摘要:Objective To observe the effect of electroacupuncture(EA) on the expression of ionotropic glutamate receptor(iGluR) subunits and their mRNAs in the lumbar segments of spinal cord in rats with neuropathic pain,so as to explore its underlying mechanism in relieving spinal hyperalgesia. Methods Thirty SD rats were randomly divided into control, model, and EA groups, with 10 rats in each. The spared nerve injury(SNI) model was established by ligature of the sural nerve after cutting off the common peroneal nerve and anterior tibial nerve.EA(2Hz,1mA) was applied to "Huantiao"(GB30) and "Weizhong"(BL40) for 30min,once daily for 7 days.Mechanical pain threshold was detected before and after modeling and before and after EA treatment. The expression levels of N-methyl-d-aspartic acid(NMDA) receptor subunits NR1 and NR2B,and AMPA receptor subunit GluR1 of iGluR and their genes were assayed by Western blot and reverse transcription polymerase chain reaction(RT-PCR) separately. Results In comparison with control group, the mechanical pain thresholds were decreased significantly on day 2,7 and day 14 following modeling in the model group(P<0.05,P<0.01).While compared with the model group, the pain threshold was increased considerably on day 14 in the EA group(P<0.01).Compared with the control group, the expression levels of lumbar spinal cord NR2B and NR2B mRNA in the model group were increased significantly(P<0.05),and those of lumbar spinal cord NR1 and NR1 mRNA,GluR1 and GluR1 mRNA in the model group increased slightly(P>0.05).In comparison with the model group, the expression levels of lumbar spinal cord NR2B and NR2B mRNA in the EA group were down-regulated remarkably(P<0.05),and those of lumbar spinal cord NR1 and NR1 mRNA,GluR1 and GluR1 mRNA in the EA group down-regulated slightly(P>0.05).Conclusion EA can significantly suppress pain reaction in rats with neuropathic pain proba-bly through down-regulating the expression of lumbar spinal cord NR2B protein and NR2B mRNA.
摘要:Objective To observe the effect of acupoint-embedement of medicated-thread and acupoint-injection of Chuanxiongzine on the expression of urokinase-type plasminogen activator(uPA) and plasminogen activator inhibitor-1(PAI-1) in the cerebral cortex in rats with cerebral ischemia-reperfusion injury(CI/RI),so as to explore its underlying mechanism in protecting the ischemic cerebral tissue. Methods Seventy-eight SD rats were randomly divided into normal control group(n=6),sham operation(sham) group(n=18),model group(n=18),acupoint injection(AI) group(n=18),and acupoint-thread-embedment(ATE) group(n=18).Rats of the latter 4 groups were randomized into 1d,3d and 5d subgroups, with 6 rats in each.CI/RI model was established by occlusion of the right middle cerebral artery(MCAO) for 30 min and reperfusion. For rats of the AI group,Chuanxiongzine(0.1mL/200g) was injected into "Baihui"(GV20) and "Dazhui"(GV14),and for those of ATE group,a piece of medicated thread containing collagen protein(extracted from the rat's tail tissue) and Chuanxiongzine + retarder was embedded into GV20 and GV14,respectively.The expression of uPA and PAI-1 in the cerebral cortex on the ischemia side was detected by immunohistochemistry. Results In comparison with the normal control group, the expression of uPA of the ischemia cerebral cortex on day 1,3 and day 5 in the model group was increased significantly(P<0.01),while the PAI-1 expression decreased remarkably in the model group(P<0.01).Compared with the 3 time-points of the model group, cortical uPA expression levels at the 3 time-points in the AI group and those of day 3 and day 5 in the ATE group were down-regulated significantly(P<0.01),whereas cortical PAI-1 expression levels at the 3 time-points in both AI and ATE groups up-regulated considerably(P<0.05,P<0.01).Comparison between AI and ATE groups showed that the expression levels of cortical uPA in the latter group on day 3 and day 5 were significantly lower than those of the former group(P<0.05),whereas the cortical PAI-1 expression levels in the latter group on day 3 and day 5 were evidently higher than those of the former group(P<0.05).But,cortical PAI-1 expression of the ATE group on day 1 was significantly lower than that of the AI group(P<0.05).No significant differences were found between the AI and ATE groups in the expression level of cortical uPA on day 1 and between normal and sham groups in both uPA and PAI-1 expression levels at the 3 time-points(P>0.05).Conclusion Both AI and ATE can down-regulate cortical uPA expression and up-regulate cortical PAI-1 expression in rats with CI/RI,which may contribute to their protective effect in reducing cerebral ischemic injury.

摘要:Objective To observe the influence of electroacupuncture(EA) of "Fengfu"(GV16) and "Taichong"(LR3) on morphological changes and apoptotic percentage of substantia nigra cells so as to explore its underlying mechanism in relieving Parkinson's disease. Methods Forty Wistar rats were randomly divided into normal control,sham-operation(sham),model and EA groups, with 10 rats in each. The Parkinson's disease model was established by micro-injection of 6-hydroxyl-dopamine(6-OHDA) into the right striatum.EA(2Hz,1mA) was applied to "Fengfu"(GV16) and "Taichong"(LR3) for 30 min, once daily for 2 weeks.HE staining,Nissl staining and immunohistochemical staining were conducted for observing the morphological changes of substantia nigra pars compacta(SNc) and striatum, and the tyrosine hydroxylase(TH) immuno-reaction(IR) positive neurons and nerve fibers. The apoptosis was detected by flow cytometry. Results Compared to the normal control and sham groups, the total number of TH-IR positive neurons and Nissl-stained cells, and OD value of TH-IR positive nerve fibers in the SNc on the 6-OHDA-lesioned side were significantly lower in the model group(P<0.01).Compared with the model group, the aforementioned 3 indexes of the EA group were increased obviously(P<0.01).The percentage of apoptosis of SNc on the 6-OHDA-lesioned side was obviously lower in the EA group than in the model group(P<0.01),but still obviously higher than in the normal group and sham group(P<0.01).Conclusion Electroacupuncture therapy can significantly increase the number of neurons of substantia nigra and the density of striatum nerve fibers, and reduce the apoptotic percentage of substantia nigra cells in the Parkinsonian rats, which may contribute to its effect in relieving Parkinson's disease.
摘要:Objective To explore the relationship between the effect of electroacupuncture(EA) and EA of different layer tissues of the acupoint area and different acupoints in upregulating mean arterial pressure(MAP) and heart rate(HR) in hypotension plus bradycardia rats. Methods A total of 200 SD rats were used in the present study.Bradycardia plus hypotension model was established by intravenous injection of 0.4% propranolol(0.4mg/100g,maintaining dosage 0.025mg/100g per minute).EA(2Hz /15Hz,5mA) was applied to 1) right "Daling"(PC7) and "Jiexi"(ST41),"Ximen"(PC4) and "Housanli"(ST36),"Quze"(PC3) and "Dubi"(ST35) which have a similar tissue structure, and are located in the upper and lower limbs and different meridians, and non-acupoint [3 mm left-superior to the "Tianshu"(ST25)],2) skin, muscle layer and periosteum part of "Ximen"(PC4),3) skin, muscle layer and periosteum of "Housanli"(ST36) for 15min.The HR and MAP were recorded by using a multi-channel physiological signal sampling-processing system. Results 1)In comparison with the model group, the percentages of the increased HR and MAP in the "Ximen"(PC4),"Quze"(PC3),"Housanli"(ST36) and "Jiexi"(ST41) groups,PC4-skin,PC4-muscle,PC4-periosteum,ST36-skin,ST36-muscle and ST36 periosteum groups, and the increased HR in the "Dubi"(ST35) group were upregulated significantly(P<0.05,P<0.01).The percentages of the increased HR and MAP were significantly higher in the "Quze"(PC3) and "Ximen"(PC4) groups than in the "Daling"(PC7) group(P<0.01),and the increased HR evidently higher in the "Housanli"(ST36) and "Jiexi"(ST41) groups than in the "Dubi"(ST35) group(P<0.01),suggesting different effects of EA stimulation of different acupoints in the same one meridian. No significant differences were found among the "Ximen"(PC4),"Quze"(PC3),"Housanli"(ST36) and "Jiexi"(ST41) groups, and between the "Daling"(PC7) and model groups, and between the non-acupoint and model groups in the rising rates of both HR and MAP(P>0.05).2) Regarding the effects of EA of different tissue layers in "Ximen"(PC4) and "Housanli"(ST36) areas, the rising rates of HR were markedly higher in the PC4-skin group than in the "Ximen"(PC4),PC4-muscle and PC4-periosteum groups(P<0.01),and considerably higher in the "Housanli"(ST36),ST36-skin,and ST36-muscle groups than in ST36-periosteum group(P<0.05).The rising rates of MAP were significantly higher in the PC4-skin and PC 4-muscle groups than in the "Ximen"(PC4) and PC 4-periosteum groups(P<0.01),and considerably higher in the ST36-skin group than in the "Housanli"(ST36),ST36-muscle and ST36-periosteum groups(P<0.01),suggesting different effects of EA stimulation of different tissue layers in the same one acupoint. No significant differences were found between the "Ximen"(PC4) and PC 4-muscle groups, among the "Housanli"(ST36),ST36-skin and ST36-muscle groups in the rising rates of HR,between the "Ximen"(PC4) and PC4-perio-steum groups, and among the "Housanli"(ST36),ST36-muscle and ST36-periosteum groups in the rising rates of MAP(P>0.05).3) The effect of the PC4-skin group was significantly superior to that of the ST36-skin group in increasing HR(P<0.01),and the effect of the PC4-muscle group was obviously stronger than that of the ST 36-muscle group in raising MAP(P<0.01),suggesting different therapeutic effects of EA stimulation of the similar tissue in different meridian-acupoints.The effects of the "Ximen"(PC4) and "Housanli"(ST36) groups in raising both HR and MAP,and that of the PC4-muscle and ST36-muscle groups in upregulaing HR,and that of the PC4-periosteum and ST36-periosteum groups in raising MAP were comparable(P>0.05).Conclusion EA of different acupoints of the same one meridian, the similar structure of different meridian acupoints and different tissue layers of the same one acupoint have their own relatively specific effects in upregulating HR and MAP in hypotension plus bradycardia rats.
