      摘要:Objective To observe the effect of electroacupuncture(EA)on the differentially expressed proteins in the spinal cord at different time courses after acute spinal cord injury(ASCI)in the rat,so as to study its underlying mechanism in improving spinal traumatic injury.Methods A total of 105 male SD rats were randomized into normal control,model-6 h,EA-6 h,model-24 h,EA-24 h,model-48 h,EA-48 h groups,with 15 cases in each.ASCI model was established by using modified Allen's method.EA(2 Hz,2-5 mA)was applied to "Mingmen"(GV 4)and "Dazhui"(GV 14)for 30 min.The injured spinal cord tissue(T10-T11)was collected 6 h,24 h and 48 h after ASCI and EA treatment,weighted and stored under-80 ℃ till detection.Two-dimensional gel electrophoresis(2-DE)was used to separate total proteins of the spinal tissue,followed by protein extraction and quantitation,2-D gel image analysis,matrix assisted laser desorption ionisation time-of-flight mass spec-trometry(MALDI-TOF-MS),and databases-searching for identification of the differentially-expressed proteins.Results A total of 10 differentially expressed proteins were identified in the present study.At 6 h,compared with control group,of the 5 types of spinal differential proteins,4 were upregulated in the expression after ASCI,and the rest one was downregulated;while after EA,ASCI-induced expression changes in 4 of the 5 differential proteins were reversed.At 24 h after ASCI,7 types of differential proteins were identified.Compared with control group,6 differential proteins were upregulated,and the rest one was downregulated in model group.Compared with model group,ASCI-induced expression changes in 6 of the 7 differential proteins were reversed by EA.At 48 h after ASCI,a total of 8 types of differential proteins were identified.Compared with control group,6 differential proteins were upregulated in the expression,and the rest two downregulated in model group.Compared with model group,ASCI-induced expression changes in 5 of the 8 differential proteins were reversed by EA.Along with the increased time and treatment,24 h vs 6 h,two more differential proteins were identified,i.e.,nucleoside diphosphate kinase and triosephosphate isomerase 1(TPI 1).48 h vs 24 h,3 more differential proteins were identified,i.e.,dihydrolipoamide dehydrogenase,malate dehydrogenase 1,and glyceraldehyde-3-phosphate dehydrogenase(GAPDH);but two proteins disappeared,i.e.,nucleoside diphosphate kinase,and ubiquitin-conjugating enzyme E2N.The identified differential proteins involving the effects of EA in regulating cellular energy metabolism,DNA repair,cellular generation,differentiation,apoptosis,etc.Conclusion Proteome analysis indicates that in ASCI rats,some differentially expressed proteins involving energy metabolism,neuronal apoptosis reduction,protein-degradation inhibition may contribute to the effect of EA in repairing the traumatic spinal tissue.

      摘要:Objective To observe the influence of preventive acupuncture(PA)and preventive moxibustion(PM)at "Guanyuan"(CV 4)on the immune function in natural climacteric rats.Methods A total of 160 female SD rats were randomized into control,PA and PM groups,the former one group was further divided into 10 month(mon),12 mon,14 mon and 16 mon subgroups,and the later two groups were further divided into 12 mon,14 mon and 16 mon subgroups,with 16 rats in each.In addition,other 16 female SD rats aged 3.5 mon were used as the young control(YC)group."Guanyuan"(CV 4)was punctured with an acupuncture needle and the needle was retained for 20 min,or given with one ignited moxa-cone from the age of 10 mon on.The treatment was conducted twice every week,8 weeks altogether.The expression of HSP 70 and HSP 70 mRNA of the spleen tissue was detected by using immunohistochemistry and in situ hybridization respectively,and serum IL-2 and TNF-α contents were assayed by using radio-immunoassay.Results In comparison with YC group,1)the expression of spleen HSP 70 and HSP 70 mRNA increased significantly in 10 mon control(mon-C),12 mon-PM and 12 mon-PA groups,and 14 mon-PA group(only HSP 70 mRNA)(P<0.05,P<0.01);2)HSP 70 expression decreased remarkably in 14 mon-C,16 mon-C and 16 mon-PA groups(P<0.05,P<0.01);3)IL-2 contents decreased evidently in 12 mon-C and 14 mon-C groups,and TNF-α contents increased obviously in 12 mon-PM,12 mon-PA and 16 mon-C groups(P<0.05).In comparison with the corresponding same age control groups,HSP 70 and HSP 70 mRNA expression increased significantly in 12 mon-PM and 12 mon-PA groups,14 mon-PM and 16 mon-PM(HSP 70 only),14 mon-PA(HSP 70 mRNA only)groups(P<0.05,P<0.01);IL-2 level of 12 mon-PM group,and TNF-α contents of 12 mon-PM and 12 mon-PA groups increased markedly(P<0.05,P<0.01).No significant differences were found between PM and PA groups in most different age groups(P>0.05).Conclusion Both preventive acupuncture and preventive moxibustion can upregulate the expression of spleen HSP 70 and HSP 70 mRNA and serum IL-2 and TNF-α levels,which may contribute to their effects in enhancing the immune function in menopausal rats.

      摘要:Objective To study the effect of electroacupuncture(EA)on liver cytochrome P4501A1 immunoactivity,superoxide dismutase(SOD)and malondialdehyde(MDA)contents in nonalcoholic fatty liver disease(NAFLD)rats.Methods Thirty male SD rats were randomized into normal control(n=10),model(n=10)and EA(n=10)groups.NAFLD model was established by feeding the animal with high-fat forage for 8 weeks.EA(1.6-2 Hz,1-4 mA)was applied to bilateral "Zusanli"(ST 36),"Fenglong"(ST 40),"Sanyinjiao"(SP 6)and "Taichong"(LR 3)for 15 min,once daily for 4 weeks.Then the rats anesthetized with ether were killed for collecting liver tissue.Following homogenate and centrifugalization of the partial liver tissue,the supernatant was collected for assaying superoxide dismutase(SOD)and malondialdehyde(MDA)contents by xanthinoxidase chromometry and thio-malonylurea chromometry respectively.The other partial liver tissue samples were fixed in 10% formalin,followed by paraffin imbedding and sectioning(4 μm),and staining with streptavidin-perosidase methods respectively for displaying hepatic pathological changes and cytochrom P4501A1 immunoreaction.Results Compared with control group,hepatic SOD content of model group was significantly lower(P<0.05),and MDA level and cytochrome P4501A1(CYP1A1)integrated optic density(IOD)value were obviously higher in model group(P<0.05).In comparison with model group,liver SOD level of EA group increased considerably(P<0.05),while MDA level and CYP1A1 IOD of EA group decreased evidently in EA group(P<0.05).Conclusion EA can effectively reduce lipid peroxidation and up-regulate CYP1A1 expression in nonalcoholic fatty liver tissue,which may contribute to its effect in improving fatty liver.

      摘要:Objective To observe the effect of electroacupuncture(EA)of "Zusanli"(ST 36)and "Sanyinjiao"(SP 6)on collagen-induced arthritis(CIA)and its underlying mechanism in regulating the secretory function of knee-joint synovial cells in CIA rats.Methods Thirty-six Wistar rats were equally randomized into control,model and EA groups.CIA model(rheumatoid arthritis)was duplicated by intradermal injection of Bovine type Ⅱ collagen into the back of the anesthetized rats.EA(2 Hz,2 mA)was applied to "Zusanli"(ST 36)and "Sanyinjiao"(SP 6)for 30 min,once a day for 30 days.The paw volume was measured by using a Plethysmometer and hot water tail-flick tests(50 ℃)were conducted for detecting the rats' pain threshold(PT)before and after the treatment.The contents of PGE2,IL-1β and TNF-α in supernatant of the cultured joint synoviocytes were detected by enzyme linked immunosorbent assay(ELISA).Results In comparison with control group,the swelled paw volume,the declined PT,PGE2,TNF-α and IL-1β contents in knee-joint synoviocyte supernatant were significantly higher in model group(P<0.05).Compared with model group,PT increased obviously(P<0.05);paw volume,supernatant PGE2,TNF-α and IL-1β contents decreased obviously(P<0.05)in EA group.Conclusion EA of ST 36 and SP 6 has a pronounced therapeutic effect in relieving knee joint pain and inflammation in CIA rats,which may be related to its effect in regulating the secretory function of the knee-joint synoviocytes.

      摘要:Objective To observe the effect of acupuncture and moxibustion on the conditional position preference(CPP)and prefrontal cortical ultrastructure in heroin re-addicted rats.Methods Thirty-two Wistar rats were randomly divided into normal,model,medication and acu-moxibustion(acu-moxi)groups,with 8 cases in each.Re-addiction model was established by repeated intramuscular injection of heroin into the hindlimbs.Rats of medication group were treated with intragastric administration of Methadone during detoxification.For rats in acu-moxi group,acupuncture needle was inserted into "Baihui"(GV 20)and moxibustion treatment was used to bilateral "Shenshu"(BL 23)for 30 min during detoxification.Rats in normal group were given with intramuscular injection of normal saline during addiction.CPP tests were conducted once daily in a spatial place preference box for 8 days.On the 39th day of the experiment,the rats under anesthesia were killed for sampling the prefrontal cortex(PFC)tissue on an ice plate,then,the ultrastructure of the neurons was observed by using a transmission electron microscope.Results After modeling,the rats' staying duration in the dark-box was shortened obviously(P<0.01).After the treatment,the staying duration in dark-box in acu-moxi group was significantly longer than that in model and medication groups(P<0.01).Compared with normal control group,electron-microscopic results indicated that in PFC tissues of model and medication groups,vacuoles and edema of cytoplasm at different degrees,enlargement of the rough endoplasmic reticulum(REPR),vacuoles or disappearance of mitochondria,reduction of ribosome in number,and widening of the nuclear intermembrance interstice were seen.While in acu-moxi group,the number of mitochondria and ribosome increased slightly,the mitochondrial cristae was clear,the number of REPR increased relatively but dilated slightly,and the intermembrance space was basically normal.Conclusion Acu-moxibustion can improve heroin re-addicted rats' conditional position preference and has a protection effect on the cerebral cortical neurons.

      摘要:Objective To observe the effect of electroacupuncture(EA)on changes of ultrastructure and proliferation of astrocytes in the marginal zone of the cerebral ischemia locus in rats at different time courses so as to explore its underlying mechanism in the treatment of cerebral ischemia(CI).Methods Ninety Wistar rats were randomized into sham operation(sham),model and electroacupuncture(EA)groups which were further divided into 1 h,1 d,3 d,7 d,and 21 d subgroups,with 6 cases in each.CI model was established by occlusion of the middle cerebral artery with heat-coagulation.EA(4/20 Hz,2-3 V,1-3 mA)was applied to "Baihui"(GV 20)and "Dazhui"(GV 14)for 30 min once daily.The ultrastructure changes of astrocytes were observed by using transmission electronic microscope,and glial fibrillary acidic protein(GFAP)expression of astrocytes was displayed by using immuno-fluorescentdyeing and observed under laser confocal scanning microscope(LCSM).Results Following CI,the astrocytes were swelling in structure and increased in number in CI model group,while compared with model group,the degree of swelling of astrocytes was decreased obviously in EA group.The mean fluorescence intensity values of GFAP on day 1,day 3,day 7 and day 21 post-modeling in model group increased significantly in comparison with sham group(P<0.01),while those of GFAP from day 1 to day 21 post-modeling in EA group were significantly lower than those in model group(P<0.01,P<0.05),suggesting an inhibitory effect of EA on CI-induced upregulation of GFAP expression.ConclusionEA of GV 20 and GV 14 can relieve CI-induced ultrastructural injury of astrocytes in the rat,which is closely with its effect in inhibiting CI-induced overexpression of GFAP in astrocytes of the ischemic brain tissue.

      摘要:Objective To observe the effect of electroacupuncture(EA)of "Xiusanzhen" [bilateral "Yingxiang"(LI 20)+"Yintang"(GV 29)] on learning-memory ability and hippocampal somatostatin(SS)and arginine vasopressin(AVP)contents in vascular dementia(VD)rats.Methods Forty SD rats were randomly divided into control,VD model,VD plus olfactory bulb lesion(OBL,destroyed by electro-coagulation)and EA groups,with 10 cases in each.VD model was established by 4-vessel occlusion.Morris maze tests were conducted for evaluating the rats' learning and memory ability.EA(1-3 mA,80/100 Hz)was applied to bilateral LI 20 and GV 29 in VD+OBL and EA groups for 10 min,once daily(except Saturdays and Sundays)for 6 weeks.The contents of SS and AVP in hippocampus tissue were measured by radioimmunoassay.Results In comparison with control group,the average escape latency(AEL)prolonged significantly and the target-platform crossing times(TPCT)decreased remarkably in VD model group(P<0.01).In comparison with VD model group,the AEL was shortened and TPCT increased remarkably in EA group(P<0.01).No significant differences were found between VD model and VD+OBL groups in the AEL and the TPCT(P>0.05).Hippocampal SS and AVP contents in VD model group were significantly lower than those in control group(P<0.05),while those in EA group were markedly higher than those in VD model group(P<0.05).No significant differences were found between VD model and VD+OBL groups in SS and AVP contents(P>0.05).Conclusion EA of "Xiusanzhen" can improve VD rats' learning-memory ability,which may be related to its effects in raising hippocampal SS and AVP contents and to the intact olfactory pathway.

      摘要:Objective To observe the effect of electroacupuncture(EA)of "Baihui"(GV 20)and "Yamen"(GV 15)on the permeability of blood-brain barrier for exogenous nerve growth factor(NGF)and its relevance to PKC pathway in cerebral ischemia(CI)rats.Methods Forty-eight SD rats were randomized into sham-operation(sham),model,NGF,EA,NGF+EA,and NGF+H7+EA groups.CI model was established by repeated occlusion of the bilateral carotid arteries and reperfusion.NGF(10 μg/kg)was administrated by intravenous injection(vena caudalis);while H 7 [1-(5-isoquinolinesulfonyl)-2-mehtylpoperazine,an inhibitor of protein kinase C(PKC),1 mg/kg] was given by intraperitoneal injection.EA(100 Hz,2 mA)was applied to "Baihui"(GV 20)and "Yamen"(GV 15)for 20 min.NGF content in the brain tissue was assayed by enzyme-labeled immunosorbent assay(ELISA).Results In comparison with sham group,cerebral NGF content had no apparent change in model group(P>0.05).Comparison among the abovementioned 6 groups showed that cerebral NGF level in NGF+EA group was significantly higher than those in sham,model,EA and NGF groups(P<0.01).No significant differences were found among sham,model,NGF,EA and NGF+H7+EA groups and between NGF+EA and NGF+H7+EA groups in cerebral NGF contents(P>0.05).Conclusion EA of GV 20 and GV 15 can raise the permeability of blood-brain barrier for NGF to increase its level in the cerebral tissue in CI rats,favoring the regeneration and repair of nerves,which has no clear relevance to PKC pathway.

      摘要:Objective To observe the effect of electroacupuncture(EA)on peripheral blood endothelial progenitor cell(EPC)counts,vascular endothelial growth factor(VEGF)level and total nitric oxide synthase(TNOS)and inducible nitric oxide synthase(iNOS)activity in cerebral ischemia-reperfusion injury(CI/RI)rats.Methods A total of 72 male rats were randomly and evenly assigned to normal control,sham-operation(sham),model and EA groups which were further divided into 24 h,48 h and 72 h subgroups,with 6 cases in each.Acute focal cerebral ischemia model was established by occlusion of the middle cerebral artery(MCAO,120 min)and reperfusion.EA(2/15 Hz,1 mA)was applied to "Quchi"(LI 11)and "Zusanli"(ST 36)for 30 min,once daily.Peripheral blood was collected from abdominal aorta for detecting EPC count by using flow cytometry,serum VEGF level by using enzyme-linked immunosorbent assay(ELISA),and serum TNOS and iNOS activity by spectrophotometry,respectively.Results Compared with the corresponding normal control subgroups,blood EPC counts,serum TNOS and iNOS activity and serum VEGF content at 24 h,iNOS activity and VEGF level at 48 h,and EPCs and iNOS at 72 h in model subgroups all increased significantly(P<0.01,P<0.05).In comparison with the corresponding model subgroups,EPC count at 24 h and 72 h,and TNOS activity at 24 h in EA subgroups decreased considerably(P<0.01,P<0.05);while EPC and VEGF levels at 48 h in EA subgroup increased evidently(P<0.05,P<0.01).No significant differences were found among normal,sham,model and EA subgroups in serum TNOS activity at 48 h and 72 h(P>0.05).Conclusion EA of LI 11 and ST 36 can suppress CI/RI induced increase of blood EPC count and serum TNOS activity,and upregulate serum VEGF level,which may contribute to its effect in relieving CI/RI.

      摘要:Objective To observe the effect of acupuncture on the fatigue degree in patients with chronic fatigue syndrome(CFS).Methods Seventy CFS patients were equally randomized into control and treatment groups according to randomized block design.Acupuncture was applied to Baihui(GV 20),Danzhong(CV 17),Zhongwan(CV 12),etc.,for patients in treatment group,and to non-acupoints(2 cm respectively to the abovementioned acupoints)for those in control group.The treatment was given once every other day,14 times altogether.The fatigue degree and the therapeutic effect were assessed by Chalder's fatigue scale(FS).Results A total of 64 cases(32/group)were finished in this study.After the treatment,the physical FS(5.0±2.4 vs 6.8±1.5),mental FS(1.8±1.8 vs 3.1±1.5)and the total FS(6.8±3.8 vs 9.9±2.5)in treatment group,physical FS(5.0±2.5 vs 6.4±1.5)and the total FS(7.5±3.4 vs 9.6±2.8)in control group decreased significantly compared with pre-treatment(P<0.01,P<0.05).There was no marked change in mental FS(2.5±1.6 vs 3.2±1.6)in control group after the treatment(P>0.05).Comparison between two groups showed no significant differences in the 3 indexes(P>0.05).Conclusion Acupuncture can relieve CFS patients' physical and mental fatigue and the therapeutic effect of acupuncture of acupoints is relatively better than that of non-acupoints in reducing mental fatigue.

      摘要:Objective To observe the clinical therapeutic effect of electroacupuncture(EA)of Jiaji(EX-B 2)plus focus-encircled needling for promoting the crust formation of herpes zoster and analgesia.Methods Eighty cases of herpes zoster patients were equally randomized into EA group [treated with EA of Ashi-point,Jiaji(EX-B 2),Zhigou(SJ 6)and Houxi(SI 3),once daily for 10 times] and medication group(treated with valaciclovir hydrochloride 300 mg/time,b.i.d.and vitamin B1 10 mg/time,t.i.d.,10 days).The pain severity was evaluated by using visual analogous scale(VAS)method.The time when the cutaneous scabbing area was equal or over 50% was recorded.Results After the treatment,of the two 40 cases in EA and medication groups,30(75.0%)and 15(37.5%)were cured,7(17.5%)and 12(30.0%)improved,3(7.5%)and 13(32.5%)failed,with the total effective rates being 92.5% and 67.5%,respectively.The therapeutic effect of EA was significantly superior to that of medication(P<0.01).VAS scores of both groups reduced significantly(P<0.01).Both the VAS score and the crust formation time of EA group were significantly lower than those of medication group(P<0.01).Conclusion EA of Jiaji(EX-B 2)in combination with focus-encircled needling is effective in facilitating the crust formation and pain relief in the treatment of herpes zoster,and the effect of acupuncture is superior to that of medication.

      摘要:Objective To observe the effect of combined treatment of apoplectic upper-limb dyscinesia with scalp-acupuncture,body-acupuncture and abdominal-acupuncture in stroke patients.Methods A total of 222 stroke patients were randomized into treatment group(n=133)and control group(n=89)according to the random number table method.For patients of treatment group,scalp-point used was Motor Area(MS 6);body acupoints were Hegu(LI 4),Quchi(LI 11),Shousanli(LI 10),etc;and abdominal acupoints Zhongwan(CV 12),Qihai(CV 6),etc.For patients in control group,only the same body acupoints were used.Acupuncture treatment was given once daily for 5 weeks.The therapeutic effects were evaluated by Fugl-Meyer motor function assessment(FMA)and Barthel-index(BI)scores.Results In comparison with pre-treatment,FMA score and BI score increased significantly 5 weeks,1 and 3 months in treatment group,1 and 3 months in control group(P<0.01).Comparison between two groups showed that both FMA score and BI score of treatment group were markedly higher than those of control group 5 weeks,1 and 3 months after the treatment(P<0.01).Conclusion Alternative administration of scalp-acupuncture,body-acupuncture and abdominal acupuncture can effectively improve stroke patients' upper-limb motor function,and its effect is obviously better than that of simple body acupuncture.

      摘要:The descriptions about the traveling routes of meridians in ancient Chinese literatures directly reflect the understanding and recognition of the ancient Chinese physicians on "meridians" concept at that time.Therefore,the changes of meridian traveling routes in different ancient Chinese periods certainly reflect the changed connotations of meridians.Beginning from va-riations of the starting and ending points,direction and traveling routes of book Ling Shu Jing(Miraculous Pivot)and the "eleven meridians" in the Silk Book,the authors of the present article hold that the main factors influencing the variation of meridian traveling pathways are 1)development of the pulse diagnosis,2)ancient Yin-Yang and math ideology,3)ancient surface anatomy knowledge,and 4)formation of the concepts of Qi,Xue(blood),and Ying-Wei circulation.The authors also analyze the significant evolution of meridian concept and connotation from the "eleven meridians" to the "twelve regular meridians".

      摘要:Non-specific connective tissue(fascia connective tissue)plays an important role in the mechanical signal transduction of acupuncture.Acupuncture needle manipulation-induced mechanical stress has a certain effect on the fibroblasts and cytoskeleton in the nonspecific connective tissue(including loose connective tissue and fat tissue)in morphology,histochemistry and biochemistry.For example,acupuncture-needle manipulation can make the fibroblast deformed,the cytoskeleton remodeled and result in the release of biochemical materials from the connective tissue.The present review summarizes new results of studies on the effect of acupuncture needle manipulation from cytobiology,imageology and physiology;and holds that making clear the transduction pathways of acupuncture mechanical stress signals in the connective tissue and its impact on the organism possesses an important significance in revealing the mechanism of acupuncture underlying clinical therapeutic effects.

      摘要:The characteristics and advantages of acupuncture therapy for apoplexy have been widely approved in China.However,the factors affecting the curative effect are various.More and more researchers think highly of the acupuncture needle-retaining duration.The present article makes a discussion on the significance of needle-retaining duration for improving stroke patients' symptoms and signs;and reviews the recent progresses,the extant problems and the academic significance about the studies of the abovementioned topic by combining the authors' previous work.Generally,a systemic study on this topic is rather deficient up to now,and is definitely necessary for further improving clinical therapeutic effect in the light of the requirements of evidence-based medicine.
