摘要:Objective To observe the cardioprotective effect of electroacupuncture(EA)stimulation at"Neiguan"(PC 6)in mice with myocardial ischemia(MI)and to explore the involvement of cardiac beta-adrenergic receptors(β-AR)in the cardioprotective effect of EA intervention. Methods Adult male C 57BL/6mice of wild type andβ1/β2-AR double-knockout(β1/β2-AR-/-)mice were randomly and respectively divided into control group and EA group, with 8mice in each group. These mice were subject to procedures of basic control, fatigue swimming and fatigue swimming+EA.The model was established by fatigue swimming for inducing acute MI.EA(2Hz,0.5mA)was applied to bilateral"Neiguan"(PC 6)for 30min,once daily for 7 days in both EA groups. The ST-segment of electrocardiogram(ECG)of standard limb leadⅡ,heart rate were recorded by using a biophysical amplifier and the arrhythmia score was assessed according to Curtis and Walker's methods(1988).Results Selfcomparison showed that, compared with the baseline, the amplitude of ECG-STⅡsegment was obviously increased in swimming fatigue C 57BL/6mice(P<0.01)and further obviously increased inβ1/β2-AR-/-mice(P<0.01),suggesting an acute MI in C 57BL/6mice and a worsened MI inβ1/β2-AR-/-mice.Simultaneously,the heart rate was markedly decreased(P<0.05), and the score of arrhythmia obviously increased in both C 57BL/6andβ1/β2-AR-/-mice(P<0.05).Compared with the modeling procedures,ECG-STⅡ amplitude and arrhythmia score were significantly decreased in C 57BL/6 mice of the EA group(P<0.01,P<0.05),rather than in theβ1/β2-AR-/-mice of the EA group(P>0.05).In addition, heart rate levels of both C 57BL/6mice andβ1/β2-AR-/-mice had no significant differences between control and EA groups(P>0.05).ConclusionEA at"Neiguan"(PC 6)can protect the heart from swimming fatigue-induced MI in C 57BL/6mice not inβ1/β2-AR-/-mice,suggesting an involvement ofβ1/β2-AR in the protective effect of EA.
摘要:Objective To observe the effect of deep electroacupuncture(EA)stimulation of"Huantiao"(GB 30)on the functional and pathological changes and nerve growth factor(NGF)expression of the damaged sciatic nerve in rats,so as to study its mechanisms underlying reliving sciatica. Methods Forty-eight SD rats were randomly divided into normal, model, deep EA and shallow EA groups(n=12in each group). The sciatic nerve injury model was established by mechanical clamp of the sciatic nerve stem. For deep and shallow EA,the acupuncture needles were inserted into GB 30about 16mm and 7mm,respectively. The EA treatment was given 20min,once daily for 14days.The evoked potentials of the injured sciatic nerve stem responding to electrical stimulation were recorded by using a biophysiological experimental system for calculating the motor conduction velocity. Pathological changes of the sciatic nerve were displayed by H.E.stain.The expression of NGF and Fos proteins was detected by immunohistochemistry. Results In comparison with the normal group, the conduction velocity and the amplitude of the evoked potentials of the sciatic nerve were significantly decreased in the model group(P<0.05).Following EA,both conduction velocity and amplitude of the evoked potentials in the deep EA group, and the conduction velocity in the shallow EA group were considerably increased(P<0.05).The therapeutic effects were significantly better in the deep EA group than that in the shallow EA group(P<0.05).No significant differences were found between the shallow EA and model groups in the amplitude of evoked potentials(P>0.05),and no significant changes of latencies of the evoked potentials in the four groups(P>0.05).In the model group, the disorganized nerve fibers axons, myelin and Schwann cells of the damaged sciatic nerve were found, which became milder in the EA groups particularly in the deep EA group. In regard to the NGF and Fos immunoactivity of the injured sciatic nerve, the expression levels of both NGF and Fos proteins were obviously higher in the model group than in the normal group(P<0.05).After EA stimulation,NGF expression was further significantly up-regulated in both deep and shallow EA groups(P<0.05),and Fos protein expression was notably down-regulated in the deep and shallow EA groups(P<0.05).The expression of NGF was significantly higher and Fos protein was obviously lower in the deep EA group than in the shallow EA group(P<0.05).ConclusionDeep EA stimulation of GB 30can improve the pathological changes and function of the injured sciatic nerve in the rat, which is closely associated with its effects in up-regulating NGF expression and down-regulating Fos expression. The deep EA is relatively better in the therapeutic effect. These facts may be one of the mechanisms of EA in relieving sciatica in clinic practice.

摘要:Objective To observe the effect of acupotomy, electroacupuncture(EA)or round-sharp acupuncture needle intervention on the expression of Bcl-2,Bax and Caspase-3proteins in the rectus femoris in rabbits with knee ostarthritis(KOA), so as to explore their mechanisms underlying improvement of braking-induced joint damage from the cellular apoptosis. Methods Forty-five New Zealand rabbits were equally and randomized into control group, model group, acupotomy(AP)group,EA group and round-sharp acupuncture needle(RSAN)group(n=9in each group). The knee-joint injury model was established by fixing the left knee joint in extention position with plaster bandage.EA(2Hz/100Hz,3mA,20min each time)was applied to the left"Yanglingquan"(GB 34)-"Yinlingquan"(SP 9)and left"Neixiyan"(EX-LE 4)-"Waixiyan"(ST 35)for rabbits in the EA group. The EA treatment was given once daily,3times a week,3weeks in total. For rabbits of the AP group,a needle-knife was held to insert into the front edge of the midpoint, the starting point and the stopping point of the left medial collateral ligamen, lateral collateral ligament and the patellar ligament of the knee to make a loosening manipulation for 5times in a session of treatment, once a week,3times altogether. For rabbits of the RSAN group,a round-sharp needle was performed in the same way to the needle-knife including the stimulation point, the manipulation method and treatment sessions. At the end of the experiment, the left rectus femoris was taken out for detecting the expression of Bcl-2,Bax and Caspase-3proteins with Western blot.ResultsIn comparison with the control group, the passive range of motion(PROM)level was significantly decreased 4,8and 12weeks after modeling(P<0.01),and the expression levels of Bax and Caspase-3proteins in the rectus femoris were considerably upregulated in the model group(P<0.05),while the ratio of Bcl-2/Bax was notably down-regulated(P<0.05)in the model group.Compared with the model group, the PROM level at week 12after modeling in the AP,EA and RSAN groups were significantly increased(P<0.01);while Bax and Caspase-3expression levels in both AP and RSAN groups were considerably downregulated(P<0.05).No significant differences were found among the five groups in Bcl-2expression levels(P>0.05),and between the EA and model groups in Bax and Caspase-3expression levels and the ratio of Bcl-2/Bax(P>0.05).ConclusionAP,RSAN and EA interventions are effective in improving the knee-joint motion range in KOA rabbits, and this effect of both AP and RSAN is closely associated with their actions in lowering the expression of Bax and Caspase-3proteins of the rectus femoris and in raising ratio of Bcl-2/Bax protein(reducing muscular cellular apoptosis). The mechanism of EA intervention in improving PROM may be different.
摘要:Objective To observe the effect of different intensities of manual acupuncture(MA)stimulation on mechanical pain thresholds(PTs)and the expression of phosphorylated extracellular signal-regulated kinases(p-ERK)in lumbar spinal dorsal horn regions in rats with neuropathic mirror-image pain,so as to explore its mechanisms underlying analgesia.MethodsForty male SD rats were equally and randomly divided into control, spinal nerve ligation(SNL)model, mild MA-stimulation,and strong MA-stimulation groups.Neuropathological pain model was established by ligature of the spinal nerve(L 5).Three days after the SNL,bilateral"Huantiao"(GB 30)were stimulated by rotating the thin(0.22mm×13mm)or thick(0.3mm×13mm)filiform needles at a frequencies of 60times/min or 180times/min and at an angle of 180°or 360°for 2min for rats in the mild and strong MA-stimulation groups, respectively, followed by remaining the needle in place for 30min.The mechanical PTs were measured before and after SNL.The expression of p-ERK protein in bilateral dorsal horn regions of the lumbar spinal cord(L 4-L 6)was detected by Western blot. Results In comparison with the control group, the mechanical PTs were significantly decreased beginning from the 3rd day on after SNL on the affected side and from the 7th day on after SNL on the healthy hindpaw(P<0.05),simultaneously,p-ERK protein expression levels of dorsal horn regions on both sides of the spinal cord were considerably up-regulated on the 12th day(P<0.05).Compared with the model group, the PTs of the affected hindpaw and the healthy hindpaw were significantly increased on the 7th and 12th day in the strong MA-stimulation group(P<0.05,P<0.01),whereas pERK expression levels in the bilateral spinal dorsal horn regions were obviously down-regulated in the strong MA-stimulation group(P<0.05).No significant differences were found between the model and mild MA-stimulation groups in the PTs of bilateral hindpaws and p-ERK expression levels of the bilateral spinal dorsal horn regions(P>0.05)except the PTs of the healthy hindpaw on 7th day(P<0.05).Conclusion Strong MA-stimulation can alleviate neuropathic mirror-image pain in SNL rats, which is closely related to its effect in down-regulating the expression of p-ERK in the bilateral spinal dorsal horn regions.
摘要:Objective To investigate the chemical expression of the primary sensory and motor neurons associated with "Hegu"(LI 4)area by using neural tracing and fluorescent immunohistochemical techniques. Methods A total of 4SD rats were used in the present study. After anesthesia, the rats received microinjection of 5μL of Alexa Fluor 594conjugated cholera toxin subunit B(AF 594-CTB)into the right"Hegu"(LI 4)area for observing the distribution of the related primary sensory neurons in dorsal root ganglia(DRGs)and motor neurons in the spinal cord(C 1-C 5).Three days after the microinjection, the rats were anesthetized and transcardiacally perfused with 4% paraformaldehyde, followed by routine section of the DRGs and spinal cord tissues. The labeled neurons were observed by light microscope and their chemical characteristics determined by calcitonin generelated peptide(CGRP)fluorescent immunohistochemical method. Results All the AF 594-CTB labeled neurons in DRGs and spinal cord were located on the side of the tracer injection. The labeled sensory neurons distributed in DRGs of cervical(C)5to thoracic(T)1segments with a high concentration in C 7DRG in which 73.5%sensory neurons were shown to be CGRP-positive. The labeled motor neurons were found to be in the dorsolateral region of the spinal anterior horns from C 6to T 1segments,with a high concentration at C 8in which 100% motor neurons were shown to be CGRP-positive.Conclusion Acupoint LI 4is innervated by sensory neurons from DRGs of C 5-T 1and motor neurons from anterior horns of the same segmental spinal cord in rats. Among them,73.5% of the sensory neurons and 100% of the motor neurons are CGRP-positive,suggesting an involvement of CGRP in acupuncture stimulation-LI 4induced therapeutic effects.
摘要:Objective To observe the effect of acupuncture stimulation of"Tianshu"(ST 25)on electrical and mechanical activities of jejunum smooth muscle at different phases of interdigestive migrating motor complex(MMC)in normal rats,so as to explore its mechanism underlying regulation of intestinal function. Methods A total of 16adult SD rats were used in the present study. After 18-24hours' fasting(with free access to water),the rats were subjected into experimental observation.Electromyogram and intestinal mechanical motor were respectively recorded by using implanted metal electrodes and strain gauge(sutured to the serosal surface of the jejunum)and biophysiological amplifier system.Manual acupuncture stimulation was applied to ST 25for about 1min at phaseⅠ,Ⅱ andⅢ of MMC,respectively. Results Following manual acupuncture stimulation of ST 25,the mean frequencies and amplitudes of both fast and slow waves of electrical activities and muscular contraction tension of the jejunum smooth muscle at phaseⅡandⅢof MMC were significantly decreased compared to their baseline levels(P<0.01).No significant changes were found in the mean frequency and amplitude of fast wave of electrical activities of the jejunum smooth muscle at phaseⅠ of MMC after acupuncture stimulation of ST 25(P>0.05).Conclusion Acupuncture stimulation of ST 25can inhibit the jejunum electrical and mechanical activities at phaseⅡandⅢof MMC in normal rats, probably via somato-sympathetic reflex.

摘要:Objective To observe the effect of electroacupuncture(EA)stimulation of"Ganshu"(BL18)on expression of substance P(SP)in the gastric mucosa and hypothalamus tissues and hippocampal 5-hydroxytryptamine(5-HT)content in rats with depression+gastric ulcer,so as to explore its mechanism underlying improvement of depressive gastric ulcer.MethodsA total of 60SD rats were equally randomized into normal,model,EA-BL18and non-acupoint groups. The depression+gastric ulcer model was established by chronic unpredictable mild stress(CUMS)paradigm for 21days in accordance with Willner and colleagues' methods(1987),and injection of 90% glacial acetic acid(0.01mL)into the sub-serous layer near the gastric antrum under anesthesia on day 10after starting CUMS.Following modeling,EA(4Hz/15Hz,2V)was applied to bilateral"Ganshu"(BL 18)for 20min,once daily for 2weeks,with one day's interval between two weeks. The non-acupoint was located about 2.5 cm lateral to the umbilicus and stimulated by EA similar to EA-BL18.The rats' locomotor ability was assessed by open field tests(crossing and rearing numbers). The gastric ulcer index was calculated according to Guth's method, and hypothalamic and gastric antrum SP immunoactivity was determined by immunohistochemistry and hippocampal 5-HT content was detected by ELISA. Results After modeling, compared to the normal group, both crossing number and rearing number of open field tests on day 21 and 34were significantly decreased in the model group(P<0.01),while compared to the model group, both crossing and rearing numbers on day 34were considerably increased in the EA-BL18group(P<0.01)rather than in the non-acupoint group(P>0.05).Following modeling, the gastric ulcer index,SP expression levels in the gastric antrum and hypothalamus tissue were obviously increased in the model group(P<0.01),and notably decreased in the EA-BL18group(P<0.01)but not in the nonacupoint group(P>0.05).The hippocampal 5-HT content was remarkably lower in the model group than in the normal group, but obviously higher in the EA-BL18group than in the model group(P<0.01).No significant difference was found between the model and non-acupoint groups in the hippocampal 5-HT content(P>0.05).Conclusion EA of"Ganshu"(BL18)can promote locomotor and reduce gastric ulcer index in depression+gastric ulcer rats, which may be related to its effects in lowering gastric and hypothalamic SP immunoactivity and in raising hippocampal 5-HT content.
摘要:Objective To observe the effect of electroacupuncture(EA)stimulation of"Mingmen"(GV 4)on the bone morphogenetic protein-2(BMP-2)content and biomechanics in osteoporosis rats so as to explore its mechanism underlying improvement of osteoporosis. Methods Fifty female SD rats were randomized into sham operation(sham),model,EA-GV 4, EA-non-acupoint(non-acupoint)and estrogen(medication)groups, with 10rats in each group.Postmenopausal osteoporosis model was established by removing the rats' bilateral ovaries under anesthesia.EA(2Hz/15Hz,1.0mA)was applied to"Mingmen"(GV 4)or non-acupoint for 20min,once daily for 30times,with one day's interval between every two 10times.Rats of the medication group were lavaged with Pentanoic Acid Estradiol(25μg/mL,2mL/500g),once every day(the dosage of estradiol was adjusted according to their body weight)continuously for 1month.Rats of the model and sham groups experienced the fixing and fastening procedures as the other rats in the EA and medication groups. After intervention, the BMP-2expression level of the femoral bone tissue, and bone biomechanical values were determined by immunohistochemistry and three-point bending tests, respectively. Results ①In comparison with the sham operation group, the femoral biochemical maximum load and fracture load values were significantly decreased in the model group(P<0.05).While compared with the model group, the biochemical maximum load and fracture load values were obviously increased in the EA-GV 4and medication groups(P<0.05),but not in the non-acupoint group(P>0.05).② Compared with the sham group, the femoral BMP-2expression of model group was significantly decreased(P<0.05),compared with the model group, the expression of BMP-2of GV 4and medication groups significantly increased(P<0.05).Conclusion EA-GV 4intervention can improve bone biomechanical changes in osteoporosis rats.
摘要:Objective To observe the effect of electroacupuncture(EA)stimulation of"Baihui"(GV 20)-"Yintang"(EX-HN 3)on changes of learning-memory ability and hippocampal neuron structure in chronic stress-stimulation induced depression rats. Methods Forty-eight SD rats were randomly divided into normal, model,EA and medication(Fluoxetine)groups, with 12rats in each group. The depression model was established by chronic unpredictable mild stress stimulation(swimming in 4℃ water, fasting, water deprivation, reversed day and night, etc).Treatment was applied to"Baihui"(GV 20)and"Yintang"(EX-HN 3)for 20min,once every day for 21days.For rats of the medication group,Fluoxetine(3.3mg/kg)was given by gavage(p.o.),once daily for 21days.The learning-memory ability was detected by Morris water maze tests. The pathological and ultrastructural changes of the hippocampal tissue and neurons were assessed by H.E.staining,light microscope and transmission electron microscopy, respectively. Results Compared to the normal group, the rats' body weight on day 14and day 21after modeling was significantly decreased in the model group(P<0.01),the escape latency and total swimming distance in the 4 quadrants on day 10and 21were significantly increased in the model group(P<0.01).In comparison with the model group, the body weight on day 14and 21were significantly increased(P<0.05),and the escape latency and total swimming distance levels were significantly decreased in the EA group(P<0.01),suggesting an improvement of learning-memory ability.Observations of light microscope and transmission electron microscope showed that modeling induced pathological changes such as reduction in hippocampal cell layers, vague and broken cellular membrane, and ultrastructural changes of hippocampal neurons including swelling and reduction of mitochondria and mitochondrial crests were relived after EA and Fluoxetine treatment. Conclusion EA intervention can improve the learning-memory ability and relieving impairment of hippocampal neurons in depression rats, which may be one of its mechanisms underlying bettering depression.
摘要:Objective To observe the effect of electrocupuncture(EA)intervention at different time-points of post-modeling on behavior and hippocampal monoamine neurotransmitter noradrenalin(NE),dopamine(DA)and 5-hydroxytrypamine(5-HT)contents in vascular dementia(VD)mice,so as to study its mechanism underlying improvement of VD.Methods A total of 60Kunming mice were randomized into sham-operation control(n=20),VD model(n=20),EA-day(D)-1(EA treatment was given from the 1st day on after modeling,n=10),EA-D-3(EA was given from the 3rd day on after modeling,n=10)groups. VD model was established by occlusion of the bilateral cervical common arteries and reperfusion.EA(2Hz/80Hz)was applied to "Baihui"(GV 20),"Dazhui"(GV 14),"Zusanli"(ST 36)and"Geshu"(BL 17)for 10min,once daily for 15days.Hippocampal NE,DA and 5-HT contents were assayed by fluorospectrophotometry. The mouse's learning-memory ability was assessed by step-down tests. Results In comparison with the sham-operation control group, the learning-memory ability(marked increase of reaction time and error times, decrease of step-down latency)was apparently lowered in the model group(P<0.01).The hippocampal NE,DA and 5-HT contents were significantly lower in the model group than in the sham-operation group(P<0.01). Compared with the model group, the mice's learning-memory ability(marked decrease of reaction time and error times, increase of step-down latency)was significantly increased in EA intervention groups(P<0.01),and hippocampal NE,DA and 5-HT levels were significantly increased(P<0.01),and the effect of EA-D-3group was obviously better than that of the EA-D-1group(P<0.05,P<0.01).Conclusion EA can improve the VD mice's learning-memory ability, which is closely related to its effects in up-regulating hippocampal NE,DA and 5-HT contents, and the effect of later EA intervention after modeling is better.
摘要:Acupoint compatibility is not only a foundation for formulating acupoint recipes in clinical practice, but also a core of meridian and acupoint theory. Many studies have revealed that the application of multiple-acupoints combination in the treatment of different clinical conditions or illnesses may lead to three outcomes,i.e.,synergistic effect, antagonistic effect and no effect. Therefore, the application of multiple acupoints combination is definitely not a simple result of 1+1=2,or 2-1=1in the management of many clinical disorders, and rather, the reasonable combination of multiple acupoints is a non-liner,dynamic and complicated issue. It has been well documented that the complex biological network consists of modules/communities,a set of nodes that are more densely inter-connected among themselves than with the rest of the network, which can be detected by community detection according to their modularity. After analyzing the current status and development trends of acupoint comparability researches in both China and other countries of the world, the authors of the present paper put forward some ideas and methods for the research of correlation, composing characteristics and dynamically evolutionary regularities of acupoint combination from the macroscopic and local aspects, dynamic and static angles on the basis of the theory of community structure theory of complex networks.
摘要:The old theory of midnight-noon ebb-flow for selection of acupoints in acupuncture treatment of clinical disorders is mechanical in the calculation of the time(hours and days)of flowing and ebbing of qi and blood in terms of the heavenly stems and earthly branches, thus,it has been called considerable questions by many Chinese physicians in different historical periods. The author of the present paper holds that the time calculation is merely a method, only the symptom-differentiation and innovation ideas are the key points for the acupuncture treatment.Following summarizing up the key points of this acupuncture management, the present paper thinks that on-time and time-point selection,acupoint-open and acupoint-closing,academic schools and clinical choice-making are critical for clinical practice. It denotes that despite the midnight-noon ebb-flow acupuncture management has its special calculation method, but the calculation always differs from person to person, from time to time, and from place to place, and the best treatment is conducted according to syndrome-differentiation.Thus,when applying the midnight-noon ebb-flow acupuncture method,we should not adhere to mechanical calculation, and rather, thoroughly understand the conventional management regularities and make corresponding changes in the treatment of different clinical conditions,i.e.,inherit the predecessors' excellent academic thoughts on one hand, and conduct innovation research and application on the basis of current natural and social environments on the other hand.

摘要:Up to now,in the treatment of stroke patients by acupuncture therapy, three main representative achievements involving scalp acupuncture intervention,"Xing Nao Kai Qiao"(restoring consciousness and inducing resuscitation)acupuncture technique and nape acupuncture therapy have been got.Regarding their neurobiological mechanisms, the scalp acupuncture therapy is based on the functional localization of the cerebral cortex,"Xing Nao Kai Qiao"acupuncture therapy is closely related to nerve stem stimulation, and the nape acupuncture therapy is based on the nerve innervation of the regional neck-nape area in obtaining therapeutic effects. In fact, effects of these three acupuncture interventions are all closely associated with the modern neuroanatomy. In the treatment of post-stroke spastic paralysis, cognitive disorder and depression with acupuncture therapy, modern neuroanatomical knowledge should be one of the key theoretical basis and new therapeutic techniques should be explored and developed continuously.
摘要:Sports fatigue belongs to the category of functional deficiency-syndrome according to the theory of traditional Chinese medicine. The moxibustion therapy has a long history and possesses a definite therapeutic effect in the prevention and treatment of sports fatigue. In the present paper, the authors reviewed development of researches on the effects of moxibustion intervention in the prevention and treatment of sports fatigue in recent 5years.Results of researches showed that moxibustion intervention can 1)eliminate free radicals and reduce oxidative damage;2)increase energy(glycogen)supply to delay the production of fatigue;3)raise serum testosterone level(relieve exercise-induced neuroendocrine disorder)and reduce post-sports fatigue; 4)raise the anaerobic exercise ability, reduce the accumulation of metabolic products in the body and strengthen the endurance capacity of the skeletal muscle;and 5)improve ischemic cardiac function, and suppress cardiomyocyte apopotosis, etc. However,we should further strengthen our investigations on the moxibustion therapy in the ancient classical literature and sum up academic thoughts of different academic schools in the successive dynasties, put emphasis on the large sample randomized controlled clinical trails, establish united treatment standards, etc.,and provide much evidence for effectively treating sports fatigue in the future.
