      摘要:Objective: To observe the analgesic effect of repeated electroacupuncture (EA) with different intervals in a day in adjuvant arthritis (AA) rats so as to find the best time interval of EA for analgesia. Methods: A total of 96 SD rats were evenly randomized into normal, model and EA groups which were further divided into 3 h , 6 h , 12 h and 24 h subgroups, with 8 cases in each subgroup. AA model was established by subcutaneous injection of Freund’s Complete Adjuvant ( 0.1 mL ) into the right hind paw. EA (15/ 100 Hz , dense-sparse waves, 2- 4 V ) was applied to the right “Kunlun” (BL 60)-“Xuanzhong” (GB 39) for 20 min every time, with intervals of 3 h , 6 h , 12 h and 24 h everyday and continuously for 6 days. The pain grade (according to Xu’s method 1 ) and pain threshold (PT, the latency of radiant heat irradiation induced leg withdrawal reaction) of AA rats were determined after EA treatment. The rat’s hypothalamus tissue was sampled to cut into sections (5- 6 μm ) to be stained with in-situ hybridization histochemistry for detecting proopiomelanocortin (POMC) and proenkephalin (PENK) mRNA expression. Results: Contrasted with 3 h , 6 h , 12 h and 24 h of control group, the pain grades of both model and EA groups were apparently higher than those of normal control group, while those of EA group were lower than those of model group. Compared with control group, the PT values of model group ( 3 h , 6 h , 12 h and 24 h subgroups) and EA group ( 3 h and 6 h subgroups) were significant lower (P<0.01, 0.05), while comparison between model group and EA group showed that the PT values of EA group were higher ( 3 h , 6 h and 12 h subgroups) or significantly higher ( 24 h subgroup) than those of model group (P<0.01). In EA group, the PT value of 24 h subgroup was significantly higher than those of 3 h , 6 h and 12 h subgroups (P<0.05, 0.01). The POMC mRNA and PENK mRNA expression-positive cells of the 4 subgroups of both model and EA groups were significantly more than those of control group (P<0.01); and those of the 4 subgroups of EA group were significantly more than those of model group (P< 0.01). In EA group, the POMC mRNA and PENK mRNA expression-positive cells of 24 h subgroup were significantly more than those of 3 h , 6 h and 12 h subgroups (P<0.01, 0.05). Conclusion: Repeated EA has a definite analgesic effect and can obviously promote POMC mRNA and PENK mRNA expression in AA rats, and the effects of 24 h subgroup is the best.

      摘要:Objective: To observe the effect of electroacupuncture (EA) on visceral pain and investigate the significance of the expression of c-fos and glial fibrillary acidic protein (GFAP) in medullary visceral zone (MVZ) in experimental visceral pain rats. Methods: Thirty-two male SD rats were randomized into control, visceral pain (VP), EA+saline and EA+VP groups, with 8 rats in each group. Visceral pain model was established by intraperitoneal injection of 0.6% acetic acid ( 10 mL/kg ). EA ( 50 Hz , 2- 5 V , intermittent waves) was applied to bilateral “Zusanli”(ST 36) for 30 min followed by establishment of VP. Saline ( 10 mL/kg ) was given (i.p.) to the rats after EA. Immuno-histochemical single or double staining methods (ABC) were used to show the immunoactivity of c-fos and GFAP in MVZ including nucleus of solitary tract (NTS) and dorsal motor nucleus of the vagus nerve (dmnx). The degree of visceral pain was evaluated by visceral pain index (VPI), according to the Schmauss exponential method. Results: Pain reactions (writhing reactions: abdominal muscular contraction, hind-paw twitching, etc) happened in rats of model group and EA+VP group after injection of acetic acid, being more serious in the former group, and no such reactions were found in control and EA+saline groups. Fos-immunoreaction (IR)-positive neurons of VP, EA+saline and EA+VP groups found mainly in NTS and dmnx were significantly more than those of control group (P<0.05), and in comparison with VP group, c-fos-IR-positive neurons of EA+saline and EA+VP groups were significantly fewer (P<0.05). The optical greyvalues of GFAP-IR-positive microglial cells found mainly in NTS and dmnx in VP, EA+saline and EA+VP groups were markedly higher than that of control group (P<0.05); while compared with VP group, the grey values of GFAP activity of EA+saline and EA+VP groups were significantly lower (P<0.05). Compared with VP group, VPI of EA+VP group was significantly lower (P<0.01) . Conclusion:EA has a marked antinociceptive effect in visceral pain model rats, which may be related to its effect in down-regulating c-fos and GFAP expression in MVZ.

      摘要:Objective: To investigate the effects of electroacupuncture (EA) on the expression and content of nuclear factor-κB in hippocampus of rats with cerebral ischemia/reperfusion (CI/R). Methods: A total of 120 SD rats were randomly divided into normal group (n=10), sham operation group (n=27), model group (n=36) and EA group (n=36), and the lateral 3 groups were evenly divided into 24 h , 48 h and 72 h subgroups. CI/R model was established by middle cerebral artery occlusion(MCAO)and reperfusion with the suture in MCA. EA ( 2 Hz , 1 mA , continuous waves) was applied to “Dazhui”(GV 14) and bilateral “Neiguan”(PC 6) for 30 min , once 3 h after CI/R, followed by once every 12 h . The hippocampus tissue was taken out to be cut into sections ( 5 μm ). The expression and content of nuclear factor (NF)-κB-p65 protein in hippocampus were assayed by immunohistochemistry and Western blotting. Results: In comparison with normal group, NF-κB-p65 expression-positive cells of hippocampal CA1 area in 24 h of model group, and every 48 h and 72 h subgroup of model and EA groups were significantly more ( P< 0.05, 0.01). Compared with 24 h , 48 h and 72 h subgroups of model group, NF-κB-p65 expression-positive cells in the corresponding subgroups of EA group were significantly fewer (P<0.01). The grey values of NF-κB-p65 protein in the 3 subgroups of model group were significantly higher than those of normal group and sham-operation groups (P<0.01); while the grey values of NF-κB-p65 in the 3 subgroups of EA group were markedly lower than those in the corresponding 3 subgroups of model group (P<0.05). Conclusion: EA can reduce the expression and content of nuclear factor-κB-p65 in hippocampus of rats with CI/R and prevent it from being transported to the nucleus of the neurons in hippocampal CA1 area, which may contribute to the protective effect of EA on ischemic neurons.

      摘要:Objective: To observe the effect of herbal-cake-separated moxibustion on the expression of vascular cell adhesion molecule (VCAM)-1 in hyperlipemia (HLP) rabbits with concomitant aterosclerosis (AS) so as to explore the mechanism of moxibustion in improving HLP. Methods: Seventy-five New Zealand species rabbits were divided into control (n=13), model (n=14), direct-moxibustion (n=14), herbal-cake-separated moxibustion (n=14), and medication (“Xuezhikang”, a herbal drug for lowering blood lipid, n=13) groups. HLP with AS model was established by using immunologic injury method and high-fat forage feeding. Moxibustion was applied to “Juque” (CV 14), “Tianshu” (ST 25), “Xinshu” (BL 15), “Ganshu”(BL 18), etc. 30 min every acupoint, once daily, continuously for 6 weeks. The expression of VCAM-1 mRNA in the aorta wall was assayed with reversed transcription polymerase chain reaction (RT-PCR) technique. Results: Compared with control group, the expression of VCAM-1 mRNA in model group was up-regulated significantly(P<0.01); while in comparison with model group, VCAM-1 mRNA expression in herbal-cake-separated moxibustion, direct-moxibustion and medication groups was down-regulated obviously(P<0.01), suggesting a favorable regulation of the 3 methods on VCAM-1 mRNA expression. No significant difference was found among the 3 treatment groups (P>0.05). Conclusion: Moxibustion can down-regulate HLP-induced enhancement of VCAM-1 mRNA expression, which likely contributes to its effect in delaying the development of aortic endothelial AS in rabbits with HLP combined with AS.

      摘要:Objective: To study the mechanism of “Shuanggu Yitong” acupuncture therapy in the treatment of postmenopausal osteoporosis (PMO). Methods: A total of 60 female SD rats (aged 9-10 months) were randomized into control group, routine-acupoint group “Pishu” (BL 20), “Weishu”(BL 21), “Shenshu”(BL 23) and “Qihaishu”(BL 24), “Shuanggu” group “Guanyuan”(RN 4) and “Zusanli”(ST 36), “Yitong” group “Shenshu”(BL 23), “Geshu”(BL 17) and “Dazhu”(BL 11), and “Shuanggu Yitong” group “Guanyuan”(RN 4), “Zusanli”(ST 36), “Shenshu”(BL 23), “Geshu”(BL 17) and “Dazhu”(BL 11), with 12 cases in each group. PMO model was established by removing the bilateral ovaries and fed normally for 90 days. Electroacupuncture (EA, 8- 20 Hz , about 1 mA ) was respectively applied to the aforementioned acupoints for 15 min , once a day and continuously for 8 weeks. The interval between every 6 days' treatments was one day. At the end of the study, the rats were deeply anesthetized and killed for taking out the right tibial tissue which was cut into sections (10 μm). The expression of osteoprotegerin (OPG) mRNA was assayed by using reversed transcription-polymerase chain reaction (RT-PCR) technique. Results: Compared with model control group, the expression levels of OPG mRNA in the 4 acupuncture groups were all up-regulated significantly (P<0.05), and the integral value of the absorbed optical density of “Shuanggu Yitong” group was significantly higher than those of routine-acupoint group,“ Shuanggu” group and “Yitong” group (P<0.05). No significant differences were found among routine-acupoint group, “Shuanggu” group and “Yitong” group (P>0.05). Conclusion: Acupuncture can up-regulate the expression level of OPG mRNA in osteoporosis rats, and the effect of “Shuanggu Yitong” group is markedly superior to that of routine-acupoint acupuncture, “Shuanggu” and “Yitong” groups.

    • Effect of Acupuncture on Serum IgE Level in Guinea Pigs with Asthma

      摘要:Objective: To observe the effect of acupuncture of “Dazhui” (GV 14) at different frequencies on serum IgE in guinea pigs with asthma. Methods: A total of 40 guinea pigs were randomly divided into control, model, 1 d -acupuncture (once everyday), 2 d -acupuncture (once every other day) and 3 d -acupuncture (once every 3 days) groups, with 8 cases in each group. Asthma model was established by intraperitoneal injection of 10% Ovalbumin suspension, followed by inhaling the nebulized 1% Ovalbumin plus normal saline for 30 sec. “Dazhui” (GV 14) was punctured with a filiform needle, with the needle retained for 30 min , once every day, every other day and every 3 days respectively in the 3 acupuncture groups. Serum IgE was determined by radioimmunoassay. Results: In comparison with control group, serum IgE level in model group increased significantly (P<0.01), while compared with model group, IgE levels of the 3 acupuncture groups decreased considerably (P<0.05, 0.01, 0.001), and that of 2 d -acupuncture group was significantly lower than that of 1 d -acupuncture group (P<0.05). No significant difference was found between 1 d -acupuncture and 3 d -acupuncture groups (P>0.05) in serum IgE content. Conclusion: Acupuncture of GV-14 can significantly lower serum IgE in asthma rats, which may contribute to its effect in relieving bronchial asthma.

      摘要:Objective: To explore the relationship between calcium ion and meridian phenomenon. Methods: A total of 10 female goats (30- 35 kg ) tranquilized by muscular injection of chlorpromazine hydrochloride ( 25 mg ) were used in the present study. Electrical potential values (reflecting the concentration) of Ca 2+ in the tissues at “Ganshu”(BL 18), “Dachangshu”(BL 25), “Guanyuanshu” (BL 26) of the Bladder Meridian and 3 control points 3 cm lateral to the abovementioned 3 acupoints were detected by using a needle-type calcium sensor. One minute after insertion of the calcium sensor, the electrical potentials of Ca 2+ in the tissues at “Ganshu”(BL 18), “Dachangshu”(BL 25), “Guanyuanshu” (BL 26) of the Bladder Meridian and 3 control points 3 cm lateral to the abovementioned 3 acupoints were detected by using a needle-type calcium sensor. One minute after insertion of the calcium sensor, the electrical potentials of Ca 2+ were determined once every 5 sec, continuous for 3 min . Results: The values of electrical potential of calcium ion in the aforementioned 3 acupoints were all significantly higher than those in the 3 non-acupoints (P<0.01 or P<0.05), which suggests an enrichment phenomena of calcium ion in meridian-points. Conclusion: Calcium ion concentration in BL-18, BL-25 and BL-26 is significantly higher than that of non-acupoint, which may be one of the specificities of acupoint. It suggests that the determination of concentration of calcium ion could be regarded as an important physicochemical marker in identifying acupoints.

    • Measurement of the Temperature in Deep Tissues along the Governor Vessel

      摘要:Objective: To explore the mechanism of the formation of Infrared Radiant Track along Meridian (IRRTM) course. Methods: The temperature of the skin and subcutaneous deep tissues in 5 points (GV-11, GV-4, GV-5, GV-6 and the point at the thoracic segment 8) along the Governor Vessel and the bilateral control points (non-acupoints, 2 cm to the abovementioned 5 points of the Governor Vessel) was measured in 32 volunteers by using temperature transducer and infrared imaging system. At the same time, the image of infrared radiant track over the back of these subjects was recorded. Results: The temperature values at the skin and in the deep tissues along the Governor Vessel were higher than those of the bilateral non-meridian points (P<0.05, 0.01), suggesting a higher energy metabolism rate in the tissues along the meridian which may contribute to the formation of IRRTM. No significant differences were found between the left and right non-meridian points. In 10 subjects, results of infrared imaging showed a higher temperature band along the course of the Governor Vessel (whole course in 7 cases, and more than half course in 3 cases). Conclusion: A higher temperature exists in the tissues along the Governor Meridian course. It suggests that the meridian is possibly a channel for the transformation and transmission of materials, energy and signals in the regulation of functional activities of the human body.

      摘要:Objective: To explore the mechanism of acupuncture therapy in improving infant dysaudia and aphasis. Methods: Fifty-three children with dysaudia and aphasis caused by medicines were randomly divided into acupuncture group (31 cases) and control group (22 cases). Sishenzhen 4 points 1.5 cun left, right, superior and inferior respectively to Baihui (GV 20), Niesanzhen (point 1: 2 cun straight superior to the ear-tip; point 2 and 3: 1 cun lateral to point 1), Ersanzhen Tinggong (SI 19), Tinghui (GB 2) and Wangu (GB 12), Fengchi (GB 20), Yamen (GV 15), Hegu (LI 4), etc were punctured, once daily, 6 times a week. Infant patients in control group were ordered to take Flunarizine Hydrochloride ( 5 mg /time/d), ATP (20 mg/time, t.i.d.), Vit B_1 ( 20 mg /time, t.i.d.) and Vit B_ 6 ( 20 mg /time, t.i.d.). Both groups were treated for 4 months. Brainstem auditory evoked potentials (BAEP) induced by a brief click stimulus (110- 125 dB , 9 Hz ) to one ear was recorded by using surface electrodes typically placed at the vertex of the scalp and ear lobes and Kepotin EP/EMG Instrument before and after the treatment. The waveform peaks were labeled as Ⅰ-Ⅴ. Results: Compared with pre-treatment, the latency values of peak Ⅰ and Ⅴ of acupuncture group decreased significantly (P<0.05); while comparison between two groups after the treatment showed that the latency values of peak Ⅰ and Ⅴ of acupuncture group were significantly lower than those of control group (P<0.05). The interphases of peak Ⅰ-Ⅲ and peak Ⅲ-Ⅴ presented a similar tendency to those of peak latency, suggesting thatthe effect of acupuncture in improving auditory disorder is significantly better than that of medication. No significant differences were found between two groups in peak Ⅲ latency and interphaseⅠ-Ⅴ (P>0.05). Conclusion: Acupuncture has a good effect in lessening the latency and interphase of BAEP, which may be responsible for its effect in improving hearing and language disorders in children with dysaudia and aphasis.

      摘要:Objective: To observe the effect of acupuncture combined with patent herbal drug (Zhen Ren Yi Zhi Bao, Treasured Drug for improving intelligence) on the intelligence quotient (IQ) and adaptive behavior quotient (ADQ) in infant patients with Down's Syndrome. Methods: Fifty child patients with Down's Syndrome (mongolism) were randomly divided into acupuncture plus medication (n=30) and medication (n=20) groups. For patients of acupuncture plus medication group, Zhisanzhen (point 1: the cross-point between the forehead hair border and the midline of the head; point 2 and 3: 3 cun left and right to point 1), Sishenzhen 4 points 1.5 cun left, right, superior and inferior respectively to Baihui (GV 20), Naosanzhen Naohu (GV 17), bilateral Naokong (GB 19), Shouzhizhen Neiguan (PC 6), Shenmen (HT 7) and Laogong (PC 8), Hegu (LI 4), etc were punctured, once daily, 5 times a week. For medication, patients of two groups were asked to take Zhen Ren Yi Zhi Bao, 2-3 granules/time (b.i.d.), with 1 month being a therapeutic course, two courses altogether. The interval between two courses was one month. IQ and ADQ tests were conducted before and after the treatment. Results: After the treatment, both IQ and ADQ in acupuncture plus medication group all increased obviously (IQ: 60.58±7.46 vs 51.27±5.55; ADQ: 61.96±6.67 vs 52.42±5.47, P<0.01), while in medication group, both IQ and ADQ increased significantly (IQ: 56.40±6.92 vs 51.55±5.90; ADQ: 58.35±6.24 vs 53.25±5.27, P<0.01). Comparison between two groups showed that the increased fraction of IQ and ADQ of acupuncture plus medication group were significantly higher than those of medication group (P<0.01). Conclusion: Acupuncture combined with Zhen Ren Yi Zhi Bao can significantly improve IQ and ADQ of patients with Down's syndrome, and the therapeutic effect of acupuncture plus medication is markedly better than that of simple medication.

      摘要:Objective: To compare the therapeutic effects of scalp-acupuncture, body-acupuncture and body-acupuncture combined with scalp-acupuncture for stroke. Methods: Two hundred and thirty stroke patients were divided into scalp-acupuncture group (112 cases),body-acupuncture group (51 cases), and body- plus scalp-acupuncture group (joint treatment group, 67 cases) according to a random number table. Scalp points used were Motor Area (MS 6), Sensory Area (MS 7), Foot-motor Sensory Area (MS 8), etc for hemiplegia and facial palsy; Motor Area (MS 6), Speech Area (MS 9), etc for aphasia; and body acupoints used were Jianyu (LI 15), Hegu (LI 4), Geshu (BL 17), Huantiao (GB 30), Zusanli (ST 36), Fengchi (GB 20), etc. In treating stroke patients at the early stage, acupoints on the healthy side were punctured first and stimulated with reducing method, followed by puncturing acupoints on the affected side and using mild stimulation maneuver. The treatment was given once daily in the 1 st therapeutic course and once every other day in the 2 st therapeutic course and once every other day in the 2 nd course, continuously for 15 days (one therapeutic course) and the interval between two courses was 7 days. After two courses of treatment, the therapeutic effect was analyzed. Results: After treatment, of the 112, 51 and 67 cases in scalp-acupuncture, body-acupuncture and joint treatment groups, 40, 12 and 34 were cured basically; 48, 22 and 28 experienced marked improvement; 22, 15 and 4 were effective; 2, 2 and 1 failed, with the effective rates being 98.2%, 96.0% and 98.5% respectively. Rank test showed that the curative effect of joint treatment group was significantly superior to that of scalp-acupuncture group and body-acupuncture group (P<0.01), and CT-scanning-shown absorption rates (marked absorption + absorption rates) of the hemorrhagic and infarct loci in joint treatment group were markedly higher than those in scalp-acupuncture and body-acupuncture groups (P<0.01). Conclusion: The curative effect of scalp plus body acupuncture is significantly superior to that of simple scalp- and simple body-acupuncture groups in the treatment of stroke patients.

      摘要:Objective: To observe the effect of electroacupuncture (EA) of Shaoze (SI 1) on hypolactation so as to explore an effective approach for promoting lactation. Methods: A total of 80 cases of hypolactation parturients were randomly divided into treatment group (n=40) and control group (n=40) in accordance with a program edited by Visual Basic 6.0 software. Patients of control group were treated by EA ( 9 V , 20 Hz , intermittent pulses) of Shangyang (LI 1), while those of treatment group treated by EA of Shaoze (SI 1,an empirical acupoint for improving hypolactation). The treatment was conducted once daily, with 5 sessions being a therapeutic course, 2 courses altogether, followed by making a one-month's follow-up. The lactigenous volume of each parturient was detected before, after the treatment and the day of follow-up by squeezing breast repeatedly till emptying the milk. Venous blood samples were collected in the early morning before lactation for assaying the content of prolactin with radioimmunoassay. Results: After the treatment, of the every 40 cases in treatment and control groups, 30 (75.0%) and 2 (5.0%) were cured, 8 (20.0%) and 7 (17.5%) were effective, 2 (5.0%) and 31 (77.5%) failed, with the effective rates being 95.0% and 22.5% respectively. The therapeutic effect of treatment group was significantly superior to that of control group (P<0.001). The lactation volume and prolactin contents at the end of the treatment and 1 month after the treatment in treatment group were significantly higher than those in control group (P<0.001). Conclusion: EA of Shaoze (SI 1) can effectively improve hypolactation parturients' symptoms, raise serum prolactin level, and is thus an effective approach for reliving hypolactation.

      摘要:Objective: To observe the clinical therapeutic effect of abdominal acupuncture therapy for simple obesity. Methods: Forty-one female simple obesity patients were randomized into sham-acupuncture group (n=15) and acupuncture group (n=26), and other 15 normal women were made up of normal control group. Abdominal points used were 8 points 2 cun, 3 cun and 4 cun (upper, lower, left, right, left-upper, left-lower, right-upper and right-lower sites) to the umbilicus respectively, combined with Zusanli (ST 36) and Sanyinjiao (SP 9). The treatment was conducted once every other day, with 20 times being a therapeutic course. Simultaneously, the patients were asked to restrict their diet. For patients of sham-acupuncture group, the acupuncture needles (specially-made retractable needle with dull tip) were just touch the skin and the manipulations were the same to those for acupuncture group. Body mass index (BMI) and the ratio of waist circumstance and body height (WC/BH) were detected before and after the treatment. Results: Before treatment, both BMI and WC/BH of sham-acupuncture and acupuncture groups were significantly higher than those of normal control group. After one course of treatment, no significant changes were found in these two indexes in patients of sham-acupuncture group (P>0.05); while compared with sham-acupuncture group, both BMI and WC/BH decreased significantly (P<0.01), suggesting that acupuncture could effectively reduce the obesity patient's body weight and WC/BH. Conclusion: Abdominal acupuncture therapy is an effective remedy for reducing simple obesity patient's body weight.

      摘要:In the present paper, the authors introduce the origin and development of scalp acupuncture, the theoretical basis including literature description, “functional orientation-projection hypothesis of the cerebral cortex” and comprehensive regulation, and bio-holographic theory; and review clinical treatment and researches on scalp-acupuncture treatment of cerebral hemorrhage in recent years. Scalp acupuncture originates from clinical practice in ancient China, and its theory is based on some descriptions in book Huangdi Neijing (The Yellow Emperor's Internal Classic). In 1970's, scalp-acupuncture therapy was set up under the inspiration of micro-acupuncture therapies (as ear-acupuncture, etc) combined with the study achievements of modern neuroscience and bio-holographic principles. Scalp acupuncture has achieved definite curative effect in the treatment of brain-originated paralysis and was expanded to be applied to the treatment of acute intracerebral hemorrhage in 1980's. Initial 8 clinical trials (randomized controlled trials included) of scalp-acupuncture treatment of acute intracerebral hemorrhage showed a good curative effect, which is closely associated with its favorable regulation on patho-physiological activities and blood brain barrier, regional cerebral blood flow, neuroelectrophysiology, immune function, and biochemical parameters, etc. Therefore, it is extremely urgent and necessary to carry out randomized, controlled and multi-centre clinical and experimental studies to identify its clinical effect and to clarify the related mechanisms.

      摘要:In the present paper, the author comprehensively analyzes results of 34 papers about clinical and experimental researches on acupuncture treatment of coronary heart disease (CHD, myocardial ischemia) published in recent 15 years, and concludes that one of the mechanisms of acupuncture in improving clinical symptoms and signs of CHD patients is its favorable regulation on blood rheology, multiple bio-active substances, substances for anti-oxidation, monoamines substance, intracellular singling, myocardial enzymes, energy metabolism, blood sugar, blood lipid, etc., which provides new evidence for acupuncture treatment of myocardial ischemia.
