      摘要:Objective To study the bio-molecule basis of correlativity between Heart Meridian and hypothalamus from gene differential expression. Methods Twenty-eight healthy Sprague-Dawley male rats were divided into 4 groups randomly: normal control, model, Heart Meridian-electroacupuncture (EA) and Lung Meridian-EA groups, with 7 cases in each. Acute myocardial ischemia (AMI) model was duplicated by occlusion of the left anterior descending branch (LAD) of the coronary artery under anesthesia (3% Nemb, 40 mg/kg). EA (1.1 mA, 2 Hz, duration of waves 300 μs) was applied to bilateral "Shenmen" (HT 7)-"Tongli" (HT 5, Heart Meridian), "Taiyuan" (LU 9)-"Lieque" (LU 7, Lung Meridian) separately for 20 min, once daily for 3 days. Then, hypothalamus tissue was taken for differentially expressed gene profiling analysis (Affymetrix GeneChip Rat Genome U 230 Affymetrix). Results Compared with model group, among the differentially expressed genes above 2 times in Heart-Meridian-EA group and Lung-Meridian-EA group, 190 and 57 were up-regulated,34 and 26 down-regulated separately. Compared with Lung-Meridian-EA group, among the 175 differentially expressed genes above 2 times in hypothalamus tissues of Heart-Meridian-EA group, 147 were up-regulated and 28 down-regulated. Conclusion In comparison with EA of Lung Meridian, a different gene expression profile of hypothalamus tissue is involved in the effect of EA of Heart-Meridian on acute myocardial ischemia.

      摘要:Objective To observe the effect of acupuncture on p 130 expression in neocortex and hippocampus of Senescence Accelerated Mouse/Prone (SAMP) 10 so as to explore its underlying molecular mechanism in treating dementia and aging-related diseases. Methods Thirty SAMP 10 were randomly divided into model, acupuncture, and non-acupoint groups with 10 cases in each, and 10 SAM/Resistant (SAMR)1 were used as normal control group. The needle manipulation of "Yi Qi Tiao Xue, Fu Ben Pei Yuan" (Reinforcing qi and blood, and strengthening the body resistance) was applied to "Shanzhong" (CV 17), "Zhongwan" (CV 12), "Qihai" (CV 16), "Xuehai" (SP 10) and "Zusanli" (ST 36), once daily for two weeks. p 130 expression of neocortex and hippocampus was assayed by western blotting technique. Results In SAMR 1 control group, p 130 expression in the neocortex and hippocampus exhibited an age-related stable upregulation from month 2 to 12, being significantly from the 6th month (neocortex) on and 8th month (hippocampus) on respectively (P<0.05,0.01). In SAMP 10 group, hippocampal and neo-cortical p 130 expression exhibited an age-related downregulation change in the same period, fluctuating in about 6th month (downregulated remarkably in hippocampus, P<0.01; upregulated markedly in neocortex, P<0.01). In comparison with SAMR 1 (8 months old) group, p 130 expression in hippocampus in SAMP 10 model group was upregulated significantly (P<0.05); while that in neocortex downregulated considerably (P<0.05). The expression of p 130 in neocortex in acupoint-acupuncture group was significantly higher than that in SAMP 10 model and non-acupoint acupuncture groups (P<0.05). No significant differences were found between acupoint-and non-acupoint acupuncture groups in hippocampal p 130 expression, and between non-acupoint acupuncture and SAMP 10 model groups in neocortical p 130 expression (P>0.05). Conclusion Along with the increase of aging, p 130 expression in both hippocampus and neocortex increases gradually in SAMR 1, while decreases in SAMP 10, in spite of fluctuating at month 6 in age. Acupuncture can beneficially regulate the aberrant expression of p 130 in neocortex of 8-month-old SAMP 10.

      摘要:Objective To observe the effect of preventive acupuncture and moxibustion of "Guanyuan" (CV 4) on uterus in ovariectomized (OVX) rats. Methods A total of 80 female SD rats were randomly divided into 5 groups: normal control, sham-operation (sham), model, preventive acupuncture (PA) and preventive moxibustion (PM) groups, with 16 cases in each. PA (with the needle retained for 20 min after insertion) and PM (one moxa-cone/rat) were applied to "Guanyuan" (CV 4) separately before ovariectomy, two times a week, 4 weeks altogether. Then, ovariectomy was performed on rats of model, PA and PM groups. Uterus tissue was taken under anesthesia for homogenate (10 rats/group) or sectioning (5-6 μm, 6 rats/group). Uterus estradiol (E2), progestone (P) were detected with radioimmunoassay; superoxide dismutase (SOD), nitric oxide synthase (NOS), and malondialdehyde (MDA) were assayed with immunoturdidimetry. Partial uterus slices were stained with H&E method for observing morphological changes, or stained with immunohistochemical method or with in situ hybridization method for displaying estrogen receptor (ER)-α and ER-α mRNA expression separately. Results After ovariectomy, the rat’s uterus presented obvious squamous metaplasia, hyperplasy, and thickening of the endomembrane, decrease in glands and blood vessels and increase in fibrous connective tissue, etc; while the situation was evidently better in PA and PM groups. In comparison with normal control group, E2, P contents and SOD, NOS activities of uterus tissue in model group decreased significantly (P<0.01), while MDA in model group increased evidently (P<0.01). Compared with model group, uterus P content in PA and PM groups increased obviously (P<0.05), while uterus MDA decreased apparently (P<0.01). Compared with normal control group, the expression of ER-α and ER-α mRNA in model group was downregulated markedly (P<0.01); after PA and PM, both ER-α and ER-α mRNA expression increased obviously (P<0.01). No significant differences were found between PA and PM groups in the above mentioned 7 indexes (P>0.05). Conclusion Preventive acupuncture and moxibustion of CV4 can postpone the structural degeneration of uterus in OVX rats, which may be related to their effects in modulating the secretion of uterus E2 and P, upregulating the expression of ER-α and ER-α mRNA, and improving the anti-oxidative ability.

      摘要:Objective To observe the effect of acupuncture(Acu) on bone metabolism and serum estradiol (E2) in ovariectomized (OVX) rats for studying its underlying mechanism in treating osteoporosis. Methods Forty female SD rats of six months were randomized into sham operation (sham), model, Acu and Diethylstibestrol (DES) groups, with 10 cases in each. Postmenopausal osteoporosis model was established by removing ovaries under anesthesia. In Acu group, bilateral "Dazhu" (BL 11), "Shenshu" (BL 23) and "Pishu" (BL 20) were punctured and stimulated for 30 minutes, once daily for 60 days. Rats of DES group were drenched with saline+DES (22.5 μg/ml) 1 ml/100 g, once daily for 60 days. At the end of experiments, blood samples were collected from femoral artery for assaying serum alkaline phosphatase (ALP), tartrate-resistant acid phosphatase (TRAP) contents by using biochemistry, and serum bone gla protein (BGP) and E2 levels by immunoradioassay. Results Compared with sham group, uterus wet weight, serum E2 content in model group decreased significantly (P<0.01); while body weight, serum ALP, BGP and TRAP levels in model group increased significantly (P<0.01, 0.05). Compared with model group, uterus wet weight and serum E2 content in Acu and DES groups increased significantly (P<0.01); while body weight, serum ALP, BGP and TRAP levels decreased considerably (P<0.01). No significant differences were found between Acu and DES groups in serum E2, ALP, BGP and TRAP levels (P>0.05). Conclusion Acupuncture can suppress abnormal increase of body weight and decrease of serum E2 level, and significantly downregulate serum ALP, BGP and TRAP levels in OVX rats, which may contribute to its effect in relieving osteoporosis.

      摘要:Objective To explore the underlying mechanism of electroacupuncture (EA) in relieving morphine withdrawal syndrome in rats. Methods Forty SD rats were randomly divided into normal control group, model group 1, model group 2 and EA group. Morphine withdrawal syndrome model was established by muscular injection of morphine (5 mg/kg on the 1st day, progressively increasing everyday till 100 mg/kg on the 20th day) in the hind limbs. Then, rats of model group 1 were anesthetized (10% chloraldurat) to be killed on the 21st day, and those of model group 2 killed on the 27th day. EA (2/100 Hz, 2-4 mA) was applied to bilateral "Zusanli" (ST 36) for 30 min, once a day for 7 days. The rat’s thymus was removed (after anesthesia), cut into sections (4 μm) and stained with immunohistochemical method for displaying the expression of apoptotic promoters Bax, Fas, Fas Ligand (FasL) as well as anti-apoptotic peptide Bcl-2. Results Compared with normal group, Bcl-2 immunoreaction (IR)-positive cell number of model group 1 and group 2 decreased significantly while Bax, Fas and FasL IR-positive cell number and Bax/Bcl-2 in two model groups increased considerably (P<0.01). In comparison with model group 2, Bcl-2 IR-positive cell number of model group 1 decreased significantly (P<0.05), Bax, Fas and FasL IR-positive cell number and Bax/Bcl-2 of model group 1 were significantly higher (P<0.01, 0.05). After EA, in comparison with model group 2, Fas and FasL IR-positive cell number and Bax/Bcl-2 decreased significantly (P< 0.01, 0.05), and Bcl-2 IR-positive cell number increased markedly in EA group (P<0.05); in comparison with model group 1, Bcl-2 IR-positive cell number increased significantly (P<0.01), while Bax, Fas and FasL IR-positive cell number and Bax/Bcl-2 decreased evidently in EA group (P<0.01). Conclusion EA at "Zusanli" (ST 36) can inhibit morphine-induced downregulation of Bcl-2 and upregulation of Fas and FasL expression, which may contribute to its effect in resisting thymus apoptosis in morphine withdrawal rats.

      摘要:Objective To study the effect of electroacupuncture (EA) of "Zusanli" (ST 36), "Hegu" (LI 4) and/or "Sanyinjiao" (SP 6) on immune functions in gastric carcinoma rats after operation. Methods Wistar rats were randomly divided into normal control, ST36, LI4, SP6, ST36+LI4, ST36+SP6, LI4+SP6, ST36+LI4+SP6, non-acupoint (about 10 mm lateral to ST36) and model groups, with 6 cases in each. Gastric carcinoma model was made by intraperitoneal injection of Walker-256 cloned strain (0.1 ml, 2×107 cells). EA (2-100 Hz, 1-3 mA) was applied to these acupoints for 30 min, once daily for 7 days. Serum IgG, IgM, IgA, C3 and C4 contents were detected with simple immunodiffusion method; and CD4+ and CD8+ levels were measured by flow cytometry. Results In comparison with normal control group, serum IgG, IgM, IgA , C3, C4 and CD8+ contents, and CD4+/CD8+ in model group decreased significantly (P<0.05, 0.01); while CD8+ content in model group increased remarkably (P<0.01). Compared with model group, serum IgG, IgM, IgA , C3, C4, CD4+, CD8+ and CD4+/CD8+ in non-acupoint group had no significant changes (P>0.05), while most of these indexes in EA groups (ST36, LI4, SP6, ST36+LI4, ST36+SP6, LI4+SP6, ST36+LI4+SP6) increased considerably (P<0.05, 0.01) except CD8+ level (decreased significantly, P<0.05, 0.01). No significant differences were found among 7 EA groups (P>0.05), but the effects of ST36+LI4+SP6 group were slightly better than those of the other 6 EA groups. Conclusion EA of "Zusanli" (ST 36), "Hegu" (LI 4) or "Sanyinjiao" (SP 6) or two of them or these 3 acupoints can obviously enhance the immune function of post-surgery rats with gastric carcinoma.

      摘要:Objective To observe the effect of electroacupuncture (EA) on pain threshold and contents of excitatory amino acids (EAA) in dorsal root ganglia (DRG) and spinal cord in rats with neuropathic pain. Methods Fifty SD rats were randomly divided into control (C),model (M), sham-model (SM), EA, and sham-EA groups, with 10 cases in each. Neuropathic pain (spared nerve injury, SNI) model was established by cutting off the right common peroneal nerve and proso-tibial nerve (with the sural nerve reserved intact). Before and after surgery, the mechanical pain threshold (MPT) and thermal pain threshold (TPT) were measured respectively on the injured side under consciousness state. EA (2 Hz, 1-3 mA, adding 1 mA/10 min) was applied to "Huantiao" (GB 30) and "Weizhong" (BL 40) on the affected side for 30 min. For rats of sham-EA group, filiform needles were inserted into GB30 and BL40 simply without manipulation or electrical stimulation. The treatment was given once daily for 7 days. On the 15th day, the rats were sacrificed for sampling right L4-L6 DRG and spinal cord. The contents of neurotransmitters, glutamate (Glu) and aspartic acid (Asp) in DRG and spinal cord were detected with high performance liquid chromatography (HPLC). Micro-dialysis technique was used to collect the dialysate from the spinal cord, homogenated for measuring EAA by HPLC. Results In comparison with control group, after SNI, MPT decreased significantly from the 1st day on in model group. Compared with model group, on the 15th day, MPT increased significantly in both EA and sham-EA groups (P<0.05, 0.01),but MPT of sham-EA was significantly lower than that of EA group (P<0.05). No significant changes were found in TPT in 5 groups (P>0.05). It suggested that sham EA still had an analgesic effect in spite of being lower than that of true EA. Compared with control group, the contents of Glu and Asp in the spinal cord tissue and micro-dialysate in model group increased significantly after SNI (P<0.01). In comparison with model group, the contents of Glu in DRG and spinal cord tissue and micro-dialysate, and Asp in the spinal cord tissue and micro-dialysate in both sham-EA and EA groups decreased considerably (P<0.05, 0.01). Dialysate Glu and spinal cord tissue Asp contents in EA were remarkably lower than those in sham-EA group (P<0.01, 0.05). Conclusion EA has a significant analgesic effect in SNI rats, which is closely related with its effect in inhibiting the release of Glu and Asp from dorsal horns of the spinal cord and lowering contents of EAAs.

      摘要:Objective To explore the anti-aging effect of moxibustion of "Zusnali" (ST 36) and "Xuanzhong" (GB 39) in D-galactose induced aging mice. Methods Thirty-three female Kunming mice (3 months old) were randomly divided into model group (n=11),saline control group (n=10), and moxibustion group (n=11). In addition, another 10 female mice (16 months old) were used as the physio-senility control (PSC) group. Senescent mouse model was established by subcutaneous injection of D-galactose (120 mg/kg·d-1) for 42 days. From the 13th day on after the first injection, moxibustion (3 cones/acupoint) was applied to bilateral ST36 and GB39, once every other day for one month. At the end of the treatment, the mice were killed under anesthesia for collecting cerebral cortex and cerebellar cortex separately. NO content and NOS activity were assayed by using nitrate reductase method and chemico-chromatometry respectively after homogenate and centrifugalization. Results Compared with saline control group, cerebral NO contents and NOS activity in physio-senility group and model group, cerebellar NO content in model group, cerebellar NOS activity in physio-senility and model groups increased considerably (P<0.05, <0.01). In comparison with model group, cerebral and cerebellar NO contents and NOS activity of moxibustion group lowered significantly (P<0.05,0.01). Conclusion Moxibustion of ST36 and GB39 can significantly suppress aging induced increase of both NO content and NOS activity in cerebral and cerebellar cortical tissues in aged mice, suggesting an effective anti-aging effect of moxibustion.

      摘要:Objective To observe the effect of electro-warmed needle (EWN, of Inner-Mongolian medicine) on fatigue rats’ behavior, hypothalamic-pituitary-adrenocortex (HPA) axis activity and immune system so as to reveal its neuro-endocrino-immune mechanism. Methods Male SD rats were randomized into control (n=20), model (n=20) and EWN (n=19) groups. Fatigue model was established by forcing the rat to swim in a water pool till exhaustion, once daily, continuously for 21 days. "Dinghui" (central spot over the bregmatic bone) and "Xinxue" (the center of the depression beneath the 7th thoracic vertebra) were punctured with silver needles which were warmed electrically by using a MLY-Ⅰ Electrical Needle-warming Apparatus, once every 3 days, 7 sessions altogether. On the 21st day of modeling, swim-exhaustion duration (SED), and immobility time and struggle times in tail suspension test were measured. Twenty-four hours after the last swim, the rats’ serum TNF-α, ACTH and corticosterone contents were detected by enzyme linked immunosorbent assay (ELISA). Results Compared with control group, SED, immobility time and struggle times in tail suspension test in model group decreased, increased and lowered respectively and significantly (P<0.01, 0.05); while in comparison with model group, the first 2 indexes of EWN group increased and lowered respectively and significantly (P<0.05). No significant difference was found between EWN and model groups in struggle times (P>0.05). Compared with control group, serum TNF-α, ACTH and corticosterone contents in model group increased significantly (P<0.01), while in comparison with model group, the 3 indexes of EWN group were significantly lower (P<0.01, 0.05). Conclusion EWN treatment can reduce fatigue-induced increase of serum TNF-α, ACTH and corticosterone levels, and raise motor ability, suggesting a favorable regulation of HPA axis and immune function after EWN and improvement of fatigue in fatigue rats.

      摘要:Objective To evaluate the effect of electroacupuncture (EA) of Sanyinjiao (SP 6) on perimenopausal syndrome (PMS). Methods Multicentral randomized controlled trials and single blind test were adopted in the present study. A total of 157 PMS patients were randomly divided into EA group (n=81) and medication group (n=76) . EA (2/100 Hz, 8-10 mA) was applied to bilateral Sanyinjiao (SP 6), 3 times a week and for 3 months (mon). Patients of medication group were treated by oral administration of nylestriol, 2 mg/time, twice a mon for 3 mon. For PMS patients, medroxyprogesterone (6 mg/d, for 10 days) was added from the third mon on after the treatment. The therapeutic effect was evaluated by using "symptoms-signs score scale", and changes of serum estradiol (E2), follicle stimulating hormone (FSH) and luteotrophic hormone (LH) were detected by chemiluminescence immune assay. Kupperman index was determined before and after the treatment. Results In comparison with pre-treatment, Kupperman index of EA group decreased significantly (P<0.01). After the treatment, contents of serum FSH and LH of EA group decreased significantly (P<0.01), while serum E2 contents of EA and medication groups increased significantly (P<0.01). Serum LH and E2 levels of EA group were significantly lower than those of medication group (P<0.05). No significant differences were found between two groups in Kupperman index, markedly-effective rates and total effective rates (P>0.05). Conclusion EA of Sanyinjiao (SP 6) is able to regulate serum E2, FSH and LH levels and effectively improve per-imenopausal syndrome.

      摘要:Objective To observe the influence of acupuncture of Shenmen (HT 7) and Yanglao (SI 6) on neuro-images in different cerebral regions in voleenteer subjects. Methods Twelve healthy young voleenteer participants were enrolled in this research. fMRI scans were taken in random order in a block design (one for baseline and two for acupuncture stimulations). During stimulation phase, an acupuncture needle inserted in right HT7 or SI6 was twirled at an angle of 180° and a frequency of about 1.5 Hz for 35.6 ms. Main parameters for fMRI were TR/TE/FA=3 560 ms/50 ms/90°. Acupuncture sensations were evaluated by means of visual analog scale (VAS). The collected images were analyzed by using SPM 2 for exploring inter-group differences in blood oxygen level dependent (BOLD) responses. Results When manipulating the needle, both the operator and the subject had a feeling of Deqi. Increases in BOLD signal (group analysis, corrected, 0.05, K≥10) were found mainly in right postcentral gyrus of frontal lobe (BA 2, BA 1, BA 43), left inferior frontal gyrus (BA 47), secondarily, in the right inferior parietal lobule (BA 40), right inferior frontal gyrus (BA 44), left superior temporal gyrus (BA 22) and right insula (BA 40) after acupuncture at HT7; and chiefly in left inferior parietal lobule (BA 40), right inferior frontal gyrus (BA 45, BA 46), secondarily in the left middle temporal gyrus and inferior temporal gyrus (BA 37) as well as the left superior frontal gyrus (BA 10) after acupuncture of SI6.       摘要:Verses on Acu-moxibustion Treatment have played an important role in the pervasion and spread of indications of acupoints. Abundant verses are widely cited in different versions of the textbook Acupuncturology for university and college students in China. Some contents about clinical indications of acupoints are even listed in partial verses. However, one important issue should be noted that the source of some indications of acupoints summarized in these verses needs being investigated, and the indications of some acupoints have been seldom mentioned in literature. For example, Sizhukong (TE 23) is used to treat toothache, Diwuhui (GB 42) used to treat tinnitus, Shangqiu (SP 5) used to treat jaundice, Chongmen (SP 12) selected to treat metrorrhagia, metrostaxis and abnormal vaginal discharge, Sanjiaoshu (BL 22) chosen to treat dysentery, etc. These descriptions (indications of these acupoints) need being verified in clinical practice, and thus should be used cautiously. Moreover, the verse is a type of barriers of literature in which some errors may occur in the course of spread from generation to generation and are also inherited in verses. For instance, Fubai (GB 10) is employed to treat scrofula, Yanggang (BL 48) employed to treat jaundice, Yishe (BL 49) used to treat vomiting and difficulty in swallowing, Shiguan (KI 18) taken to treat sterility, Yamen (GV 15) taken to treat epilepsy, etc. Therefore, the editors of teaching materials for acu-moxibustion should take a very cautious attitude when using such kinds of contents in verses.

    • Identification of Meridian-acupoint Diagrams and Meridian Diagrams

      摘要:In acu-moxibustion literature, there are two kinds of diagrams, meridian-acupoint diagrams and meridian diagrams. Because they are very similar in outline, and people now have seldom seen the typical ancient meridian diagrams, meridian-acupoint diagrams have been being incorrectly considered to be the meridian diagrams for a long time. It results in confusion in acu-moxibustion academia. The present paper stresses its importance in academic research and introduces some methods for identifying them correctly. The key points for identification of meridian-acupoint diagrams and meridian diagrams are: the legend of diagrams and the drawing style of the ancient charts. In addition, the author makes a detailed explanation about some acu-moxibustion charts which are easily confused. In order to distinguish meridian-acupoint diagrams and meridian diagrams correctly, he or she should understand the diagrams’ intrinsic information as much as possible and make a comprehensive analysis about them.

      摘要:The acupoint is one of the basic elements of the acupuncture prescription. Just as thinking over different properties of every Chinese materia medica in a prescription, we should also sufficiently consider the characteristics of each acupoint in a prescription of acupuncture or moxibustion. Chinese materia medica have their own action-tendencies of lifting, lowering, floating and sinking, properties of cold, hot, warm and cool, five flavors of sour, bitter, sweet, acrid and salty, target-meridian distribution, different toxicity and actions, etc. What are the properties of acupoints? The authors of the present paper summarize them by simulating the characters of Chinese materia medica. It is found that acupoints also have functions in regulating lifting, lowering, floating and sinking of Qi, inducing cold and heat effects, producing different needling sensations; and pertain to different meridians (meridian distribution). And acupoint stimulation may produce adverse effects under certain specific conditions, etc. Full understanding of the properties of acupoints is definitely helpful in improving clinical therapeutic effects and shortening therapeutic courses by reasonably selecting and combining related acupoints.

      摘要:PET-CT system is an innovative hybrid system combining Positron Emission Tomography (PET) with the fastest Computed Tomograph (CT) in one single instrument, which realizes a better integration of structure and function. This system represents the future of development of nuclear medicine imaging. When applied to revealing the underlying central mechanism of acupuncture effect, PET-CT is very helpful not only in revealing its substantial basis as neurotransmitters, etc. but also in finding the intrinsic regularities between efficacies and changes of images. It provides a new stool for scientific research. In the present paper, the authors make an introduction about its characteristics in techniques, advantages, points for paying attention and possible progresses achieved in the studies about central mechanisms of acupuncture.

      摘要:Acute ischemic cerebrovascular disease is one of the critical diseases seriously endangering human health. Acupuncture therapy, an effective treatment method for many types of disorders has been generally acknowledged. In recent years, many scientific researchers have studied the relationship between the effects of acupuncture in relieving cerebral ischemia-induced sequelae and the stimulating parameters. The acupuncture stimulating parameter includes the frequency of electroacupuncture (EA), the frequency of acupuncture treatment, and the acquired quantity of stimulation, etc for clinical patients and experimental animals. It was found that different stimulating parameters may have different efficacies. Current research results provide a good basis not only for analysis of the factors of acupuncture-produced effects, but also for determination of the optimal combination of stimulating parameters. However, acupuncture therapeutic effect involves multiple factors and multiple levels, and current quantitative acupuncture parameter researches have been mainly restricted to animal experiments. Hence, more researches in which statistics specialists take part are definitely needed.
