摘要:Objective To observe the effect of acupuncture serum on the number of apoptosis of the cultured hippocampal neurons with seizure-like discharges and the expression levels of endoplasmic reticulum stress-inducible molecular chaperones glucose-regulated protein 78(GRP 78),C/EBP homologous protein(CHOP)and cysteine protease protein-12(Caspase-12),so as to reveal its protective mechanism on seizure-induced injury of hippocampal neurons.Methods A regular primary culture of neurons derived from the hippocampus of the newly-born SD rats was conducted for 10 days,then these cultured neurons that displayed seizure-like discharges in Mg(2+)-free medium were divided into normal extracellular fluid(medium)group(normal),Mg(2+)-free medium were divided into normal extracellular fluid(medium)group(normal),Mg(2+)-free medium group,acupuncture serum group and non-acupuncture serum group(n=30).Blood examples were taken from pentylenetetrazol(i.p.i.)-induced acute convulsion adult SD rats underwent manual acupuncture stimulation of"Baihui"(GV 20)and"Dazhui"(GV 14,once daily for 7days)to prepare acupuncture serum.Hippocampal neuronal cultures were prepared from hippocampal tissue isolated from 24h-old SD rats.The isolated neurons were incubated normally in Dulbecco's Modified Eagle Medium(DMEM)/Nutrient Mixture F 12(1∶1)for 10 days,followed by exposure to the extracellular fluid{composed of NaCl,KCl,CaCl_2,MgCl_2,HEPES [4-(2-hydroxyethyl)-1-piperazineethanesulfonic acid],D-glucose and aminoethanoic acid(pH 7.3)}for3hand returned to normal culture for normal group,by exposure to Mg(2+)-free medium group,acupuncture serum group and non-acupuncture serum group(n=30).Blood examples were taken from pentylenetetrazol(i.p.i.)-induced acute convulsion 12(1∶1)]plus acupuncture serum(9∶1)for acupuncture serum group,and by exposure to Mg(2+)-free extracellular fluid for 3hand returned to DMEM/F 12(1∶1)plus non-acupuncture serum(9∶1)for non-acupuncture serum group.The cell apoptosis was assessed at 2,12 and 48hafter application of acupuncture serum or non-acupuncture serum by using TUNEL method and the expression levels of GRP 78,CHOP and Caspase-12 proteins of the cultured cells at the 3time-points detected using Western blot.Results The number of apoptotic hippocampal neurons was significantly higher in the Mg(2+)-free extracellular fluid for 3hand returned to DMEM/F 12(1∶1)plus non-acupuncture serum(9∶1)for non-acupuncture serum group.The cell apoptosis was assessed at 2,12 and 48hafter application of acupuncture serum or non-acupuncture serum by using TUNEL method and the expression levels of GRP 78,CHOP and Caspase-12 proteins of the cultured cells at the 3time-points detected using Western blot.Results The number of apoptotic hippocampal neurons was significantly higher in the Mg(2+)-free medium group than in the normal medium group at 2,12 and 48h(P<0.01),and considerably decreased in the acupuncture serum group(but not in the non-acupuncture serum group)relevant to the Mg(2+)-free medium group than in the normal medium group at 2,12 and 48h(P<0.01),and considerably decreased in the acupuncture serum group(but not in the non-acupuncture serum group)relevant to the Mg(2+)-free medium group at the same timepoints after application of acupuncture serum(P<0.05,P<0.01).The expression levels of GRP 78 protein at 2h,CHOP and Caspase-12 proteins at 2,12 and 48hwere significantly up-regulated in the Mg(2+)-free medium group at the same timepoints after application of acupuncture serum(P<0.05,P<0.01).The expression levels of GRP 78 protein at 2h,CHOP and Caspase-12 proteins at 2,12 and 48hwere significantly up-regulated in the Mg(2+)-free medium group relavant to the normal medium group(P<0.05,P<0.01).After application of acupuncture serum to the culture medium,the expression levels of GRP 78 protein at the three time-points were significantly increased in comparison with the Mg(2+)-free medium group relavant to the normal medium group(P<0.05,P<0.01).After application of acupuncture serum to the culture medium,the expression levels of GRP 78 protein at the three time-points were significantly increased in comparison with the Mg(2+)-free medium group(P <0.01,P<0.05),while those of CHOP at 12 and 48 h,and Caspase-12 at the three time-points were notably down-regulated(P<0.05,P<0.01).No significant differences were found between the non-acupuncture serum and Mg(2+)-free medium group(P <0.01,P<0.05),while those of CHOP at 12 and 48 h,and Caspase-12 at the three time-points were notably down-regulated(P<0.05,P<0.01).No significant differences were found between the non-acupuncture serum and Mg(2+)-free medium groups in the expression levels of GRP 78,CHOP and Caspase-12 proteins at the three time-points(P>0.05).Conclusion Acupuncture serum can significantly reduce apoptosis of the cultured hippocampal neurons,which may be related to its effects in increasing the expression of GRP 78 protein and down-regulating the expression of CHOP and Caspase-12 proteins,suggesting an important role of acupuncture serum in maintaining the stability of the neuronal endoplasmic reticulum.
摘要:Objective To explore the potential mechanism of acupuncture stimulation of "Taichong"(LR 3)and"Neiguan"(PC 6)in spontaneous hypertension rats(SHR)by investigating its effects on blood pressure and contents of aspartic acid(Asp)and glutamic acid(Glu)in the rostral ventrolateral medulla(RVLM)region.Methods A total of 75 SHR were randomized into model group,Taichong(LR 3)group,Neiguan(PC 6)group,LR 3+non-acupoint group and LR 3 + PC 6group(n=15rats in each group),and 15 Wistar rats of the same age were used as the normal control group.The filiform needles were inserted into the abovementioned acupoints and non-acupoint,twirled for a while and then retained for 30 min.The treatment was conducted once per day,15 times in total.The rats' tail blood pressure was examined on day 1,3,7and 15 after acupuncture treatment by using a non-invasive blood pressure monitor.At the end of experiment,the contents of Asp and Glu in the RVLM were detected by high-performance liquid chromatography with UV technique.Results On day 1,3,7and 15 after the acupuncture treatment,the raised systolic blood pressure(SBP),diastolic blood pressure(DBP)and mean arterial blood pressure(MBP)of PC 6,LR 3,LR 3 + non-acupoint and LR 3 + PC 6groups were all decreased significantly compared with the model group(P<0.05).On day 15,the effect of the LR 3 + PC 6group was significantly superior to that of the LR 3,PC 6,and LR 3 +non-acupoint groups in reducing the levels of SBP,DBP,and MBP(P<0.05).Compared with the normal control group,the contents of Asp and Glu in RVLM were significantly higher in the model group(P<0.05).After acupuncture intervention,both Asp and Glu levels in the PC 6,LR 3,LR 3 + non-acupoint,and LR 3 + PC 6groups were markedly lower in comparison with the model group(P<0.05),and those of the LR 3 + PC 6group were notably lower than those in the LR 3and PC 6groups(P<0.05).Conclusion Acupuncture stimulation of PC 6,LR 3,LR 3 + non-acupoint and LR 3 + PC 6(in particular)is effective in lowering blood pressure in spontaneous hypertension rats,which may be associated with its effects in lowering Asp and Glu contents in RVLM.
摘要:Objective To observe the effect of electroacupuncture(EA)on pancreatic glucagon-like peptide-1receptor(GLP-1R),pancreatic and duodenal homeobox 1(PDX-1)protein expression and blood glucose in type 2diabetes rats,so as to explore the underlying mechanism of EA treatment in improving type 2diabetes.Methods Sprague-Dawley rats were randomly divided into blank control group,model group,"Weiwanxiashu"(EX-B 3)group,"Xinshu"(BL 15)group,and "Shenshu"(BL 23)group,12 rats in each group.Diabetes model was established by feeding the rat with high fat and high sugar diet combined with intraperitoneal injection of streptozotocin(35mg/kg).All the EA groups received 2Hz,2mA continuous wave treatment for 20 min everyday,6times per week lasting for 4weeks.Fasting blood glucose was measured by Roche glucometer before and after treatment.Hematoxylin and eosin(HE)staining and Masson staining were used to detect pancreas morphology.GLP-1Rand PDX-1protein expressions in the pancreas were detected by Western blot.Results Compared to the blank control group,fasting blood glucose was significantly increased in the model group(P<0.01),accompanied with shrunken islet area,reduced nucleus counts of isletβcells,and compensatorily enlargedβcell nucleus.Compared to the model group,EA intervention significantly reduced fasting blood glucose level only in the EX-B 3group(P<0.05),partly restored pancreas morphology and nucleus counts of isletβcells in the EX-B 3,BL 15,and BL 23 groups.Compared to the blank control group,GLP-1Rand PDX-1expressions were decreased in the model group(P<0.01),while EA treatment could obviously increase GLP-1Rexpression in the EX-B 3(P<0.01),BL 15(P<0.01)and BL 23(P<0.05)groups compared with the model group.The expression of GLP-1Rin the BL 15 group was the highest among the three EA groups(P<0.05,P<0.01),and that in the EX-B 3group was higher than in the BL 23group(P<0.05).There were no signifincant differences in the expression of PDX-1protein among the three EA groups(P>0.05).Conclusion EA treatment at EX-B 3can reduce blood glucose via regulating pancreas function,increasing pancreatic GLP-1Rexpression,and partly restoring the morphology of pancreas.

摘要:Objective To investigate the effects of different electroacupuncture(EA)parameters for the treatment of asthenozoospermia in rats.Methods One hundred and five male Sprague-Dawley rats were randomly divided into 2 Hz-EA treatment daily in 3dgroup(n=9),sham-EA group(n=10),model group(n=10);2Hz-EA treatment every other day in 5dgroup,sham-EA group,model group(8rats in each group);2Hz-EA treatment every other day in 9dgroup,sham-EA group,model group(10rats in each group);100Hz-EA treatment every other day in 9dgroup(n=7),sham-EA group(n=8),model group(n=7).Asthenozoospermia model was established by intragastric administration of ornidazole(ORN,400mg·kg-1·d-1)once daily till the end of treatment.EA treatments(2Hz or 100Hz)were applied to"Shenshu"(BL 23,bilateral),"Zusanli"(ST 36,bilateral)for 30 min,intensity of 1-2-3mA(increasing 1mA per 10min),once a day or once every other day for 3times or 5times.Sham-EA groups were treated with similar procedure except that the output leads of the stimulator were disconnected.The sperm density,viability,motility,the number of grade A sperm,and grade A+B sperm were examined by computer-assisted sperm analysis.Results(1)2Hz-EA treatment daily in 3d:compared with the model group and the sham-EA group,2Hz-EA treatment once daily had no significant effect on all of the sperm motility indexes in the asthenozoospermic rats(P>0.05).(2)2Hz-EA treatment every other day in 5d:compared with the model group,EA treatment could increase the sperm motility(P<0.05),the number of grade A sperm(P<0.05),and the number of grade A+B sperm(P<0.05)in the asthenozoospermic rats.However,compared with the sham-EA group,EA treatment could only improve the number of grade A+B sperm(P<0.05).(3)2Hz-EA treatment every other day in 9d:compared with both the model group and the sham-EA group,EA treatment could markedly improve the sperm viability(P<0.001),the sperm motility(P<0.001),the number of grade A sperm(P<0.001),and the number of grade A+B sperm(P<0.001)in the asthenozoospermic rats.(4)100Hz-EA treatment every other day in 9days:compared with both the model group and the sham-EA group,all of the sperm indexes in the asthenozoospermic rats including the sperm viability(P<0.001 vs.the model group,P<0.05 vs.the sham-EA group),the sperm motility(P<0.001 vs.the model group,P<0.01 vs.the sham-EA group),the number of grade A sperm(P<0.01)and the number of grade A+B sperm(P<0.01)also could be improved after EA treatment.Unexpectedly,none of the EA treatment had significant influence on the sperm density in the asthenozoospermic rats.Conclusion Both 2Hz-EA and 100Hz-EA treatment once every other day for 5times in 9dhad a therapeutic effect on asthenozoospermia by improving the sperm viability and the sperm motility in the rats.
摘要:Objective To observe the effect of electroacupuncture(EA)stimulation of"Fenglong"(ST 40),"Sanyinjiao"(SP 6)plus manual acupuncture(MA)stimulation of"Shuigou"(GV 26)and "Baihui"(GV 20)on Caspase-3protein expression in the cerebral cortex of rats with hyperlipemia and cerebral ischemia(HL-CI),so as to reveal its mechanisms underlying improvement of HL-CI.Methods Forty-five rats were randomly divided into normal control,sham operation,model,EA group I(EA+MA was given for 14 days,i.e.,7days before CI,and 7days more after HL-CI)and EA group II(EA+MA was given for only7 days after HL-CI),with 9rats being in each group.The HL-CI model was established by feeding the animals with high fat forage for 6weeks and then making an occlusion of the unilateral middle cerebral artery by regional application of quantitative paper adsorbing 50% FeCl3solution(10μL).Rats of the sham operation group were treated with the same procedures only without application of FeCl3 solution.For rats of the EA group I,EA(1-3mA,2Hz/100Hz)was applied to bilateral acupoints SP 6and ST 40(for 20min),and MA stimulation applied to GV 26 and GV 20.EA was conducted once daily for 7days after 6weeks' high fat forage feeding,and EA+MA intervention was conducted once daily for 7days after CI modeling.For rats in the EA group II,EA+MA was applied to the same 4acupoints once a day for 7days only after CI modeling.The neurological impairment was assessed by Zea Longa's scoring.The blood sample was taken from the abdominal aorta for measuring the contents of serum cholesterol(CHO),triglyceride(TG)and high-density lipoprotein cholesterol(HDL-C)and low-density lipoprotein cholesterol(LDL-C).Pathological changes of the cerebral cortex were observed after H.E.staining,and the expression of cerebro-cortex Caspase-3was analyzed by immunohistochemistry.Results Following modeling,the neurological score,CHO,TG and LDL-C contents,and the number of Caspase-3positive cells as well as Caspase-3immunoactivity level were significantly increased in the model group(P<0.05),while serum HDL-C level was obviously decreased(P<0.05).After the treatment,the increased neurological score,CHO,TG and LDL-C contents,and the number of Caspase-3positive cells and Caspase-3immunoactivity level were considerably decreased in the EA group I and II(P<0.05)while the decreased HDL-C level was notably increased relevant to the model group(P<0.05).The effects of the EA group I were notably superior to those of EA group II in down-regulating the neurological score,CHO,TG and LDL-C levels and the expression of Caspase-3protein(P<0.05).No significant differences were found between the normal control and sham operation groups in the neurological scores 20 min and 7days after modeling and Caspase-3expression levels(P>0.05).H.E.staining showed a reduction of the apoptotic cells and inflammatory cells in both EA group Ⅰ and Ⅱ.Conclusion Both EA and EA+MA interventions can improve neurological function in HL-CI rats,which may be related to their effects in adjusting the levels of serum lipids and down-regulating the expression of cell apoptosis-related Caspase-3protein in the ischemic cortex.Moreover,the cerebral ischemia injury may be lightened by EA-lowering hyperlipemia first.
摘要:Objective To observe the effect of electroacupuncture(EA)on expressions of nerve growth factor(NGF)and growth arrest-specific protein 7(Gas 7)in the right ventral posterolateral nucleus(VPL)in the focal cerebral ischemia(FCI)rats,so as to explore its possible mechanism underlying improvement of secondary injury of FCI.Methods Forty-eight SpragueDawley rats were randomly divided into normal group,sham operation group,model group and electroacupuncture(EA)group(n=12in each group)by random number table.The FCI model was made by occlusion of the right middle cerebral artery(MCAO)with thread embolus.One week after MCAO,EA(2Hz,2V)was performed on "Baihui"(GV 20)and left"Zusanli"(ST 36)for30min,once daily for successively 21 days.The expressions of NGF and Gas 7in the right VPL were detected by immunohistochemistry(n=6in each group)and Western blot(n=6in each group),respectively;meanwhile Nissl staining was conducted to show VPL neurons.Results Nissl staining showed that the structure of right VPL was clear and complete,and the nuclei were centered and clear in the normal group and sham operation group;the VPL neurons were deeply stained,and the nuclei were pyknotic in the model group;the morphology of neurons in the EA group was similar to that of the normal group.The results of immunohistochemistry and Western blot were consistent.Expressions of NGF and Gas 7proteins in the right VPL were not of significant differences between the sham operation group and the normal group(P>0.05).Compared with the normal group,the expressions of NGF and Gas 7proteins in the VPL of the ischemic side were significantly increased in the model group(P<0.05).After treatment,the expression levels of NGF and Gas 7 proteins were further up-regulated in the EA group in comparison with the model group(P<0.05).Conclusion The enhanced expressions of NGF and Gas 7in the ischemic VPL of FCI rats may be involved in the neuroprotection and repairing;EA can significantly up-regulate the expressions of NGF and Gas 7in VPL of the ischemic side,which may contribute to its effect in improving secondary thalamic impairment of FCI.

摘要:Objective To observe the different effects of electroacupucnture of"Neiguan"(PC 6)grouping with other acupoints on myocardial ischemia(MI)injury,so as to provide evidence for clinical acupoint formulas.Methods Forty Wistar male rats were randomly divided into normal,model,EA 1[bilateral"Neiguan"(PC 6),bilateral"Zusanli"(ST 36),"Guanyuan"(CV 4)],and EA 2 [bilateral"Neiguan"(PC 6),bilateral"Shenmen"(HT 7),"Danzhong"(CV 17)]groups,10 rats in each group.The MI model was established by subcutaneous injection with ISO solution(5mL/kg),once per day for 7days.Both EA 1and EA 2groups were subjected to EA(2Hz/100 Hz,1mA)for 10 min before subcutaneous injection,once daily for 21 days.Electrocardiogram(ECG)was recorded after treatment.Serum cTnT was detected by biochemistry.SOD,GSH-Px and MDA in myocardial tissues of left ventricular were measured by ELISA,and the ultrastructural changes of myocardial cells in the left ventricular were observed by electron microscope.Results Compared with the normal group,the amplitude of S-T segment and HR of ECG,cTnT and MDA contents were significantly increased in the model group(P<0.01)accompanied with seriously destroyed ultrastructure in the myocardial tissue,while SOD and GSH-Px levels were lower than those in the normal group(P<0.01).However,EA treatments reversed all the abnormal changes in the model group(P<0.05).There was no significant difference between EA 1and EA 2groups in the amplitude of S-T segment and HR(P>0.05);cTnT and MDA levels in the EA 1group were lower than those in the EA 2group(P <0.05),while SOD and GSH-Px levels were higher than those in the EA 2group(P<0.05),accompanied with lighter degree of ultrastructure damage in the myocardial tissue in the EA 1group.ConclusionEA can prevent myocardial ischemic injury,and the effect of EA 1treatment was better than that of EA 2.
摘要:Objective To observe the ethology and expression of transient receptor potential vanilloid-1(TRPV 1)in colon of irritable bowel syndrome(IBS)rats treated by electroacupuncture(EA)at"Yintang"(GV 29)and"Tianshu"(ST 25),so as to explore the different effectiveness of different acupoints and its related mechanism underlying improvement of the symptoms of somatopsychic illness.Methods Thirty-two neonatal Wistar rats were randomly divided into blank control group,model group,Yintang(GV 29)group,and Tianshu(ST 25)group(n=8in each group).IBS model was established by neonatal maternal separation and acetic acid enema combined with colon and rectum expansion stimulation.Rats of the GV 29 and ST 25 groups were given EA treatment at age of 9weeks old,20 minutes,once every other day for 5times.Latency of the 1st time contraction wave and numbers of contractive wave in 90 swere recorded to evaluate abdominal visceral sensitivity by abdominal withdrawal reflex.Horizontal and vertical movements were observed to assess the emotional and psychological behavior of rats by open field test.TRPV 1expression in colon was detected by immunohistochemistry method.Results Compared with the black control group,latency of the 1st time contractive wave was significantly shortened and the wave numbers in 90 swere increased significantly in the model group(P<0.01),while the numbers of vertical and horizontal movements were also significantly reduced(P<0.01),accompanied with increased TRPV 1expression in the colon(P<0.05).Compared with the model group,After EA intervention,the levels of the above-mentioned 5indexes were significantly reversed in the GV 29 and ST 25groups(P<0.01,P<0.05),except the number of horizontal movement in the ST 25group(P>0.05).The expression level of TRPV 1was significantly lower in the ST 25 group than in the GV 29group(P<0.01).Conclusion EA at GV 29 and ST 25 can alleviate the abdominal pain and improve the mental and emotional disorders in IBS rats.GV 29 has a better effect on relieving the depressivelike psychoemotional behavior of IBS rats,while ST 25 is more effective in treating abdominal pain.There is significantly different influence on TRPV 1expression in colon between these two treatment groups,which may contribute to their different effect in pain relieving.
摘要:Objective To observe the effect of acupoint injection on eosinophils(EOS)counts and expression of eotaxin in nasal mucosa of allergic rhinitis(AR)rats,so as to reveal its mechanism underlying improving AR.Methods Twenty-four Sprague-Dawley(SD)rats were randomly divided into normal,model and acupoint injection groups(n=8in each group).The AR model was established by intraperitoneal injection of ovalbumin sensitization.Bilateral"Yingxiang"(LI 20)and"Yintang"(GV29)were selected for acupoint injection of the mixture solution of lidocaine,dexamethasone,and transfer factor(0.1mL/acupoint)on the 1st,5th,9th,and 13 th day after AR model established,a total of four times.EOS in the nasal mucosa was counted under light microscope after HE staining.Protein and mRNA expressions of eotaxin in the nasal mucosa were detected by immunohistochemical and RT-PCR methods,respectively.Results Compared with the normal group,EOS counts,protein and mRNA expressions of eotaxin in the nasal mucosa were significantly higher in the model group(P<0.05).Compared with the model group,EOS counts,protein and mRNA expressions of eotaxin in the nasal mucosa were significantly lower in the acupoint injection group(P<0.05).Conclusion Acupoint injection can reduce the nasal mucosa inflammation by suppressing the protein and mRNA expressions of eotaxin,decreasing the infiltration and gathering of EOS in the nasal mucosa.
摘要:Objective To observe the expression of catechol-O-methyltransferase(COMT)in inferior colliculus and auditory cortex of guinea pigs with age-related hearing loss(AHL)induced by D-galactose,so as to explore the possible mechanism of electroacupuncture(EA)underlying preventing AHL.Methods Thirty 3-month-old guinea pigs were randomly divided into control group,model group and EA group(n=10in each group),and ten 18-month-old guinea pigs were allocated as elderly group.The AHL model was established by subcutaneous injection of D-galactose.EA was applied to bilateral"Yifeng"(SJ 17)and"Tinggong"(SI 19)for 15 min in the EA group while modeling,once daily for 6weeks.After treatment,the latency of auditory brainstem response(ABR)Ⅲ wave was measured by a brain-stem evoked potentiometer.The expressions of COMT in the inferior colliculus and auditory cortex were detected by Western blot.Results Compared with the control group,the latencies of ABRⅢwave were significantly prolonged and the expressions of COMT in the inferior colliculus and auditory cortex were significantly decreased in the model group and the elderly group(P<0.05).After the treatment,the latency of ABRⅢ wave was significantly shortened and the expressions of COMT in the inferior colliculus and auditory cortex were significantly increased in the EA group in comparison with the model group(P<0.05).Conclusion EA at"Yifeng"(SJ 17)and "Tinggong"(SI 19)can improve the hearing of age-related deafness in guinea pigs,which may contribute to its effect in up-regulating the expression of COMT in the inferior colliculus and auditory cortex.
摘要:Objective To observe the influence of moxibustion on serum interleukin-17(IL-17)and tumor necrosis factor alpha(TNF-α)levels in collagen-induced arthritis(CIA)rats,so as to study its mechanism underlying improvement of rheumatoid arthritis.Methods A total of 40 male Wistar rats were used in the present study,and 8rats were randomly selected as a normal control group.The other 32 rats were modeled.The primary immunity emulsion was made with mixed TypeⅡchicken collagen and complete Freund's adjuvant,and 0.3mL emulsion(containing 0.3mg collagen)was injected equally into left pelma,tail root and the back.Seven days after the primary immune,the same procedure was conducted to induce the secondary immunity,and the emulsion was made with mixed TypeⅡ chicken collagen and incomplete Freund's adjuvant.The whole course of modeling lasted 21 days.And then 24 CIA rats were randomly divided into model group,medication group and moxibustion group(n=8in each group).For those of the moxibustion group,suspended moxibustion with 20 mm distance above"Zusanli"(ST 36)and"Kunlun"(BL 60)was performed for 20min/acupoint,once daily,alternately on left and right hind limbs for 10 consecutive days.For those of the medication group,gavage of methotrexate(0.1mg/100g)was administrated once every 5days,and totally two times.Left ankle joint diameter and body weight were detected,and X-ray of left tarsus was observed in each group before and after modeling or after treatment.Serum levels of IL-17 and TNF-αwere determined by ELISA kits.Results After modeling,the left ankle diameter and serum concentrations of IL-17 and TNF-αincreased(P<0.05),and the body weight decreased(P<0.05)in the model group compared to the control group,combined with the tarsus soft tissue swelling,joint space narrowing,bone destruction seen from the tarsal X-ray.After intervention,the ankle diameter,the serum IL-17 and TNF-αlevels decreased(P<0.05),and the body weight increased(P<0.05)in both medication and moxibustion groups compared to the model group;meanwhile the tarsus soft tissue swelling and the bone deformity turned to be improved.There were no significant differences between the medication group and the moxibustion group in above mentioned indexes(P>0.05).Conclusion Moxibustion is effective in CIA rats,and the mechanism may be related to the reduction of serum IL-17 and TNF-αlevels.
摘要:Objective To observe the effect of acupoint application on transforming growth factor(TGF)-β1/Smad 3signaling pathway,so as to reveal its mechanism underlying improvement of airway remodeling in chronic asthma mice.Methods BALB/c mice were randomly divided into normal group,model group,sham acupoint application group(sham group),acupoint application group,and dexamethasone group(n=8in each group).The chronic asthma model was established by intraperitoneal injection of ovalbumin sensitization.The acupoint application group was treated with plaster composed with White Mustard Seed,Rhizoma Corydalis,Unprocessed Radix Kansui,and Herba Asari,on bilateral"Feishu"(BL 13),"Xinshu"(BL 15)and"Geshu"(BL 17)points,6hours a day for 14 days.The sham group was treated with plaster containing vaseline on bilateral BL 13,BL 15 and BL 17,6hours a day for 14 days.The dexamethasone group was treated with dexamethasone 1mg·kg-1·day-1 intraperitoneally,once daily for 14 days.At the end of treatment,airway resistance was detected by mouse lung function instrument.Patho-logical changes of airway were observed under light microscope after hematoxylin-eosin staining,and the morphological parameters were measured by image analysis software.Immunohistochemistry method was used to detect the expression of TGF-β1and Smad 3in the lung tissue of mice.Results Compared with the normal group,the airway resistance,the total bronchial wall area(WAt)/lumen perimeter(Pi),the smooth muscle wall area(WAm)/Pi,and the expression of TGF-β1and Smad 3in the lung tissues were significantly higher in the model and sham groups(P<0.01).Compared with the model group and the sham group,the airway resistance,WAt/Pi,WAm/Pi,the expression of TGF-β1and Smad 3in the lung tissues were decreased in the acupoint application group and the dexamethasone group(P<0.05).There were no significant differences between the acupoint application group and the dexamethasone group in the above mentioned indexes(P>0.05).Conclusion Acupoint application can improve airway remodeling by down-regulating the expression of TGF-β1/Smad 3in the airway of chronic asthmatic mice.
摘要:Objective To observe the effect of moxibustion at"Feishu"(BL 13)and"Shenshu"(BL 23)on peripheral blood T cells and serum interleukin in rats with asthma,so as to explore its immunological mechanisms in relieving asthma.Methods Thirty SD rats were randomly divided into normal,model and moxibustion groups(10rats/group).Asthma model was established by intraperitoneal injection of ovalbumin and aluminum hydroxide,and ovalbumin inhalation.Moxibustion was applied to bilateral"Feishu"(BL 13)and"Shenshu"(BL 23)for 30 min,once daily for 14 d.The pathologic changes of the lung tissue were detected by H.E.staining.The levels of CD 3+,CD 4+,CD 4+,CD 8+,CD 8+T cells in whole blood were detected by flow cytometry.Serum IgE,IL-1β,IL-1Ra contents were assayed by ELISA(enzyme-linked immunosorbent assay).Results The thickened muscle of bronchial wall,mucosal edema,infiltration of inflammatory cells and partial destruction of the alveolar structure were found in the model group.While in the moxibustion group,the structure of the alveolar wall was complete,the morphology of the bronchioles was regular,and there were no mucus plug formation and epithelial cell shedding in the lumen.The contents of blood CD 8+T cells in whole blood were detected by flow cytometry.Serum IgE,IL-1β,IL-1Ra contents were assayed by ELISA(enzyme-linked immunosorbent assay).Results The thickened muscle of bronchial wall,mucosal edema,infiltration of inflammatory cells and partial destruction of the alveolar structure were found in the model group.While in the moxibustion group,the structure of the alveolar wall was complete,the morphology of the bronchioles was regular,and there were no mucus plug formation and epithelial cell shedding in the lumen.The contents of blood CD 8+and CD 3+and CD 3+T cells,and serum IgE and IL-1βin the model group were obviously higher than those in the normal group(P<0.01).Compared with the model group,the levels of blood CD 8+T cells,and serum IgE and IL-1βin the model group were obviously higher than those in the normal group(P<0.01).Compared with the model group,the levels of blood CD 8+T cell,serum IgE and IL-1βin the moxibustion group were all decreased(P<0.05,P<0.01),meanwhile the levels of blood CD 4+T cell,serum IgE and IL-1βin the moxibustion group were all decreased(P<0.05,P<0.01),meanwhile the levels of blood CD 4+T cell and serum IL-1Ra were increased(P<0.01,P<0.05).Conclusion Moxibustion can reduce the levels of serum IgE and IL-1β,and increase the level of serum IL-1Ra,which may play an important role in the treatment of asthma.

摘要:Objective To observe the clinical efficacy of modified acupuncture at Renying point(ST 9)for patients with cervical spondylosis of vertebral artery type and its influence on velocity of cervical blood flow.Methods Fifty-nine cases of vertebral artery type cervical spondylosis were randomly divided into control group(n=30)and treatment group(n=29).Both groups were acupunctured at ST 9,with routine acupuncture technique used in the control group and modified technique in the treatment group,respectively.All cases received two courses of treatment,each course covered consecutive 6once-perday treatments.Before and after treatment,transcranial Doppler(TCD)was used to measure the systolic peak blood flow velocity(Vs)of left vertebral artery(LVA),right vertebral artery(RVA)and basilar artery(BA),and the scores of"cervical vertigo symptoms and functional assessment scale"(CVSFAS)were also assessed,separately.Results CVSFAS scoring,Vs of LVA,RVA and BA after treatment showed significant improvement compared with those before treatment(P<0.01,P<0.05).The efficacy of the treatment group in the above mentioned indexes was superior to that of the control group(P<0.05).The total effective rate of the treatment group was 93.1%(27/29),superior to 70.0%(21/30)of the control group(P <0.05).Conclusion The modified acupuncture method at ST 9is clinically effective in the treatment of cervical spondylosis of vertebral artery type via increasing the Vs of vertebral-basilar artery,improving the local blood circulation and relieving pain.
摘要:Objective To observe the clinical effect of vitalstim electroacupuncture(EA)combined with swallowing rehabilitation training in the treatment of stroke patients with dysphagia.Methods A total of 80 stroke patients with dysphagia were randomized into treatment and control groups(n=40in each group).Patients of the control group were treated by regular medication for anti-platelet aggregation and anti-coagulation,lipid-lowering,neuroprotection,blood glucose control and blood pressure control,etc.and swallowing function rehabilitation training,and those of the treatment group treated by EA stimulation of Fengchi(GB 20),Jinjin(EX-HN 12)and Yuye(EX-HN 13)with a Vitalstim Electrostimulator and manual acupuncture stimulation of Lianquan(CV 23),Tiantu(CV 22)in combination with regular medication plus swallowing function training as those mentioned in the control group.The EA and manual acupuncture stimulation treatment was conducted once daily,6times a week and 4weeks altogether. The therapeutic effect was assessed by using Kubota swallowing ability test(6levels),dysphagia subscale(0-6scores)of the neurological deficit degrees,videofluorography(VFG)assessment(markedly effective,effective and invalid,for evaluating the function and symmetry state of the swallowing movements),and the MOS Item Short Form Health Survey(SF-36,8minor items of two major aspects in physiological function,mental health,emotional function,social function and overall health)for assessing the patients' daily-life quality.Results After the treatment,the dysphagia score of the treatment group was significantly lower than that of the control group(P<0.05).VFG outcomes showed that,of the two 40 patients in the control and treatment groups,16 and 23experienced a marked improvement,20 and 15were effective,4and 2were ineffective,with the markedly effective rate being 40.0% and 57.5%,respectively.The daily-life quality scores for physiological function,mental health,emotional function,social function and overall health were all notably increased after the treatment in both groups,particularly in the treatment group(P<0.05).The therapeutic effects of the treatment group were remarkably superior to those of the control group in improving dysphagia(showed by dysphagia score and VFG outcomes)and life quality.Conclusion EA treatment combined with swallowing function rehabilitation training is effective in improving swallowing ability and daily-life quality in stroke patients with dysphagia.

摘要:Objective To observe the clinical effectiveness of acupuncture intervention in weight reduction by modulating body compositions in obesity patients.Methods A total of 71 obesity patients during weight-loss procedure were allocated to acupuncture+nutrition-consultation group(n=40)and simple nutrition-consultation group(n=31).The patients of the acupuncture +nutrition-consultation group were treated by acupuncture stimulation of Zhongwan(CV 12),Xiawan(CV 10),Tianshu(ST 25),Wailing(ST 26),Qihai(CV 6),Guanyuan(CV 4),etc.for 30 min,once every other day,3times per week,12 times altogether,and also given with weekly nutrition consultation(including subjective query,objective measurement,analysis,program for nutrition support)at the same time.The patients of the simple nutrition-consultation group were treated by only weekly nutrition consultation for 4weeks.Before and after the treatment,the patients' body weight,body mass index(BMI),fat mass,percentage of body fat,muscle mass,protein quality,water quality and bone mass were measured by using a composition analyzer.Results After 4weeks' treatment,the body mass,BMI,fat mass and fat percentage in both acupuncture+nutrition-consultation and simple nutrition-consultation groups were significantly decreased(P<0.01),while the weight levels of muscle,protein,bone and water content had no apparent changes(P>0.05).The therapeutic effect of acupuncture+nutrition-consultation group was markedly superior to that of the simple nutrition-consultation group in increasing the improved degrees of body weight,BMI,fat mass and fat percentage(P<0.01).Conclusion Acupuncture plus nutrition consultation is effective in reducing body mass,fat mass and percentage of body fat in obesity patients.
摘要:Objective To review systematically the clinical effects of spastic paralysis after stroke treated with acupuncture at meridian sinew("Jingjin",musculotendon).Methods"Meridian sinew""stroke"and"spasm"were taken as the key words to retrieve from the Chinese National Knowledge Infrastracture Database(CNKI),Chongqing VIP Chinese Science and Technology Periodical Database(VIP),Chinese Biomedical Library(CBM),Wanfang Data,PubMed and the Cochrane Library.The Cochrane"risk of bias"tool was used to conduct the methodological quality evaluation to the literature.RevMan 5.3software was adopted for Meta-analysis.Results Totally,13papers were included,with 820patients involved.In reference to Cochrane Reviewers’Handbook 5.0.2,the randomized controlled trial(RCT)risk of bias was assessed and it was unclear for all of the 13papers.The results of Meta-analysis showed that the clinical effect was improved with acupuncture at meridian sinew as compared with normal acupuncture technique[(1)total effective rate:OR=3.86,95%CI(2.67,5.57),Z=7.20,P<0.00001;(2)modified Ashworth spasm scale:OR=4.54,95%CI(2.91,7.10),Z=6.64,P<0.00001;(3)evaluation of limb motor function with Fugl-Meyer score:MD=4.18,95%CI(-0.59,8.94),Z=1.72,P=0.09>0.05].The publication bias of included papers was not obvious and therefore it could be neglected in the impact on the combined effect size.Conclusion Acupuncture at meridian sinew is effective in the treatment of spastic paralysis after stroke.The total clinical effect and the improvement in muscular tone with acupuncture at meridian sinew are better than those with normal acupuncture technique.The quality of the included literature is not high generally.Hence,it is necessary to have more clinical studies with high-quality and strict design.

摘要:Electroacupuncture(EA)has been extensively used in clinical treatment of obesity.The effectiveness relies not only on the correct diagnosis and rational treatment,but also on the voluntary cooperation of patients,that is the satisfactory compliance.The patient compliance refers to his/her submissiveness and executive ability to medical advice,which is the indispensable factor for the clinical efficacy.The strength of it depends on the patient himself/herself,and moreover is improved through some therapeutic strategies.Based on the emphasis of individual treatment in the real world,the practical randomized controlled trial(RCT)study pays attention to patient's compliance at the beginning of trial design.In this paper,in terms of the influential factors and control strategies,the compliance of simple obesity patient in the practical RCT study was summarized systematically.
摘要:As an analogy,aprototype of"bell-striking"is proposed in the present paper for exploring the basic properties,major elements,and potential mechanisms of acupuncture stimulation.On the strength of analysis on the physiological basis of acupuncture effect,several fundamental aspects of acupuncture are summarized as a)the body-surface stimulating characters,b)general and local effects,and c)triggering the auto-regulative function of the organism,which mimics the "bell-striking"response.Namely,when stroke,bell will chime,otherwise,chiming will not be heard.During analyzing special contents of acupuncture theory,its formative background should not be separated,and it is improper to take,modern medical theory of the human body as the guiding thinking way for researching the ancient Chinese medical literature.
