of CFA,n=3),the exudation of the joint cavity,synovium edema,and infiltration of mononuclear cells of EA group(n=6) were improved remarkably.In comparison with control group,4 days after injection of CFA,spinal COX-2 mRNA expression and COX-2 immuno-reaction positive cell number of model group increased significantly(P<0.05);6 days after EA,no significant differences were found between control and EA groups in both COX2 mRNA expression and COX-2 immuno-reaction positive cell number(P>0.05),indicating that EA could down-regulate the expression of COX-2 mRNA and COX-2 protein in the spinal cord.Conclusion: EA has a remarkable analgesic effect and can promote the repair of inflammatory tissue,which may be related to its functions in down-regulating the expression of COX-2 mRNA and COX-2 protein in the spinal cord.

      摘要:Objective: To observe the effect of electroacupuncture(EA) on pain threshold,paw swelling and serum T-lymphocyte subsets in adjuvant-induced arthritis(AA) rats so as to study the mechanism of EA in cellular immunity.Methods: Thirty male Wistar rats were divided into control,model and EA groups using random number table method,with 10 rats in each group.AA model was established by injection of complete Freund adjuvant(0.1%,(0.1 mL)) into the subcutaneous tissue of the right hindpaw.EA((4/60 Hz),dense-sparse pulses,(1 mA)) was applied to bilateral Jiaji(EX-B 2) points of L_3 and L_5 for 30 minutes,once daily,continuously for 7 days.The pain threshold(latency of paw withdrawal induced by radiant heat irradiation),paw swelling rate [(paw volume of post-injection of CFA-that of pre-injection)/that of pre-injection ×100%] and serum T-lymphocyte subsets(CD_4+ and CD_8+ and CD_8+) were detected respectively.Results: Compared with normal control group,the paw volume(PV) and paw swelling rates of model group and EA group increased significantly while the paw withdrawal latencies on the focus side shortened markedly one day after injection of CFA(P<0.01).On the 7+) were detected respectively.Results: Compared with normal control group,the paw volume(PV) and paw swelling rates of model group and EA group increased significantly while the paw withdrawal latencies on the focus side shortened markedly one day after injection of CFA(P<0.01).On the 7(th) day,PV and paw swelling rate of model group were still significantly higher than those of control group(P<0.01);while in comparison with model group,both PV and paw swelling rate decreased considerably(P<0.05) and the paw withdrawal latency increased markedly in EA group(P<0.05).It indicated that EA could effectively relieve pain and swelling of the focal hindpaw.In comparison with control group,the percentages of CD_4(th) day,PV and paw swelling rate of model group were still significantly higher than those of control group(P<0.01);while in comparison with model group,both PV and paw swelling rate decreased considerably(P<0.05) and the paw withdrawal latency increased markedly in EA group(P<0.05).It indicated that EA could effectively relieve pain and swelling of the focal hindpaw.In comparison with control group,the percentages of CD_4+ and CD_8+ and CD_8+ of model group decreased apparently(P<0.05),and CD_4+ of model group decreased apparently(P<0.05),and CD_4+/CD_8+/CD_8+ increased significantly(P<0.05).Comparison between model and EA groups showed that the percentage of CD_8+ increased significantly(P<0.05).Comparison between model and EA groups showed that the percentage of CD_8+ increased evidently(P<0.01) and the ratio of CD_4+ increased evidently(P<0.01) and the ratio of CD_4+ to CD_8+ to CD_8+ decreased significantly in EA group(P<0.05).Conclusion: EA of Jiaji(EX-B 2) has anti-inflammatory and analgesic effects,which maybe related to EA induced favorable regulation on T-lymphocyte subsets in AA rats.

      摘要:Objective: To observe the effect of acupuncture of auricular concha area on blood pressure(BP) and to explore its mechanism in lowering BP.Methods: Fifteen spontaneous hypertension rats(SHR) and 22 normal SD rats anesthetized with urethane((1.0 g/kg)) were subjected into the present study.Mean blood pressure of the common cervical artery and activities of the cervical vagus trunk were recorded during electroacupuncture(EA,75-(100 Hz),dense-sparse waves and duration of(2 min)) or manual acupuncture stimulation(about 2 revolutions/s) of the auricular concha and left "Neiguan"(PC 6).BP and electrical activities of the left vagus and sympathetic nerve were recorded in some rats.Atropine(1%,(0.05 mg/kg)) was injected into the carotid artery for observing the effect of EA on BP. Results: In SHR and normal rats,electrical stimulation and manual acupuncture stimulation of both left and right auricular concha could apparently lower blood pressure of the carotid artery in comparison with the basic values before stimulation(P<0.001);correspondingly,when manual acupuncture stimulation of both left and right auricular concha induced decline of mean pressure,the vagal activity increased and sympathetic discharges decreased clearly.Manual acupuncture stimulation of "Neiguan"(PC 6) could also significantly lower BP,but the effect was remarkably weaker than that of stimulation of auricular concha(P<0.001).In vagotomized and atropine-administrated rats,the effect of acupuncture disappeared,while electrical stimulation of vagus could produce auricular concha-acupuncture-like effects in lowering BP and heart rate.Conclusion: Acupuncture of auricular concha has a definite hypotensive effect in both SHR and normal rats,which depends on the integrity of the vagus in both function and structure.

      摘要:Objective: To observe the effect of electroacupuncture(EA) of "Baihui"(GV 20) and "Taixi"(KI 3) on changes of behavior activities and concentrations of monoamines transmitters in hypothalamic tissues of rats with chronic fatigue syndrome(CFS).Methods: Thirty-six male Wistar rats were evenly randomized into control,model and EA groups.CFS model was established by intramuscular injection of hydrocortisone(20 mg/kg,1 time/d,9 d) and withdrawal of hydrocortisone,and forced swim in cool water(30 min/1 time/d,21 days).EA(2/20 Hz,9 V) was applied to "Baihui"(GV 20) and "Taixi"(KI 3) for(30 min),once daily and continuously for 21 days.Open-field test,rat's tail suspension immobility test and exhausted swimming test were used to check the rats' behavioral changes and double antibody sandwich enzyme-linked immunosorbnent assay(ABC-ELISA) was used to determine concentrations of noradrenaline(NE),serotonin(5-HT) and dopamine(DA) in hypothalamus tissues.Results: Compared with control group,the crossing and rearing activity times,the duration of tail-suspension immobility and the duration of exhausted swim,and the contents of NE,5-HT and DA of hypothalamus tissues in model group were all significantly lower(P<0.05);while the values of the abovementioned 7 indexes of EA group were all significantly higher than those of model group(P<0.05).No significant differences were found between EA group and control group in these 7 indexes(P>0.05).It showed that EA could improve behavioral activities of CFS rats and raise the contents of monoamines neurotransmitters in the hypothalamus.Conclusion: EA of "Baihui"(GV 20) and "Taixi"(KI 3) can ameliorate behavior activities of CFS rats and reverse the declined contents of NE,5-HT and DA of hypothalamus tissue.

      摘要:Objective: To observe the effect of electroacupuncture(EA) on the activity of superoxide dismutase(SOD) and the content of lipid peroxide(LPO) in the compressed lumbar nerve roots so as to explore the mechanism of acupuncture in the treatment of nerve root compression syndrome.Methods: In this study,42 SD rats were randomized into six groups: control group,model group and EA group which were further divided into 15 d and 30 d subgroups respectively,with 7 cases in each subgroup.Nerve root compression model was established by embedding a small piece of hard silicon film near the right lumbar 5(L_5) nerve root beneath the subdural space.EA((2 Hz),2-(3 mA),continuous waves) was applied to right "Huantiao"(GB 30) and "Yanglingquan"(GB 34) for(20 min),once every other day.On the 15(th) and 30(th) and 30(th) after the treatment,the rats were killed separately for collecting the nerve root samples.SOD activity and LPO content of the compressed nerve root tissue were assayed by means of pyrogallol autoxidation inhibition method and LPO fluorescence assay respectively.Results: Compared with control group,the SOD activity decreased significantly,and LPO contents increased obviously in(15 d) and(30 d) subgroups of model group(P<0.01).While compared with(15 d) and(30 d) subgroups of model group,SOD activity of the 2 subgroups of EA group increased significantly(P<0.01) in spite of being still evidently lower than that of control group(P<0.01).LPO contents in the 2 subgroups of EA group were considerably lower than those of the 2 subgroups of model group(P<0.05,0.01).No significant difference was found between(30 d) subgroup of EA group and that of control group in LPO level(P>0.05).Conclusion: EA of"Huantiao"(GB 30) and "Yanglingquan"(GB 34) can effectively raise SOD activity and lower LPO content in the compressed lumbar nerve root which may contribute to its therapeutic effect in improving clinical symptoms of lumbar nerve compression syndrome.

      摘要:Objective:To observe the effect of different frequencies of electroacupuncture(EA) of "Zusanli"(ST 36) on the expression of somatostatin(SOM) mRNA in rats' amygdaloid nucleus.Methods: Thirty SD rats were randomized into control,2 Hz-EA and 128 HzEA groups with 10 cases in each group.Left "Zusanli"(ST 36) was punctured and stimulated electrically for 30 min by setting the parameters being 2 Hz and 128 Hz in frequency,3 V in strength,and 1 ms in the duration of waves.After EA, the rats were transcardically perfused with 1% paraformaldehyde and 1.25% glutaraldehyde and 0.1 mol/L phosphate buffer solution.The brain tissues were sampled to cut into sections(40 μm).The expression of SOM mRNA of amygdaloid nucleus was detected by using in situ hybridization technique.Results: Compared with control group,the number of SOM mRNA expression positive neurons and the grey-density values of SOM mRNA expression in amygdaloid nucleus on the bilateral sides increased significantly in 2 Hz and 128 Hz EA groups(P<0.05,0.01).There were significant differences between two EA groups in the number of SOM mRNA expression positive neurons and the grey-density values.Conclusion: Both 2 Hz and 128 Hz EA of "Zusanli" can up-regulate the expression of SOM mRNA in the rat's amygdaloid nucleus.The high frequency of EA has a more obvious effect.

      摘要:Objective: To study the protective mechanism of electroacupuncture(EA) of "Zusanli"(ST 36) in improving gastric mucosa injury in rats.Methods: Thirty Wistar rats were randomized into control,model and EA groups with 10 cases in each group.Gastric mucosal injury model was made by intragastric perfusion of anhydrous alcohol(0.4 mL).EA(4/20 Hz in frequency,0.5 ms in pulse-duration,0-8 V instrength) was applied to bilateral "Zusanli"(ST 36) for(20 min).Gastric mucosal blood flow(GMBF) was detected with a blood flow detector,and gastric mucosal lesion index(GMLI) calculated according to Guth's criteria.Serum nitric oxide(NO) and plasma endothelin(ET) contents were assayed with nitrate reductase method and enzyme-linked immunoassay respectively.Results: Following perfusion of anhydrous alcohol,gastric mucosal lesion was very evident and GMBF decreased remarkably in model group(P<0.01),after EA,GMLI decreased considerably and GMBF increased significantly(P<0.01).Compared with control group,serum NO level decreased significantly and plasma ET increased considerably in model group(P<0.01);while compared with model group,serum NO increased significantly and plasma ET decreased significantly in EA group(P<0.01).Conclusion: EA of "Zusanli"(ST 36) can effectively relieve alcohol-induced gastric mucosal injury,which maybe be related to its regulative effects on blood NO and ET.

      摘要:Objective: To study the correlation among acupoints,meridians and cerebral structures with BOLD-based fMRI technique.Methods: Twenty-four healthy volunteer subjects(20 men,4 women;age range 21-48 years) participated in this study and were divided into Guangming(GB 37),Yanglingquan(GB 34),Zusanli(ST 36) and Futu(ST 32) groups with 6 cases in each group.These acupoints(left side) were punctured with filiform needles(0.3 mm×40 mm) and manipulated with hand at a frequency of 200 revolutions/min for 15 sec.The signal data [blood oxygen level depended(BOLD) signals] of the brain were collected by using GE1.5 T Signa/Horizon LX Superconductor Nuclear Magnetic Resonance Imaging System.The signal intensities with the needle at rest before,during and after the needle manipulation periods were processed by using a "functional neuroimage software".Results: After acupuncture stimulation of the left GB-34 and GB-37,the signal intensity was increased(by 3.31±1.2%) in contralateral occipital lobe(visual cortex,7 cases),frontal lobe(8 cases),temporal lobe(5 cases),basal nerve ganglion(5 cases),pons(3 cases);ipsilateral frontal lobe(2 cases) and temporal(1 case);and bilateral occipital lobe(5 cases),frontal lobe(2 cases),temporal lobe(2 cases) and basal nerve ganglion(1 case).Acupuncture of ST-36 and ST-32 resulted in signal increase(4.28±1.5%) in the contralateral hypothalamus(6 cases),hippocampus(8 cases) and frontal lobe(7 cases);ipsilateral hypothalamus(2 cases),hippocampus(2 cases) and frontal lobe(3 cases);and bilateral hypothalamus(3 cases),hippocampus(2 cases) and frontal lobe(1 case).Conclusion: Manual acupuncture at acupoints of the same one meridian results in activation of BOLD signals in the same cerebral regions,suggesting a certain specific correlation between the acupoints of a certain meridian and the cerebral structure.

      摘要:Objective: To evaluate the influence of different acupuncture manipulations on bladder volume in female urethral syndrome(FUS) patients.Methods: Eighty-one cases of FUS patients were randomly divided into electroacupuncture(EA) plus hand-acupuncture group(observation group,n=40) and hand-acupuncture group(control group,n=41) by using random table method.Main acupoints used were ①Guanyuan(CV 4),Dahe(KI 12),Shuidao(ST 28) and Sanyinjiao(SP 6);②Shenshu(BL 23),Huiyang(BL 35),Zhonglushu(BL 29) and Weizhong(BL 40).The two groups were used in turn,combined with other acupoints according to the syndromes.After insertion,the acupuncture needles were manipulated with reinforcing method and twirled continuously for about 1 min every acupoint,then kept in place for 20 min.The treatment was conducted once every other day.The bladder volume(BV) and maximumurine flow rate(MUFR) were detected with Bonito Urine Dynamic Detector.Results: After 10 treatments,the BV and MUFR of both observation group and control group increased significantly in comparison with their individual values of pretreatment(P<0.05),and the difference values between post-and pre-treatment of the two indexes of observation group were apparently higher than those of control group(P<0.05).Conclusion: Both EA plus hand-acupuncture and simple hand-acupuncture can effectively improve the bladder volume and urine flow rate in female urethral syndrome patients and the effect of the former is significantly superior to that of simple hand-acupuncture.

      摘要:Objective: To observe clinical therapeutic effect of acupoint injection of compound Methylthioninum Chloride(CMC) for treating post-herpetic neuralgia(PHN).Methods: Eighty PHN patients were divided into treatment group(n=40)and control(acupuncture) group(n=40)with random number table method.For patients in treatment group,CMC injection((10 mL)) composed of 0.5% Bupivacaine Hydrochloride((3 mL)),Triamcinolone injection((15 mg)),VitB_(12)(1 mg),VitB_6 injection((100 mg)),CMC((0.02 mg)) and normal saline((3 mL)) was injected into back-shu-points on the first lateral line of the Bladder Meridian and its upper and lower back-shu-points according to the location of herpes zoster,2-(3 mL/point),followed by injecting oxygen((10 mL)) into the same point slowly,once every week,3 times altogether.For patients of control group,the same back-shu points were used and punctured with filiform needles,combined with acupuncture of Zhigou(TE 6),Yanglingquan(GB 34),etc.,once daily,continuously for 3 weeks.Results: Self-comparison showed that after the treatment 6-point-behavoral rating scale(BRS-6) scores of two groups both decreased significantly(P<0.001),and the BRS-6 score of treatment group was significantly lower than that of control group(P<0.01).Of the 40 cases in treatment and control groups,32(80%) and 3(7.5%) cases were cured,6(15.0%) and 7(17.5%) markedly effective,1(2.5%) and 19(47.5%) effective,1(2.5%) and 11(27.5%) failed,with the total effective rates being 97.5% and 72.5% respectively.The therapeutic effect of treatment group was significantly superior to that of control group(P<0.001).Conclusion: The therapeutic effect of point-injection of compound Methylthioninum Chloride is significantly better than that of acupuncture therapy in the treatment of PHN patients.

      摘要:In the present paper,the authors review recent progress in acupuncture-resisting brain aging in recent ten years from clinical treatment and studies and animal experiments.In clinical,acupuncture could improve the intelligence and social ability of vascular dementia(VD) and aged dementia(AD) patients.In basic research,acupuncture was proved to have definite effects in improving cognitive ability,raising cortical excitability and cerebral blood flow,regulating neurotransmitter level,resisting free radical and inflammation,lessening apoptosis,potentiating anti-stress ability,modulating lipometabolism and blood trace element level,and regulating the expression of some related genes.The research findings provide insight into the popularization of acupuncture therapy in clinical treatment of aged dementia and settle a good foundation for further study.
