      摘要:Objective To observe the effect of electroacupuncture(EA) on the profiles of hippocampal whole-genome expression in depression rats so as to study its underlying mechanism in relieving depression.Methods Thirty male SD rats were randomized into normal,model and EA groups(n=10/group).Depression model was established by chronic unpredictable stress stimulation(forced ice-water swimming,electric shock,tail-clamping,etc.) and lonely raising for 21 days.EA(2 Hz,1 mA) was applied to "Baihui"(GV 20) and "Yintang"(EX-HN 3) for 15 min,once daily for 7 days.The rats' ethological activities(horizontal and vertical movements) were detected with Open-field test in Morris labyrinth,and 1% sucrose solution consumption was detected.After termination of the treatment,the rats were killed and their hippocampal tissues were taken out for analyzing the whole-genome expression with Roche-NimbleGen chips(Rattus norvegicus 12×135 Karray).Results Compared with normal group,the distance of horizontal movement,rearing times,and sucrose consumption at day 21 and day 28 of model group decreased considerably(P<0.01).In comparison with model group,the sucrose consumption at day 28 in EA group increased significantly(P<0.05).Results of gene expression microarray displayed that compared with normal group,of the down-regulated and up-regulated genes due to modeling,101 and 125 turned to normal in their expression,respectively following EA treatment.The down-regulated genes involve energy and substance metabolism,protein biosynthesis,etc.,and the up-regulated genes involve blood clotting,immunity,inflammation,apoptosis,etc.Conclusion EA of GV 20 and EX-HN 3 can normalize chronic stress stimulation induced down-and up-regulated expression of many genes in depression rats,which may contribute to its clinical effect in relieving depression.

      摘要:Objective To observe the effect of electroacupuncture(EA) of bilateral "Futu"(LI 18) on the immunoactivity of substance P(SP),NK-1 receptor(R),cyclooxygenase-1(COX-1),5-HT 1 AR and 5-HT 2 AR in the cervical spinal dorsal horn of rats with neck incision pain,so as to explore its underlying mechanism in relieving incision pain.Methods A total of 40 Wistar rats were randomly divided into control,model(incision pain),EA-Futu(LI 18),and EA-Zusanli(ST 36)-Yanglingquan(GB 34,EA-ST 36-GB 34) groups(n=10/ group).A 1.5 cm longitudinal incision was made along the middle of the neck in pentobarbital-anesthetized rats.Pain threshold(PT) was measured using radiant heat.EA(1-2 mA,2 Hz/15 Hz) was applied to bilateral LI 18,ST 36-GB 34 for 30 min.The expression of SP,NK-1 R,COX-1,5-HT 1 AR and 5-HT 2 AR in the spinal cord(C 1-C 4) tissue were detected with immunohistochemistry.Results After the operation,the thermal PT was decreased obviously in comparison with normal control group(P<0.05).Compared with pre-operation,the PT of EA-Futu(LI 18) group increased markedly after treatment(P<0.05),and no significant changes were found in PT in EA-ST 36-GB 34 group(P>0.05).The immunoactivity of SP,NK-1 R,COX-1 in the superficial dorsal horn in model group was increased significantly and that of 5-HT1 AR and 5-HT2 AR significantly and mildly,separately compared with control group(P<0.05).In comparison with model group,the immunoreactivity of SP,NK-1 R and COX-1 was decreased considerably,and that of 5-HT1 AR and 5-HT2 AR was upregulated remarkably in EA-LI 18 group(P<0.05).No significant changes were found in the immunoactivity of SP,NK-1 R,COX-1,5-HT1 AR and 5-HT2 AR in EA-ST 36-GB 34 group(P>0.05).Conclusion EA-Futu(LI 18) can significantly suppress pain reaction of neck incision in the rat,which is closely associated with its effects in downregulating the expression of SP,NK-1 R,COX-1 and upregulating that of 5-HT1 AR and 5-HT2 AR in the spinal dorsal horn.

      摘要:Objective To observe the activation effect of histamine on mast cells in the acupoint area and its analgesic effects similar to acupuncture stimulation,so as to reveal the underlying mechanism of acupoint injection therapy.Methods A total of 60 SD rats were divided into control,model,normal saline(NS),histamine-injection and acupuncture groups(12 rats/group).Acute adjuvant-induced-arthritis(AIA) model was duplicated by injection of complete Freunds' Adjuvant(0.05 mL) into the left ankle articular cavity.For rats of saline and histamine groups,normal saline(50 μL) and histamine(50 μL,100 μg/mL) were injected into the left "Zusanli"(ST 36) acupoint area.For rats of acupuncture group,manual acupuncture(lifting-thrusting and twisting needle intermittently) was applied to ST 36 for 20 min.Paw withdrawal(pain threshold,PT) was detected by radiant heat irradiation of the hindpaw.Skin and muscle tissues of ST 36 area were sampled to be fixed in formalin,then,sliced(5 μm),and stained with Toluidine Blue,separately,followed by counting the degranulation rate of mast cells under microscope.Results Compared with normal control group and pre-modeling,PT of model,NS,acupuncture and histamine groups decreased significantly(P<0.01),while in comparison with model group,PT of acupuncture and histamine groups increased considerably(P<0.01),suggesting an induction of analgesic effect after acupuncture and histamine injection.The degranulation rates of acupuncture and histamine groups were remarkably higher than those of model and NS groups(P<0.01),suggesting an activation of mast cells following regional acupuncture stimulation and histamine injection.Conclusion Both manual acupuncture and histamine injection of ST 36 can relieve pain in AIA rats,which may be closely related to its effect in activating mast cells in acupoint area.It suggests that the mast cells are a new target for acupoint injection therapy.

      摘要:Objective To observe the effect of electroacupuncture(EA) of "Neiguan"(PC 6) and "Taichong"(LR 3) on mechnical activities of the heart in acute myocardial ischemia(AMI) rabbits.Methods Fifty chinchilla rabbits were randomized into normal control,model,EA-PC 6,EA-LR 3 and non-acupoint groups(n=8/group).AMI model was duplicated by intravenous injection(femoral vein) of posterior pituitrin(3 U/kg).The maximal ascending velocity(+dp/dt max),maximal descending velocity(-dp/dt max) and left ventricular systolic pressure(LVSP) of the heart were recorded by using Biopac Bio-signal system.EA(1 mA,2 Hz) was applied to PC 6,LR 3 and non-acupoint for 10 min.Results Compared with normal control group,+dp/dt max,-dp/dt max and LVSP of the heart in model group reduced significantly(P<0.01);while in comparison with model group,left ventricular +dp/dt max and-dp/dt max at 10 min after EA in EA-PC 6 and EA-LR 3 groups increased markedly(P<0.01),and LVSP of EA-PC 6 group increased remarkably(P<0.05).No significant differences were found between EA-non-acupoint group and model group in the above-mentioned 3 indexes(P>0.05).Conclusion Both EA of PC 6 and LR 3 points can improve AMI induced reduction of the left ventricular myocardial mechanical activities in the rabbit,and the therapeutic effect of EA-PC 6 is superior to that of EA-LR 3.

      摘要:Objective To observe the effect of catgut embedding(CE) at "Shimen"(CV 5) on the endometrial receptivity,pinopodes expression of the endometrial surface and leukaemia inhibitory factor(LIF) immunoactivity during periimplantation period.Methods Thirty Kunming mice were randomly divided into control,CE of "Shimen"(CV 5,CE-CV 5) and medication groups(n=10/group).For mice of CE-CV 5 group,a piece of catgut was embedded into the subcutaneous tissue of "Shimen"(CV 5) on the day when the vagina-discharged epistom was found(pregnancy day 1,Pd 1).On Pd 4,mifepristone(0.1 mL,diluted in propylene glycol) was subcutaneously injected into CV 5 in rats of medication group.The expression of endometrial pinopodes(sampled on Pd 5) was observed by scanning electron microscopy.The immunoactivity of LIF of the endometrium was detected by immunohistochemistry.Results Compared with control group,the mean optical density(OD) values of LIF in CE-CV 5 and medication groups decreased significantly(P<0.05),suggesting an downregulation of the endometrial LIF expression.No significant difference was found between CE-CV 5 and medication group in LIF expression(P>0.05).Electronic microscopic observation showed 1) abundant mature pinopodes on the endometrial surface,with a large number of well-stacked ecphyma in control group;2) fewer pinopodes and well-arranged smooth epithelial cells on the endometrial surface in catgut embedding group;and 3) no pinopodes found in the endometrium of medication group.Conclusion Catgut embedding at "Shimen"(CV 5) can suppress the establishment of the mouse endometrial receptivity,which is possibly related to its effect in inhibiting the expression of endometrial pinopodes and LIF.

      摘要:Objective To observe the effect of electroacupuncture(EA) on contents of myocardial nitric oxide(NO),nitric oxide synthase(NOS),and intracellular Ca2+ levels in experimental myocardial ischemia-reperfusion injury(MI/RI) rats,so as to provide some evidence for acupuncture treatment of cardiovascular disorders and for the correlation between the acupoints and the internal organs.Methods Fifty male Wistar rats were randomly divided into sham-operation(sham),model,Neiguan(PC 6),Lieque(LU 7),and Hegu(LI 4) groups(n=10/group).MI/RI model was established by occlusion of the descending anterior branch of the left coronary artery for 40 min and reperfusion for 60 min.EA(30 Hz/100 Hz,2-4 mA) was applied to the bilateral PC 6,LU 7 and LI 4 for 20 min.The myocardial tissue of the ischemic area was taken,homogenated and centrifuged,separated for assaying NO and NOS activity by using spectrophotometer and their contents with nitrate reductase colorimetric assay.Another part of the myocardium tissue was processed to be cell suspension(added with Fluo-3/AM dye) for assaying intracellular freei concentration under the laser confocal microscope.Results Compared with sham group,myocardial NO and NOS contents of model group decreased mildly and significantly(P<0.05),while myocardial fluorescence intensity(content) of intracellular Ca2+ increased evidently(P<0.01).In comparison with model group,the contents of myocardial NO and NOS were upregulated considerably in PC 6 group(P<0.05),and intracellular Ca2+ content in PC 6 group reduced significantly(P<0.01).No significant differences were found between LU 7 and model groups and between LI 4 and model groups in the abovementioned 3 indexes(P>0.05).Conclusion EA of PC 6 can upregulate myocardial NO and NOS contents and downregulate myocardial intracellular Ca2+ level in MI/RI rats,which may contribute to its effect in relieving myocardial injury.Neiguan(PC 6) and the heart have a relatively specific correlation.

      摘要:Objective To observe the effect of electroacupuncture(EA) on the expression of glucose transporter-1(GLUT-1) of hippocampal microvascular endothelial cells in focal cerebral ischemia(CI) rats.Methods Sixty male Wistar rats were randomly divided into sham-operation,model and electroacupuncture(EA) groups.CI model was established by middle cerebral artery occlusion with a piece of thread embolus.EA(4 Hz/20 Hz,1 mA) was applied to bilateral "Neiguan"(PC 6) for 30 min,once daily for 15 days.GLUT-1 expression in the hippocampus was detected with immunohistochemistry and Western blot,respectively.Results After the treatment,compared with the sham-operation group,the expression of GLUT-1 in microvascular endothelial cells of the hippocampus in model group shown by both histochemical and Western blot techniques decreased apparently(P<0.05).While in comparison with model group,the expression of GLUT-1 in EA group was upregulated significantly(P<0.05).Conclusion EA of PC 6 can effectively upregulate GLUT-1 expression in the hippocampus in cerebral ischemia rats,which may contribute to its effect in improving CI by raising the glucose transportation of microvascular endothelial cells.

      摘要:Objective To observe the effect of electroacupuncture(EA) on heart rate(HR),arrhythmia duration,and plasma vasoactive intestinal peptide(VIP) level in ventricular tachycardia(VT) rats so as to study its underlying adjustment mechanism.Methods A total of 70 SD rats were randomly divided into normal control,model(VT),"Daling"(PC 7,EA-PC 7) and "Taiyuan"(LU 9,EA-LU 9) groups.VT model was duplicated by injection of CsCl(2 mol/L,1.6 mmoL/kg,femoral vein).EA(2 Hz/15 Hz,1 mA) was applied to "Daling"(PC 7) and "Taiyuan"(LU 9) for 5 min after inserting acupuncture needles.BL-410 Biofunctional System was used to record the ECG of the standard limb lead Ⅱ,and plasma VIP content assayed by using radioimmunoassay.Results Following intravenous injection of CsCI,HR increased significantly in model,EA-PC 7 and EA-LU 9 groups(P<0.05).Compared to model group,HR at 5 min,10 min and 15 min after administration of CsCI and the duration of arrhythmia in EA-PC 7 group decreased considerably(P<0.01).No significant differences were found between EA-LU 9 and model groups in HR and the duration of arrhythmia(P>0.05).Compared with normal group,the content of plasma VIP in model group decreased apparently(P<0.05),while in comparison with model group,plasma VIP level in EA-PC 7 group increased considerably(P<0.01).Conclusion Electroacupuncture of "Daling"(PC 7) can effectively suppress CsCI-induced tachycardia which may be closely associated with its effect in reducing plasma VIP level.

      摘要:Objective To observe the therapeutic effect of acupuncture combined with blood-letting therapy for knee osteoarthritis(KOA) and to analyze the synergism of the blood-letting therapy.Methods Sixty-three cases of KOA patients were randomized into a treatment group(n=31)and a control group(n=32).Patients of treatment group were treated with routine acupuncture at Ashi-points,Dubi(ST 35) and Neixiyan(EX-LE 4),Zusanli(ST 36),Yanglingquan(GB 34),etc.on the affec-ted side,3 times a week for 4 weeks,in combination with blood-letting(at the superficial veins around popliteal fossa) once a week for 4 weeks.Patients of control group were treated with routine acupuncture only.Western Ontario and McMaster Universities osteoarthritis index scores(WOMAC) and clinical therapeutic effects served as the objective indexes for evaluating the efficacy of blood-letting therapy.Results After the treatment,of the 32 and 31 KOA patients in control and treatment groups,4(12.50%) and 5(16.13) were under control in their symptoms,14(43.75%) and 17(54.84%) experienced marked improvement,10(31.25%) and 6(19.35%) had an improvement,and 4(12.50%) and 3(9.68%) were failed,with the total effective rates being 87.50% and 90.32% respectively.No significant difference was found between the two groups in the therapeutic effect(P>0.05).In comparison with pre-treatment,WOMAC scores,pain index,and physiological integral scores in treatment and control groups after the first and the last treatment decreased significantly(P<0.001),and their joint stiffness index of the last treatment also lowered considerably(P<0.001).The difference values of the integral scores between post-and pre-treatment in WOMAC,pain index,and physiological function of treatment group were remarkably higher than those of control group(P<0.05).Conclusion Acupuncture combined with blood-letting therapy can effectively improve KOA patients' physiological function and reduce joint pain especially in the early stage.

      摘要:Objective To observe the effect of electroacupuncture(EA) in combination with small dose of clozapine on clinical symptoms of refractory schizophrenia patients so as to evaluate its validity and security.Methods Eighty schizophrenia patients were randomized into medication group and EA+medication group(n=40/group).Patients of medication group were treated with conventional oral administration of clozapine(50-100 mg/d to 200-500 mg/d,for 8 weeks) and those of EA+ medication group treated by EA of Baihui(GV 20) and bilateral Taiyang(EX-HN 5) and oral administration of clozapine(50 mg/d to 100-150 mg/d).EA was given to the patients,3 times a week for 8 weeks.Scores of the Positive and Negative Symptom Scale(PANSS) and the Untoward Effect Symptom Scale(TESS) were used to assess the therapeutic effect.Results Following treatment,PANSS scores for positive symptoms and the common psychiatric symptoms of the medication and EA+medication groups all decreased evidently from the 2rd week on(P<0.05,P<0.01),and those of the negative symptoms reduced obviously from the 4th week on(P<0.05).Comparison between two groups showed no significant differences in PANSS scores for positive and negative symptoms and the common psychiatric symptoms after the treatment(P>0.05).But TESS score of EA+medication group was evidently lower than that of medication group(P<0.01),suggesting an apparently fewer untoward effects in EA+medication group.Of the two 40 cases in medication and EA+medication groups,6(15%) and 5(12.5%) were cured,8 and 8(20%) experienced marked improvement in their symptoms,17(42.5%) and 16(40%) had an improvement,9(22.5%) and 11(27.5%) failed,with the effective rates being 77.5% and 72.5% separately.Conclusion Both medication and EA+medication can improve refractory schizophrenia patients'clinical symptoms,but the later has fewer untoward effects.

      摘要:Objective To explore the feasibility of judgment of the lifting-thrusting acupuncture manipulation by using "quality control figure" measurement,so as to provide a possible reference for demonstration in acupuncture teaching and scienti-fic research.Methods A total of 13 high-professional title acupuncturists and teachers and 100 college students participated in the present study.The so-called "quality control figures" of lifting-thrusting acupuncture skill were established by collecting operation data from 13 acupuncturists and teachers with an "Acupuncture Manipulation Parameter Detector System".A total of 100 college students who have learnt acupuncture and massage learnings were asked to perform the same acupuncture manipulation after training,followed by giving a mark with conventional manipulation method and "quality control figure" method(scores),respectively.In addition,the correlation between these two scoring methods was analyzed.Results The average score of conventional scoring of lifting-thrusting manipulation was(81.03±8.40) points and that of "quality control figure" scoring was(80.60± 7.48) points in those college students after training.No significant difference was found between these two scoring methods(P>0.05).But a positive correlation was found between them(r =0.6943,P<0.01),suggesting a positive feasibility for judging the lifting-thrusting acupuncture manipulation with "quality control figure" measurement.Conclusion A positive correlation exists between the conventional scoring and "quality control figure" scoring for lifting-thrusting acupuncture manipulation,suggesting a feasibility in judging this acupuncture manipulation with "quality control figure" measurement.

      摘要:Channel-based syndrome(pattern) differentiation has played a leading role in acupuncture practice since the time of Huangdi Nei Jing(The Yellow Emperor's Inner Classic).In the past several decades,however,the clinical application of channel-based syndrome differentiation has begun to diminish,even presenting a substitutional trend by syndrome differentiation of Zangfu organs.The authors elaborate the importance of channel-based pattern differentiation in acupuncture clinic by analyzing its theory from Huangdi Nei Jing and by summing up the methods,clinical significance and application of the basic diagnostic methods-inspection,auscultation and olfaction,interrogation,pulse-feeling and palpation,and measurement of imbalance of the cutaneous electrical and thermal parameters of meridians and acupoints.

      摘要:Acupuncture combined with drugs has been widely used in the treatment of various clinical conditions and can produce a stronger or more beneficial curative effect than either separate acupuncture or chemical drug does.However,the mechanisms underlying the additive or synergetic effects of acupuncture plus drugs are not well understood so far.Based on careful review and analysis of the literature accumulated,the authors proposed several aspects possibly involving the additive or synergetic effects produced by acupuncture plus drugs.For instance,acupuncture stimulation at acupoints may influence the drug concentration in blood and/or in target organ and the bioavailability of the drug via modulating the drug absorption,distribution and metabolism in the body.In addition,the curative effect of drug may also be strengthened by acupuncture stimulation via altering the signaling pathways of the targeted specific receptors and via potentiating the resultant responsiveness or sensitivity of the cells in the target organ in responding to the drugs administered.The ideas mentioned above may raise the novel clues for further studying the mechanisms underlying acupuncture-produced increase of the curative effects of medicines.

    • Evaluation on Acupuncture Treatment of Primary Insomnia

      摘要:Acupuncture treatment of insomnia has been reported for many years,but its validity remains controversial up to now.In the present review,the authors took "Acupuncture","Insomnia" as the key words to do a literature search from databases in both Chinese and English via computer retrieval system and also conducted a hand-made retrieval from journals of Chinese medicine as Chinese Acu-moxibustion,and collections of theses of the related academic conferences.Then,they made a systemic evaluation on the original articles from methodology,diagnosis,criteria for the enlisted and excluded patients,intervention methods of acupuncture and control groups,standards for assessing the curative efficacy,follow-up visit,etc.Of the retrieved 80 clinical articles,20 are RCT(randomized controlled trails) composed of 1907 cases,including 4 high-quality trails and 16 lower-quality trails.Regarding the assessment of the therapeutic effect,measuring scales are often adopted in overseas studies,while in domestic researches,self-drawn standards are frequently used.In conclusion,there have had no high-quality clinical trails about acupuncture treatment of primary insomnia in China at the present,and the related evaluating methods could not definitely confirm the efficacy of acupuncture in relieving insomnia.Therefore,a strict and scientific clinical trail scheme being in line with evidence-based medicine is urgently needed in the coming studies on acupuncture treatment of primary insomnia.

      摘要:In the present paper,the authors review progresses of researches on the protective effect of acupuncture in resisting acute myocardial ischemia-reperfusion injury in patients and animals from 1) resisting injury of oxygen-derived free radicals,2) reducing intracellular Ca2+ overload,3) relieving dysfunction of vascular endothelial cells of the myocardium,4) regulating immune functional activity and reducing inflammatory injury,5) reducing cardiocyte apoptosis,6) upregulating the expression of heat shock protein(HSP 70) in the myocardium,and 7) regulating activities of multiple cardiovascular protection factors and intracellular signaling pathways.The abovementioned results are mainly obtained from experimental animal studies.For this reason,the authors hold that the coming researches on acupuncture therapy for cardiovascular disorders should pay more attention to its clinical application,for example,controlled blood-pressure decline induced lower perfusion of the coronary artery,cardiac surgery(percutaneous tranluminal coronary angioplasty,intracardiac operation underdirect vision) etc.
