      摘要:Objective To study the effect of electroacupuncture(EA) of Stomach Meridian acupoints on changes of the related protein phosphorylation in the gastric mucosa involving the repair of the injured gastric mucosa in the rat.Methods A total of 20 SD rats were divided into control,model,acupoint and non-acupoint groups,with 5 cases in each.Gastric mucosa injury model was established by intragastric perfusion of dehydrated alcohol(0.8 ml/100 g).EA(4 Hz/50 Hz,1-3 mA) was applied to "Zusanli"(ST 36),"Liangmen"(ST 21) and "Sibai"(ST 2),and the corresponding non-acupoints,once daily for 7 days.Gastric mucosal ulcer index(UI) was measured,and the profiling of the protein phosphorylation of interest in gastric mucosa cells was detected by antibody microarrays.Results In comparison with control group,the gastric mucosal UI of model group was significantly higher than that of control group(P<0.01),while the UI of EA group was significant lower than those of model group and non-acupoint group(P<0.01),suggesting a recovery of the injured mucous membrane.Findings of protein array analysis indicated that a total of 720 kinds of phosphorylated proteins(≥1.5 folds)were found in the injured gastric mucosa.Compared to model group,the phosphorylation levels of 100 and 20 proteins were up-regulated,and 16 and 9 proteins down-regulated in acupoint and non-acupoint groups,respectively.The up-regulated proteins involve cell cycle mediation,cell proliferation,anti-apoptosis,and the regulation of the cell adhesion,cytoskeleton protein,inflammation,immune activities,etc.while the down-regulated proteins involve the down-regulation of the phosphorylation levels of the apoptosis proteins,adhesion proteins and cytoskeleton proteins,etc.In comparison to non-acupoint group,the phosphorylation levels of 100 proteins were up-regulated and 16 proteins down-regulated in acupoint group.Conclusion EA of acupoints of the Stomach Meridian can promote repair of the injured gastric mucosa,which may be related to its effects in regulating the levels of phosphorylation of many signaling proteins.

      摘要:Objective To observe the effect of electroacupuncture(EA) on the expression of amyloid precursor protein(APP) and amyloid β-protein(Aβ) in hippocampal CA 1 area and cerebral cortex in APP 695 V 717 I transgenic mice,so as to investigate its underlying mechanism in improving Alzheimer's disease(AD).Methods APP 695 V 717 I transgenic mice were randomly divided into model and EA groups,with 6 cases in each.The other 6 negatively-transgenic mice(C 57 BL/6 J) were used as the normal control group.EA(2 Hz/100 Hz,3 mA) was applied to "Baihui"(GV 20) and "Yongquan"(KI 1) for 15 min,once every other day for 3 months.The learning-memory ability of mice was measured by Y-type maze test and the expression levels of APP,Aβ,and choline acetyltransferase(ChAT) in the cerebral cotex and hippocampal CA 1 area were detected by immunohistochemistry.Results In comparison with control group,the times of training for gaining correct reaction and the expression levels of Aβ and APP in both cerebral cortex and hippocampal CA 1 area in model group increased significantly(P<0.05,P<0.01),while the expression level of ChAT of cerebral cortex in model group decreased considerably(P<0.05).Compared to model group,the times of training for gaining correct reaction and the expression levels of APP in both cerebral cortex and hippocampal CA 1 area,and Aβ in CA 1 area of EA group lowered remarkably(P<0.05),while those of ChAT in cerebral cortex and hippocampal CA 1 area of EA group increased evidently(P<0.05,P<0.01).Conclusion Electroacupuncture can improve the learning-memory capacity of the APP transgenic mice,which may be related to its effects in increasing the production of acetylcholine and lowering the levels of APP and Aβ in the brain.

      摘要:Objective To observe the effect of electroacupuncture(EA) on changes of cyclooxygenase(COX)-2 mRNA and protein expression after inflammation occurrence at the back by application of human recombinant IL-1β(hr IL-1β) plus air-pouch in rats,so as to analyze the relativity between EA-induced changes of COX-2 expression and proinflammatory cytokines regulative pathway.Methods Ninety female Wistar rats were randomized into control,model,and EA groups with 30 cases in each.Inflammatory model was established by subcutaneous implantation of a sterilized hollow teflon cylinder(sac,1.5 ml in volume) at the back and intrasaccal injection of hr IL-1β(10 ng/ml).EA(2 Hz,1 mA) was immediately applied to "Quchi"(LI 11) for 30 min.Exudate in the sac was withdrawn at various intervals after hr IL-1β-injection for assaying COX-2 mRNA expression with RT-PCR and protein expression with Western Blotting.Results In comparison with control group,one hour(h) after hr IL-1β-injection,COX-2 mRNA and protein expression levels were up-regulated significantly in model group(P<0.01);while compared to model group,COX-2 mRNA and protein expression levels in EA group were down-regulated considerably(P<0.05).5 h and 24 h after hr IL-1β-injection,COX-2 mRNA expression in model and EA groups was significantly higher than that in control group(P<0.01),and no significant differences were found between model and EA groups in COX-2 mRNA expression levels(P>0.05).On the contrary,5 h after administration of hr IL-1β,COX-2 protein expression in model group was significantly higher than that in control group(P<0.05).On the 24 h after hr IL-1β-injection,no significant differences were found in COX-2 protein expression levels among control,model and EA groups(P>0.05).Conclusion EA can effectively suppress hr IL-1β-injection induced up-regulation of COX-2 mRNA and protein expression in the rats,suggesting a close relativity between the regulation of proinflammatory cytokines and EA-induced inhibition of COX-2 mRNA and protein expression.

      摘要:Objective To study the underlying mechanism of electroacupuncture(EA) in relieving epilepsy in pentylenetetrazole(PTZ)-induced epilepsy rats.Methods Twenty SD rats were randomly divided into normal control,model,EA,Nimodipine groups,with 5 cases in each.Epilepsy model was established by intraperitoneal injection of PTZ(32.0 mg/kg),once daily for 28 days.EA(100 Hz,0.6 mA) was applied to "Baihui"(GV 20) and "Dazhui"(GV 14),once daily for 7 days.For Nimodipine group,the rats were given with nimodipine(0.25 mg/kg,i.p.),once daily for 7 days.Electroencephalogram(EEG) was recorded and the fluorescence intensity of Ca2+ of the hippocampus tissue sections was detected by laser scanning confocal microscope(LSCM) after incubation in artificial cerebrospinal fluid containing Flou-3/AM(10 μmol/L) and pluronic F-127(5 μl).Results Compared with model group,the latencies of epileptic EEG seizure prolonged obviously(P<0.05),and epileptic EEG seizure frequencies decreased significantly(P<0.05) in EA and Nimodipine groups.The fluorescence intensity of intracellular Ca2+ in hippocampus tissue in model group was obviously higher than that in control group(P<0.01).In comparison to model group,Ca2+ levels in EA and Nimodipine groups lowered considerably(P<0.05,P<0.01).No significant differences were found between EA and Nimodipine groups in the aforementioned 3 indexes(P>0.05).Conclusion EA has an obvious anti-epileptic effect,which may be closely related to its effect in downregulating the increased hippocampal Ca2+ level in PTZ-kindled epilepsy rats.

      摘要:Objective To observe the effect of electroacupuncture(EA) of "Baihui"(GV 20) and "Dazhui"(GV 14) on the ultrastructure of cerebral cortex in rats with cerebral ischemia reperfusion(CI/R) injury.Methods Fifteen SD rats were equally randomized into sham-operation(sham),model and EA groups.CI/R model was established by right middle cerebral artery occlusion for 1 h and reperfusion for 24 h.EA(20 Hz/80 Hz,1-3 mA) was applied to "Baihui"(GV 20) and "Dazhui"(GV 14) for 30 min.The ischemic cerebral cortex tissue was taken,fixed in 2.5% glutaral solution,embedded in araldite,cut into sections and stained for observing the ultrastructure changes of cortical pyramidal cells,astrocyte and blood brain barrier(BBB) under transmission electronic microscope.Results In sham group,the anatomical structure of the cerebral pyramidal cells was normal basically.In model group,the neuronal mitochondria and the capillary endothelium and the processes of the astrocyte got swelling,the mitochondrial cristae were broken,the capillary lumens became narrow or were blocked up.In EA group,the injury degree of the pyramidal cells,glial cells and BBB were lighter.Conclusion EA can reduce ischemic injury of the cerebral cortical neurons and blood brain barrier in rats with CI/R injury.

      摘要:Objective To observe the effect of electroacupuncture(EA) of "Neiguan"(PC 6) and "Zusanli"(ST 36) on c-fos expression in the nucleus of the solitary tract(NTS)and ischemic electrocardiogram(ECG) in acute myocardial ischemia(AMI) rats,so as to study the role of NTS in EA-induced improvement of AMI.Methods Thirty-six male SD rats were randomly divided into control,model,ST 36,PC 6,"Pianli"(LI 6),and "Heyang"(BL 55) groups,with 6 cases in each.AMI model was established by intraperitoneal injection of Isoproterenol(5 mg/kg).EA(4 Hz/20 Hz,1 mA) was applied to PC 6,ST 36,LI 6 and BL 55 for 20 min separately.c-fos expression in the NTS was detected by immunohistochemistry and ECG-STⅡ was recorded before and after AMI and EA.Results ① Compared to control group,ECG-STⅡ of model group elevated significantly,while in comparison with model group,ECG-STⅡ values of ST 36 and PC 6 groups decreased significantly(P<0.01),suggesting an improvement of AMI after EA.No significant changes of ECG-STⅡ were found in LI 6 and BL 55 groups(P>0.05).② The number of fos-immunoreaction(fos-IR) positive neurons in the NTS in ST 36 and PC 6 groups was significantly lower than that in model,LI 6 and BL 55 groups(P<0.01).Conclusion EA of PC 6 and ST 36 can improve ischemic ECG in AMI rats and NTS is one of the centers for integrating cardiac functional activity and information from EA of PC 6 and ST 36.

      摘要:Objective To study the underlying mechanism of electroacupuncture(EA) in relieving depression.Methods A total of 30 Wistar rats were randomly divided into normal control group(n=10),model group(n=10) and EA group(n=10).Depression model was established by lonely raising and chronic unpredictable mild stress for 21 days.EA(2 Hz,1 mA) was applied to "Baihui"(GV 20) and "Sanyinjiao"(SP 6) for 20 min,once daily for 14 days.The expression(option density) of hippocampal somatostatin(SS) and SS mRNA was detected by using immunohistochemistry and reversed transcription-polymerase chain reaction(RT-PCR) techniques respectively.Results Compared to normal control group,the crossing movement and rearing movement times of open-field test,and sucrose consumption in model group decreased significantly(P<0.01),and hippocampal SS and SS mRNA expressions were downregulated remarkably(P<0.05).Compared to model group,the crossing movement and rearing movement times in EA group increased considerably(P<0.01),and hippocampal SS and SS mRNA expressions were upregulated remarkably(P<0.05).No significant difference was found between EA and normal control groups in SS and SS mRNA expression.Conclusion EA can effectively improve depression rats' behavior activity and upregulate hippocampal SS and SS mRNA expression.

      摘要:Objective To observe the effect of electroacupuncture(EA) of "Neiguan"(PC 6) on heart rate(HR) and plasma catecholamine(CA) levels in ventricular tachycardia(VT) rats.Methods A total of 50 SD rats were randomly divided into normal control,sham-operation(sham),model,EA-PC 6 and EA-LU 7 groups,with 10 cases in each.VT model was established by intravenous injection of cesium chloride(CsCl) via femoral vein.Electrocardiogram(ECG) was recorded.EA(2 Hz/15 Hz,1 mA) was separately applied to bilateral "Neiguan"(PC 6) and "Lieque"(LU 7) for 5 minutes,followed by collecting blood samples for detecting plasma norepinephrine(NE) and epinephrine(E) levels with fluorometric method.Results In comparison with normal control group,plasma NE and E contents in model group increased apparently(P<0.01).Compared with model group,HR and plasma NE and E contents of EA-PC 6 group reduced significantly(P<0.01),while those of EA-LU 7 had no apparent change(P>0.05).Conclusion EA of "Neiguan"(PC 6) can downregulate heart rate and plasma CA level in VT rats,which may contribute to its effect in relieving tachycardia.

      摘要:Objective To observe the therapeutic effect of acupuncture("Jin's Sanzhen") treatment for autism patients with different severity.Methods A total of 202 autism children were divided into treatment group(n=118) and control group(n=84) according to their parents' will.Autism children in the treatment group were treated with "Jin's Sanzhen" therapy [for example,Niesanzhen:the point 2 cun superior to the ear tip,and those at the same level 1 cun apart on the left and right sides;Naosanzhen:Naohu(GV 17),bilateral Naokong(GB 19),etc.],while those of control group accepted special training:behavior education(cognition,speech,communication ability,life self-care ability,etc.) and sensory integration training.The treatment was given to the patients in both groups once per day,six times a week for 4 months.Results Of the 78 and 58 severe patients in the treatment and control groups,63(80.8%) and 4(6.9%) experienced marked improvement,9(11.5%) and 33(56.9%) experienced improvement,6(7.7%) and 21(36.2%) failed,with the effective rates being 92.3% and 63.8% respectively.Of the 118 and 84 patients in the treatment and control groups,after the treatment,the no symptoms,mild,moderate and severe cases were 12(10.2%) and 3(3.6%),56(47.5%) and 25(29.8%),45(38.1%) and 23(27.4%),5(4.2%) and 33(39.3%),respectively.The total score of Childhood Autism Rating Scale of treatment group was significantly lower than that of control group after the treatment in severe cases(P<0.01),showing a better therapeutic effect of the treatment group.Conclusion "Jin's Sanzhen" therapy has a better therapeutic effect for improving the autism patients symptoms at different severity.

      摘要:Objective To observe the therapeutic effect of ear-electroacupuncture(Ear-EA) on dysmenorrhea in patients with endometriosis and to explore its underlying mechanism.Methods A total of 80 endometriosis patients were randomly and equally divided into ear-EA group and body-EA group.EA(50 Hz,0.5-0.8 mA) was applied to auricular points(Uterus,Subcortex,Shenmen,Endocrine,etc.) and body acupoints [Tianshu(ST 25),Qihai(CV 6),Guanyuan(CV 4),Sanyinjiao(SP 6),Diji(SP 8),Uterus(EX-CA 1),etc.] respectively for 30 min,once every other day for 3 months.Dysmenorrhea severity score(DSS) was assessed and plasma prostaglandin(PGE2) and 6-Keto-PGF1α levels detected by radioimmunoassay.Results Compared with pre-treatment,DSS lowered significantly during the 1st and 2nd menstrual cycle in body-EA group,and during the 1st,2nd and 3rd menstruation in ear-EA group;and the DSS of ear-EA group during the 3rd menstruation was evidently lower than that of body-EA group(P<0.05).During the 3rd menstrual onset after the treatment,plasma PGE2 contents in both groups decreased obviously(P<0.01),and plasma 6-Keto-PGF1α levels increased considerably in comparison with pre-treatment(P<0.01).Comparison between two groups during the 3rd menstruation showed that plasma PGE2 level of ear-EA group was markedly lower than that of body-EA group,and 6-Keto-PGF1α level of ear-EA group was significantly higher than that of body-EA group(P<0.05).No significant difference was found between two groups in clinical therapeutic effect(P>0.05).ConclusionBoth ear-EA and body-EA can effectively relieve endometriosis-induced dysmenorrhea,and the former is superior to the later in reducing pain severity,which may be closely related to their effects in reducing plasma PGE2 and raising 6-Keto-PGF1α level.

      摘要:Objective To observe the therapeutic effect of spoon-needle cluster electrical stimulation of scalp-points for motor dysfunction of cerebral infarction(CI) patients,so as to analyze its superiority in the treatment of CI.Methods A total of 164 CI inpatients with motor disturbance were randomly divided into spoon-needle cluster(SNC) group(n=84) and filiform-needle cluster(FNC) group(n=80).Multiple filiform needles were inserted into the Yu's 7 scalp-points(Parietal region,Antero-parietal region,etc.),and manipulated for a while,followed by 6 hours' retention of the needles.Multiple spoon needles were fixed to the same scalp-points and then these scalp-points were stimulated with electric pulse for 30 min.The treatment was given once daily,6 times a week and for 4 weeks.The therapeutic effect was assessed by Fugl-Meyer assessment scale,Barthel Index and clinical neural function-defect assessment respectively.Results Compared to pre-treatment,the scores of Fugl-Meyer assessment scale and daily-life ability assessment of both SNC and FNC groups increased significantly after the treatment(P<0.01),and those of clinical neural function-defect assessment of these two groups lowered considerably(P<0.01).No significant differences were found between two groups in these 3 indexes(P>0.05).After the treatment,of the 84 and 80 cases in SNC and FNC groups,0 and 3 were cured basically,44 and 45 experienced a marked improvement,35 and 27 had an improvement,5 and 5 failed,respectively.Conclusion 〗Spoon-needle cluster electro-stimulation can effectively improve CI patients' motor dysfunction and daily-life activity ability,and is comparable to the filiform-needle cluster stimulation in clinical therapeutic effect.

      摘要:The specificity of the therapeutic effect of acupoints is an important research subject in acu-moxibustion medicine,but is short of systematic and profound literature review by using modern science and technology.The modern data mining technique provides a new reliable technique or an approach for assessing the relative specificity of the acupoint efficacy from literature reviewing angle.In order to explore the basic regularities of the specificity of acupoint efficacy,a data-library of acu-moxibustion literature should be established first,followed by setting up a data analysis model for which the acupoint prescriptions are considered to be the basis,and by establishing a data mining methodological system.

      摘要:In the present paper the authors analyze the anatomical structure of the meridian running track by using the dialectical thought and comprehensive analysis of the integrated Chinese and western medicine.It has been observed that the "Qi-passages" of the 14 meridians of Chinese medicine are located in the connective tissue among the interspace of the muscles,etc.distributing longitudinally.The "Qi-passages" of the 15 Luomai(collaterals of the meridians) are located in the connective tissue among the interspace of the muscles,etc.distributing transversally,while those of the small branches of the meridian collaterals are located in the interspace mesenchyme of the muscle bundles distributing in the whole body.The "Qi-passages" of the tiny branches of the meridian collaterals are located in the mesenchyme of the intracellular space,such as the muscle fibers in the whole body.The authors hold that the so-called "Mai Qi" of the meridian-collaterals is the liquid-Qi flowing in the vertical and horizontal tissue interspaces.The "Qi-passage" of the meridian-collaterals of Chinese medicine is the pathway of the liquid-Qi of the tissue interspaces.The structure of the meridian-collaterals is the tissue interspace.The meridian-collateral system is a regulation-control system in the human body where the Qi-passages communicate with each other,and is,in fact,the protoplasm,the liquid-Qi circulating in the tissue interspaces.

      摘要:In the present paper the authors review recent progress in the study on the underlying mechanism of acupuncture effects by using functional magnetic resonance imaging(fMRI) from 1) fMRI imaging technology,2) the meridian-collateral system of Chinese medicine,3) the factors affecting the therapeutic effect of acupuncture,4) the mechanism underlying the efficacy of acupuncture treatment,5) the experimental design methods for studying acupuncture by using fMRI,and 6) the methods for analyzing fMRI data.Research results show that fMRI technology is a new and useful approach for studying the underlying mechanism of acupuncture effect.

      摘要:Acupuncture therapy combined with medication has been widely used in clinic practice in China and proved to be effective in the treatment of many disorders.The authors of the present paper review researches on the underlying mechanism of joint application of acupuncture therapy and medication in recent decade from 1)neural regulation,2)endocrine regulation,and 3) immunological regulation.After comprehensive analysis,the authors think that the mechanism underlying the enhanced clinical efficacy of acupuncture therapy combined with drugs may be closely associated with the following factors: 1) acupuncture stimulation of acupoints leads to changes of the drug concentration in the blood and/or in the target organ and,2) the reactivity or sensitivity of the target organ tissues to the specifically effective drug elements is enhanced.
