摘要:Objective To observe the effect of acupuncture on changes of neurological function and expression of proteins in the ischemic brain region in middle cerebral artery occlusion(MCAO)rats with protein chip technique,so as to reveal the profiles of cerebral proteins related to its effectiveness in improving cerebral ischemia(CI).Methods SD rats were randomized into sham operation(sham),CI model,non-acupoint and acupoint groups(n=10in each group).The CI model was established by MCAO according to modified Longa's method.For rats of the acupoint group,"Dazhui"(CV 14),"Baihui"(GV 20)and"Shuigou"(GV 26)were punctured by twirling the filiform needles for about 1min,and repeated once again during 30 min of needle retention,which was conducted once every 12 h,and 6times altogether.The non-acupoints were about 3mm away from the above-mentioned acupoints and stimulated with the same method and same procedures.The neurological deficit score was scaled according to Longa's method.The differentially-expressed proteins(≥1.5folds in up-and down-regulation)in the ischemic region of the brain were detected by using Springbio 720 antibody chip technology.Results Compared with the sham group,the neurological deficit score was significantly higher in the model group(P<0.01),while compared with the model group,the neurological deficit scores were considerably in both acupoint and non-acupoint groups(P<0.05,P<0.01).The therapeutic effect of acupoint group was evidently superior to that of the non-acupoint group in improving neurological function(P<0.05).Compared with the sham group,the differentially-expressed proteins in the ischemic brain including 33up-regulated and 12down-regulated were found in the model group,mainly functioning in cell apoptosis,cell cycle regulation,cell differentiation and proliferation,cell cytoskeleton and connection,cell signal transduction,DNA repair and transcription factors.Compared with the model group,the differentially-expressed proteins including 12up-regulated and 31down-regulated in the acupoint group,and 15up-regulated and 17down-regulated in the non-acupoint group were detected,functioning being the same as those mentioned above in the model group.Conclusion Manual acupuncture stimulation of GV 14,GV 20 and GV 26 can improve neurological function in CI rats,which may be associated with its function in regulating the expression of many proteins in the ischemic region of the brain.

摘要:Objective To investigate the microstructure(blood vessels and nerve fibers)of the skin tissue in "Taichong"(LR 3)region for reveling morphological characteristics of acupoint.Methods Five SD rats were used in the present study.The skin tissue in the region of acupoint LR 3was taken from the dorsum of hind foot following transcardial perfusion with4% paraformaldehyde.Then,the skin samples were sagittally or horizontally cut into sections(20μm or 40μm in the thickness)to be stained with Phalloidin and calcitonin gene related peptide(CGRP)by using fluorescent histochemistry and immunohistochemistry.The labeled vascular structure and nerve fibers were observed and recorded using fluorescent microscope and laser scanning confocal microscope.Results In the skin tissue of LR 3,different types of blood vessels labeled by phalliodin,including capillaries and glomera mainly distributing in the superficial layer of the dermis,and thicker blood vessels and their sub-branches mainly existing in the deeper layer of the dermis and the subcutaneous layer,were found.In addition,CGRP positive nerve fibers were found to run parallel to the thin blood vessels or to gather around the thicker blood vessels.From the subcutaneous layer to the epidermis,blood vessels and nerve fibers coexisted and formed a stereo-network structure.Conclusion In the skin of LR 3area,there exists a stereo-network structure consisting of different types of blood vessels and nerve fibers.Although this structure is not specifically associated with acupoint area alone,this result may provide a new sight to further understand the microstructure of acupoint.

摘要:Objective To observe the effect of electroacupuncture(EA)intervention at different treatment time-points on locomotor function and neuronal apoptosis in rats with acute spinal cord injury(ASCI),so as to explore its mechanism underlying improvement of ASCI.Methods Seventy-two female Wistar rats were randomly divided into sham operation,ASCI model and EA groups which were further divided into 4time-points:1,3,7and 14d(n=6at each).The ASCI model was established by weight-drop striking the exposed spinal cord(T10)using a NYU impactor device.EA(0.4-0.6mA,100Hz)was applied to bilateral"Jiaji"(EX-B 2)for 30 min,once a day for 1,3,7and 14 days,respectively in the EA group.The Basso-BeattieBresnahan(BBB)grading score was used to assess the animals' locomotor function,and H.E.staining was used to display the pathological changes of spinal cord injury at different time points.The TUNEL staining was used to detect the nerve cell apoptosis of spinal cord tissue.Results After ASCI,the BBB scores were significantly decreased and the numbers of the apoptotic nerve cells considerably increased on day 1,3,7and 14 in the model group compared with the sham operation group(P<0.05).In addition,H.E.staining showed severe damage of the spinal cord tissue,infiltration of inflammatory cells,increase of dead nerve neurons and glia cells in the number from day 3to 7,and additional increase of vacuoles on day 14.Following EA intervention,the decrease of BBB scores and the increase of apoptotic nerve cell numbers were markedly suppressed on day 3,7and 14(P<0.05).The TUNEL staining showed a decrease of the apoptotic nerve cells from day 3on and an improvement of the injuredtissue structure from day 7to 14 in the EA group.Conclusion EA stimulation of EX-B 2can significantly improve locomotor function,and lighten the spinal cord injury in ASCI rats,presenting a time-dependant tendency,which may be associated with its effect in reducing neuronal apoptosis in the spinal cord.

摘要:Objective To observe the effect of electroacupuncture(EA)at"Zusanli"(ST 36)on Ghrelin/cAMP/PKA expression in the jejunum in rats with spleen qi deficiency syndrome,so as to reveal its underlying mechanism in improving energy metabolism.Methods Forty male SD rats were randomly divided into 4groups:normal group,spleen qi deficiency syndrome(model)group,EA group and non-acupoint group(n=10in each group).The model of spleen qi deficiency syndrome was established by improper diet and overstrain.EA(2Hz/15 Hz,0.5mA)was applied to bilateral"Zusanli"(ST 36)in the EA group and non-acupoint in non-acupoint group for 20 min,once a day for 6days.The pathologic changes of the jejunum tissue were detected by H&E staining.Ghrelin,ATP and cAMP levels in jejunum tissue were determined by ELISA.The expression levels of PKA protein in jejunum tissue were determined by Western blot.Results H&E staining showed that the intestinal villi of the model group were swelling,shortening and thickening,with a damaged or broken top-part in the model group,and basically restored to normal after EA treatment.ELISA results showed that the contents of Ghrelin,ATP and cAMP in the jejunum tissue were significantly lower in the model group than in the normal group(P<0.05),while significantly higher in the EA group than in the model group(P<0.05).Western blot results showed that the expression of PKA protein in the jejunum tissue was significantly lower in the model group than in the normal group(P<0.05),and significantly higher in the EA group than in the model group and non-acupoint group(P<0.05).Conclusion EA at ST 36 can improve the morphological changes in the jejunum of spleen qi deficiency rats,which may be associated with its effects in increasing Ghrelin,ATP and cAMP contents,and up-regulating PKA expression,leading to an increase of energy metabolism and spleen qi at last.

摘要:Objective To observe the effectiveness of grain-moxibustion in resisting oxidative stress in doxorubicin(DOX)-induced cardiomyopathy rats.Methods Sixty Sprague-Dawley(SD)rats were randomly divided into blank control group(blank group),cardiomyopathy model(model)group and grain-moxibustion(moxibustion)group,with 20 rats in each one.The cardiomyopathy model was established by intraperitoneal injection of DOX(0.5mg/100g),once a week for 4weeks.Grainmoxibustion was applied to"Guanyuan"(CV 4)and"Juque"(CV 14)for 5cones,once daily for 4weeks except the Sundays.The rats' heart rate(HR),left ventricular systolic pressure(LVSP),left ventricular diastolic pressure(LVDP),left ventricular pressure maximum rising rate(+dp/dtmax),left ventricular pressure maximum decline rate(-dp/dtmax),left ventricular pressure change time(t-dp/dtmax)were detected to evaluate the cardiac function by using aphysiological signal acquisition-analysis system.The pathological changes of the myocardium were observed by optical microscopy after H.E.staining.The activity of serum lactate dehydrogenase(LDH),creatine kinase(CK)and myocardial superoxide dismutase(SOD),and myocardial malondialdehyde(MDA)and serum cardiac troponinⅠ(cTnⅠ)contents were assayed by enzyme-linked immuno sorbent assay kit(ELISA).Results Following modeling,the HR,LVSP,+dp/dtmax,-dp/dtmax,and myocardial SOD activity were significantly downregulated(P<0.05,P<0.01),while the LVDP,t-dp/dtmax,serum LDH and CK activity and cTnⅠ content,as well as myocardial MDA content were considerably increased in comparison with the blank group(P<0.05,P<0.01).After moxibustion intervention,the decreased HR,LVSP,+dp/dtmax,-dp/dtmax,and myocardial SOD activity levels and the increased LVDP,tdp/dtmax,serum LDH and CK activity and cTnⅠcontent,as well as myocardial MDA content were remarkably suppressed in comparison with the model group(P<0.05,P<0.01).Conclusion Grain-moxibustion can significantly improve the left ventricular function,increase heart rate in cardiomyopathy rats,which may be associated with its effects in reducing myocardial oxidative stress damage by down-regulating the content of MDA and up-regulating SOD activity in the heart.

摘要:Objective To observe the effect of electroacupuncture(EA)stimulation of the Heart Meridian on electrical activities of neck sympathetic and vagal nerves in acute myocardial ischemia(AMI)rats.Methods Thirty SD rats were randomly divided into 3groups,sham operation(sham),model and EA(n=10rats in each group).The left anterior descending branch(LADB)of the coronary artery was occluded to make an acute myocardial ischemia(AMI)model.For rats of the sham group,a surgical suture was simply threaded beneath the LADB without ligation.EA stimulation(2Hz,1mA)was applied to bilateral"Shenmen"(HT 7),"Tongli"(HT 5)and the middle-point between HT 7and HT 5for 20 min.The electrical activities of the left cervical sympathetic and vagal nerves were recorded using a bipolar platinum wire electrodes and a BIOPAC MPl 50 system,and the electrocardiogram(ECG)of the standard limb lead II was recorded simultaneously.The AMI severity was evaluated by using J-spot height of ECG.Results After AMI,the heart rate(HR),J-spot of ECG,and the discharge frequency of the cervical sympathetic nerve were significantly increased in the model group compared with the sham group(P<0.01),while the discharge frequency of vagal nerve was obviously decreased(P<0.01).Following EA stimulation,the increased levels of HR,J-spot height and discharge frequency of sympathetic nerve,and the decreased discharge frequency of vagus were significantly suppressed at time-points of 1,3,5and 15 min after EA(P<0.01).In addition,the ratio of discharge frequencies of vagal/sympathetic nerves was notably down-regulated after AMI(P<0.01).After EA stimulation,the ratios of vagal/sympathetic discharge at the time-points of 1,3,5and 15 min following EA were significantly increased(P<0.01).Conclusion EA stimulation of the Heart Meridian may improve AMI by inhibiting sympathetic discharges and elevating vagal discharges in AMI rats,i.e.,coordinating activities of the autonomic nerve systems.
摘要:Objective To observe changes of urinary microprotein,and serum creatinine,urea nitrogen and uric acid levels in focal segmental glomerulosclerosis(FSGS)rats treated by mild moxibustion,so as to explore the mechanism of moxibustion underlying improvement of FSGS.Methods SD rats were randomized into normal control(normal),sham operation(sham),model,medication(Losartan),moxibustion-Shenshu(BL 23)and moxibustion-Geshu(BL 17)groups.The latter two moxibustion groups were further divided into 10 min,20min and 30 min subgroups(n=6in each group/subgroups).The FSGS model was established by unilateral nephrectomy combined with injection of Losartan into the tail vein twice.Mild moxibustion was applied to bilateral BL 17 and BL 23 for 10,20 and 30min,respectively,once every other day,for 12 weeks.The contents of urinary microglobulinα1,micro-albumin,transferring and IgG were assayed using enzyme linked immunosorbent assay(ELISA),and serum creatinine,urea nitrogen and uric acid contents(indexes of renal function)determined using an automatic biochemical analyzer.The pathological changes of the kidney tissue was observed by using a microscope after periodic acid schiff(PAS)staining.Results No significant differences were found between the normal control and sham groups in the levels of all the urinary and serum indexes(P>0.05)and pathological changes of the renal tissues.Compared with the normal control group,the contents of urinary microglobulinα1,micro-albumin,transferrin and IgG,and serum creatinine,urea nitrogen and uric acid were significantly increased in the model group(P<0.01).Following medication and moxibustion,the contents of the aforementioned 7indexes in the Losartan group,and urinary microglobulinα1,micro-albumin,transferrin and IgG,and serum creatinine levels in the moxibustion BL 23-20 min and 30 min groups,and the micro-albumin and transferrin contents in the BL-17 10 min group and IgG level in the BL-23 10 min group,the serum creatinine and urea nitrogen levels at the 3time-points of both moxibustion BL 23 and BL 17 groups,and serum uric acid in the moxibustion BL 23 30 min,and BL17 20 and 30min groups were all considerably downregulated(P<0.05,P<0.01).The therapeutic effects of moxibustion 30 min were notably better than moxibustion 10 min in reducing urinary microglobulinα1,micro-albumin,transferring and IgG levels(P<0.05,P<0.01).Results of PAS staining showed that the injury of the renal tissue as the endothelial and mesangial cellular proliferation,collagen proteinosis,interstitial fibrosis,etc were relatively milder in the Losartan and moxibustion 20 and 30min groups.Conclusion Mild moxibustion may reduce proteinuria,and improve the kidney function and pathological changes in FSGS rats,and longer duration of moxibustion is better in achieving therapeutic effect.
摘要:Objective To observe the effect of manual acupuncture intervention on serotonin transporter(5-HTT),5-HT1Areceptor(5-HT1AR),norepinephrine(NE)alpha receptor 2(NEα_2R)protein of brain tissues in rats with post-stroke depression(PSD).Methods Forty SD rats were randomly divided into normal group,model group,medication group and acupuncture group,with 10 rats in each.The PSD model was established by the combination of middle cerebral artery occlusion and chronic non-predictable mild stress.Rats of the medication group were treated by gavage of fluoxetine(2 mg/kg,once daily for 21days),and those of the acupuncture group treated by manual acupuncture stimulation of"Baihui"(GV 20),"Fengfu"(GV 16),bilateral"Shenmen"(HT 7)and"Taichong"(LR 3)for 20 min,once a day for 21 days,with one day's interval between each every7 days.The animals' neurological deficit was detected by Zea Longa's scaling,and the depression status assessed by sugar solution consumption,and the locomotor activity(crossing and rearing times in 5min)detected by open-field tests.The contents of 5-HT,NE,acetylcholine(ACh),gamma aminobutyric acid(GABA),and glutamate(Glu)of the cerebral cortex were detected using high performance liquid chromatography(HPLC)and expression levels of 5-HTT,5-HT1 AR,NEα_2R proteins of the hippocampus,raphe nuclei and locus coeruleus(LC)tissues detected using western blotting(EB),respectively.Results After modeling,the neurological deficit score was significantly increased,while the sugar solution consumption and the locomotor activities(crossing and rearing times in 5min)were notably decreased(P<0.05,P<0.001).Findings of HPLC and WB indicated that after modeling,the cerebrocortical 5-HT and NE contents and 5-HTT protein expression levels in the hippocampus,raphe nucleus and LC were obviously decreased and the cortical ACh,GABA and Glu contents as well as the expression of 5-HT1 AR and NEα_2R proteins in the hippocampus,raphe nucleus and LC considerably increased in the model group(P<0.01).Following acupuncture intervention and medication,the increased neurological deficit score,cortical ACh,GABA and Glu contents,and the expression levels of 5-HT1 AR and NEα_2R proteins in the hippocampus,rahpe nucleus and LC,as well as the decreased sugar solution consumption,crossing and rearing times of open field tests,cortical 5-HT and NE contents,as well as cortical 5-HT protein expression were all reversed in both acupuncture and medication groups(P<0.05,P<0.01).No significant differences were found between the acupuncture and medication groups in all the above-mentioned indexes(P>0.05).Conclusion Acupuncture can improve the locomotor function changes in stroke rats with depression after stroke,which may be associated with its effects in suppressing the up-regulated cortical ACh,GABA,Glu contents,the expression levels of 5-HT1 AR and NEα2R proteins in the hippocampus,raphe nucleus and LC,and the down-regulated 5-HT and NE contents in the cerebral cortex and 5-HTT protein expression levels in the hippocampus,raphe nucleus and LC.
摘要:Objective To explore the relationship between the theory of constitution in terms of Chinese medicine and clinical efficacy of acupuncture treatment via analyzling the characteristics of Deqi during acupuncture stimulation of Zusanli(ST 36)in healthy volunteers with different constitutions.Methods In the present study,a total of 527 healthy undergraduate student volunteers(267girls and 260boys)were recruited.They received questionnaire first about their constitutions in accordance with Professor WANG Qi's Classification and Determination of Constitutionin terms of Chinese medicine.Then,the subjects were asked to take a supine position on a check-bed,aqualified acupuncturist held a sterilized fifiform needle to rapidly insert it into ST 36 and manipulated the acupuncture needle with uniform reinforcing-reducing technique at a frequency of about 60-90times/min and an amplitude of 0.3-0.5cm.The status of Deqi was assessed by using visual analogue scale(VAS).ResultsNeedling sensations mainly involve soreness,numbness,heaviness,distension,dull pain and propagated sensations along meridian(PSC).During acupuncture stimulation,most subjects experienced distension sensation,accounting for 90.3%,followed by soreness,accounting for 45.9%.Except for PSC reaction,the rest 5needling sensations had no statistical difference in their occurrence rates in those subjects with different constitutions(P>0.05).The PSC appeared a higher incidence in yin-yang harmony(balanced)constitution subjects than in qi-deficiency,yang-deficiency,blood-stagnation and qi-stagnation constitution subjects(P<0.05).The intensity of soreness was obviously stronger in the balanced constitution subjects than in qi-deficiency,yang-deficiency,yin-deficiency,phlegm-damp,damp-heat and qi-stagnation constitution subjects(P<0.05),while the intensity of distension sensation was apparently stronger in the balanced constitution subjects than in simpleyang-deficiency subjects(P<0.05).Conclusion Except for PSC,the Deqi sensations of soreness,numbness,heaviness,distension and dull pain induced by acupuncture stimulation of ST 36 are comparable in the occurrence rates in healthy volunteer subjectswith different(9types of)constitutions in terms of Chinese medicine,whereas the intensity of soreness may be the important factor for acupuncture induced pain relief.A certain correlation between the constitution and acupuncture effectiveness exists,suggesting that acupuncture treatment should vary from patient to patientin clinical practice.

摘要:Objective To explore the regularity of distribution of press-sensitive acupoints on the body surface in tinnitus patients,so as to provide evidence for selecting suitable acupuncture points in the treatment of tinnitus.Methods Sixty-three cases of tinnitus patients(from 25 to 75years in age)were recruited in the present study.The patient was asked to take a sitting,supine and prone position,successively,and the operator used the thumb to moderately press the subject's head,neck,back,four limbs below the knee-and elbow joints along the three Hand-and three Foot-yang and yin meridians.Those acupoints that appeared pressing pain,or soreness-distension,numbness,or radiation feeling more than 10 times were brought into the sequence analysis by using the Excel 2010.Results A total of 131press-sensitive acupoints including 41 points with an appearance frequency of more than 10 times were found in the 63 tinnitus patients.These 41 acupoints distributing in the top three meridians were the Gallbladder Meridian of Foot-Shaoyang,Tri-energizer Meridian of Hand-Shaoyang and Spleen Meridian of FootTaiyin,and their involved regions were those below the knee-joints,bilateral sides of the head and marginal portion of the ear,sites bellow the elbow-joints,back-waist,successively in the appearance frequency.The top ten acupoints are Wangu(GB 12),Yifeng(SJ 17),Waiguan(SJ 5),Quchi(LI 11),Sanyinjiao(SP 6),Yanglingquan(GB 34),Fengchi(GB 20),Taichong(LR 3),Shousanli(LI 10)and Zulinqi(GB 41),in sequence.Conclusion The press-sensitive acupoints in tinnitus patients distribute principally in the Gallbladder Meridian of Foot-Shaoyang,and below the knee-joint,and the most sensitive acupoints are Wangu(GB 12)and Yifeng(SJ 17).These findings may provide an objective basis for clinical diagnosis and treatment of tinnitus.

摘要:Objective To analyze the characteristics and rules of acupoint selection for acupuncture treatment of insulin resistance.Methods Data collections were conducted by searching references on acupuncture treatment of insulin resistance in PubMed,CNKI,VIP data base from 1991 to 2016,and acupuncture prescription data base for acupuncture treatment of insulin resistance was established.Data mining was applied to analyze the characteristics and rules of acupoint selection.Results A total of 64 papers were recruited,and 73 acupoints were selected in these papers.It was found that the acupoints as Zusanli(ST36),Sanyinjiao(SP 6),Fenglong(ST 40)and Taichong(LR 3)were used with highest frequencies.All acupoints selected distributed in 13 meridians,especially Foot Yangming Stomach Meridian,Foot Taiyin Bladder Meridian,and Conception Vessel with a total frequency of 58.07%.The special acupoints including five-shu points,eight confluent points and back-shu points,accounted for 56.71%.Conclusion This study excavated the regular acupoint selection and acupoints compatibility for acupuncture treatment in patients with insulin resistance,giving evidence based confirming and direction for acupuncture clinical treatment.

摘要:Electroacupuncture(EA)has been demonstrated effective for pain relief.However,repeated application may decline analgesic effect of EA,which is termed EA tolerance,that reduces the clinical efficacy of EA.Therefore,it has attracted attention from researchers in recent years and the progresses include:(1)acute and chronic EA tolerance animal models have been established;(2)cross-tolerance between EA and morphine;(3)Anti-opioid substances,including cholecystokinin,orphanin FQ and angiotensin Ⅱ,have been reported to contribute to EA tolerance;(4)glutamate receptors and transporters,5-hydroxytryptamine and norepinephrine have been revealed involvement in EA tolerance;(5)cyclic adenosine monophosphate,cyclic guanosine monophosphate and Ca(2+) have been reported to be the second messengers cellularly in EA tolerance.The current EA tolerance effect lacks in-depth researches.Therefore,studies on its molecular mechanisms and signaling pathway are necessarily required.
摘要:In the present paper,the authors introduce a newly-developed"Acupuncture Needle Manipulation Training-evaluation System"based on optical motion capture technique.It is composed of two parts,sensor and software,and overcomes some shortages of mechanical motion capture technique.This device is able to analyze the data of operations of the pressing-hand and needle-insertion hand during acupuncture performance and its software contains personal computer(PC)version,Android version,and Internetwork Operating System(IOS)Apple version.It is competent in recording and analyzing information of any operator's needling manipulations,and is quite helpful for teachers in teaching,training and examining students in clinical practice.

摘要:Stress disease is a body functional disorder caused by harmful stimuli overwhelming the nonspecific adaptive capabilities.Numerous evidence proved the feasibility of acupuncture in treating stress diseases.This paper aims at exploring new perspective in the study on the underlying mechanism of acupuncture in the treatment of stress diseases.The discussion is conducted by summarizing the pathogenesis of stress diseases and mechanisms of its treatment with acupuncture,in exploring the related factors of acupuncture and stress response,and the correlation between the two from the regulatory pathway,medium,characteristics and manifestations.
摘要:Acu-moxibustion is the important component and inseparable in Chinese medicine with its unique characteristics.Just due to the distinct characteristics of acu-moxibustion,whether to standardize acu-moxibustion has been highlighted among the people in consideration of its development.In the present paper,the author introduces the characteristics of acu-moxibustion,the existence situation of its characteristics after being standardized,and the function,substance,feasibility,practice and effect,plan and strategy of acu-moxibustion standardization.
