      摘要:Objective:To observe the influence of electroacupuncture(EA)on mitochondrial ultrastructure and mitochondrial enzymatic activity in hippocampus of senescence accelerated mouse/prone 8(SAMP 8)so as to explore its underlying mechanism in the treatment of Alzheimer's Disease(AD).Methods:Twenty-four SAMP 8 mice were randomly and uniformly divided into model group and EA group and other 12 SAMR1 mice were used as control group.EA(2-100 Hz,2-4 V)was applied to"Baihui"(GV 20)and "Yongquan"(KI 1)for 30 min.Changes of learning and memory ability were measured by Morris water maze test.The ultrastructure of hippocampal mitochondria was observed by using transmission electronic microscope.The activities of cytochrome C oxidase(COX)in hippocampal mitochondria were detected by spectrophotometric method.Results:In comparison with control group,the average escape latencies of model group were significantly longer from the 1st day to the 5th day in swimming test(P<0.01),while in comparison with model group,the escape latencies of EA group at the corresponding test day were markedly shorter(P<0.05,0.01),despite being still obviously longer than those of control group.The ultrastructure of most mitochondria of EA group was basically normal,and the mitochondrial cristae were line up in order and presented mild swelling.Volume density and area density of mitochondria and COX activity of model group were significantly lower than those of control group,and these 3 indexes of EA group were significantly higher than those of model group(P<0.05,0.01).Conclusion:EA can improve AD rat's hippocampal mitochondrial structure and increase mitochondrial COX activity,which may contribute to its effect in improving the rat's learning and memory ability.

      摘要:Objective:To investigate the protective effect of electroacupuncture(EA)on ischemic cerebral neurons and the related intracellular signal transduction mechanism in rats with cerebral ischemia-reperfusion(CI-R).Methods:A total of 24 SD rats were randomized into sham-operation(n=6),model(n=6),EA(n=6)and EA+PD 98059(n=6)groups.CI-R model was established by right middle cerebral artery occlusion(MCAO)for 2 hours and reperfusion for 24 hours.EA(disperse and dense waves,80-100 Hz,1-3 mA)was applied to "Dazhui"(GV 14)and "Baihui"(GV 20)for 60 minutes.The activity of extracellular signal-regulated kinases(ERK)positive cells in cerebral cortex tissue was detected with immunohistochemical method.Lumbar intrathecal injection of PD 98059,the inhibitor of mitogen activated protein kinase(MAPK)1(MEK1,2.78 mg/kg),was conducted in rats of EA+PD 98059 group during CI-R.Results:In comparison with sham-operation group,the score of behavior test and the number of ERK immuno-reaction(IR)-positive neurons of model group decreased significantly(P<0.01,0.05),while compared with model group,the score and the number of ERK IR-positive neurons in EA group increased markedly(P<0.01).No significant difference was found between model group and EA+PD 98059 group in behavior score and the number of ERK IR-positive neurons(P>0.05).Compared with sham-operation group,the mean area density and mean numerical density of ERK IR-positive neurons in model group were significantly lower(P<0.05),while in comparison with model group,the mean area density and mean numerical density in EA group were significantly higher(P<0.01);no significant differences were found between model and EA+PD 98059 groups in the area density and numerical density(P>0.05).Conclusion:EA can reduce CI-R induced injury of cerebral neurons,which may be related to its effect in regulating ERK signaling pathway.

      摘要:Objective:To investigate the effect of electroacupuncture(EA)on the expression of neuropeptide Y(NPY)and calcitonin gene-related peptide(CGRP)in hippocampus tissue in cerebral hemorrhage rats so as to reveal its underlying mechanism in protecting ischemic cerebral neurons.Methods:Thirty Wistar rats were randomly divided into normal control,model and EA groups,with 10 cases in each.Cerebral hemorrhage model was duplicated by intracerebral injection of collagenase Ⅶ(0.8 μL,AP:1.0 mm,R:4.0 mm).EA(1 mA,2 Hz)was applied to "Shuigou"(GV 28)for 30 min,once again 24 h later.NPY and CGRP expression of hippocampus tissue was assayed with immunohistochemical method(SABC).Results:Compared with normal control group,OD values of hippocampal NPY immuno-reaction(IR)-positive and CGRP IR-positive neurons in model group increased obviously(P<0.01,0.05),while compared with model group the OD value of NPY IR-positive neurons in EA group decreased significantly(P<0.01),and the OD value of CGRP IR-positive neurons increased further considerably(P<0.01).Conclusion:EA of "Shuigou"(GV 28)can suppress cerebral ischemia-induced over expression of NPY,and potentiate the expression of CGRP IR-positive neurons in the hippocampus,balancing functional activities of NPY and CGRP to improve microcirculation in the brain tissue.

      摘要:Objective:To observe the effect of electroacupuncture(EA)of "Xiusanzhen" on anti-oxidation in cerebral ischemia-reperfusion(CI-R)injury rats and to analyze its underlying mechanism in resisting cerebral ischemic injury.Methods:A total of 30 SD rats were randomized into sham,model and EA groups,with 10 cases in each.CI-R model was established by reversible occlusion of the middle cerebral artery.EA(80-100 Hz,1-3 mA)was applied to "Xiusanzhen"(the mid-point of the nasal root,2 mm bilateral to the nasal root)for 60 min.The levels of catalase(CAT),glutathione peroxidase(GSH-Px)and total anti-oxidation capability(T-AOC)in cerebral cortex tissue were determined by using spectrophotometry and chromatometry respectively.Results:Compared with control group,the levels of CAT,GSH-Px and T-AOC in model group were markedly lower(P<0.01);while compared with model group,the levels of CAT,GSH-Px and T-AOC increased significantly in EA group(P<0.01).Conclusion:EA of "Xiusanzhen" has definite effects in raising CI-R-induced decrease of CAT,GSH-Px activity and T-AOC in the cerebral cortex,which may contribute to its effect in reducing ischemic cerebral damage.

      摘要:Objective:To observe the effects of warming-promotion needling(WPN)on the levels of plasma calcitonin gene-related peptide(CGRP)and endothelin(ET)contents in vascular dementia(VD)rats so as to investigate its underlying mechanism in improving VD.Methods:A total of 50 Wistar rats were randomized into normal control,model,medication,WPN,and needle-twirling groups,with 10 cases in each.VD model was established by intraperitoneal injection of sodium nitroprusside(2.5 mg/kg)in combination with repeated occlusion of the bilateral common carotid arteries and reperfusion.For rats in medication group,Nimodipine(0.0108 g/kg)was given(oral perfusion),and for animals in WPN and needle-twirling groups "Dazhui"(GV 14),"Baihui"(GV 20)and "Shuigou"(GV 26)were punctured manually by manipulating the needle with WPN and common twirling methods respectively,60 s for each acupoint.The treatment was given once daily for 15 days.Plasma CGRP and ET contents were measured by radioimmunoassay.Results:Compared with normal control group,plasma CGRP content in model group decreased considerably(P<0.01),while plasma ET level in model group increased significantly(P<0.01).In comparison with model group,CGRP contents of WPN,needle-twirling and medication groups increased significantly(P<0.05,0.01),while plasma ET levels of WPN,needle-twirling and medication groups decreased significantly(P<0.01).The therapeutic effects of WPN group were significantly superior to those of needle-twirling group in raising CGRP content and lowering ET level(P<0.01);and the therapeutic effect of medication group was markedly superior to that of needle-twirling group in lowering ET content(P<0.01).No significant differences were found between WPN and medication groups in raising plasma CGRP and lowering ET level,and between needle-twirling and medication groups in raising CGRP content(P>0.05).Conclusion:Warming-promotion needling can effectively lower plasma ET and raise plasma CGRP level in VD rats,which may contribute to its clinical effect in improving VD patients' memory-learning ability.

      摘要:Objective:To observe the effect of electroacupuncture(EA)on serum leptin contents and the expression of leptin receptor in the hypothalamus in simple obesity rats so as to explore the underlying mechanism of acupuncture in reducing weight.Methods:A total of 45 male Wistar rats were randomly divided into normal control,model and EA groups with 15 cases in each.Obesity model was established by feeding the animal with high fat forage.EA(10 Hz,1.5 V)was applied to "Zusanli"(ST 36)and "Zhongwan"(CV 12)for 10 min,once daily for 14 days.The body weight,body length and Lee's index were measured once every 10 days.Serum leptin contents were measured by radioimmunoassay,and leptin receptor expression was detected with immunohistochemistry.Results:In comparison with normal control group,the body weight,Lee's index and serum leptin content of model group increased significantly(P<0.05,0.01),while in comparison with model group,these 3 indexes in EA group decreased markedly(P<0.05,0.01).Compared with normal control group,the number of leptin receptor immuno-reaction(IR)-positive cells in the hypothalamus in model group decreased considerably(P<0.01),while compared with model group,the number of IR-positive cells increased significantly in EA group(P<0.01).It suggests that EA-stimulation induced improvement of leptin resistance may contribute to its effect in reducing body weight.Conclusion:EA can lower serum leptin level and upregulate the expression of leptin receptor in the hypothalamus in obesity rats.

    • Placebo Control in the Design of Acupuncture Clinical Trials

      摘要:Placebo controls have often been used in standard clinical studies in order to minimize the effects of psychological interference.However,no ideal placebo(including false needle technique)has been designed for the purpose of acupuncture research.As a result,many of the clinical studies of acupuncture do not follow proper criteria for designing a conventional clinical trial.Understanding the differences of the placebo effect in acupuncture and pharmaceutical drug research is the key to answering this challenge.To do this,the concept of placebo,sham point,acupuncture point specificity and meridian all need to be properly considered in order to avoid a failure in clinical design.The sham points control in the previous acupuncture studies often obeyed the definition of the placebo.Being different from drugs which act upon pre-designed specific receptors,acupuncture may have various effects on multiple systems,organs and a lot of cells and receptors,multiple transmitters,hormones,etc..For this reason,the results of acupuncture treatment,such as pain relief and relaxation,have been criticized as a "placebo" effect.However,evidence from fMRI and other studies strongly suggest that acupuncture affects multiple psychological and emotional control centers of the brain,such as the limbic system and the prefrontal cortex.This may result in a series of functional modulations,which involves pain and stress management.The results suggest that the placebo effect could be induced by non-specific physical stimulation of acupuncture and may favor the patient's health recovery to a certain degree.From this point of view,acupuncture may be a unique remedy for "mind-body" disorders.The current placebo control design in clinical trials of acupuncture needs to be reevaluated.

      摘要:Traditional Chinese Medicine(TCM)and acupuncture are ancient medical theories that combine the experience of clinical practice of TCM and acupuncture with ancient Chinese philosophy.Although basic and clinical researches have been carried out to explore the effectiveness and possible mechanisms of TCM and acupuncture treatment,many aspects of TCM and acupuncture in daily patient care still need to be optimized under the framework of TCM and acupuncture.The greatest geneticist Gregor J.Mendel had shown us that great science like Mendel's Law can be developed with a careful planning of a large sample collection,accurate observation,data recording,innovative data analysis and mathematic modeling.Learning from Mendel,TCM and acupuncture can take advantage of bioinformatics in genomic and post-genomic era to advance TCM and acupuncture according to their own theories.In recent years,many new data mining and statistical analysis methods developed with bioinformatics platforms provide tools for scientists to investigate diseases and treatments using less of a reductionism approach and more of a systemic approach.Bioinformatics can provide an effective platform to improve the accuracy of data collection in TCM and acupuncture clinic.It can facilitate large sample collections and provide computational tools for data mining and statistical analysis.It is a systems approach to advance TCM and acupuncture in genomic and post genomic era.

    • Research on the Substantial Basis of Acupuncture Efficacy

      摘要:The present paper retrospects the research history of acupuncture and moxibustion in China,particularly expounds the background,definition,theoretical basis,goal,technological means and the life science value of the research on the substantial basis of acupuncture efficacy based on the researches about the mechanism of acupuncture actions in the past decades of years,and studies about serum proteome in recent years.The research on the substantial basis of acupuncture effects should take the essence of the life phenomena as the starting point of strategy,and the response genes and response proteins of acupuncture efficacy as the primary technical tactics for explaining the underlying biological mechanisms of acupuncture effects.This research project has higher theoretical altitude and clear academic innovation,and important strategic significance for maintaining the leading position in the field of acupuncture research in the world and for owning originally creative independence intellectual property rights.Moreover,this article also puts forward a few methods for the application of achievements of researches on the substantial basis of acupuncture effects.This research project may have a very active significance for unfolding the future developmental direction of acupuncture research.

      摘要:During general anesthesia for some specific surgical cases which need to lower blood pressure,hypotension induced lower blood-perfusion of tissues and the subsequent blood pressure-raising and reperfusion may induce an imbalance between tissue perfusion and oxygen metabolism,probably leading to different degrees of injury of the brain,heart,liver,gastrointestinal tract,etc..Nowadays,simple acupuncture anesthesia has been become into compound anesthesia of acupuncture and drugs,and transcutaneous electrical acupoint stimulation(TEAS)has been already used as one principal measure for acupuncture anesthesia.It has been demonstrated that acupuncture stimulation has a protective effect on the important organs as brain,heart,liver,kidney,etc.,and there exists a central dependability between acupuncture analgesia and blood pressure regulation.In addition,it has been found that during controlled lowering of blood pressure by TEAS in combination with isoflurane general anesthesia,both oxygen metabolism in tissues and performance of the circulation system are rather stable.Along with the increasing of cranial operation and major-arthroplasty,TEAS which possesses important organ protective effect during controlled-blood pressure lowering under general anesthesia has a good applicable prospect.

      摘要:The present paper expounds the view-point that "traditional learning of acu-moxibustion is impossible to directly become the starting-point for contemporary evidence-based scientific study by using reductionism as a reasonable method",puts forward that the knowledge which is transformed from the effective techniques of current acupuncture clinical practice,and accords with the methodological characters of the investigation field of modern experimental studies is the starting-point of the experimental studies of acu-moxibustion,and holds that traditional learning system of acu-moxibustion may still exist for a long-term,and can't have a cross-talk with evidence-based experimental studies on the same level in a longer period of time;and the tension between the traditional acu-moxibustion learning and the experimental acu-moxibustion learning is probably one of the power for accelerating the development of acu-moxibustion learning.

      摘要:Acupuncture pretreatment has been shown to attenuate ischemic arrhythmias.In order to provide scientific explanations about the mechanism of anti-arrhythmic effect of acupuncture pretreatment,the current progress of researches on arrhythmia and acupuncture pretreatment was reviewed from 1)clinical significance of acupuncture pretreatment in the treatment and prevention of myocardium ischemia and ischemic arrhythmias;and 2)the similarity between acupuncture pretreatment and ischemic preconditioning.In addition,the authors of the present paper make a systemic analysis on the possible mechanism of acupuncture and acupuncture pretreatment underlying the anti-arrhythmic effect from intracellular calcium activities,gap junction between cardiac myocytes,and electrical conductance between cardiac myocytes.The present paper attempts to provide some possible cues for studies on acupuncture pretreatment of ischemic arrhythmia and break a new pathway for clinical treatment.
