      摘要:Objective: To observe the effect of moxibustion at "Zusanli"(ST 36)-"Liangmen"(ST 21) on proliferation and apoptosis of gastric mucosal cells and the relationship between the effect of moxibustion and the expression of heat shock protein 70(HSP70) mRNA so as to explore the underlying molecular biological mechanism of moxibustion in accelerating the repair of the injured gastric mucosa. Methods: Sixty SD rats were randomly assigned to control,model,moxibustion-ST 36+ST 21 and moxibustion non-acupoint groups with 15 cases in each group.Gastric ulcer model was established by fasting(24 h) and forced water(20 ℃) immersion(10 h).Moxibustion was applied to unilateral ST 36,ST 21 or non-acupoint(about 1 cm lateral to ST 21 and ST 36) for about 30 min(4 cones/point),once daily and continuously for 8 days.At the end of the experiments,the rat's gastric mucosa was sampled for examining gastromucosal injury index(ulcer index,UI,GUTH's method),apoptosis index(apoptosis reagent kit),proliferating cell nuclear antigen(PCNA,immunohistochemical method) activity,transforming growth factor α(TGF-α) content(radioimmunoassay),and HSP70 mRNA expression(reversed transcription-polymerase chain reaction,RT-PCR) separately. Results: Compared with control group,gastric mucosal UI,apoptosis index and HSP70 mRNA expression of model group increased significantly(P<0.01,0.05),while TGF-α content and PCNA numerical density decreased remarkably(P<0.01,0.05).While in comparison with model group,UI and apoptosis index of ST 36+ST 21 group lowered considerably(P<0.01,0.05),and TGF-α level,PCNA numerical density,and HSP70 mRNA expression of ST 36+ST 21 group increased remarkably(P<0.01).No significant differences were found between model and non-acupoint groups in UI,TGF-α leveland apoptosis index,and between ST 36+ST 21 and non-acupoint groups in PCNA numerical density(P>0.05). Conclusion: Moxibustion at "Zusanli"(ST 36)+"Liangmen"(ST 21) has a protective effect on gastromucosa in stress gastric ulcer rats,which is closely related to its actions in promoting synthesis of TGF-α and proliferation of gastromucosal cells,suppressing gastromucosal cell apoptosis,and upregulating HSP70 mRNA expression.

      摘要:Objective: To analyze the effect of electroacupuncture(EA) on myocardial cellular signal transduction of G-protein in myocardial ischemia(MI) rats so as to explore the molecular pathway of EA against MI injury. Methods: Twenty male SD rats were randomized into normal,model,EA-normal and EA-model groups,5 rats in each group.MI model was established by occlusion of the descending anterior branch of the left coronary artery under anesthesia with 10% chloral hydrate(0.36 mL/100 g).EA (1.3 mA,2 Hz,300 μs in the duration of pulse) was applied to "Shenmen"(HT 7) for 20 min,once daily continuously for 3 days.Three days later,the myocardial tissue sample was taken for shifting out related G-proteins and corresponding genes by using gene chip technology. Results: In comparison with model group,after EA of "Shenmen"(HT 7),782 differentially expressed genes were found,of which,328 were upregulated and 454 downgrelated;compared with normal group,426 differentially expressed genes were found in EA-normal group,among them,217 genes were upregulated and the other 209 downregulated in the expression.Further analysis showed that,21 genes were associated with G-protein signal transduction pathway,15 genes(L01115.1,Gng11,Adcy2,Calm3,Ppp3ca,Adcy5,Gnao,Prkcc,Arhgef1,RGD:619921,Pccb,Prkcd,Ppp3cb,Gng5,Gnai1) had an apparent expression in the 4 groups,4 genes(Gnaz,Prkcb1,Adcy3 and Adcy8) had no marked changes,and the rest 2(Gng8 and Prkar2b) were differentially expressed genes.Of the 2 differentially expressed genes,Gng8 displayed weaker and stronger expression in EA-normal and EA-model groups respectively, and had no expression in normal and model groups,suggesting being closely related to EA-HT 7;while Prkar2b showed no expression in normal and EA-normal groups,and had a detectable expression in model and EA-model groups,suggesting being related to MI. Conclusion: EA-HT 7 can affect gene expression of ischemic myocardiocytes;among the differentially expressed genes being closely associated with G-protein signaling pathway,Gng8 and Prkar2b may play an important role in EA-induced protective action on myocardial ischemia.

      摘要:Objective: To explore the effect of electroacupuncture(EA) of acupoints of the Pericardium Meridian of Hand-Jueyin on changes of mitochondria in myocardial ischemia/reperfusion(MI/R) injury rats so as to explore the protective mechanism of acupuncture on ischemic myocardiocytes. Methods: Fifty male Wistar rats anesthetized by 10% urethane(1 g/kg) were evenly randomized into control,model,EA-"Neiguan"(PC 6),EA-"Ximen"(PC 5),EA-"Zhigou"(TE 6) groups.Under artificial respiration,MI/R model was established by occlusion of the left descending anterior branch(DAB) of the coronary artery for 40 min,followed by reperfusion for 60 min.EA(30-100 Hz in frequency,20-30 V in strength,dense-sparse waves) was applied to the above mentioned acupoints for 20 min,two times altogether.Then,the myocardial sample(below the ligation site of DAB) of the left cardiac ventricle was taken for observing the morphological changes of mitochondria under transmission electron microscope. Results: Before occlusion of DAB,ECG-ST_Ⅱ values of the 5 groups had no significant differences(P>0.05),while after occlusion of the coronary artery,ECG-ST_Ⅱ of model and 3 EA groups increased considerably.Compared with model group,ST values of EA-"Neiguan"(PC 6) and EA-"Ximen"(PC 5) groups were significantly lower from 20 min to 40 min after occlusion,and 30 min after reperfusion(P<0.05),showing that EA could remarkably reduce MI/R induced myocardial injury.Correspondingly,in model group,mitochondrial swelling,reduction in the number of the mitochondrial cristsae,and broken cristae with disordered arrangement and vacuous changes were found,and in EA-"Zhigou"(TE 6) group,these pathological changes were similar.But in EA-"Neiguan"(PC 6) group,and EA-"Ximen"(PC 5) group,the situations were much better. Conclusion: EA stimulation of acupoints of the Pericardium Meridian can improve ischemic ECG and reduce MI/R induced structural injury of myocardial mitochondria in rats,which may contribute to its protective effect on the ischemic myocardial cells.

      摘要:Objective: To investigate the effect of electroacupuncture(EA) on plasma soluble intercellular adhesion molecule(sICAM)-1 and endothelin(ET)-1 contents in cerebral ischemia-reperfusion(CI/R) rats for revealing the underlying mechanism of acupuncture in preventing CI/R induced vascular endothelium injury. Methods: A total of 40 SD rats were randomly divided into normal control(n=10),sham-operation(n=10),model(n=10),and EA(n=10) groups.CI/R model was established by electrocoagulation of the right external carotid artery and occlusion of the middle cerebral artery(MCA) with a nylon thread(burned global end) for 30 min and reperfusion under anesthesia(10% chloral hydrate,300 mg/kg).EA(4/16 Hz,1-3 V) was applied to "Baihui"(GV 20) and "Shuigou"(GV 26) for 30 min,and once again 12 hours later.Twenty-four hrs after CI/R,blood sample was collected by excising the rat's eyeballs for assaying plasma sICAM-1 and ET-1 contents with enzyme linked immunosorbent assay(ELISA) and radioimmune methods respectively. Results: In comparison with control group,both sICAM-1 and ET-1 contents in model group increased significantly(P<0.01),while in comparison with model group,these two indexes of EA group decreased significantly(P<0.05).No significant differences were found between control and sham-operation groups in these two indexes. Conclusion: EA intervention can effectively reduce CI/R induced vascular endothelia injury,which may contribute to its action in clinically improving cerebral ischemia.

      摘要:Objective:To observe the impact of acupuncture on morphological changes of retinal cells after intraperitoneal injection of N-methyl-N-nitrosourea(MNU) in the rat.Methods: Twenty 50-day-old female SD rats were randomized into control(n=4),model(n=8),and acupuncture(n=8) groups.Retinal degeneration model was established by intraperitoneal injection of MNU(50 mg/kg) in the rats of the later two groups.Bilateral "Xinming"(behind the ear,the mid-point of the folding region inferior to the ear's lower angle),and "Jingming"(BL 1) were punctured once daily,continuously for 7 days.Following finishing the experiments,the rat's eyeballs were taken out to be immersed in 4% paraformaldehyde,embedded in paraffin,sectioned,stained with hematoxylin and eosin stain,and observed by using light microscope and transmission electron microscope(TEM).Results: Compared with normal control group,the outer nuclear layer(ONL) thickness and the total thickness of the retina in model group showed thinning significantly(P<0.01),and the cells' arrangement was in disorder;when compared with model group,the ONL thickness and the total thickness in acupuncture group were markedly bigger(P<0.01) and the cellular arrangement was relatively well-arranged.Results of TEM showed smaller(withered) cellular body,condensed nuclear chromatin,swelling of the mitochondria with severer vacuole,etc in model group,while in acupuncture group,the apoptosis of the photoreceptor cells was apparent but the injured degree was much lighter than that of model group.Conclusion: Acupuncture can protect retinal photoreceptor cells from MNU-induced injury in rats.

      摘要:Objective: To compare the analgesic effects of electroacupuncture(EA) at different stimulating intensities and different treatment intervals so as to explore a suitable set of therapeutic parameters for chronic pain.Methods: A total of 70 Wistar rats were randomized into model(n=10),1 mA-EA(n=30) and 5 mA-EA(n=30) groups which of the later two groups were further divided into 1 time(t)/day(d),1 t/3 d and 1 t/week(w) EA subgroups,with 10 cases in each subgroup.Chronic constrictive injury(CCI) pain model was established by loosely ligating the sciatic nerve near the proximal side of the bifid site.Paw withdrawal latency(PWL) was detected once every other day by using radiant heat source to radiate the rat's bilateral hind-paws.The difference values between the affected limb and the healthy limb were used to judge changes of the pain threshold.EA (2/15 Hz,1 mA,5 mA) was respectively applied to bilateral "Zusanli"(ST 36)-"Yanglingquan"(GB 34) for 30 min,once everyday,every 3 days and every week in different subgroups.Results: 1) When 1 mA stimulating current was used,in comparison with model group,the mean difference values of PWL in 1 t/d-EA,1 t/3 d-EA and 1 t/w-EA subgroups from the 1st day to 21st day to 21st day after EA were significantly lower(P<0.05);while comparison among the 3 EA subgroups showed that the mean difference values of PWL in 1 t/3 d and 1 t/w subgroups were significantly higher than those of 1 t/d group(P<0.05),indicating that 1 t/d-EA,1 t/3 d-EA and 1 t/w-EA at the lower intensity all had a marked analgesic effect and that of 1 t/d-EA was the best.In addition,the accumulative effect in 1 t/3 d and 1 t/w subgroups was apparent along with the increase of the therapeutic times.2) When 5 mA stimulating current was used,compared with model group,the mean difference values of PWL in 1 t/d-EA subgroup and in 1 t/w-EA subgroup from the 1st day after EA were significantly lower(P<0.05);while comparison among the 3 EA subgroups showed that the mean difference values of PWL in 1 t/3 d and 1 t/w subgroups were significantly higher than those of 1 t/d group(P<0.05),indicating that 1 t/d-EA,1 t/3 d-EA and 1 t/w-EA at the lower intensity all had a marked analgesic effect and that of 1 t/d-EA was the best.In addition,the accumulative effect in 1 t/3 d and 1 t/w subgroups was apparent along with the increase of the therapeutic times.2) When 5 mA stimulating current was used,compared with model group,the mean difference values of PWL in 1 t/d-EA subgroup and in 1 t/w-EA subgroup from the 1st day to 21st day to 21st day were significantly lower(P<0.05),displaying that at the higher stimulating current,1 t/d-EA and 1 t/w-EA subgroups had apparent analgesic effects,while 1 t/3 d-EA had no marked analgesic effect.3) When EA treatment was given once everyday,the mean difference values of PWL in 1 mA group decreased significantly faster(P<0.05),meaning that the analgesic effect of lower stimulating intensity EA was significantly better than that of higher stimulating intensity in CCI rats.Conclusion: In the treatment of neurogenic pain with EA,both lower and higher stimulating intensities have an apparent analgesic effect,the effect of lower intensity is remarkably superior to that of higher intensity;when lower intensity EA is applied,the analgesic effect of 1 t/d is the best,and the accumulative effect is definite;when the higher intensity EA applied,the analgesic effect of 1 t/d and 1 t/w is the best.

      摘要:Objective: To investigate the effect of electroacupuncture(EA)on spinal interleukin-6(IL-6) protein expression in neuropathic pain rats.Methods: Twenty-four adult SD rats were evenly and randomly divided into sham-operation group,model group and EA group.Chronic constrictive injury(CCI) model was established by ligating the sciatic nerve with catgut under anesthesia(10% chloral hydrate,300 mg/kg).EA(2 Hz,1 mA) was applied to "Weizhong"(BL 40) and "Huantiao"(GB 30) for 30 min,once on the 1st,4st,4th and 7th and 7th day after operation separately.The mechanical(von Frey filaments) and thermal(heat radiation) pain thresholds were measured before operation and on the 7th day after operation separately.The mechanical(von Frey filaments) and thermal(heat radiation) pain thresholds were measured before operation and on the 7th day after the operation.The expression of IL-6 in the spinal cord(L_4-5) tissues was assayed by immunohistochemical technique.Results: Before operation,no significant differences were found among the 3 groups in both mechanical pain threshold and thermal pain threshold of the bilateral hind-limbs(P>0.05).In comparison with sham-operation group,after CCI,both the mechanical and thermal pain thresholds of model group on the affected hind-limb lowered significantly,appearing mechanical allodynia and thermal hyperalgesia.Compared with model group,the mechanical and thermal pain thresholds of EA group increased significantly after EA(P<0.05).Correspondingly,the average optical density value of IL-6 immunoreactive(IR) positive cells in the ipsilateral spinal dorsal horn of model group was significantly higher than that of sham-operation group and EA group(P<0.05).It indicated that EA could markedly suppress CCI-induced allodynia and hyperalgesia and increase of IL-6 expression in the spinal cord.Conclusion: EA analgesia may be related to its resultant downregulation of IL-6 expression in the dorsal horn of the spinal cord in neuropathic pain rats.

      摘要:Objective: To observe changes of vagal electrical activity after electroacupuncture(EA) at "Zusanli"(ST 36).Methods: Thirty male SD rats(220±20 g) were evenly and randomly divided into control group,EA group,and non-acupoint group.After anesthesia(20% urethane,1 g/kg,i.p.),the rat's bilateral cervical vagus nerves were isolated and ligated on the middle part;then,two pairs of bipolar platinum electrodes were respectively placed at the cephalic and caudal ends of the ligated site for recording the discharges of the afferent and efferent fibers of the vagus nerve(covered with mineral oil).The electrical signals from the electrodes were amplified,filtered(100-1000 Hz),and fed to a computer for recording and analysis.EA(3 Hz,2-4 V) was applied to bilateral "Zusanli"(ST 36) and non-acupoints(about 5 mm beside ST 36) for 30 min separately.Results: Both afferent and efferent vagal activity of control group and non-acupoint group had no apparent changes(P>0.05) during the experimental course.In EA group,about 7.5 min after EA of "Zusanli"(ST 36),both the frequency and the peak values of the efferent vagal electrical activity increased significantly in comparison with pre-stimulation(P<0.01).However,this situation did not happen in the afferent vagal activity.Conclusion: EA of "Zusanli"(ST 36) can increase the efferent activity of the vagal nerve,but does not influence the afferent activity.

      摘要:Objective: To observe the therapeutic effect of acupuncture combined with biofeedback therapy in the treatment of outlet obstruction constipation. Methods: A total of 45 patients with outlet obstruction type constipation were randomly divided into treatment(acupuncture combined with biofeedback therapy) group(n=25) and control(biofeedback therapy only) group(n=20).In treatment group,bilateral Tianshu(ST 25),Zusanli(ST 36),Shangjuxu(ST 37),Baihui(GV 20),Dachangshu(BL 25) were used as the main acupoints in combination with Hegu(LI 4),Zhongwan(CV 12),etc.Biofeedback therapy was conducted by training the patient to repeatedly make rectal contraction and relax activities for 45 min,once daily.The treatment was given once everyday,with 5 sessions being a therapeutic course,and continuously for 3 courses.The interval between two courses was 2 days.The number of defecation,shape of stool,difficulty degree of defecation,and intra-rectal pressure were observed and detected. Results: Following the treatment,self-comparison of these two groups indicated that the times of defecation/week,scores of stool shape and the mean intrarectal pressure increased significantly(P<0.01),the score of difficulty degree of defecation lowered considerably(P<0.01),no significant differences were found between two groups in these indexes(P>0.05).One month after the treatment,the therapeutic effects(long-term effect) of treatment group were significantly superior to those of control group in raising the times of defecation/week,score of stool shape,and lowering the difficulty degree of defecation and maximum tolerable volume of intra-rectal pressure(P<0.05). Conclusion: Both acupuncture and biofeedback therapies can better the symptoms of outlet obstruction constipation patients,and the long-term therapeutic effect of acupuncture is markedly superior to that of biofeedback therapy.

      摘要:Objective: To observe the clinical therapeutic effect of electroacupuncture(EA) at Jianyu(LI 15) for scapulohumeral periarthritis of prophase adhesion type. Methods: In accordance with the requirements of evidence-based medicine,in the present study,unified criteria for diagnosis and assessment of therapeutic effects were adopted,various treatment parameters were fixed,and changes of shoulder pain index(visual analogue scale,VAS) and ranges of shoulder-joint movement were taken as the observed indexes.A total of 216 cases of scapulohumeral periarthritis of prophase adhesion type were randomized into EA group(108 cases) and medication group(108 cases).For patients of EA group,Jianyu(LI 15) was punctured and stimulated electrically with frequency of 100 Hz and 2 Hz and strength of (10±2) mA for 30 min,and those patients of medication group were ordered to take Brufen(0.6 g/day).The treatment was given once daily,with 7 days being a therapeutic course.Results: After one course of treatment,EA treatment gained a good therapeutic effect with the total effective rate and cure+markedly effective rate being 92.59% and 78.70% respectively which were significantly higher than those(82.41% and 59.25% separately) of medication group(P<0.05).Self-comparison of the two groups showed significant decrease in pain index and marked increase in the score of shoulder-joint movement(P<0.01).The decrease rate of the pain index and the improving rate of the shoulder-joint movement in EA group were 63.21% and 22.64% separately,being also slightly higher than 60.16% and 16.95% of medication group.Conclusion: EA of LI 15 can relieve scapulohumeral periarthritis patients' pain and improve the range of shoulder-joint movement,and its therapeutic effects are significantly better than those of Brufen.Thus EA of LI 15 is a better remedy for scapulohumeral periarthritis of prophase adhesion type.

      摘要:In the present paper,the author makes a comparison about Chinese and South-Korean original research articles on acupuncture analgesia(AA) collected by SCI and Medline from the number of papers,impact factor of the published journals,frequency of citation of papers,the researchers' output,cooperation among research institutes,types of articles,and the quality and validity of methodology.It is found that 1) in comparison with South-Korean articles,Chinese theses are more in the number and earlier in the publication time,but the quantity of Chinese papers changes bigger from 1994 to 2004,while that of South-Korean theses is relatively stable;2) the impact factor of Chinese researchers' papers is dramatically different from paper to paper,while that of South-Korean theses is relatively similar;3) Chinese authors' paper output is higher,while the cooperative researches in South-Korean research institutions are more;4) the South-Korean authors generally hold an altitude of "organized skepticism",but majority of Chinese authors do not.It is concluded that the research on AA in China is languishing while that in South-Korea is flourishing;the general idea and tendency about research on AA are unfavorable to the long-term development of Chinese Acupuncturology in China.

      摘要:Through systemic collection,sorting and analysis on acu-moxibustion prescriptions for stroke treatment in ancient Chinese medical literature,we concluded that the ancient prescriptions function in inducing resuscitation to restore consciousness in the treatment of emergency and may be divided into "comprehensive prescription" type,"Da Jie Jing Fang" (Prescription of acupoints selected according to the running course of meridian and the arrangement sequence) type and "kidney-qi strengthening prescription" type.Counterevidence from these prescriptions showed that the pathogenesis of stroke was insufficiency of yang and invasion of evil wind,and the insufficiency refers to deficiency of the kidney-yang.These rules had been applied to clinical practice and proved effective.In addition,for preventing stroke the related acupoints should be selected in accordance with the concrete conditions,rather than focusing only on Zusanli(ST 36) and Juegu(GB 39).The present research may have certain significance in guiding clinical practice to solve some complicated problems in acupuncture treatment of stroke.

      摘要:Diabetic Peripheral Neuropathy(DPN) is one of the most important factors for disabled patients with diabetes.So far,there have been no ideal methods to treat in Western medicine.In this article,the authors review clinical treatment of DPN by acupuncture and moxibustion in recent ten years from 1) simple therapies including hand acupuncture,moxibustion,electroacupuncture(EA),acupoint-injection,etc;2) comprehensive therapies,such as acupuncture combined with moxibustion,acupuncture combined with cupping,acupuncture with Chinese medicinal herbs,acupuncture with acupoint plaster,and EA with acupoint-injection;and 3) clinical researches.Clinical practice demonstrates that acu-moxibustion therapies are effective in the treatment of DPN and more researches are needed.
