摘要:Objective To observe the effect of"Huayu Tongluo"(Blood-stasis Dispersing and Meridian-collateral Dredging)moxibustion on the delayed memory and expression of Nestin and Doublecortin(DCX)proteins in the hippocampus in vascular dementia(VD)rats in the view of neurogenesis produced by intracerebral transplantation of neural stem cells(NSCs)and endothelial progenitor cells(EPCs).Methods Healthy male Wistar rats were randomized into control group,VD model group,NSCs+EPCs group and NSCs+EPCs moxibustion group.The VD model was established by using a modified 2-vessels occlusion method,and neurogenesis was produced by transplantation of NSCs+EPCs(2×106 cell/10 μL)into the lateral ventricle for rats of the NSCs+EPCs groups 3 days after successful VD-modeling.Moxibustion was applied to"Dazhui"(GV 14),"Baihui"(GV 20)and"Shenting"(GV 24)once daily for 21 days with an interval of one day between every two 7 days.The Morris Water Maze was used to test the rat's delayed memory ability before and 24 hafter the treatment.The expression of Nestin and DCX proteins in the hippocampus tissues was detected using double-labeled immunofluorescence technique.Results Following modeling,Morris Water Maze tests showed that the average escape latency of location navigation task was significantly prolonged in VD rats(P<0.008)and the times of target platform crossing(spatial probing task)within 120 swere remarkably reduced in VD rats(P<0.008).Compared with pre-treatment in the same one group,the escape latency of NSCs+EPCs and NSCs+EPCs moxibustion groups were considerably reduced(P<0.05),and the average times of target platform crossing of the NSCs+EPCs moxibustion group were markedly increased(P<0.05).The effect of NSCs+EPCs moxibustion was evidently superior to that of simple NSCs+EPCs in shortening the escape latency(P<0.008).The expression levels of Nestin protein were significantly higher in the NSCs+EPCs moxibustion group after 1 and 3 period treatment than those in the NSCs+EPCs group(P<0.05).Conclusion Moxibustion intervention is able to improve the delayed memory in VD rats,which may be related to its effect in up-regulating the expression of hippocampal Nestin and DCX proteins within 15 days via accelerating neurogenesis.
摘要:Objective To observe the effect of electroacupuncture(EA)combined with Gastrodin on learning-memory ability and expression of silent information regulator 2 homologous protein 1(SIRT 1)and peroxisome proliferator activated receptorγcoactivator(PGC-1α)of hippocampal CA 1 region in Alzheimer's disease(AD)rats,so as to explore its mechanism underlying improvement of AD.Methods Sixty male SD rats were randomly divided into normal control(normal),sham operation(sham),model,EA,Gastrodin and EA+ Gastrodin groups(n=10 in each).The AD model was established by intraperitoneal injection of D-Galactose(120 mg·kg(-1)·d(-1)·d(-1))combined with bilateral hippocampal injection ofβamyloid 1-40(Aβ1-40).EA was applied at"Baihui"(GV 20),"Dazhui"(GV 14)and"Zusanli"(ST 36)for 30 min,once daily for 4 weeks.For rats of the Gastrodin group and EA+ Gastrodin group,intraperitoneal injection of gastrodin(10 mg/kg)was conducted once daily for 4 weeks.Morris water maze tests were used to assess the rat's learning-memory ability.Nissl staining was used to assess the morphological changes of neurons in the hippocampal CA 1 area.The expression of SIRT 1 and PGC-1αof hippocampal CA 1 region was measured by immunohistochemical staining.Results 1)Morris water maze tests showed that,compared with the normal and sham group,the escape latency was significantly prolonged(P<0.05),and the percentage of platform quadrant residence duration and the platform crossing times were considerably decreased in the model group(P<0.05).After the intervention,the escape latency was obviously shortened(P<0.05),and the percentage of platform quadrant residence duration and the platform crossing times were markedly increased in the EA,Gastrodin and EA+Gastrodin groups relevant to the model group(P<0.05).2)Nissl staining showed that,in comparison with the normal group or sham group,the number of cells in the hippocampal CA 1 area was decreased and the arrangement was disorganized in the model group.The number of cells in CA 1 area was relatively higher in the3 treatment groups than in the model group.3)The expression levels of SIRT 1 and PGC-1αproteins in the hippocampal CA 1 area were significantly down-regulated in the model group than in the normal and sham groups(P<0.05).After the intervention,the expression levels of SIRT 1 and PGC-1αin the EA,Gastrodin and EA+Gastrodin groups were significantly up-regulated compared with the model group(P<0.05).The effects of EA+Gastrodin were significantly superior to those of simple EA and simple Gastrodin in shortening the escape latency,up-regulating the expression levels of SIRT 1 and PGC-1αas well as in increasing the percentage of platform quadrant residence time and platform crossing times(P<0.05).Conclusion Both EA and Gastrodin can improve the learning-memory ability of AD rats,which may be related to their effects in up-regulating the expression of SIRT 1 and PGC-1αand reducing neuronal injury in the CA 1 region of hippocampus,suggesting aprotective role of EA on hippocampal neurons.The effect of EA combined with Gastrodin is markedly better than that of EA and Gastrodin alone.

摘要:Objective To observe the effect of acupuncture on the expression of silent information regulator factor 2-related enzyme 1(SIRT 1)and nuclear factor-κB(NF-κB)in rats with acute cerebral ischemia(ACI),so as to explore its mechanisms underlying improvement of ACI.Methods One hundred female SD rats were randomly divided into 5 groups:normal control(normal),sham-operation(sham),model,non-acupoint and acupoint,with 20 rats in each.The ACI model was established by occlusion(electric coagulation)of the middle cerebral artery after craniotomy."Baihui"(GV 20)and "Shuigou"(GV 26)or non-acupoints were punctured with filiform needles which were retained for 30 min after rotating for 1 min.The treatment was conducted once after modeling and 24 hthereafter.The cerebral infarct volume was measured after 2,3,5-triphenyltetrazolium chloride(TTC)staining.The contents of interleukin-1β(IL-1β),IL-6 and IL-8 in the serum and ischemic brain tissue were detected by radioimmunoassay,and the expression of protein SIRT 1 and NF-κB p 65 in the ischemic brain tissue was detected by immunoblotting.Results The brain infarction volume was obvious in the model group in comparison with the normal group(P<0.01),but was markedly reduced in the acupoint group relevant to the model group(P<0.05).The contents of IL-1β,IL-6 and IL-8 in the serum and ischemic brain tissues,and the expression level of cerebral NF-κB p 65 protein were significantly increased(P<0.01),and the expression of cerebral SIRT 1 was obviously decreased in the model group compared with the normal group(P<0.01).Following acupuncture intervention,the increased IL-1β,IL-6 and IL-8 contents and NF-κB p 65 expression were considerably down-regulated,and the decreased SIRT 1 expression was markedly up-regulated in the acupuncture acupoint group(P<0.05).No significant changes were found in the levels of all the indexes mentioned above between the normal and sham groups(P>0.05).Conclusion Acupuncture intervention at acupoints can reduce the ischemic infarction volume in ACI rats,which may be associated with its effects in down-regulating the levels of IL-1β,IL-6 and IL-8 in the serum and ischemic brain tissue,and in regulating cerebral SIRT 1/NF-κB signaling.
摘要:Objective We have repeatedly demonstrated that electroacupuncture(EA)of"Neiguan"(PC 6)can improve myocardial ischemia in rats.The present study was designed to investigate the metabolomic profile of peripheral blood serum and myocardium involving EA-induced improvement of myocardial ischemia-reperfusion injury(MIRI)in rats by using nuclear magnetic resonance spectroscopy.Methods Thirty male SD rats were equally randomized into blank control,model and EA groups.Rats of the control group were only banded for 20 min,once a day for 7 days.The MIRI model was established by occlusion of the anterior descending branch of the left coronary artery for 40 min,followed by reperfusion for 60 min,and rats of the model group were banded as those in the control group.EA(10 Hz/50 Hz,1 mA)was applied to bilateral PC 6 for 20 min,once daily for 7 days.The blood samples and left ventricular myocardial tissues were collected for assaying the profiles of differential metabolites using 1 H nuclear magnetic resonance(1 H NMR)spectroscopy and multivariate statistical analysis such as the principal components analysis(PCA),partial least squares-discriminant analysis(PLS-DA)and orthogonal PLS-DA(O-PLS-DA)with SIMCA-P software 12.0.Results A total of 19 differential metabolites(17 down-regulated,2 up-regulated)in the serum and 14 differential metabolites(13 down-regulated and 1 up-regulated)in the ischemic left myocardium were identified after MIRI.Of the 19 serum differential metabolites,amino acids(leucine,isoleucine,valine,alanine,lysine,glycine,glutamine),3-hydroxy butyric acid(3-HB),lactic acid,acetate,N-acetyl glycoprotein(NAc),acetone,acetoacetate,succinate,polyunsaturated fatty acids(PUFA),creatine,glycerophosphocholine(GPC)were down-regulated;while low density lipoprotein(LDL),LDL/very low density lipoprotein(LDL/VLDL)and glucose obviously up-regulated.Of the 14 myocardial differential metabolites,amino acids(alanine,lysine,glutamate,glutamine,aspartate,taurine,glycine,threonine),GPC,creatine,lactic acid,adenosine monophosphate(AMP),nicotinamide adenine dinucleotide(NAD+)were significantly decreased,and glucose was up-regulated.Following EA treatment,most of the decreased serum differential metabolites except acetone,acetoacetate and PUFA,and the increased serum LDL,LDL/VLDL and glucose recovered,basically close to the control level;and the decreased myocardial creatine,GPC and NAD+ were also apparently up-regulated and the increased myocardial glucose was down-regulated.But,myocardial threonine and AMP still presented a decreasing state.Although the pattern of myocardial differential metabolites of the EA group had a trend to be close to the control group,the significant difference still existed,while the metabolic pattern of serum metabolites in the EA group was close to that of the control group.Conclusion EA stimulation of PC 6 can regulate serum or/and myocardial metabolites as amino acids,carbohydrates,lipids,etc.in MIRI rats,of which both serum and myocardial creatine,GPC and glucose may be jointly confer a favorable potential for EA-induced improvement of MIRI.

摘要:Objective To assess the effect of electroacupuncture(EA)on chronic inflammatory response of Leydig cells in aged rats with low testosterone,so as to investigate its underlying mechanism of anti-male reproductive aging.Methods Twenty-four 20 months old SD rats were randomly divided into EA,medication and aged control groups(n= 8 in each),and other 8 young SD rats(2 months of age)were used as the youth control group.EA(2 Hz/100 Hz,1 mA)was applied to"Guanyuan"(CV 4)and bilateral"Shenshu"(BL 23)for 15 min,once daily for 8 weeks except the weekends.The medication group received abdominal subcutaneous injection of testosterone propionate(mg·kg(-1)·3d(-1)·3d(-1))for 8 weeks.The aged control group and the youth control group received subcutaneous injection of 0.9% normal saline,with the same dose and same treatment frequency as those of the medication group.The rats'physical power was assessed according to the exhausted swimming duration,and the levels of serum total testosterone(TT)and free testosterone(FT)were determined by ELISA.The pathological changes of the testis tissue were detected by using H.E.staining,and the immunoactivity of cyclooxygenase-2(COX-2)in Leydig cells was detected by immunohistochemistry.The expression levels of nuclear factor-κB p 65(NF-κB p 65),COX-2,interleukin-1β(IL-1β)and tumor necrosis factor-α(TNF-α)proteins in the testis tissues were determined by Western blot.Results Before and after treatment,the exhaustive swimming duration and the levels of serum TT and FT in the aged control group were significantly lower than those of the youth control group(P <0.01).After the treatment,the exhaustive swimming duration and serum TT and FT in the EA and medication groups were notably higher than those in the aged control group(P<0.01).HE staining showed that the incompleteness of basement membrane of spermatogenic tubules,reduction of spermatogenic cells and supporting cells and irregularity of Leydig cells in the testis tissue of the aged rats were relatively milder after EA intervention.Compared with the youth control group,the expression levels of NF-κB p 65 and COX-2,IL-1βand TNF-αin the testicular tissue were significantly higher in the aged control group(P<0.01),while in compared with the aged control group,the expression levels of NF-κB p 65,COX-2,IL-1βand TNF-αproteins were significantly down-regulated in the EA group(P<0.01).Conclusion EA intervention can improve the physical power of the aged rats with low testosterone,which may be related to its effects in up-regulating TT and FT levels,and in reducing chronic inflammatory response in the testis tissue.

摘要:Objective To observe the effect of different strength of electroacupuncture(EA)stimulation on gastrointestinal motility and Ras homolog gene family member(RhoA)/Rho associated coiled-coil forming protein kinase(ROCK)signaling in diabetic gastroparesis(DGP)rats,so as to reveal the underlying mechanisms of EA for improving DGP.Methods Sixty SD rats were randomly and equally divided into blank control,DGP model,weak EA,medium EA,and strong EA groups(n=12 rats in each).The DGP model was established by intraperitoneal injection of streptozotocin(STZ,55 mmol/kg,2%)and high-sugar and high-fat fodder feeding for 8 weeks.EA(0.12,0.24,0.36 mA,20 Hz/100 Hz)was applied to"Zusanli"(ST 36),"Sanyinjiao"(SP 6)and"Liangmen"(ST 21)for 20 min,once daily for 15 successive days.Blood glucose levels were measured weekly with blood glucose meter and blood glucose test paper.Fecal phenol red excretion method was used to display gastric emptying and small intestinal propulsion function.The expression of RhoA protein in the gastric antral smooth muscle tissue was detected by immunohistochemistry and Western blot(WB),separately,and that of ROCK,myosin phosphatase target subunit 1(MYPT 1)and phosphorylated(p)-MYPT 1 proteins in gastric antrum detected by WB.Results Compared with the blank control group,the gastric emptying rate and small intestine propulsion rate of the model group were significantly decreased(P<0.05),and the blood glucose level was remarkably increased(P<0.05).Moreover,the expression levels of RhoA,ROCK,MYPT 1 and p-MYPT 1 proteins in the gastric antrum were significantly down-regulated relevant to the control group(P<0.05).After administration of EA,the decreased gastric emptying rate and intestinal propulsion rate,and the down-regulated expression of RhoA,ROCK,MYPT 1 and p-MYPT 1 proteins were significantly increased in the strong,medium and weak EA stimulation groups(P<0.05).Comparison among the3 EA groups showed that the strong stimulation was significantly superior to weak stimulation in up-regulating the expression of RhoA,ROCK,MYPT 1 and p-MYPT 1 proteins,and obviously superior to the medium stimulation in up-regulating RhoA and MYPT 1 protein levels(P<0.05),while the medium stimulation was significantly stronger than the weak stimulation in up-regulating the expression of ROCK,MYPT 1 and p-MYPT 1 proteins(P<0.05).There were no significant differences among the 3 EA groups in up-regulating the gastric emptying rate and small intestinal propulsion rate,and between the strong stimulation and medium stimulation in the expression levels of ROCK and p-MYPT 1 proteins(P>0.05).Conclusion Electroacupuncture stimulation of ST 36-SP 6-ST 21 at 0.12,0.24 and 0.36 mA can promote the gastrointestinal motility in DGP rats,which may be associated with its effects in enhancing RhoA/ROCK signaling in the gastric antral smooth muscle at different degrees.
摘要:Objective To observe the effect of electroacupuncture(EA)stimulation of Heart and Lung meridians on the expression of hyperpolarization-activated cyclic nucleotide gated potassium channel 2(HCN 2)mRNA and protein in the myocardium of the left ventricle in acute myocardial ischemia(AMI)rats,so as to underlying its mechanism in improving ischemic myocardial injury.Methods A total of 120 male SD rats were randomly divided into normal control,model,EA-Heart Meridian(HT)and EA-Lung Meridian(LU)groups(n=30 rats in each group).The AMI model was established by occlusion of the anterior descending branch of coronary artery.EA(1 mA,10 Hz)was applied to bilateral"Shenmen"(HT 7)or"Taiyuan"(LU 9)and the spot about 1 mm apart from the HT 7 or LU 9 on the axopetal end for 15 min on the second day after modeling,once a day for 7 days.The left ventricular myocardium tissue was sampled for determining the expression levels of HCN 2 mRNA and protein with qPCR and Western blot,separately.Results After AMI,both HCN 2 mRNA and protein expression levels in the model group were significantly decreased in comparison with the normal control group(P<0.01).Following EA intervention,the expression levels of HCN 2 mRNA and protein expressions were significantly up-regulated relevant to the model group(P<0.01).The effects were more obvious in the EA-HT group than in the EA-LU group,and the expression levels were higher in the left ventricular myocardium tissue sampled on the next day after the treatment than those in the tissue sampled immediately after the treatment(P<0.01).Conclusion Both EA stimulation of Heart Meridian and Lung Meridian can improve the expression of HCN 2 mRNA and protein in the myocardium in AMI rats,which existing a delayed effect and meridian specificity.
摘要:Objective To evaluate the influence of superficial needling on electroencephalogram(EEG)in patients with insomnia by using nonlinear kinetics measure.Methods Six patients(3 male and 3 female)suffering from insomnia for more than a month were recruited in the present study.EEG data before and during superficial acupuncture stimulation of Shangen point(the midpoint between the bilateral inner canthus)was recorded using an electroencephalograph for analyzing the correlation dimensionality(D 2)and approximate entropy(ApEn)of different cerebral regions.Results During superficial needling,the D 2 data of the right-forehead(Fp 2),anterior area of the left temple(F 7)and the anterior region of the right temple(F 8)were significantly increased(P<0.05),and ApEn data at the right anterior forehead(Fp 2),left occipital region(O 1),and F 8 region markedly decreased relevant to pre-acupuncture(P<0.05).Both paring test and trend analysis of D 2 data showed no significant changes.Only an obvious increase of D 2 was found in the Fp 2 region,suggesting a coherence of EEG activities in stability and synchronization during acupuncture treatment.Clustering analysis of D 2 data of various cerebral regions displayed a relatively concentrated tendency,particularly in the Fp 2,the right forehead(F 4)and F 8 regions.Factor analysis of ApEn data showed a significant change in Fp 2,F 8,O 1 and right occipital(O 2)regions.It suggests that during superficial needling stimulation of Shangen point,the EEG signals were lowered in complexity,and improved in synchronization,stabilization and ordering,favoring sleep at last.Conclusion Superficial needling of Shangen point can make the EEG signals synchronized in insomnia patients,suggesting an improvement of sleeping.
摘要:Objective To observe the effect of electroacupuncture(EA)combined with neural mobilization(NM)in the treatment of occipital neuralgia.Methods A total of 62 occipital neuralgia patients were randomized into EA group(19 cases),NM group(22 cases)and EA+NM group(21 cases).EA was applied at acupoint-pairs as Yuzhen(BL 9)-Tianzhu(BL 10),Fengchi(GB 20)-Wangu(GB 12),etc.NM intervention consisted of occipital muscle group mobilization,C2 spinous process mobilization,cervical joint passive movement management mobilization,etc.,was performed at the impaired cervical spine segment.The two methods were used in combination for patients in the EA+NM group.All the treatment was given once a day for 2 weeks.Before and after treatment,the visual analogue scale(VAS)and the 6-point(1-6 points)behavioral rating scale(BRS-6)of headache were used to assess the severity of pain.The therapeutic effect was evaluated according to the"Criteria for Diagnosis and Cure-Improvement of Clinical Conditions"formulated by State Administration of Traditional Chinese Medicine of the People's Republic of China in 1994.Results After treatment,both VAS and BRS-6 scores were significantly lower than those before treatment in each of the three groups(P<0.05),and were significantly lower in the EA+NM group than in the simple EA and simple NM groups(P<0.01,P<0.05).The total effective rates were 78.95%(15/19)in the EA group,68.18%(15/22)in the NM group,and 90.48%(19/21)in the EA+NM group,with an obviously better therapeutic effect being in the EA+NM group relevant to each of the other two treatment groups(P<0.05).Conclusion EA,NM and EA combined with NM can improve symptoms of patients with occipital neuralgia,and EA+NM has a synergic analgesic effect for occipital neuralgia.

摘要:Objective To summarize the regularity of application of Lianquan(CV 23)in clinical practice in Chinese ancient times through analysis of ancient traditional Chinese medical(TCM)literature.Methods A total of 60 books involving CV 23 from the 1 156 ancient TCM books listed in the fifth edition of Encyclopedia of Traditional Chinese Medicine were collected by using CV 23 as the main keyword and"Sheben""Benchi",and"Jieben"(the other names of CV 23 in TCM)as the supplementary keywords and analyzed systematically.A database was then constructed from the collected data,including the related types of disorders or symptoms,acupoint recipes,and methods of needling and moxibustion,contraindications,etc.Results A total of196 articles related to the application of CV 23 from 60 ancient classical books were collected in accordance with the inclusive criteria.Among them,155 articles are referred to the indications of CV 23,35 to types of disorders such as asthma,cough,tongue swelling with difficulty in speaking,protracted tongue,acute contraction of tongue root,vomiting,spasm syndrome,stroke,aphtha,problems of mouth and teeth,throat problems,etc.of the internal medicine,surgery,pediatrics,and five-sense organs;64 items are referred to the application of single CV 23,91 to CV 23-included recipes containing 111 adjunct acupoints,and 78 to stimulation of CV 23 with acupuncture needle,moxibustion,pricking blood,and fire needle.Moreover,of the 111 adjunct acupoints,the most commonly used are Shaoshang(LU 11),Tiantu(CV 22),Hegu(LI 4),Yuye(EX-HN 13),Zhongchong(PC 9),etc.Conclusion Lianquan(CV 23)is mainly used for glossopharyngeal problems chiefly by syndrome-meridian differentiation.The supplement of complementary acupoints or five-shu points in combination with CV 23 has a synergistic effect.Moxibustion(3 moxacones in general)is often employed,and the needling depth is usually about 7.5 mm.The common contraindication of CV 23 is severe tongue swelling.

摘要:Objective To explore the regularity and characteristics of clinical application of scalp acupuncture therapy for different types of clinical conditions so as to provide a reference for clinical practice.Methods In the present study,"head acupuncture"and"scalp acupuncture"were used as the keywords to search papers and academic dissertations having definite standards for diagnosis and therapeutic effect assessment and published in journals and academic conferences collected in database China National Knowledge Internet(CNKI)from January 1 of 1959 to December 31 of 2016,followed by constructing a database after sorting,screening,recording,extracting,and statistical analysis by using a computer.Then,the data mining was conducted for summarizing the indications of disease categories,involved medical departments,needle-insertion methods,needle manipulation techniques,academic schools,and clinical efficacy of scalp acupuncture therapy.Results As a result,a total of 587 papers met our including criteria were analyzed.The scalp acupuncture therapy was widely employed in the treatment of various clinical conditions of different departments,with the application frequency being the internal medicine(438 times),surgery(75 times),pediatrics(44 times),etc.Of the indicated 102 types of clinical problems,55 belong to the internal medicine,constituting of 53.92%,particularly the cerebral apoplexy and its sequelae with the top application frequency being 102 and 115 times,respectively.In terms of needle inserting methods mentioned in partial papers(most papers do not mention),fingernail-pressing-aided needle insertion,needle-twirling insertion,fingers-squeezed-needle insertion,particularly the swiftly rotating needle insertion and rapid needle-propelling insertion were most commonly used.Regarding the needle manipulation method,rapid needle twirling technique was frequently employed,usually at a frequency of approximately 200 times per min.In regard to the academic schools,JIAO Shun-fa's scalp acupuncture system was most frequently used,followed by the international standardized scalp acupuncture.The therapeutic effect of scalp acupuncture is effective in the treatment of different conditions of various departments,especially those of the dermatology and gynecology.Conclusion Scalp acupuncture has superiority in the treatment disorders of the internal medicine and has been demonstrated to have positive effects for many types of problems,particularly for apoplexy and its sequelae.Rapid needle-propelling insertion and rapid needle-twirling technique are often employed.
