摘要:Objective To observe the effect of electroacupuncture(EA) on the learning-memory ability in Alzheimer's disease(AD) rats.Methods Thirty SD rats were equally randomized into control,model and EA groups.AD model was established by injecting β-amyloid(Aβ25-35,10μg) into the bilateral dentate gyri of the hippocampal CA 1 area(AP-3.5mm,ML±2.0mm,DV 2.7mm).EA(4Hz,1-2mA) was applied to "Baihui"(GV 20),"Dazhui"(GV 14),bilateral "Shenshu"(BL 23) and bilateral "Yongquan"(KI 1) for 30min,once daily for 7 days.The learning-memory ability was detected by using step-down test.Long term potentiation(LTP) of hippocampal CA 1 area was recorded by using tungsten microelectrodes after high frequency stimulation(HFS) conditioning of the cortical anterior perforated substance.Results In AD rats,the error number and total error time of step-down test were increased significantly(P<0.05,P<0.01),and the slope of the excitatory post-synaptic potential(EPSP),and the amplitude and area of population spikes(PS) at 30min,60min,90min and 120min after high-frequency stimulation of the cortical anterior perforated substance lowered markedly in comparison with sham operation(control) group(P<0.05,P<0.01).After EA treatment,the error number and total error time decreased significantly,the slope of EPSP,and the amplitude and area of PS from 30min to 120min after high-frequency stimulation increased considerably in compa-rison with model group(P<0.05,P<0.01),suggesting an improvement of the synaptic transmission after EA.Conclusion EA can improve AD rats' learning-memory ability and raise the slope of EPSP and the amplitude of PS,which may contribute to its effect in relieve symptoms of AD in clinic.

摘要:Objective To observe the effect of the eye-acupuncture therapy on serum and colonic substance P(SP) and vasoactive intestinal peptide(VIP) contents in rats with irritable bowel syndrome(IBS) so as to explore its underlying mechanism.Methods Forty male Wistar rats were equally randomized into control group,IBS model group,eye-acupuncture group and medication(Pinaverium bromide,7.5mg/kg,twice daily,intragastric administration) group.IBS model was established by giving the rat with chronic stress stimulation(cold-water swimming,tail clamping,electrical shock,etc.) for 18 days.Eye-acupuncture of Xiajiao(Low Energizer) Area,Pi(Spleen) Area,Gan(Liver) Area and Dachang(Large Intestine) Area was given to the rat 20min,twice daily for 7d.Histopathological changes of the colon tissue were displayed by HE staining;and serum and colonic SP and VIP contents were detected by enzyme linked immunosorbent assay(ELISA).Results No significant difference was found among 4 groups in the histopathological changes of the colon.In comparison with normal control group,both serum and colonic SP and VIP contents in model group increased significantly(P<0.01),while compared with model group,those in eye-acupuncture and medication groups lowered considerably(P<0.01).Conclusion Eye-acupuncture can reduce serum and colonic SP and VIP contents in IBS rats,which may play a role in relieving IBS in eye-acupuncture clinic.
摘要:Objective To observe the effect of electroacupuncture(EA) on the expression of spinal glial fibrillary acidic protein(GFAP),tumor necrosis factor-α(TNF-α) and interleukin-1β(IL-1β) in chronic constriction injury(CCI) rats.Methods Seventy-two SD rats were randomized into sham operation(sham),CCI(model) and EA groups(n=24/group).The mechanical and thermal pain thresholds were measured by using Von Frey filaments and radiant-heat irridiation separately.The immunoactivity of GFAP of spinal dorsal horn(L4-L5) was assessed by immunohistochemistry,and the expression of spinal TNF-α mRNA and IL-1β mRNA was detected by real time-PCR.Results Compared with pre-CCI and sham group,both mechanical and thermal pain thresholds decreased considerably in rats with CCI(P<0.05),and in comparison with model group,those of EA group increased markedly(P<0.05).Compared with sham group,GFAP immunoactivity(mainly in the lamina Ⅰ-Ⅱ of the spinal dorsal horn),TNF-α mRNA and IL-1β mRNA expression in the ipsilateral spinal cord on the CCI side in model group increased consi-derably(P<0.05),while compared with model group,the expression of GFAP,TNF-α mRNA and IL-1β mRNA in EA group was down-regulated remarkably(P<0.05).Conclusion EA can effectively suppress CCI-induced up-regulation of expression of spinal GFAP,TNF-α mRNA and IL-1β mRNA,which may contribute to its effect in reducing mechanical allodynia and thermal hyperalgesia in neuropathic pain rats.
摘要:Objective To observe the effect of electroacupuncture(EA) on cortical spreading depression(CSD) and contents of plasma calcitonin gene-related peptide(CGRP) and substance P(SP) in migraine rats.Methods Thirty male SD rats were equally randomized into control,model and EA groups.Migraine model was established by topical application of KCl(3mol/L) immersed in a piece of filter paper to the cerebral cortex(parietal lobe,6mm posterior to the Bregma and 5mm to the sagital fissure) after exposure of the skull(in reference to Michael' method).KCl stimulation evoked CSD potentials(3mm rostral to the Bregma,and 2mm to the sagital fissure) were recorded by using a glass microelectrode.For rats of control group,filter paper containing 0.9% NaCl was applied to the same parietal cortex area.EA(1mA,2Hz/100Hz) was applied to bilateral "Yanglingquan"(GB 34) and "Taichong"(LR 3) for 30min.The contents of plasma CGRP and SP were assayed by radioimmunoassay.Results CSD was induced 3-5 min after application of KCl to the parietal lobe.The average amplitude of model group was(-25.13±1.23)mV,and that of EA group was(-19.19±1.53)mV,displaying a significant reduction of CSD amplitude after EA(P<0.01).Comparison among 3 groups showed that both plasma CGRP and SP contents in model group were significantly higher than those of control group(P<0.001,P<0.01),while compared with model group,plasma CGRP and SP levels in EA group decreased considerably(P<0.05,P<0.001),suggesting an inhibitory effect of EA on pain-producing substance.Conclusion EA of GB 34 and LR 3 can effectively suppress KCl provoked cortical spreading depression and plasma CGRP and SP levels in the rat,which may contribute to its effect in relieving migraine in clinic.

摘要:Objective To observe the effect of electroacupuncture(EA) on Calbindin D28K(CB) expression and apoptosis in hippocampus in hyperlipemia rats with concomitant cerebral ischemia(CI).Methods Forty male SD rats were rando-mized into control,hyperlipemia+CI(model),acupuncture Ⅰ,and acupuncture Ⅱ groups,with 10 cases in each.Hyperlipemia model was established by feeding the animals with high fat forage for 6 weeks and CI model established by occlusion of the unila-teral middle cerebral artery.For rats of acupuncture Ⅰ group,EA(1-3 mA,15 Hz) was applied to bilateral "Sanyinjiao"(SP 6) and "Fenglong"(ST 40) for 20 min every time,once daily for 10 days before CI.For rats of both acupuncture Ⅰ and Ⅱ groups,after CI,EA was applied to SP 6,ST 40,"Baihui"(GV 20) and "Shuigou"(GV 26) were punctured with filiform needles and stimulated by twirled the needle with hand continuously for 1 min,once daily for 7 days.The apoptotic cells of hippocampal CA 3 area were displayed by TUNEL(terminal deoxynucleotidyl transferase biotin-dUTP nick end labeling) method.CB antibody staining with immunohistochemistry was used to show CB-positive cells which were observed by optic microscope.Results In comparison with control group,the percentage of apoptotic cells of the hippocampal CA 3 area in model group increased considerably(P<0.01),while the number of CB immunoreaction(IR)-positive cells in the same area decreased significantly(P<0.05).Compared with model group,the percentages of apoptotic cells of CA 3 area in acupuncture Ⅰ and acupuncture Ⅱ groups lowe-red remarkably(P<0.01),while the numbers of CB IR-positive cells in the later two groups increased significantly(P<0.01).The therapeutic effect of acupuncture Ⅰ group was apparently superior to that of acupuncture Ⅱ group in lowering the percentage of apoptotic cells and upregulating CB IR-positive cell number(P<0.05).Conclusion Acupuncture can effectively lessen neuronal apoptosis of the hippocampus CA 3 area which may be closely related to its effect in upregulating the expression of CB in hyperlipemia+CI rats.Moreover,the cerebral ischemia injury degree may be lightened by acupuncture-lowering hyperlipemia first.

摘要:Objective To study the peripheral and central neural involvements of acupoint "Taixi"(KI 3) by using Horseradish Peroxidase(HRP).Methods Ten male SD rats were used in the present study.5μL of 1% HRP was injected into the excavate site between the apex of medial malleolus and the Achilles tendon of the left hind foot,a corresponding site of acupoint "Taixi"(KI 3) in the human body.After 36 surviving hours,the rat's brain,spinal cord and dorsal root ganglia(DRG) of the lumbar segments(L) were dissected from the perfused experimental animals,and their tissue sections were examined with 3,3',5,5'-tetramethyl-benzidine histochemistry for revealing HRP-labeling.Results All the HRP-labeling appeared ipsilaterally to the injection side.Numerous HRP-labeled neurons were detected in the DRGs of L4-L6 segments with their number being L5>L4>L6.A longitudinal column of HRP-labeled motoneurons was found in the dorsolateral and mediolateral portions of the spinal cord,distributing in the lamina Ⅸ from the caudal L4 to the rostral L6.Additionally,the transganglionic HRP-labeled central projection axonal terminals were found to be dense in the central part of laminae Ⅰ-Ⅱ from L4 to the rostral L6,and to scatter in the central part of gracile nucleus.Conclusion HRP-labeled primary afferent and efferent innervating acupoint "Taixi"(KI 3) are DRGs of L4-L6,the dorsolateral and mediolateral motoneuron columns of L4-L6,and the centrally projecting axonal terminals of laminae Ⅰ-Ⅱ of the spinal cord and the gracile nucleus.
摘要:Objective To observe the effect of electroacupuncture(EA) of different acupoints on activities of the car-diac sympathetic nerve in rabbits with acute myocardial ischemia(AMI).Methods A total of 40 Chinchilla rabbits were rando-mized into normal control,model,Shenmen(HT 7),Taixi(KI 3) and non-acupoint groups(n=8/group).AMI model was established by intravenous injection of pituitrin(3U/kg).EA(1.1mA,2Hz) was applied to bilateral HT 7,KI 3 and non-acupoint(the bulge site of the triangular muscle) for 10min.The rabbit's cardiac sympathetic discharges were recorded by BIOPAC under anesthesia.Results In comparison with control group,the heart rate(HR) and the frequency of cardiac sympathetic discharges(CSD) decreased significantly after AMI(P<0.01).At the moment of cease of EA,at 20min after EA,HR and CSD frequency in HT 7 group were significantly higher than those of model group(P<0.01),suggesting a favorable inhibition of EA of HT 7 on the decreased HR and CSD.No significant differences were found between KI 3 and model groups,and between KI 3 and non-acupoint groups in both HR and CSD at any time courses(P>0.05).Conclusion EA of HT 7 can correct AMI-induced decrease of both HR and CSD in the rabbit,while EA of KI 3 and non-acupoint have not this effect,suggesting a relatively specific efficacy of EA of different acupoints on cardiac sympathetic activities.
摘要:Objective To observe the effect of electroacupuncture(EA) on hippocampal structure in splenic asthenia pedo-rats.Methods A total of 15 SD male rats were randomly assigned to normal control group(n=5),model group(n=5) and EA group(n=5).Splenic asthenic syndrome model was established by intragastric administration of rhubarb and intraperitoneal injection of Reserpine for 14d.EA(1mA,3Hz/15Hz) was applied to bilateral "Zusanli"(ST 36) and "Sanyinjiao"(SP 6) for 20min,once a day for 14 days.The cellular structure of hippocampus was observed by light microscope and transmission electron microscope.Results Optical microscopic observation showed that in normal control group,the cellular nucleus was distinct,and the granular cell layer well-arranged and tight.In model group,the intracellular space was widened,and the granular cell layer was out of order in the arrangement.In EA group,the cellular nucleus and the granular cell layer were nearly normal.Results of the electronic microscope showed that cells in model group had a karyopyknosis with irregular appearance and clear incisure,and some of them presented dissolving and necrotic phenomena;and those in EA group were milder in injury,had nearly-normal nucleus with visible nucleoli and relatively-intact nuclear membrane.Regarding the cellular plasma,in comparison with rich normal organelles of control group,the mitochondria in model group were swelling,with vague,dissolved and broken cristae,while in EA group,majority of the organelles were well-kept,and slightly dissolved mitochondrial cristae found.In regard to the synaptic structure,in comparison with control group,synaptic apomorphosis and swelling mitochondria were found in model group.While in EA group,milder swelling and hydropic degeneration were seen.Different from the distinct pre-and post-synaptic membrane and synaptic vesicles of control group,the synaptic clefts and vesicles of model group were vague in the profile;while those in EA group were nearly-normal.Conclusion Electroacupuncture can effectively relieve splenasthenic syndrome induced pathohistological changes of neurons of the hippocampus in the rat.
摘要:Objective To observe the analgesic effect of transcutaneous electrical acupoint stimulation(TEAS) in assisting general anesthesia(GA) for radical operation of breast carcinoma so as to explore its clinical application value.Methods Sixty patients scheduled for radical operations of the breast carcinoma were randomly and equally divided into GA group and TEAS+GA group.For GA,Propofol [4.5μg/mL,target concentration;(3.0±0.5)μg/mL during operation] was used in combination with Sufentanil(0.25μg/kg) and Cis-atracurium amine(0.2mg/kg).TEAS(5-10mA,2Hz/100Hz) was applied to Hegu(LI 4)-Laogong(PC 8) and Neiguan(PC 6)-Waiguan(SJ 5) on the affected side of the body for 30min.Heart rate(HR),mean aterial pressure(MAP),tube removing time and postoperative complications were recorded.Plasma β-endorphin content was assayed by radioimmunoassay.Results After anesthesia,patients with reduction in MAP in GA group were significantly more than those in TEAS+GA group(P<0.05),and their HR decreased remarkably(P<0.05).After tracheal intubation,MAP in TEAS+GA group kept relatively stable,which was superior to that of GA group(P<0.05).In comparison with GA group,the patients' analepsia time and tube withdrawal time of TEAS+GA group were significantly shorter;and the dosage of analgesics of TEAS+GA group was significantly smaller than that of GA group(P<0.05).Patients with complications of pain,restlessness,drowsiness,nausea and vomitting were obviously fewer in TEAS+GA group than those in GA group.Compared with GA group,plasma beta-endorphin content in TEAS+GA group increased considerably 30min after TEAS,5min after skin-incision and after operation(P<0.05).Conclusion TEAS combined with general anesthesia for breast radical carcinoma operation can help keep a stable blood pressure during surgery,reduce the dosage of analgesics and strengthen pain relief.The analgesic effect of TEAS may be related to its effect in up-regulating plasma beta-endorphin level.
摘要:Objective To observe the effect of electroacupuncture(EA) of different acupoints on electroretinogram(ERG) and cerebral visual evoked potentials(VEP) in healthy subjects so as to evaluate the relative specificity of the function of acupoints.Methods A total of 33 healthy adult volunteers were observed in the present study.The subject was asked to lie on a testing bed for a while,two disk electrodes were respectively fixed to the suborbital region and the occiput(the crossing point,5cm apart from the right median line of the head and 5cm above the ear),and the reference electrode was fixed to the medial side of the earlobe for recording ERG and cerebral VEP separately.The testing room was asked to keep faint light during the whole recording process.A flash stimulation(frequency 1 Hz) was applied to the subject's eye(with a distance of about 50 cm between the light source and the tested eye,and the other eye was covered by an eyepatch).EA(0.5 Hz,0.2 ms and 3-4 V) was applied to Guangming(GB 37),Neiguan(PC 6) and Zusanli(ST 36) for 30 min respectively in different testing days(with the interval being 3 days at least).ERG and cerebral VEP were recorded by using Polygraphy(RM-6200).Results Following EA of Guangming(GB 37),Neiguan(PC 6) and Zusanli(ST 36),the changing values(increase and decrease) of the amplitude of b waves of ERG and P100 waves of cerebral VEP were(19.68±12.61)% and(12.84±14.08)%,(6.09±14.06)% and(5.61±8.01)%,(2.39±3.52)% and(5.43±8.22)%,respectively,while those of b waves of ERG and P100 waves of cerebral VEP of GB 37 were significantly bigger than those of PC 6 and ST 36(P<0.01).Conclusion EA of Guangming(GB 37) can significantly change the amplitude of both ERG and cerebral VEP in volunteer subjects,which is markedly bigger than those of EA of Zusanli(ST 36) and Neiguan(PC 6),suggesting a relative specificity of the effect of EA of different acupoints.

摘要:Objective To observe the changes of electrical property of the 12 Source-points in encephaloma patients undergoing surgery.Methods A total of 116 encephaloma patients and 60 healthy people who signed the informed consent were enlisted in the present study.The regional cutaneous electric resistance(CER) of the bilateral 12Yuan(Source)-points was measured in the afternoon(14:00-16:00) before and one week after surgery under room temperature [(22±3)℃,(55±10)% in humidity] by using "Meridian Energy Analysis Device".Results In comparison with normal subjects,CER values of the 12 Source-points on both sides of the body in encephaloma patients were significantly lower(P<0.01).Before surgery,CER values of Wangu(SI 4),Taibai(SP 3),Taichong(LR 3) and Chongyang(ST 42) on the left side were significantly higher than those of the isonym points on the right side(P<0.01,P<0.05),suggesting an imbalance of the bilateral Small Intestine,Spleen,Liver and Stomach Meridians.After surgery,CER values of Shenmen(HT 7),Wangu(SI 4),Yangchi(SJ 4) and Taibai(SP 3) on the left side were markedly higher than those of the isonym points on the right side(P<0.05,P<0.01),suggesting an imbalance of the bilateral Heart,Small Intestine,Trienergizer and Spleen Meridians.Comparison between pre-and post-surgery showed that CER values of Taiyuan(LU 9),Daling(PC 7),Taibai(SP 3),Taichong(LR 3),Taixi(KI 3),Jinggu(BL 64),Qiuxu(GB 40) and Chongyang(ST 42) on both sides,and Wangu(SI 4) on the right side lessened obviously post-surgery(P<0.01,P<0.05),suggesting an imbalance of the bilateral meridians found in more meridians,including yang meridians entering the brain as the Bladder,Gallbladder,Stomach Meridians.Conclusion In encephaloma patients,the CER values of some Source-points on the left side are significantly higher than those on the right side,suggesting an imbalance of the bilateral meridians in functional activities.
摘要:Objective To compare the therapeutic effects of different acupuncture methods in improving the myodynamia and neuronfunction of patients with acute cerebral infarction(ACI).Methods A total of 90 ACI patients were randomized into ear-acupuncture,scalp-acupuncture and body-acupuncture groups,with 30 cases in each.For patients of ear-acupuncture group,the main otopoints used for penetrative needling were Zhen(MA-AT)-Nie(MA-AT)-E(MA-AT) on the affected side in combination with Jian(MA-SF 4)-Suogu(MA-SF 5) and Zhou(MA-SF 3)-Wan(MA-SF 2)-Zhi(MA-SF 1) for upper-limb paralysis,and with Tun(MA-AH 5)-Zuogushenjing(MA-AH 6),Kuan(MA-AH 4)-Xi(MA-AH 3),and Xi(MA-AH 3)-Huai(MA-AH 2)-Zhi(MA-AH 1) for lo-wer-limb paralysis,and body acupoints as Jianyu(LI 15),Hegu(LI 4),Huantiao(GB 30),Taixi(KI 3),etc.For patients of scalp-acupuncture group,scalp-points used were Dingnie Qianxiexian(MS 6) and Dingnie Houxiexian(MS 7) on the healthy side,and combined with body acupoints(the same as those mentioned above).For patients of body-acupuncture group,only body acupoints were used.The treatment was given once daily for 14 days.Results Comparison among 3 groups showed that the increased myodynamia of both upper and lower limbs in ear-acupuncture and scalp-acupuncture groups was significantly superior to that of body-acupuncture group(P<0.01).The neurofunctional deficit scores of ear-acupuncture and scalp-acupuncture groups were significantly lower than that of body-acupuncture group(P<0.01) after the treatment.In comparison with pre-acupuncture,the neurofunctional deficit scores of the 3 groups lessened considerably after the treatment(P<0.01).Of the 30 cases in each of ear-acupuncture,scalp-acupuncture and body-acupuncture groups,0,1 and 0 were cured basically,21,18 and 4 experienced marked improvement,9,11 and 20 were improved,0,0 and 6 failed,respectively.The therapeutic effects of ear-acupuncture and scalp-acupuncture were obviously superior to that of body-acupuncture group(P<0.01).No significant differences were found between ear-acupuncture and scalp-acupuncture groups in myodynamia improvement,neurofunctional deficit scores and clinical curative effect(P>0.05).Conclusion Acupuncture can effectively improve ACI patients' clinical symptoms and the therapeutic effect of ear-acupuncture and scalp-acupuncture was superior to that of simple body acupuncture.
摘要:By using "Acupuncture Manipulation Information Analyzing System",the traditional reinforcing and reducing manipulations of acupuncture described in classical Chinese medical books were imitated and their parameters analyzed.It was found that in the oscillogram,the differences between the reinforcing and reducing manipulations(rapid-slow reinforcing-reducing skill,lifting-thrusting reinforcing-reducing skill) are the bigger duration of descending branch(t1) and smaller duration of ascending branch(t2) for the former,and shorter t1 and longer t2 for the later,due to slow insertion and rapid withdrawal of the acupuncture needle for the reinforcing,and rapid insertion and slow withdrawal of needle for the reducing.While for twirling reinforcing,t1t2 for predominantly-rightward twirling were found.In addition,it is supposed that those parameters as the amplitude(s),frequency(f) and duration of one cycle(t) of the oscillogram may be related to acupuncture stimulating quantity,while t3(duration of the plateau segment),t4(duration of the horizontal plane segment) and their variance may be associated with the stability of the acupuncture manipulation.Due to the limited literal recordation,some other traditional acupuncture manipulations can not simulate indeed,because we don't know how to perform those kinds of acupuncture manipulation.This just reflects the importance of objectivity of acupuncture manipulations.
摘要:Combining the functional metabolic imaging and anatomical imaging,PET-CT stands for the highest level of current nuclear medical imaging techniques,and has been applying increasingly in acupuncture studies.However,owning to complexity of the brain function and sensitivity of brain metabolism,the lower reproducibility of cerebral function imaging even may reverse many results.This has been provoked more and more attention by researchers.Its main cause is intimately related to poor experimental methodology.For this reason,the present paper raises the quality control of the research on mechanisms of acupuncture in the process of application of PET-CT techniques from 1) the included standards of participants,2) the preparation of participants during the scanning process of PET-CT,3) the setting of the scanning parameters,4) the used machine and the imaging reagent,5) the analysis of dada,6) the standardization of acupuncture manipulation,and 7) the designs of the acupuncture operation,needling opportunity and scanning opportunity,hoping to offer some beneficial information for the coming researches.

摘要:In the present paper,the authors review development of studies on the mechanism underlying the correlation between acupoints/ meridians and internal organs in recent 10 years from 1) doctrine of "dynamic acupoint";2) effect of one acupoint-stimulation on multi-viscera,3) effect of multi-acupoints/multi-meridians stimulation on one single internal organ,4) specific reactions of visceral disorders on the body surface,5) the therapeutic effect of acupoint-injection being better than muscular injection,6) effect of acupuncture on the expression of some known intracellular genes/proteins of the related viscera,7) effect of acupuncture on the expression or phosphorylation of whole genes/whole proteins of the related viscera,8) peripheral pathways connecting acupoints and the associated internal organ,9) central pathways connecting acupoints and the related viscera,and 10) overview of the similar researches in foreign countries.In addition,the authors also make a preliminary analysis on the research significance,current problems and the thoughts in the coming studies.Utilizing advanced knowledge and techniques of systems biology to study "the correlation between acupoints/meridians and internal organs" will greatly promote the development of acupuncturology.

摘要:The acupuncture manipulation is an important factor for achieving a good therapeutic effect in acupuncture practice.The authors review recent studies on acupuncture manipulation and its qualification from a) differences of the therapeutic effects of acupuncture manipulation,b) quantification of acupuncture manipulations,and c) instruments for delivering and sampling the analogued parameters of different acupuncture manipulations,computer analysis software,etc.Most studies revealed that different acupuncture manipulations had different clinical efficacies.Meanwhile,researches on the quantification of acupuncture manipulations and some related instruments also have made a bigger progress.Nevertheless,there still exist many problems,such as difficulties of various acupuncture techniques in unification in clinic,stronger personal subjectivity in performing manipulations,etc.We should overcome these problems and conduct more researches,so as to raise the clinical curative effect further.
