摘要:Objective To observe the influence of acupuncture on neuroendocrine in rats with primary dysmenorrhea,so as to explore its underlying mechanism in relieving dysmenorrhea.Methods Forty female Wistar rats were equal randomized into control,model,acupuncture and Extractum Leonuri Inspissatum(ELI) groups,with 10 cases in each.Primary dysmenorrhea model was established by intragastric perfusion of diethylstilbestrol(3 mg/kg),once daily for 12 days."Guanyuan"(CV 4),bilateral "Sanyinjiao"(SP 6) and "Diji"(SP 8) were punctured once daily for 7 days.ELI(80 g/kg) was given to rats of ELI group via gastrogavage,once daily for 7 days.On the 12th day,the rats of 4 groups were given with oxytocin(14 U/kg,i.p.) for inducing dysmenorrhea.The latency and incidence rate of writhing within 30 min were observed.The serum levels of follicle stimulating hormone(FSH),luteinizing hormone(LH),estradiol(E2),and progesterone(P) were detected by radioimmunoassay,and the expression of gonadotropin-releasing hormone(GnRH) mRNA in hypothalamus and ovary,GnRH receptor(GnRH-R) mRNA in pituitary,estrodiol receptor(ER) mRNA and progesterone receptor(PR) mRNA in uterus were determined using RT-PCR.Results After modeling,the writhing latency shortened considerably and the writhing times,serum FSH,LH and E2 levels increased apparently,and serum P content decreased markedly(P<0.05).Compared with model group,the writhing times,serum FSH,LH,and E2 contents in acupuncture and ELI groups reduced significantly(P<0.05),while the writhing latency and serum P contents in acupuncture and ELI groups increased remarkably(P<0.05).Compared with control group,hypothalamic GnRH mRNA,ovary GnRH mRNA,pituitary GnRH-R mRNA and uterus ER mRNA expressions in model group were up-regulated significantly,while uterus PR mRNA was down-regulated markedly in model group(P<0.05).In comparison with model group,hypothalamic GnRH mRNA,ovary GnRH mRNA,pituitary GnRH-R mRNA and uterus ER mRNA expressions in acupuncture and ELI groups were suppressed significantly(P<0.05),while uterus PR mRNA expressions in the later two groups were up-regulated obviously(P<0.05).Conclusion Acupuncture of CV4,SP6 and SP8 can effectively relieve dysmenorrhea in the rat,which may be closely related to its effects in regulating neuroendocrine activities and the related receptor expression of hypothalamus-pituitary-ovary axis.

摘要:Objective To study the efficacy of combined application of deproteinised calf serum injection(DCSI) and acupuncture in treating cerebral ischemia(CI) in rats.Methods Ninety Wistar rats were randomly divided into control,CI model,Acupunct(acupuncture),DCSI,Acupunct+DCSI groups,with 18 cases in each group.CI model was duplicated by middle cerebral artery occlusion(MCAO).DCSI(80 mg/kg) was given via the rat's tail vein,once daily for 7 days.Acupuncture was applied to "Baihui"(GV 20),"Shuigou"(GV 26),"Neiguan"(PC 6),"Hegu"(LI 4),"Taichong"(LR 3) and "Zusanli"(ST 36),once daily for 7 days.The neurological deficit score(NDS),cerebral infarct volume(CIV),water and lactic acid(LA) contents in cerebral tissues were measured.Results In comparison with control group,NDS,CIV,cerebral water content and LA in model group increased significantly(P<0.05);while compared with model group,NDS,CIV,cerebral water content and LA in Acupunct,DCSI and Acupunct+DCSI groups decreased significantly(P<0.05).Comparison among DCSI group,Acupunct group and DCSI+Acupunct group showed that NDS,CIV,cerebral water and LA contents of Acupunct+DCSI group were significantly lower than those of the former two groups(P<0.05).No significant differences were found between DCSI and Acupunct groups in the above-mentioned 4 indexes(P>0.05).Conclusion Combined application of DCSI and acupuncture has a definite effect in improving cerebral ischemia and is superior to that of simple acupuncture or DCSI alone.

摘要:Objective To observe the effect of electroacupuncture(EA) on the contents of hippocampal calcitonin gene-related peptide(CGRP) and neuropeptide Y(NPY) in cerebral ischemia(CI) rats.Methods Forty SD rats were randomly divided into normal control,sham operation(sham),model and EA groups with 10 cases in each.CI model was duplicated by middle cerebral artery occlusion.EA(2 Hz,1 mA,continuous waves) was applied to "Shuigou"(GV 26) for 30 min,once daily for 5 days.Hippocampal tissue was taken for assaying the contents of CGRP and NPY with radioimmunoassay.Results Compared with sham group and normal control group,hippocampal CGRP content in model group decreased apparently(P<0.01),while NPY content of model group increased significantly(P<0.01).Compared with model group,hippocampal CGRP in EA group increased considerably(P<0.01) and NPY content decreased obviously(P<0.01).Conclusion Electroacupuncture of GV26 can effectively adjust hippocampal CGRP and NPY contents in CI rats,which may contribute to its effect in improving cerebral ischemia.
摘要:Objective To investigate the influence of electroacupuncture(EA) on 5-serotonin(5-HT) and acetyl cholinesterase(AChE) activities in hippocampus tissue in chronic stressed-induced depression rats.Methods Thirty-two Wistar rats were randomized into normal control group(n=10),model group(n=11) and EA group(n=11).Chronic depression model was established by lonely raising,and chronic unpredictable mild stress for 21 days.EA(2 Hz,1 mA) was applied to "Baihui"(GV 20) and "Sanyinjiao"(SP 6) for 20 minutes,once daily for 14 days.5-HT and AChE expression of hippocampal CA 4 region was detected by using immunohistochemistry,and their integrated optical density(IOD)values were determined by using image analysis system.Results Compared with normal control group,IOD values of 5-HT and AChE immuno-reaction(IR) positive neurons in hippocampal CA 4 region of model group decreased significantly(P<0.05).In comparison with model group,the IOD values of 5-HT and AChE IR positive neurons of CA 4 region in EA group increased significantly(P<0.05).No significant differences were found between EA and normal control groups in IOD values of both 5-HT and AChE IR positive neurons.Conclusion EA can upregulate 5-HT and AChE expression in hippocampal CA 4 region in depression rats,which may contribute to its effect in relieving depression.

摘要:Objective To observe the effect of electroacupuncture(EA) of different acupoint groups on mean arterial pressure(MAP) and heart rate variability(HRV)in hypotension rats.Methods A total of 70 Wistar female rats were randomized into model,"Neiguan"(PC 6)-"Daling"(PC 7),"Waiguan"(SJ 4)-"Yangchi"(SJ 5),"Tianshu"(ST 25)-"Wailing"(ST 26),"Dachangshu"(BL 25)-"Qihaishu"(BL 24),"Baihui"(GV 20)-"Qianshencong"(EX-HN),and"Guangming"(GB 37)-"Xuanzhong"(GB 39)groups,with 10 cases in each.Hypotension model was established by bleeding(2.5-3 ml) through carotid artery.Immediately after bleeding,EA(2/15 Hz,1 mA,30 min) was applied to the above-mentioned acupoints of the 6 EA groups.MAP and ECG were recorded,then HRV was analyzed by using software Chart 5.2.1.Results MAP declined significantly after bleeding.Compared with model group,MAP values of PC6-PC7 and GB37-GB39 groups increased significantly 15 min and 30 min after bleeding,and those of ST25-ST26 and GV20-EX-HN groups also increased apparently 30 min after bleeding(P<0.05).No marked effects were observed in EA of SJ4-SJ5 and BL25-BL24 groups(P>0.05).The effect of PC6-PC7 and GB37-GB39 groups was significantly superior to that of SJ4-SJ5,BL25-BL24 and GV20-EX-HN groups in elevating MAP 30 min after bleeding(P<0.05).Compared with model group,heart rate(HR) of PC6-PC7 group was increased significantly 15 min and 30 min after bleeding(P<0.05.).After bleeding,low frequency(LF),LF/high frequency(HF) and very low frequency(VLF) increased apparently.Compared with model group,LF,LF/HF and VLF of PC6-PC7 and GB37-GB39 groups decreased significantly 30 min after bleeding(P<0.05),suggesting a better balancing effect of EA of PC6-PC7 and GB37-GB39 on the activities of sympathetic and vagus nerves.Conclusion EA of different acupoint groups has its own relative specificity in regulating MAP and HR in the rat,and PC6-PC7 EA and GB37-GB39 EA have a better efficacy,which is closely associated with their effects in balancing activities of the autonomic nerve system.

摘要:Objective To compare the effects of electroacupuncture(EA) stimulation of "Gastric Problem Formula" ["Neiguan"(PC 6),"Zhongwan"(CV 12) and "Zusanli"(ST 36)] on electrical activities of neurons of medullary subnucleus reticularis dorsalis(SRD) of the rat.Methods Experiments were performed on SD rats anesthetized by 10% urethane(1.0-1.5 g/kg).Under artificial ventilation and immobilization(2% gallamine triethiodide,5 ml/kg),extracelluar discharges were recorded by glass micropipettes inserted into SRD of the medulla(Caudal to the Obex:-1.0——2.0 mm,L/R: 0.5-l.5 mm,H: 0.6-1.5 mm below the medullary surface).Following getting stable recordings,the acupuncture needle inserted in "Neiguan"(PC 6) was manipulated moderately(twisting range: 120°,frequency: 80 times/min) for 30 s.The same manipulation was performed at "Zhongwan"(CV 12) and "Zusanli"(ST 36) separately.Results After moderate acupuncture stimulation,in the recorded 13 SRD neurons,the discharges were increased from(1.67±0.35) spikes/s to(17.55±1.40) spikes/s for "Neiguan"(PC 6),from(2.35±0.43) spikes/s to(16.96±1.83) spikes/s for "Zhongwan"(CV 12),and from(1.83±0.37) spikes/s to(17.39±1.58) spikes/s for "Zusanli"(ST 36).No apparent differences were found among 3 acupoints in their responses to manual manipulation.Conclusion Moderate acupuncture stimulation of "Neiguan"(PC 6),"Zhongwan"(CV 12) and "Zusanli"(ST 36) can effectively activate spontaneous discharges of SRD neurons in the rat.
摘要:Objective To observe the effects of acupoint-nerve block on manual-acupuncture(MA) and electroacupuncture(EA) analgesia and mast cells degranulation in adjuvant arthritis(AA) rats in order to analyze the underlying peripheral mechanism of MA and EA analgesia.Methods A total of 80 SD rats were randomized into control,AA model(M),normal+lidocaine(NL),EA,EA-ST36+Lidoc(L+EA),Dubi(ST 35)+Lidoc+EA-ST36(DL+ZEA),Xiajuxu(ST 39)+Lidoc+EA-ST36(XL+ZEA),MA,Lidoc+MA-ST36(L+MA),and ST35+Lidoc+MA-ST36(DL+ZMA) groups,with 8 cases in each.AA model was established by injection of complete Freund's adjuvant(0.05 ml) in the rat's right hind-paw under anesthesia.Lidoc hydrochloride(2%,0.05 ml) was injected into ST36 or ST35 separately for blocking the local nerve.Paw withdrawal latency(pain threshold,PT) and mast cell degranulation ratios(DGR) were detected.EA(2/100 Hz,0.5 mA,1 mA,1.5 mA) or MA was applied to ST36.Results Compared with model group,PT values of EA and MA groups increased significantly(P<0.05,P<0.01) and mast cell DGR of EA and MA groups were significantly higher(P<0.01).Both PT and DGR of NL group were basically not affected by ST36-Lidoc-injection in comparison with model group(P>0.05).PT values of L+EA and DL+ZEA groups were significantly lower than those of control and EA groups(P<0.01),and comparable to those of model and NL groups(P>0.05).PT of XL+ZEA group was significantly higher than those of model and NL groups(P<0.01),and comparable to that of EA group(P>0.05).Similar situations were found in MA-related groups.Mast cell DGR of ST36 area in L+EA,DL+ZEA and XL+ZEA groups were comparable to that of EA group(P>0.05),and such was the case in MA-related groups.Conclusion After nerve block of the local and proximal acupoints rather than the distal acupoint innervated by the same nerve,the analgesic effect of both MA and EA of ST36 is obviously attenuated;but no marked influences on MA-and EA-induced mast cell degranulation are found.
摘要:Objective To study the innervating character of tissues around the acupoints and along the meridian course and to analyze the reflex activity correlation between acupuncture points in a given meridian in the rat.Methods Forty Wistar and 15 SD rats were used in this study.Electrical activities of microfilaments of the afferent nerves(deep tibial nerve,peroneal nerve and the lateral cutaneous nerve of the leg) were observed for identifying their receptive field and the type of nerve fibers.Nerve stem-antidromic stimulation induced Evan's blue extravasation method was employed to compare the difference of the nerve ending distribution in the acupoint area and non-acupoint area.The reflex activities evoked by electric stimulation of the acupoint were used to analyze the interrelation between acupoint and meridian.Results Findings showed that a great majority of the afferent nerve endings supplying the tibialis anterior/rectus femoris muscle and the foot skin distributed in an uneven pattern in the sites being in accord with acupoints or with the orbit of meridians.Antidromic stimulation of C-fibers in the deep tibial nerve evoked extravasation of Evan's blue from the plasma into the interstitial fluid,blue foci appeared at the acupoints of the Stomach Meridian and along the orbit of the Stomach Meridian.The special distribution of the afferent nerve endings in the acupoint was also associated with the special reflex activity originating from the acupoints of the muscle group of a given meridian.ConclusionThe acupoint is an excitable muscle/skin-nerve complex with greatest concentration of nerve endings.The meridian consists of acupoints that possess a close interaction in physiological reflex activities.
摘要:Objective To research whether opiate receptors in the hypothalamic arcuate nuclei(ARC) participate in the inhibitory effect of electroacupuncture(EA) of "Neiguan"(PC 6) on ventricular arrhythmia in the rabbit.Methods This research consists of three parts.1) Effect of electrolytic lesion of hypothalamic ARC on EA-induced suppression of heart ventricular extrasystole(HVE): HVE model was established by electric stimulation(0.4-0.8 mA,wave-duration 0.5 ms,50 Hz,5-10 s every 5 min) of hypothalamic midline region(R/L: 0.5-1.5 mm;H:-5——4.5 mm) in 4 rabbits.Electrolytic lesion(5 mA,20 s) was induced by delivering a direct current to ARC region(A 1,L/R: 0.5 mm,H:-7 mm) through a stainless steel electrode(anode).EA(0.25 mA,5 Hz,pulse-duration 0.5 ms) was applied to "Neiguan"(PC 6) for 3 min,4 times altogether.(2) Effect of microinjection of morphine in ARC area on HVE: After inducing HVE,morphine(2 μg/μl) was injected into hypothalamic ARC region,and changes of HVE number were observed in 12 rabbits(6 for morphine group and the other 6 for normal saline group).(3) Effect of ARC microinjection of naloxone on EA-induced suppression of HVE: After inducing stable HVE,microinjection of naloxone(2 μg/μl) or same dose of normal saline was performed into ARC region in 6 rabbits respectively,followed by EA of PC6.Results Electric stimulation of hypothalamic midline region could induce continuous HVE.After EA,it decreased significantly in the occurrence rate(P<0.01).Following ARC region lesion,the effect of EA in inhibiting HVE disappeared.After microinjection of morphine into ARC region,the occurrence rate of HVE reduced apparently(P<0.01),while after microinjection of naloxone,the inhibitory effect of EA vanished.No significant changes were found in the occurrence rate and the inhibitory rate of HVE after microinjection of naloxone or normal saline in the ARC region(P>0.05).Conclusion Hypothalamic ARC and it's opium receptors participate in the inhibitory effect of EA on hypothalamic midline region electro-stimulation-induced HVE in the rabbit.

摘要:Objective To explore the effect of "Xiusanzhen" [bilateral "Yingxiang"(LI 20) + "Yintang"(EX-HN 3)] on learning-memory ability and choline acetyl transferase(ChAT) and acetyl cholinesterase(AChE) activity of hippocampus tissue in Alzheimer Disease(AD) rats.Methods A total of 40 Sprague Dawley rats were randomly divided into control,AD model,AD+olfactory-bulb lesion(OBL,induced by electrocoagulation method)+EA and "Xiusanzhen"(AD+EA) groups.AD model was established by 4-vessel occlusion.Morris water maze tests were conducted for evaluating the rat's learning-memory ability.The activity of ChAT and AChE of hippocampus tissue was detected by colorimetric method.Results In comparison with control group,the average escape latency(AEL) prolonged significantly and the target-platform crossing times decreased remarkably in AD model group(P<0.01).In comparison with AD model group,the AEL shortened and the target-platform crossing times increased remarkably in AD+EA group(P<0.01).No significant differences were found between AD model and AD+OBL+EA groups in AEL and the target-platform crossing times(P>0.05).The ChAT and AChE activity of hippocampus in AD model group was significantly lower than that of control group(P<0.05),while that of ChAT and AChE in AD+EA group was significantly higher than that in AD model group(P<0.05).No significant difference was found between AD model and AD+OBL+EA groups in ChAT and AChE activity of hippocampus(P>0.05).Conclusion EA of "Xiusanzhen" can markedly raise the learning-memory ability in AD rats,which may be related to its effect in increasing the ChAT and AChE activity of hippocampus in AD rats.Its therapeutical effect also depends on the intact of the olfactory pathway.

摘要:Objective To observe the effect of transcutaneous acupoint electrical stimulation(TAES) on serum superoxide dismutase(SOD) activity,malondialdehyde(MDA) and S100β contents in craniotomy patients for studying its cerebral protection mechanism.Methods Fifty patients scheduled for neurosurgery were randomly divided into TAES group(n=25) and control group(n=25) with randomized block method.For patients of TAES group,TAES was applied to bilateral Hegu(LI 4) and Quchi(LI 11),Zusanli(ST 36) and Sanyinjiao(SP 6) from 30 minutes on before anesthesia to the end of operation.Patients of control group were anesthetized with sevoflurane inhalation and intermittent(i.v.) of sulfenany and vecurnium bromide.Blood samples were taken for assaying serum SOD activity,MDA and S100β contents with purinase oxydasis,biochemiluminescence and enzyme linked immunosorbent assay separately.Scores of cognitive ability were given by using Mini Mental State Examination(MMSE).Results In comparison with pre-anesthesia,serum SOD activity decreased significantly 1 h after craniotomy in control group,at the end of operation in both control and TAES groups(P<0.05,P<0.01),and increased markedly 48 h after operation in control group(P<0.05).Serum MDA in control group increased significantly 48 h after operation,while that in TAES group reduced apparently 24 h after operation(P<0.01).Serum S100β content in TAES group decreased remarkably 48 h after operation(P<0.01).Serum SOD activity of TAES group was significantly higher than that of control group 24 h after operation(P<0.05).Compared with control group,serum MDA contents of 24 h and 48 h after operation and serum S100β levels at 1 h after craniotomy and 48 h after operation were markedly lower in TAES group(P<0.01,P<0.05).No significant differences were found between two groups in the cognitive function scores(P>0.05).Conclusion TAES can increase serum SOD activity and reduce MDA and S100β levels in patients undergoing craniotomy,which may contribute to its effect in reducing lipid peroxidation induced cerebral injury.But its impact on the patient's cognitive function needs study further.

摘要:Objective To observe the therapeutic effect of point injection of Danshen(Red-sage-root) for treating ischemic necrosis of femoral head in patients.Methods A total of 62 hips of ischemic femoral head necrosis were randomly divided into point-injection group(32 hips) and core decompression(CDC) group(30 hips) according to the random digits table method.Red-sage-root(freeze-dried) solved in 5 ml normal saline solution was injected into Huantiao(GB 30) and Juliao(GB 29) on the affected side,2.5 ml for each point,3 times a week,for 6 consecutive months.Patients of CDC group were treated by core decompression surgery.The therapeutic effect was evaluated by the "Evaluating Method for Adult Avascular Necrosis of Femoral Head"(issued in Dandong in 1995).Results Compared with pre-treatment,scores of pain severity of two groups from the 1st month to the 6th month,joint-motion range,daily life activity and walking distance scores of point-injection group from the 1st month to the 6th month increased significantly(P<0.01,P<0.05);while those of joint-motion range,daily life activity and walking distance in CDC group had no apparent improvement(P>0.05).The difference values between post-and pre-treatment in the scores of pain severity,joint-motion range,daily life activity and walking distance in point-injection group were all significantly higher than those in CDC group(P<0.01).Conclusion Point-injection of red-sage-root can effectively relieve the pain severity,improve the hip joint-motion function and daily life activity in ischemic femoral head necrosis patients.

摘要:Studies show that the inflammatory reaction plays an important role in cerebral ischemia/reperfusion injury.Post-ischemic inflammation is a dynamic process involving a series of complicated interactions among various inflammatory cells and molecules.Recently,researches on the effect of acupuncture in inhibiting post-ischemic inflammation had been conducted and achieved favorable results.Authors of the present paper review the recent development of studies on the underlying neuroprotective mechanism of acupuncture in inhibiting post-ischemic inflammation in the brain tissue from 1) regulation of inflammatory cytokines and mediators,2) inhibition of leucocytic infiltration,3) modulation of the activated state of glial cells,and 4) suppression of the expression of the related transcription factors.Moreover,combining the key target point,peroxisome proliferator-activated receptor-γ(PPAR-γ) mediating the process of post-ischemic inflammatory cascade response,this paper discusses the future research trend.Joint application of acupuncture and medication(including Chinese herbal medicines) for mediating the key target of post-ischemic inflammation cascade should be studied in both clinical practice and basic experiments.

摘要:In the present paper,the authors make a summary on the clinical application of acu-moxibustion therapy in the treatment of gouty arthritis in recent 10 years.Acupuncture needles often used are filiform needle,red-hot needle,moxibustion-warmed needle and three-edged needle.Clinical studies have showed that acupuncture therapy has a definite efficacy in relieving gouty arthritis and has its clinical characteristics,such as faster efficacy,lower relapse rate,etc.in comparison with medication.
