摘要:Objective To observe the effect of electroacupuncture(EA) and moxibustion of"Dazhui"(GV 14) on the proliferation levels of the splenetic CD4+ CD25+ CD25+ regulatory T cells(Tregs) of H_(22) tumor-bearing mice in vitro.Methods Forty eight Balb/c mice were randomized into control,model,moxibustion and EA groups,with 12 cases in each.H_(22) tumor-bearing model was set up by hypodermic injection of H_(22) tumor cells(0.2 ml,1×10+ regulatory T cells(Tregs) of H_(22) tumor-bearing mice in vitro.Methods Forty eight Balb/c mice were randomized into control,model,moxibustion and EA groups,with 12 cases in each.H_(22) tumor-bearing model was set up by hypodermic injection of H_(22) tumor cells(0.2 ml,1×107 cells/ml).EA(2 Hz,2 mA) was applied to"Dazhui"(GV 14) and left"Huantiao"(GB 30) for 20 min,and moxibustion was applied to"Dazhui"(GV 14) 2 moxa-cones every time. The treatment was given from the 27 cells/ml).EA(2 Hz,2 mA) was applied to"Dazhui"(GV 14) and left"Huantiao"(GB 30) for 20 min,and moxibustion was applied to"Dazhui"(GV 14) 2 moxa-cones every time. The treatment was given from the 2(nd) day on after innoculation of tumor cells,once every other day,6 times altogether.After the treatment,the mice were killed by peeling off the eyeball and blood samples were collected to be separated into serum.Then, Tregs of the spleen tissues of Balb/c mice in different groups were isolated by using megnetic activated cell sorting(MACS) system to be cultured independently,and co-cultured with EA-treated serum and moxibustion-treated serum separately in culture fluid for 96 h,added with (nd) day on after innoculation of tumor cells,once every other day,6 times altogether.After the treatment,the mice were killed by peeling off the eyeball and blood samples were collected to be separated into serum.Then, Tregs of the spleen tissues of Balb/c mice in different groups were isolated by using megnetic activated cell sorting(MACS) system to be cultured independently,and co-cultured with EA-treated serum and moxibustion-treated serum separately in culture fluid for 96 h,added with 3H-tritiate thymidine(TdR) in the culture-fluid 12 h before the end of culture,followed by collecting the cells and detecting their proliferation levels(count per minute,cpm) by using a lipid scintillation device.Results The proliferation level of Tregs in model group was elevated significantly compared to normal control group(P<0.05),while in comparison with model group,those of Tregs of EA and moxibustion groups decreased considerably(P<0.01).After separate application of the diluted acupuncture-treated serum and moxibustion-treated serum at 1:1 and 1:8(not 1:16 and 1:32) to the cultured Tregs, their proliferation levels(cpm) in EA and moxibustion groups were obviously upregulated in comparison to those of normal control group(P<0.05),and the cpm in EA group was significantly higher than that in model group(P<0.05),suggesting a different action mechanism between acupuncture-moxibustion treatment and serum stimulation.Conclusion EA of"Dazhui"(GV 14) and"Huantiao"(GB 30) and moxibustion of"Dazhui"(GV 14) can effectively downregulate the proliferation level of the cultured splenetic Tregs of the tumor bearing mice.EA-treated serum and moxibustion-treated serum diluted at 1:1 and 1:8 can evidently upregulate the proliferation level of Tregs in vitro.
摘要:Objective To observe the effect of electroacupuncture(EA) on the levels of lymphocyte proliferation rate, spleen and serum interleukin-2(IL-2),serum tumor necrosis factor-α(TNF-α),so as to analyze its immuno-regulative function in adjuvant arthritis(AA) rats.Methods A total of 40 Wistar rats were randomized into normal control,model,Dazhui(GV 14), Mingmen(GV 4) and non-acupoint groups.Adjuvant arthritis model was established by subcutaneous injection of Freund's complete adjuvant into the paw.EA was applied to"Dazhui"(GV 14),"Mingmen"(GV 4) and non-acupoint(mid-tail) for 20 min, once every other day,8 times altogether.Paw volume(swelling degree of the voix pedis) was detected;T and B lymphocyte transformation rates(LTR),and IL-2 activity of the spleen tissue were assayed with MTT chromatometry,and serum IL-2 and TNF-αcontents were determined by radioimmunoassay.Results Compared to normal group,the paw volume and serum TNF-αcontent of model group increased significantly(P<0.01,P<0.05),and T- and B-lymphocyte proliferation rates and spleen IL-2 activity in model group decreased considerably(P<0.01).In comparison with model group,paw volumes of GV 14,GV4,and non-acupoint groups and serum TNF-αcontent of GV 14 group lowered significantly(P<0.01,P<0.05);while T- and B-lymphocyte proliferation rates,spleen IL-2 activity in GV 14,GV 4,and non-acupoint groups,as well as serum IL-2 content in GV 14 and GV 4 groups increased remarkably(P<0.01).Paw volumes of both GV 14 and GV 4 groups were significantly lower than that of non-acupoint group(P<0.05);T- and B-lymphocyte proliferation rates and IL-2 activity of GV 4 group were significantly higher than those of non-acupoint group(P<0.05).No significant differences were found between GV 14 and GV 4 groups in paw volume, T- and B-lymphocyte proliferation rates,spleen IL-2 activity,serum IL-2 and TNF-αcontents,and between GV 14 and nonacupoint groups in T- and B-lymphocyte proliferation rates,as well as among GV 14,GV 4 and non-acupoint groups in serum IL-2 and TNF-αcontents(P>0.05).Conclusion Electroacupuncture of GV 14 and GV 4 induced immunoregulation on lymphocyte, IL-2 and TNF-αmay contribute to its anti-inflammation effect in AA rats.

摘要:Objective To explore the involvement of dopamine(DA) and its metabolites in the extracellular fluid of the striatum on the resisting action of electroacupuncture(EA) for acute cerebral ischemia-reperfusion(CI/R) injury.Methods A total of 50 SD male rats were randomly divided into normal control,sham-operation(sham),sham+EA,CI/R model,CI/R+EA groups.CI/R model was induced by right middle cerebral artery occlusion(MCAO) for 90 min and reperfusion for 225 min.EA (2 Hz,3 mA,continuous waves) was applied to"Fengchi"(GB 20) for 20 min,with a 10 min rest,and given once again for 20 min.The concentrations of DA and its metabolites[3,4- dihydroxy-phenyl acetic acid(DOPAC) and homovanillic acid(HVA)] in the extracellular fluid of the striatum were determined by microdialysis and high performance liquid chromatography(HPLC). Results The extracellular levels of DA,DOPAC and HVA in normal group,sham group and sham+EA group had no significant changes during the whole study.The extracellular level of DA peaked 15-45 min after Cl and 0-30 min after CI/R(P<0.05). A rising trend was also observed 120 min following CI/R.DOPAC level peaked 15 min and 75 min after CI and 75 min after CI/R. After EA,DA level was significantly lower in EA group than that in model group 90 min after CI/R(P<0.05),and no peak was observed again.After EA,DOPAC level maintained its basic level,and no peak was found after CI/R.The changing trend of HVA was similar to that of DOPAC,and the extracellular HVA level in EA + model group was significantly lower than that in model group 75 min after CI/R.Conclusion EA can decrease the accumulation of DA and its metabolites in the extracellular fluid of the striatum, which may contribute to its effect in preventing the brain from CI/R injury.

摘要:Objective To observe the effect of electroacupuncture(EA) on hippocampal nitric oxide(NO)-cyclic guanosine 3,5-monophosphate(cGMP) signaling pathway in depression rats in order to explore the underlying mechanism of EA in improving depression.Methods Thirty male SD rats were equally randomized into control,model and EA groups.Depression model was established by using chronic unpredictable mild stress stimulation(forced ice-water swimming,electric shock,tailclamping, etc.) combined with lonely raising for 21 days.EA(2 Hz,0.6 mA,20 min) was applied to"Baihui"(GV 20) and"Yintang"(EX-HN 3),once daily for 21 days.The expression of neuronal nitric oxide synthase(nNOS) and the content of cGMP in the hippocampus were determined by immuno-histochemistry and radioimmunoassay separately.Results Many nNOS immuno -reaction(IR)-positive granular cells were observed in the hippocampus in control group,fewer found in EA group and fewest in model group.Image analysis showed that the grey value of model group was significantly higher than that of control group (P<0.01),and that of EA group was obviously lower than that of model group(P<0.01),suggesting upregulation of nNOS expression after EA.Compared with control group the content of cGMP in hippocampus in model group showed a decreasing trend, but without significant difference between two groups(P>0.05).In comparison with model group,hippocampal cGMP content of EA group increased considerably(P<0.01),being comparable to that of control group(P>0.05).Conclusion Electroacupuncture of"Baihui"(GV 20) and"Yintang"(EX-HN 3) can upregulate the expression of nNOS and the content of cGMP in the hippocampus in depression rats,maintaining a normal activity of the NO/cGMP signaling pathway,which may contribute to its effect in relieving depression.

摘要:Objective To observe the influence of electroacupuncture(EA) on T cell immunologic function in the aged rats so as to reveal its underlying mechanism in anti-aging effect.Methods Fifty SD rats(half male and half female) were randomly divided into normal control(young rats).aging model(model),EA group,immunosuppression(IS),and EA+IS groups. Aging model(in the late 4 groups) was duplicated by subcutaneous injection of D-galactose for 40 days.Guanyuan(CV 4)and bilateral Zusanli(ST 36) were punctured and stimulated electrically(2 Hz.1 mA),15 min every time,six times a week and for four weeks.IS model was induced by i.p.i.of Cytoxan(60 mg/kg) after 26 and 27 days' EA treatment.Splenetic T lymphocyte proliferation index was assayed by Methylthiazolyldiphenyl-tetrazolium bromide(MTT) chromatometry.IL-2/IL-2 receptor(R) expression level of T cells was measured by enzyme linked immunosorbent assay(ELISA).CD8+ CD28+ CD28+-T cell subset fluorescence intensity was detected by flow cytometry.Results Compared with normal control group,the splenetic T lymphocyte proliferation index,IL-2 and IL-2R expression levels,and CD8+-T cell subset fluorescence intensity was detected by flow cytometry.Results Compared with normal control group,the splenetic T lymphocyte proliferation index,IL-2 and IL-2R expression levels,and CD8+ CD28+ CD28+-T cell density in model and IS groups decreased significantly (P<0.01).In comparison with model group,the abovementioned indexes in EA and EA + IS groups increased significantly (P<0.01),and those indexes levels of EA+IS group were also significantly higher than those in IS group(P<0.05, P<0.01).Comparison between EA and EA+IS groups showed that CD8+-T cell density in model and IS groups decreased significantly (P<0.01).In comparison with model group,the abovementioned indexes in EA and EA + IS groups increased significantly (P<0.01),and those indexes levels of EA+IS group were also significantly higher than those in IS group(P<0.05, P<0.01).Comparison between EA and EA+IS groups showed that CD8+ CD28+ CD28+ T cell expression of the later group was significantly lower than that in EA group(P<0.05).No significant differences were found between EA and EA+IS groups in T cell proliferation rate and IL-2 and IL-2R expression levels(P>0.05).Conclusion EA of CV 4 and ST 36 induced upregulation of T cell proliferation and IL-2/IL-2R.and CD8+ T cell expression of the later group was significantly lower than that in EA group(P<0.05).No significant differences were found between EA and EA+IS groups in T cell proliferation rate and IL-2 and IL-2R expression levels(P>0.05).Conclusion EA of CV 4 and ST 36 induced upregulation of T cell proliferation and IL-2/IL-2R.and CD8+ CD28+ CD28+-T cell may contribute to its antiaging and immuno-enhancement effects in the aged rats.

摘要:Objective To observe the effect of electroacupuncture(EA) of Jiaji(EX-B 2) on the contents of extracellular matrix metalloproteinase(MMP)-1,MMP-3 and tissue inhibitor of matrix metalloproteinase(TIMP)-1 in the cervical intervertebral disc(CIVD) tissue in rats with CIVD degeneration so as to explore its mechanism in relieving intervertebral disc degeneration. Methods SD rats were randomized into control(n=15),model(n=14),EA(n=13) and medication(n=14) groups. CIVD model was established according to Wang's and Shi's method.EA(10/20 Hz,1—3 mA) was applied to Jiaji (EX-B 2) for 10 min,once daily for 30 days.The rats of medication group were fed with Fenbid(15 mg/kg/d) and Jingfukang (0.5 g/kg/d) for 30 days.Extracellular matrix MMP-1,MMP-3 and TIMP-1 levels of the intervertebral disc tissue(C_(4-6)) were detected by ELISA.Results The contents of MMP-1 and MMP-3 in model group were increased significantly compared with control group(P<0.05),while TIMP-1 in model group was markedly lower than that of control group(P<0.05).Compared with model group,both MMP-1 and MMP-3 levels in EA group and MMP-1 in medication group were reduced considerably(P<0.05), while TIMP-1 of EA group increased significantly(P<0.05).No significant differences were found between medication and model groups in MMP-3 and TIMP-1 levels(P>0.05).Under optical microscope,the injured degrees of the fibrous ring and nucleus pulposus, and the outline of the intervertebral disc of both EA and medication groups were evidently lighter than those of model group,but the degeneration severity of the intervertebral disc of EA group was lighter than that of medication group.Conclusion EA of Jiaji(EX-B 2) can effectively relieve the degeneration degree of cervical intervertebral disc in the rat,which is probably related to its effect in regulating extracellular matrix system.

摘要:Objective To observe the effect of different frequency electroacupuncture(EA) on hippocampal substance P(SP) andβ-endorphin(β-EP) in rats with gastralgia so as to explore its underlying mechanism in preventing and relieving visceral pain.Method Fifty rats were randomly divided into control,sham operation,model,low- frequency-EA and high frequency -EA groups.Gastralgia model was duplicated by gastric distension(a pressure of 40 mmHg) with a latex balloon.Low(2 Hz) and high(100 Hz) frequency EA was applied to bilateral"Zusanii(ST 36)"for 30 min.Pain score was given by using Al-Chaer's method(abdominal withdrawal reflex) and the expression of hypothalamicβ-EP and SP was detected by immunohistochemistry. Results Both low- and high-frequency EA of ST 36 could significantly lower the pain score(P<0.01).and the effect of high-frequency EA was markedly superior to that of low-frequency(P<0.01).Compared with control and sham-operation groups, both SP immuno-reaction(IR) andβ-EP IR positive cells in the hypothalamus in model group increased significantly in the number (P<0.01).suggesting an upregulation of SP andβ-EP expression after occurrence of gastralgia.In comparison with model group,the number of SP IR andβ-EP IR positive cells in the hypothalamus in low frequency and high frequency EA groups increased further(P<0.01) particularly in low frequency EA group.Conclusion Both low-frequency and high-frequency EA of"Zusanli(ST 36)"can effectively relieve gastric distension-induced gastralgia in the rat,and the effect of high-frequency EA is better, which may be closely related to its actions in promoting the release and synthesis of hypothalamicβ-EP and SP.
摘要:Objective To observe the changes of cardiac function of spleen-deficiency syndrome after joint administration of acupuncture and medication.Methods A total of 48 Wistar rats were randomized into normal control,model,medication (American ginseng,AG) and acupuncture+AG groups.Spleen-deficiency syndrome model was established by monophagia method. Rats of medication group were fed with AG decoction(1 ml/100 g),once daily for 10 days.Acupuncture stimulation(uniform reinforcing and reducing method) was applied to bilateral"Zusanli"(ST 36) and"Sanyinjiao"(SP 6) for 1 min(with the needles retained for 20 min),once daily for 10 days.Then,all the rats anesthetized underwent right common carotid artery-left-cardiac catheter and right femoral arterial catheter surgery to accept detection of the left ventricular end diastolic pressure(LVEDP),left ventricular pressure(LVP),heart rate(HR),systolic blood pressure(SBP),diastolic blood pressure(DBP) and cardiac output (CO).Results Compared with normal control group,the levels of SBP,DBP,HR and CO of model group were significantly lower(P<0.05).In comparison with model group,SBP,DBP,HR and CO of medication and acupuncture+AG groups were markedly higher(P<0.05).No significant differences were found between medication and acupuncture+AG groups in SBP, DBP,HR,LVEDP,LVP and CO levels(P>0.05).Conclusion Administration of both acupuncture+AG and AG can effectively improve spleen-deficiency syndrome rats' cardiac function.
摘要:Objective To observe the therapeutic effect of electroacupuncture plus moxibustion(EA-Moxi) of combined Shu- and Mu-acupoints on the symptoms and quality of daily life in chronic superficial gastritis(CSG) patients.Methods Sixty cases were randomly divided into EA-Moxi group(n=30) and medication group(n=30).For patients of the former group.EA (50 Hz,3 mA) was applied to Zhongwan(CV 12),bilateral Weishu(BL 21),bilateral Zusanli(ST 36),in combination with bilateral Ganshu(BL 18) and Qimen(LR 14) for patients with liver-stomach disharmony syndrome,and with moxibustion of Zhongwan (CV 12),bilateral Weishu(BL 21),bilateral Zusanli(ST 36),Pishu(BL 20) and Zhangmen(LR 13) for those with spleenstomach deficiency syndrome.The treatment was administrated once a day,five days a week for 4 weeks.The patients of medication group were treated with Omeperazole(20 mg,qd.) capsules.The total course of treatment was 4 weeks.Clinical therapeutic effects were evaluated according to some related standards,and the quality of life was assessed by SF-36 questionnaire. Results After the treatment,of the 30 cases in EA-Moxi and medication groups.6(20.0%) and 3(10.0%) were cured,16 (53.3%) and 8(26.7%) experienced marked improvement,5(16.7%) and 9(30.0%) were effective,and 3(10.0%) and 10 (33.3%) failed,with the effective rates being 90.0%and 66.7%respectively.The difference of the effective rates between two groups was significant(P<0.05).Self-comparison showed a marked improvement in the quality of life including physical function (PF),role physical(RP),bodily pain(BP),general health(GH),vitality(VT),social function(SF),role emotion(RE) and mental health(MH) in both groups after the treatment.The scores of health utility(HU),RP and RE in EA-Moxi group were evidently higher than those of medication group(P<0.05,P<0.01).Conclusion EA-Moxi of combined Shu- and Mu-acupoints can effectively relieve CSG patients' clinical symptoms and their quality of life.

摘要:Objectives To observe the impact of acupoint-catgut implantation on physical agility of athletes and to probe into its underlying mechanism.Methods A total of 68 volunteer athletes were recruited in the present study.Sixteen rowers of China National Women's Racing Boat Team(CNWRBT) underwent comparative observation between pre- and post-treatment with acupoint-catgut implantation.Fifty-two athletes from Zhejiang Heavy Athletics Events Team(HAET) and Sand-beach Volleyball Team(SBVT) were randomized into control(n=26) and treatment(n=26) groups which were treated with massage (20 min,once daily for 1 year),and catgut implantation at Guanyuan(CV 4),Mingmen(GV 4),etc.(once every two weeks), respectively.In the later group,after 6 times of treatment,the catgut implantation was given once a month,continuously for 1 year.Serum testosterone and plasma hemoglobin(Hb) levels were detected before,1.5 mon,3 mon,6 mon and 12 mon after the treatment,respectively.Results 1) Compared with pre-treatment,the 2 000 m ergometer time was shortened significantly (P<0.01),and serum testosterone and plasma hemoglobin(Hb) levels were increased evidently in 16 volunteer rowers of CNWRBT after 3 months' acupoint-catgut implantation treatment(P<0.05,P<0.01).2) In comparison with control group and pre-treatment,the midium- and long-distance running scores of the athletes of Zhejiang HAET and SBVT were elevated considerably (P<0.01),serum testosterone and plasma Hb levels increased significantly(P<0.05).Conclusion Acupoint-catgut implantation can significantly improve the athletes' anti-fatigue exercise capacity,enhance their physical agility to elevate their athletics events score,which may be closely related to its effects in raising blood testosterone and hemoglobin levels.

摘要:Objective To observe the changes of symptom scores and serum IgE level after treatment with thick-needle subcutaneous penetration of Shendao(GV 11) in chronic urticaria patients.Methods A total of 60 chronic urticaria patients were randomly divided into acupuncture group(n=30) and medication group(n=30).Subcutaneous penetrative needling was applied to GV 11 with thick acupuncture needle(retained for 4 h/time,once daily,5 times/week) for patients of acupuncture group and Levocetirizine Hydrochloride tablets(5 mg/time,once daily in the first two weeks,then,once every other day in the 3(rd) and 4(rd) and 4(th) weeks,and once every 3 days in the last two weeks) were given to patients of medication group.Serum IgE content was assayed before and 2,6,12 weeks after the treatment by chemiluminescent technique.Symptom scores were obtained by"0-3 four levels assessment"method in the light of the size of the wheal and the itching severity.Results Self-comparison indicated that the symptom scores and serum IgE levels declined significantly(P<0.01)2 and 6 weeks(Wks) after the treatment in both acupuncture and medication groups,and 12 Wks after the treatment in the acupuncture group(P<0.01).Comparison between two groups showed that the symptom score and serum IgE content of acupuncture group were significant lower than those of medication group 12 Wks after the treatment(P<0.05).No significant differences were found between two groups in the symptom scores and serum IgE levels before,2 and 6 Wks after the treatment(P>0.05).A positive correlation exists between the symptom score and the serum IgE level before and after the treatment in both groups.Conclusion Thick-needle subcutaneous penetration of Shendao(GV 11) can effectively improve clinical symptoms of chronic urticaria patients,which may be closely related to its effect in lowering peripheral blood IgE level.
摘要:In the present paper,the authors sum up the same and different view points about acupuncture treatment of migraine in the ancient and modern clinical practice,and make a detailed explanation from three aspects:1) the therapeutic principles, handed down from the ancient times,and still surviving in clinical practice nowadays:2) the traditional principles recorded only in the ancient literature but given up in modern treatment;and 3) the new therapeutic principles innovated in modern clinical practice.Further comprehensive analysis reveals that the modern therapeutic principles for migraine are still relatively simple in spite of having some rather strong advantages,and some important traditional therapeutic principles have been lost unfortunately. The authors strongly recommend that the migraine,a commonly seen problem in clinic,may be managed from a new angle,i.e., solving it by puncturing acupoints of both Shaoyang and Taiyang meridians on the severer side.
摘要:Clinical practice has demonstrated that acupuncture of Jing(Well)-points is effective in the treatment of cerebral infarction,vascular dementia and other problems induced by acute or chronic cerebral ischemia.In recent years,abundant results from animal experiments of cerebral ischemic(CI) injury have preliminarily uncovered some underlying mechanisms of acupuncture of Jing points in reducing Cl injury,including 1) scavenging active free radicals,2) decreasing calcium overloading, 3) regulating inflammation-related cytokines expression,and 4) upregulating the expression of anti-apoptosis genes.However, further studies are needed because of the complexity of the development of ischemic cerebral injury.
摘要:In the present paper,the authors introduce recent progress of application of molecular biological techniques in the studies on the underlying mechanism of acupuncture therapy for 1) resisting inflammation and relieving pain,2) resisting tumors,3) improving ischemic cerebral and myocardial injury,and 4) delaying aging from gene transcription and expression(synthesis of mRNA);and for relieving sciatic nerve injury,spinal traumatic injury,and cerebral ischemia from protein expression or phosphorylation.In addition,the authors also make a preliminary analysis on the extant problems of the application of techniques of both genomics and proteomics in acupuncture research at the present and predict their applicable prospect.
