摘要:Objective To observe the effect of electroacupuncture(EA)on expression of pain sensory and affective processing-relatedμ-opioid receptor(MOR),glutamatergic AMPA receptor subunit GluA 1,extracellular signal-regulated kinase(ERK 1/2),cAMP response element binding protein(CREB)in the amygdala in chronic constrictive injury(CCI)+ negative affection(NA)rats,so as to reveal its mechanism underlying pain relief.Methods A total of 32 male Wistar rats were randomized into normal control,model,EA,and anesthesia+EA(AEA)groups(n=8in each group).The neuropathic pain NA model was established by ligation of the left sciatic nerve and repeated electrical stimulation of the paw-bottom in the pain-paired compartment.EA was applied to bilateral"Zusanli"(ST 36)and"Yanglingquan"(GB 34)for 30 min,once daily for 7days.Thermal pain threshold(paw withdrawl latency,PWL)of the bilateral paws was measured by using a Tail-Flick Unit.The expression levels of MOR and p-CREB in the central amygdala(CeA)and those of MOR,GluA 1,p-ERK 1/2and p-CREB in the right amygdala area were determined using immunofluorescence and Western blot,respectively.Results In comparison with the normal group,PWL difference(PWLD)values of the model group were significantly increased(P<0.001),and the time spent in the CPA-paired compartment was considerably decreased(P<0.001).After EA,the PWLD levels of both EA and AEA groups were apparently decreased(P<0.05),showing apain relief;and the time spent in the CPA-paired compartment was apparently increased in the EA group(P<0.05),rather than in the AEA group(P>0.05).Additionally,compared to the normal group,the expression level of MOR protein in the amygdala was remarkably increased(P<0.05)and those of GluA 1,p-ERK 2and p-CREB proteins were apparently decreased(P<0.05).After EA intervention for 7days,the expression levels of these four proteins in the EA group,and those of MOR,p-ERK 2and p-CREB in the AEA group were significantly up-regulated(P<0.001,P<0.01,P<0.05).The expressionlevel of GluA 1was significantly higher in the EA group than in the AEA group(P<0.05).Conclusion Repeated EA stimulation of ST 36-GB 34 has a definite effect in relieving both sensation and affection dimensions of pain in NA rats,which may be related to its effect in up-regulating the expression of GluA 1in the amygdala,but the effects of MOR,p-ERK 2and p-CREB need being researched further.
摘要:Objective To observe the effect of transcutaneuos acupoint electrostimulation(TAES)on ovarian serum sex hormone levels and ovarian follicle granular cell aromatase cytochrome P 450(P 450arom)protein and follicle theca cell cytochrome P 450 17α-hydroxylase/c 17-20 lyase cytochrome P 450(P 450c17α)protein expression in polycystic ovary syndrome(PCOS)rats,so as to explore its mechanisms underlying improvement of PCOS.Methods Forty SD rats were randomly divided into four groups:normal control,model,medication and TAES(10rats/group).The PCOS model was established by giving(gavage)the animals with letrozole solution(1.0mg/kg,once daily for 21 consecutive days).Rats of the medication group were treated with Clomiphene(1mg/kg)once daily for 7days,and those of the TAES group were treated with electrical stimulation(2Hz,3mA)of"Guanyuan"(CV 4)and"Sanyinjiao"(SP 6)areas for 30 min,once daily for 7consecutive days.The rats body weight and bilateral ovarian weight were detected,and the ovarian structure and follicular development degree were observed under light microscope after H.E.stain,and the serum testosterone(T),estradiol(E2),luteotrophic hormone(LH)and folliclestimulating hormone(FSH)contents were detected using radioimmunoassay.The expression of ovarian P 450arom(for production of estrogen)protein and P 450c17α(for production of androgen)protein was detected by using immunohistochemical stain and Western blot,respectively.Results The body weight,bilateral ovary weight,serum T and LH contents,and ratio of LH/FSH,and ovarian P 450c17αimmunoactivity and protein expression levels in the model group were all significantly increased compared with the normal control group(P<0.01),and the levels of serum E2 and ovarian P 450 arom immunoactivity and protein expression were significantly decreased after modeling(P<0.01).Following the treatment,the increased body weight,ovary weight,serum T and LH contents,ratio of LH/FSH,and ovarian P 450c17αimmunoactivity and protein expression levels,and the decreased ovarian P 450 arom immunoactivity and protein expression levels were reversed in the TAES group(P<0.01,P<0.05)rather than in the medication group,except serum T and ratio of LH/FSH in the medication group.No significant differences were found between the medication and TAES groups in the serum T and ratio of LH/FSH(P>0.05).In addition,the increased vesicular follicle number,the decreased corepus luteum number and the thickness of granular cell layer were markedly improved in the TAES group.Conclusion TAES intervention can reduce both body weight and ovarian weight and regulate the levels of serum sex hormones and ovarian P 450c17αand P 450 arom protein expression levels in PCOS rats,which may contribute to its effect in improving PCOS.

摘要:Objective To observe the effect of acupuncture on c-jun N-terminal Kinase(JNK)signaling in the hippocampus in rats with forced-swimming stress,so as to reveal its underlying mechanism in relieving depression-like motor response.Methods Forty-eight Sprague-Dawley rats were randomly divided into 8groups as control,control+JNK inhibitor(SP600125),model,model+SP 600125,acupuncture,acupuncture+SP 600125,Fluoxetine(an anti-depressant),and Fluoxetine+SP 600125(n=6in each group).The depression-like behavior(immobility)model was established by forcing the rat to swim in aglass-cylinder and solitary raise.Acupuncture stimulation was applied to"Baihui"(GV 20)and "Yintang"(GV 29)for 20 min before forced swimming and once again 24 hlater.The rats of the Fluoxetine and Fluoxetine+SP 600125 groups were treated by intragastric administration of fluoxetine 10mL(1.8mg)/kg before forced swimming and once again 24 hthereafter.The rats of the model+ SP 600125 and acupuncture+SP 600125 groups were treated by intraperitoneal injection of SP 600125(10mg/kg)90min before forced swimming and 30 min before acupuncture intervention,respectively.The immobility duration of rats in the water glass-cylinder was used to assess their depression-like behavior response.The expression levels of protein kinase kinase 4(MKK 4),MKK 7,JNK,and phosphorylated JNK(p-JNK)in the hippocampus were detected by Western blot.Results Compared to the control group,the duration of immobility,and the expression levels of hippocampal MKK 4,MKK 7,and p-JNK proteins were significantly increased in the model group(P<0.01).While in comparison with the model group,the duration of immobility in the model+SP 600125,acupuncture,acupuncture+SP 600125,Fluoxetine and Fluoxetine+SP 600125 groups,the expression levels of hippocampal MKK 4and MKK 7proteins in the Fluoxetine+SP 600125 group,and those of p-JNK protein in the acupuncture,acupuncture+SP 600125,model+SP 600125,Fluoxetine and Fluoxetine+SP 600125 groups were considerably decreased(P<0.05,P<0.01).No significant differences were found between the control and control+SP 600125 groups and among the model+SP 600125,acupuncture,acupuncture+SP 600125,Fluoxetine and Fluoxetine+SP 600125 groups in the duration of immobility(P>0.05),and in the expression level of p-JNK protein(P>0.05).No significant changes were found in the expression levels of JNK among the 8groups(P>0.05).Conclusion Acupuncture stimulation of GV 20 and GV 29 is effective in relieving depression-like motor response in forced-swimming stress rats,which may be closely associated with its effects in down-regulating the expression of hippocampal p-JNK protein.

摘要:Objective To investigate the protection mechanism of electroacupuncture(EA)therapy against Alzheimer's disease(AD)-like neurotoxicity induced by Isoflurane.Methods Twenty-four APPswe/PS 1dE 9double transgenic mice(one of the most extensively used transgenic mouse model of AD)and 24 littermate wild-type mice were randomly assigned into control(Con)group,isoflurane(Iso)group and EA group,respectively(n=8in each group).EA(2Hz/100 Hz,1mA)was applied to"Baihui"(GV 20)and"Yongquan"(KI 1)for 15 min,once a day for 3days.The transgenic mice were exposed to a closed box filled with 1.2%Isoflurane+30% O2+70% N2 for 4h.The animals' learning-memory ability was detected by Morris water maze test.The expression of cleaved Caspase-3in the CA 1area of hippocampus was detected by immunohistochemistry,and that of hippocampal Bcl-2and Bax proteins detected by Western blot.Results Compared with the wilde-type mice,the average escape latency of place navigation test was significantly longer,while the percentage of target-quadrant stay time and the targetplatform crossing times of spacial probe test were marked decreased in AD+Iso mice(P<0.05).After acupuncture intervention,the abovementioned changes were reversed(P<0.05).Correspondingly,compared with the AD-Con group,the number of hippocampal activated Caspase-3-positive cells and the expression of Bax protein were significantly increased in the AD-Iso group(P<0.05).After EA intervention,the increased Caspase-3-positive cell number and Bax protein expression were remarkably down-regulated in the AD-EA group,and the decreased ratio of Bcl-2/Bax in AD-Iso mice was obviously up-regulated in AD-EA mice(P<0.05).No significant changes were found in the average escape latency,the percentage of target-quadrant stay time and the target-platform crossing times,and in the number of hippocampal activated Caspase-3-positive cells,the expression levels of hippocampal Bcl-2and Bax and the ratio of Bcl-2/Bax in the three groups of wilde-type mice(P>0.05).ConclusionEA intervention can improve the learning-memory ability in AD+Isoflurane mice,suggesting a reduction of AD-like neurotoxicity,which may be associated with its actions in inhibiting the overexpression of activated Caspase-3and Bax proteins in the hippocampus.

摘要:Objective To observe the effect of electroacupuncture(EA)intervention on constipation-predominant irritable bowel syndrome(C-IBS)and colonic calcitonin gene-related peptide(CGRP)and substance P(SP)expression levels in IBS rats,so as to reveal its mechanism underlying improvement of IBS.Methods A total of 32 male Wistar rats were equally randomized into normal control,model,EA-Shangjuxu(ST 37)and EA-Dachangshu(BL 25)groups.The C-IBS model was established according to Peng's methods(2004).EA(2Hz/100 Hz,0.1-0.2mA)was applied to bilateral ST 37 and BL 25for30 min,once daily for 7days.The stool properties(hardness or softness,water content,etc.)were typed(1-7points)and the fecal water content was calculated by wet weight minus dry weight.The relative expression levels of CGRP and SP proteins in the colon tissues were detected by Western blot.Results Compared to the control group,the stool property score and stool water content were significantly lower in the model group(P<0.01).After EA intervention,both stool property scores and stool water contents were obviously higher in the EA-ST 37 and EA-BL 25 groups than in the model group(P<0.05,P<0.01).The expression levels of colonic CGRP and SP proteins were significantly higher in the model group than in the control group,and obviously down-regulated in the two EA intervention groups(P<0.01).No significant differences were found between the EA-ST 37 and EABL 25 groups in the stool property score and stool water content,and colonic CGRP and SP protein expression levels(P>0.05).Conclusion EA intervention can ameliorate the fecal property in C-IBS rats,which may be associated with its function in inhibiting the expression of colonic CGRP and SP proteins.
摘要:Objective To observe The effect of electroacupuncture(EA)stimulation of"Zusanli"(ST 36)and "Taichong"(LR 3)on intestinal motor and neurotensin(NT)levels in the plasma,hypothalamus,and gastro-antrum tissues in functional dyspepsia(FD)rats so as to reveal its mechanisms underlying improvement of FD.Methods Forty-eight SD rats were randomly divided into control,model and EA groups,with 16 rats in each group.The FD model was established by clamping the rats' tails and alternate day's feeding according to the related references.EA(2Hz/100 Hz,2mA)was applied to unilateral ST 36 and LR 3for 30 min,once daily for 14 days.Rats of the control group were only restricted.The gastric emptying rate and propulsive rate of the small intestine were detected.The content of NT in the plasma was assayed using ELISA,and the immunoactivity levels of NT in the hypothalamus,gastric antrum mucous membrane and ileum tissues were detected using immunohistochemistry.Results Compared with the control group,the gastric emptying rate and propulsive rate of the small intestine were considerably lowered in the model group(P<0.01),and the content and immunoactivity levels of NT in the plasma,hypothalamus,mucous membrane of the gastric antrum and ileum tissues were significantly increased(P<0.05).After EA intervention,the decreased gastric emptying rate and intestinal propulsive rate,as well as the increased NT content and immunoactivity levels of plasma,hypothalamus,gastric antrum and ileum were reversed(P<0.05).Conclusion EA intervention can obviously promote gastrointestinal motor in FD rats,which may be related to its function in down-regulating NT levels in the plasma,hypothalamus,gastric antrum and ileum.It suggests an involvement of NT in the brain-gut axis in EA-induced improvement of FD.
摘要:Objective To observe the protective effect of electroacupuncture(EA)intervention on the expression of brain-derived neurotrophic factor(BDNF)and tropomyosin receptor kinase B(TrkB)proteins and genes in the hippocampus areas in myocardial ischemia(MI)rats,so as to reveal its underlying mechanisms in protecting hippocampal cells under MI.Methods Eighty healthy male SD rats were randomized into sham-operation(sham),MI model,Shenmen(HT 7),HT 7-Zhizheng(SI 7)and HT 7-Xinshu(BL 15)groups(n=15in each group).The MI model was established by occlusion of the anterior descending branch of the left coronary artery.EA(2Hz,2mA)was applied to bilateral HT 7,HT 7-SI 7,and HT 7-BL 15,respectively for15 min,once per day for a week.The number of BDNF and its receptor TrkB positive cells in the left hippocampus and that of mRNAs in the right hippocampus tissue were determined by immunohistochemistry and real-time fluorescence quantitative PCR,respectively.Results In comparison with the sham group,the numbers of both BDNF and TrkB positive cells in the left hippocampus and the expression levels of BDNF mRNA and TrkB mRNA in the right hippocampus were increased slightly(P>0.05).After EA intervention,the numbers of hippocampal BDNF and TrkB positive cells and the expression levels of BDNF mRNA and TrkB mRNA were evidently up-regulated(P<0.05,P<0.01),and the effects of HT 7-SI 7and HT 7-BL 15 were obviously superior to those of simple HT 7in up-regulating BDNF and TrkB expression(P<0.01,P<0.05).No significant differences were found between the HT 7-SI 7and HT 7-BL 15 groups in increasing the number of hippocampal BDNF and TrkB positive cells and in up-regulating expression of both BDNF mRNA and TrkB mRNA(P>0.05).Conclusion EA intervention is effective in increasing the expression of hippocampal BDNF and TrkB in MI rats,which may contribute to its effect in protecting hippocampal cells from injury under MI condition.The effect of EA stimulation of HT 7-SI 7and HT 7-BL 15 was obviously superior to that of simple HT 7in up-regulating BDNF and TrkB expression.
摘要:Objective To observe the effect of acupuncture and moxibustion interventions on ethology and 5-HT/5-HIAA levels in the frontal cortex tissue in depression rats,so as to reveal their biological mechanisms underlying improvement of depression.Methods A total of 40 male SD rats were randomized into normal control,depression model,medication(Fluoxetine),acupuncture and moxibustion groups,with 8rats in each group.The depression model was established by chronic unpredictable mild stress for 5weeks.The interventions including acupuncture or moxibustion stimulation of"Baihui"(GV 20)and"Dazhui"(GV14)for 20min/d and intragastric administration of Fluoxetine(1.8mg·kg-1·d-1)beginning after 2weeks of modeling for 3weeks.The percentages of sucrose consumption was counted,the contents of 5-HT,hydroxyindole acetic acid(5-HIAA),tryptophan(Trp)in the prefrontal cortex tissue were assayed by radioimmunoassay,and the relative binding affinities(constant and capacity)of 5-HT1 Areceptor in the prefrontal cortex were measured by radioligand binding assay and Scatchard plot analysis in a computer.Results In comparison with the normal control group,the sucrose consumption percentage on the 35 th day of modeling,the levels of cerebral cortical 5-HT,5-HT/5-HIAA and 5-HT1 Areceptor binding constant were significantly down-regulated in the model group(P<0.01,P<0.05),and the cortical Trp content was obviously up-regulated in the model group(P<0.01).Compared with the model group,the sucrose consumption percentage on the 35 th day of modeling,and cortical 5-HT/5-HIAA in the medication,acupuncture and moxibustion groups,cortical 5-HT content in the medication group were significantly increased(P<0.01,P<0.05),and cortical 5-HIAA and Trp contents were obviously down-regulated in the three treatment groups(P<0.01).No significant changes were found in the cortical 5-HT content after acupuncture and moxibustion,and in the high and lower affinity of 5-HT1 Areceptor binding constant and binding capacity after medication,acupuncture and moxibustion interventions(P>0.05).The effects of acupuncture and moxibustion interventions were comparable in increasing sucrose consumption percentage on the 35 th day and 5-HT/5-HIAA,and in down-regulating cortical Trp content(P>0.05).Conclusion Both acupuncture and moxibustion can increase sucrose consumption(anti-depression)in depression rats,which may be closely associated with their effects in up-regulating 5-HT/5-HIAA and down-regulating Trp content in the frontal cortex.
摘要:Objective To observe the influence of electroacupuncture(EA)on serum TNF-α,IL-1βand Intercellular adhesion molecule 1(ICAM-1)levels in rheumatoid arthritis(RA)rats so as to explore its mechanism in relieving RA.MethodsEight Wistar rats were used as the normal control group in this study.The RA model was established by injecting mixture solution of incomplete Freund's adjuvant and type Ⅱchicken collagen into the left paw.The 24 successful RA rats were randomly divided into model,medication(Prednisolone)and EA groups(n=8in each group).EA(2Hz/100 Hz,1-2mA)was applied to"Zusanli"(ST 36)and "Kunlun"(BL 60)for 30 min,once daily for 10 days.For rats of the medication group,Prednisolone acetate(0.1mL/10g)was intragastric infused daily.The rats' left ankle diameter was measured before and after the treatment,and the contents of serum TNF-α,IL-1βand ICAM-1were assayed by ELISA.Results In comparison with the normal control group,the rats' ankle diameter,serum TNF-α,IL-1βand ICAM-1contents were remarkably increased in the model group(P<0.05);while compared with the model group,the increased ankle diameter,and serum TNF-α,IL-1βand ICAM-1contents were obviously reversed in both medication and EA groups(P<0.05),without significant differences between the medication and EA groups(P>0.05).Conclusion EA intervention is effective in relieving RA rats' inflammatory reactions by down-regulating the levels of serum TNF-α,IL-1βand ICAM-1.
摘要:Objective To observe the effect of electroacupuncture(EA)intervention on spleen T-helper 17(Th 17)and regulatory T(Treg)cell levels in mice with ulcerative colitis(UC),so as to reveal its mechanisms underlying improvement of UC.Methods Kunming mice were randomized into control,UC model and EA groups,with 8mice in each group.The UC model was established by giving the mice with 3% Dextran Sulfate Sodium(DSS)for 5days.EA(15Hz/25 Hz,0.1-0.2mA)was applied at"Guanyuan"(CV 4)and bilateral"Zusanli"(ST 36,used alternatively)for 10 min,once a day for 5days.The animals' disease activity index[DAI,=(body weight index score+stool score+bleeding score)/3;0-4points]were calculated.The pathological changes of colon tissues were observed by light microscopy after H.E.stain,and spleen Treg(CD4+CD25+Foxp3+Treg)and Th 17(CD3+CD8+IL-17+Th 17)lymphocyte levels were determined by flow cytometry.Results Compared to the control group,the DAI score and the ratio of Th 17/CD8+T cells were significantly increased,while the ratio of Treg/CD4+T cells obviously decreased in the model group(P<0.05).After EA intervention,the increased DAI score and the ratio of Th 17/CD8+T cells and the decreased Treg/CD4+T cells were reversed(P<0.05),and the inflammatory cell infiltration degree of the colon tissue was attenuated.Conclusion EA intervention can improve the UC rats' symptoms of activity state,bloody or viscidity stool and colonic inflammation,probably by regulating the balance between the spleenic Treg and Th 17 lymphocytes.

摘要:Objective To observe the influence of herbal cake-separated moxibustion on the contents of 5-hydroxy tryptamine(5-HT),dopamine(DA)and norepinephrine(NE)in the hypothalamus in rats with functional dyspepsia(FD)of syndrome of liver stagnation and spleen deficiency,so as to explore its mechanism underlying improvement of FD.Methods Sixty SD rats were randomly divided into 6groups:control,model,herbal cake-separated moxibustion,moxa-cone moxibustion,Xiaoyaosan(decoction for relieving liver stagnation)and Domperidon,with 10 rats in each group.The FD model was established by applying chronic restraint stress+excessive fatigue+irregular food+tail clipping+shaking for 21 consecutive days.Moxibustion(herbal cake-separated or moxa-cone)was applied to bilateral"Ganshu"(BL 18),"Pishu"(BL 20)and"Weishu"(BL 21),or"Zhangmen"(LR 13),"Qimen"(LR 14)and"Zhongwan"(CV 12)for 30 min,once daily for 14 d.For rats of the two medication groups,Xiaoyaosan[1mL(2g)/100g]and Domperidone[1mL(0.3g)/100g]were administrated by gavage,respectively.The contents of 5-HT,DA and NE in the hypothalamus tissue were detected by high performance liquid chromatography.Results The gastric empty rate was obviously lower in the model group than in the control group(P<0.01).After the treatment,the gastric empty rate in the herbal cake-separated moxibustion,moxa-cone moxibustion,Xiaoyaosan and Domperidone groups were significantly increased in comparison with the model group(P<0.01),but there were no significant differences among the four treatment groups(P>0.05).Compared to the control group,the contents of 5-HT,DA and NE in the hypothalamus were markedly decreased in the model group(P<0.01),while compared to the model group,the contents of hypothalamic 5-HT,DA and NE contents were significantly up-regulated in both the herbal cake-separated moxibustion and Xiaoyaosan groups(P<0.01),rather than in the moxa-cone moxibustion and Domperidone groups(P>0.05).No significant differences were found between the herbal cake-separated moxibustion and Xioayaosan groups in increasing hypothalamic 5-HT,DA and NE levels(P>0.05).Conclusion Herbal cake-separated moxibustion can promote the gastric empty rate in FD rats,which may be associated with its effects in inhibiting stress induced decrease of hypothalamic 5-HT,DA and NE levels.
摘要:Objective To observe the effect of electroacupuncture(EA)intervention combined with medication(Gastrodin)on changes of neurological function and expression of Nogo-A and Nogo-A receptor(NgR)in the frontal lobe cortex around the ischemic loci of focal cerebral ischemia(FCI)rats,so as to explore its mechanism underlying improvement of neuroregeneration of FCI.Methods Fifty male Sprague-Dawley rats were randomly divided into normal control,model,EA,medication and EA+medication groups(n=10in each group).The FCI model was induced by occlusion of the middle cerebral artery(MCAO)with thread embolus.EA was applied to the left"Quchi"(LI 11)and"Hegu"(LI 4)for 30 min,once daily for 14 days after MCAO.For rats of the medication group,Gastrodin(10mg/kg)was administrated by intraperitoneal injection,once daily for 14 days.The neurological impairment was assessed by Zea Longa's scoring.The expression of Nogo-A and NgR in the frontal lobe cortex around the ischemic loci was detected by immunohistochemistry.Results In comparison with the normal control group,cerebrocortical Nogo-A and NgR expression levels of the model group were significantly increased(P<0.05).Compared with the model group,the Zea Longa's score and Nogo-A and NgR expression levels were evidently down-regulated in the EA,medication and EA+medication groups(P<0.05).The Zea Longa's score and Nogo-A and NgR expression levels were significantly lower in the EA+medication group than in the EA and medication groups(P<0.05).Conclusion EA intervention and Gastrodin administration can down-regulate cerebro-cortical Nogo-A and NgR protein expression in FCI rats,which may contribute to their action in improving neurological impairment.The effect of EA+Gastrodin is better than simple EA or Gastrodin treatment.
摘要:Objective To observe changes of skin temperature along the Governor Vessel(GV)after moxibustion stimulation.Methods A total of 36 healthy volunteer subjects were recruited in the present study.The skin temperature of Mingmen(GV 4),Dazhui(GV 14),Shenzhu(GV 12),Zhiyang(GV 9),Zhongshu(GV 7),Yaoyangguan(GV 3)regions and their bilateral control sites(1.5 cunlateral to GV 4)was detected by using a Thermo Tracer TH 9100.Results Following moxibustion stimulation of GV 4region,a marked infrared radiant track along the central part of the back covering the GV was observed in the healthy volunteer subjects,and the skin temperature levels of both GV 4and its bilateral control points were significantly increased(P<0.001).The skin temperature of GV 4was remarkably higher than those of both left and right control points(P<0.001).After moxibustion stimulation,the average skin temperature levels of GV 9,GV 7,GV 4and GV 3were all up-regulated in comparison with pre-moxibustion(P<0.01).Conclusion Moxibustion stimulation of Mingmen(GV 4)can induce an infrared radiant track along the central part of the back covering the GV and raise skin temperature of acupoint areas of the GV.

摘要:Objective To observe the effect of acupoint injection of Lidocaine on serum IL-1β,TNF-αand T-lymphocyte subset activities in patients undergoing laparoscopic cholecystectomy(LC),so as to reveal its mechanisms underlying relieving postoperative pain and potentiating rehabilitation.Methods Eighty patients scheduled for elective LC surgery(grade I or II,according to American Standards of Association,ASA)were randomly divided into four groups,namely intravenous analgesia(IVA),right forearm-injection(forearm-I),Jiaji(EX-B 2,Thorax 8)-injection(EX-B 2-I),and Zusanli-injection(ST 36-I),with20 patients in each group.The conventional anesthetic induction and maintenance with Penehyclidine Hydrochloride,Midazolam,Sulfentanil,Propofol,Atracurium Besilate,and Remifentanil were same in all the 4groups.For patients of the forearm-I,EX-B 2-I and ST 36-I groups,5% Lidocaine was injected into the subcutaneous layer of the anterior side of right forearm near the elbow,EX-B 2and ST 36 regions,respectively.Analgesia pump(filled with Sulfentanil,Ramosetron+normal saline)was connected after the tracheal extubation.The visual analog scale(VAS)was used to assess the patient's pain reaction after tracheal extubation(T 1),and 6h(T 2),24h(T 3)and 48h(T 4)after surgery.The times of PCA pressing and the total dose of Sufentanil in the process of postoperative analgesia were recorded as well.The contents of serum IL-1βand TNF-αwere analyzed by ELISA,and the counts of CD4+and CD8+T cells were detected by flow cytometry.Results Compared with T 1in the same one group,the VAS scores at time-points of T 2,T 3and T 4after surgery of all the IVA,forearm-I,ST 36-I and EX-B 2-I groups were reduced significantly(P<0.05).The times of PCA pump pressing and the doses of the administrated Sufentanil were considerably lower in the ST 36-I and EX-B 2-I groups than in the IVA and forearm-I groups(P<0.05).In comparison with pre-anesthesia in the same one group,serum TNF-αand IL-1βcontents at T 1were remarkably increased,while the ratios of CD4+/CD8+at T 4in the 4groups were evidently down-regulated(P<0.05).The contents of serum TNF-αand IL-1βat T 3and T 4were obviously lower in both ST 36-I and EX-B 2-I groups than in the IVA and forearm-Ⅰ groups(P<0.05).No significant differences were found among the 4groups in the VAS scores at the 4time-points,in the serum TNF-αand IL-1βcontents at T 0and T 1,in the counts of CD4+and CD8+T cells and ratios of CD4+/CD8+at T 0,T 3and T 4,and between the ST 36-I and EX-B 2-I groups in all the 8indexes(P>0.05).Conclusion Acupoint injection of Lidocaine is effective in relieving pain in LC patients,which is demonstrated by reducing VAS score,PCA pump pressing times,and administrated Sufentanil dose,and may be associated with its effects in down-regulating serum TNF-αand IL-1βcontents.

摘要:Objective To observe the influence of scalp acupuncture on levels of high-sensitivity C-reactive protein(hsCRP),TNF-α,IL-6,and IL-1βlevels in patients with acute cerebral infarction(ACI),so as to investigate its mechanism underlying improvement of ACI.Methods A total of 61 patients with ACI were randomly allocated to scalp acupuncture group(n=31)and control(medication)group(n=30).The patients of the control group were routinely treated by administration of Aspirin,Danhong injection,Cytidine Diphosphate for neurotrophy,blood pressure-control and blood-fat lowering medicines,etc.,while those of the scalp acupuncture group were treated by routine treatment with the medicines mentioned above plus daily scalp acupuncture stimulation of bilateral Dingnieqianxiexian[MS 6,penetrative needling from Qianding(GV 21)to Xuanli(GB 6)]and Dingniehouxiexian[MS 7,from Baihui(GV 20)to Qubin(GB 7)].The treatment was conducted once daily for 7days.Serum hsCRP,TNF-α,IL-6,and IL-1βcontents were assayed by using enzyme linked immunosorbnent assay(ELISA).The therapeutic effects of scalp acupuncture were evaluated by using clinical neurological disfunction scale(NDS,0-45 points for consciousness,gazing,facial palsy,speech,myodynamia,walking-ability).Results(1)Of the 30 and 31cases in the control and scalp acupuncture groups,5(16.7%)and 8(25.8%)were basically controlled,9(30.0%)and 16(51.6%)experienced remarkable improvement in their symptoms,12(40.0%)and 6(19.4%)were improved,4(13.3%)and 1(3.2%)failed,with the effective rates being 86.7% and 96.8%,respectively.The increased levels of serum hs-CRP,TNF-α,IL-6and IL-1βin ACI patients were reversed on the 3rd and 7th day after scalp acupuncture treatment(P<0.05,P<0.01).(2)A positive correlation existed between the NDS score and the serum levels of hs-CRP(r=0.497,P<0.01).(3)NDS scores were obviously decreased in both groups on the 7th day after the treatment compared with their baseline data(P<0.05,P<0.01).Conclusion Scalp acupuncture treatment can improve the ACI patients' clinical symptoms,probably by reducing ACI induced inflammatory reactions.
摘要:The blind study design,particularly the double-blind study design is a very important method for diminishing placebo effect and reducing bias in clinical medical trial.Enlightened by Streitberger's and Park's sham needle design,the authors of the present paper introduce a newly designed sham needle device(Yan's sham-needle)for controlled double-blind trials of acupuncture.This sham needle device consists of needle,tube and base.The bottom of the tube is completely sealed and it can never arouse any invasive stimulation on the subject's skin when the sham needle is downward pressed on the body surface.Meanwhile,this sham device is filled with sponge which is able to simulate soft tissues of the acupoint area.By combining words suggestions or hints before trials and the same shape as verum device,this sham-needle device reduces the risk of blind-breaking and makes it possible to conduct controlled double-blind trials.Primary practice showed that this device may provide a new and practical tool for researching the placebo effect of acupuncture therapy.
摘要:Ischemic stroke is one of the commonly met diseases in clinical practice nowadays.Acupuncture therapy is widly used in the treatment of sequela of ischemic stroke in China and its mechanisms have been extensively studied in recent years.In the present paper,the authors focus on the development of studies on the mechanism of scalp acupuncture therapy in the treatment of ischemic stroke.Results indicate that scalp acupuncture intervention can 1)improve cerebral blood circulation to promote regional energy metabolism,2)up-regulate expression of glial cell-line derived neurotrophic factor(GDNF),etc.,possibly promoting proliferation and differentiation of neural stem cells in the focal cerebral cortex and hippocampus,3)reduce contents of excitatory amino acid and increase level of gamma-aminobutyric acid(GABA)to lower neurogenic toxicity,and relieve cerebral injury,4)ease cerebral vascular immunoinflammatory reactions,5)regulate blood lipid metabolism to resist cerebral free radical damage,and 6)inhibit cerebral cortical apoptosis.However,these results only revealed very limited intrinsic mechanisms of scalp acupuncture in improving ischemic stroke.Further studies using comprehensive techniques of multi-disciplines as molecular biology,electrophysiology,etc.are definitely needed.
摘要:Peripheral nerve injury is a common clinical disease.Acupuncture therapy has been demonstrated to be effective in improving nerve injury in clinical practice,but its underlying mechanisms in promoting tissue repair basically remain unknown.In the present paper,the authors reviewed some descriptions of traditional Chinese medicine on peripheral nerve injury and treatment,and recent development of researches on acupuncture treatment of it in both clinical practice and animal studies.Clinical trials demonstrated that acupuncture treatment can relieve nerve injury induced pain,ameliorate both sensory and motor functions.Experimental studies showed that acupuncture stimulation may promote nerve repair by reducing desquamation of medullary sheath of nerve fibers,inhibiting apoptosis of nerve cells,and up-regulating expression of myelin basic protein,Slit-1protein and gene,etc.In addition,acupuncture intervention may also improve the microenvironment of neural regeneration including increase of the proliferation and differentiation of Schwann cells and release of various types of neurotrophic factors.However,its mechanisms underlying accelerating rehabilitation of peripheral nerve injury need being researched further.
