摘要:Objective To observe the effect of moxibustion of acupoints of the Governor Vessel on the levels of cellular autophagy,βamyloid protein(Aβ)immunoactivity,and expression of LC3-Ⅰ,LC3-Ⅱ,p62 and p-P70 S6 Kproteins in the hippocampal tissue of APPswe/PS1 de9(APP/PS1)double-transgenic Alzheimer's disease(AD)mice,so as to reveal its underlying mechanisms in improving AD.Methods APP/PS1 double-transgenic AD mice were randomly divided into AD model,moxibustion,autophagyinducer(Rapamycin)and autophagy-inhibitor(3-MA)+moxibustion groups(n=10 in each group),and other 10 C57 BL/6 Jmale mice(the same age)were used as the normal control group.Herbal-cake(made of Chuanwu [Radix Aconiti Praeparata])partitioned moxibustion was applied to"Baihui"(GV20),moxibustion was applied to"Fengfu"(GV16)and"Dazhui"(GV14),all for 20 min,once daily for 2 weeks,with one day's off between two weeks.For mice of the autophagy-inducer and 3-MA+moxibustion groups,Rapamycin(2 mg·kg-1·d-1)and 3-MA(1.5 mg·kg-1·d-1)were separately administered by intraperitoneal injection for 2 weeks.The cognitive ability was examined by Morris water maze tests,and the ultrastructural changes(including autophagic lysosomes,etc.)of hippocampal neurons were observed by using transmission electron microscopy.The immunoactivity of cerebral cortex and hippocampal Amyloidβpeptide 1-42(Aβ1-42)was detected by immunohistochemistry,and the expression levels of hippocampal LC3-Ⅰ,LC3-Ⅱ,p62 and p-P70 S6 Kproteins were detected by Western blot.Results After modeling,the escape latency of Morris water maze tasks was prolonged in the model group than in the normal control group(P<0.05)and obviously shortened in the moxibustion and autophagy-inducer groups(not the autophagy-inhibitor group)than in the model group(P<0.05).Results of transmission electron microscope showed deformed,irregular or atrophic neurons with rough and incomplete and fuzzy nuclear membrane,and decreased intracellular autophagosomes in the hippocampus in the model group,and partial irregular,atrophic neurons with more autophagic vesicles and lysosomes in the moxibustion group.The expression levels of Aβ1-42 in both cerebral cortex and hippocampus tissues,and LC3-Ⅰ,p62 and p-P70 S6 Kproteins in the hippocampus were considerably up-regulated in the model group relevant to the normal control group(P<0.01),and evidently down-regulated in both moxibustion and autophagy-inducer groups(not the autophagy-inhibitor group)than in the model group(P<0.01),while that of hippocampal LC3-Ⅱ protein and LC3-Ⅱ/Ⅰratio levels were obviously down-regulated in the model group relevant to the normal control group(P<0.01),and significantly up-regulated in both moxibustion and autophagy-inducer groups(not the autophagyinhibitor group)than in the model group(P<0.01).Conclusion Moxibustion can improve the cognitive ability of APP/PS1 double-transgenic AD mice,which is associated with its effects in promoting hip-pocampal and cerebral cortex autophagy level,and down-regulating the expression levels of Aβ1-42,LC3-Ⅰ,p62 and p-P70 S6 Kproteins in the hippocampus.
摘要:Objective To observe the effect of electroacupuncture(EA)on neurological behavior and activity of Tolllike receptor 2/nuclear factor kappa B(TLR2/NF-κB)signaling of the ischemic cerebral area in cerebral ischemia-reperfusion injury(CIRI)rats,so as to explore its mechanisms underlying improvement of CIRI.Methods A total of 120 male SD rats were randomly divided into blank control,sham operation,model,EA and EA+NF-κB inhibitor(Pyrrolidine Dithiocarbamate Hydrochloride,PDTC,EA+PDTC)groups which were further divided into 3,7,14 and 28 dsubgroups(n=6 in each subgroup).The CIRI model was established by occlusion of the middle cerebral artery for 90 min,followed by reperfusion.EA(1-20 Hz,6 V)was applied to"Shuigou"(GV26),"Neiguan"(PC6),"Sanyinjiao"(SP6)and"Weizhong"(BL40)for 30 min,once a day for 28 days.For rats of the EA+PDTC group,PDTC solution(120 mg/kg)was intraperitoneally injected on the 3 rd day after successful modeling and before EA intervention.The neurological deficit severity(Zea Longa score)was assessed 3,7,14 and 28 days after modeling.The expression levels of TLR2,Interleukin-1 receptor-associated kinase(IRAK)and NF-κB mRNAs in the ischemic penumbra region of brain tissue were detected by real-time fluorescence quantitative PCR.Results Following modeling,the neurological deficit scores were significantly increased from the 3 rd day on after CIRI(P<0.05),the expression levels of TLR2 mRNA on day 3,7,14 and 28,and IRAK mRNA on day 3 and 7,as well as NF-κB mRNA on day 3,7 and 14 were significantly up-regulated in the model group relevant to the blank control group(P<0.05).After EA intervention,the neurological deficit scores were significantly decreased in the EA group on day 3,7 and 28 and in the EA+PDTC group on day 3,7,14 and 28 in comparison with those of the model group(P<0.05).In addition,the expression levels of TLR2 mRNA and NF-κB mRNA on day3,7 and 14 in the EA group,and on day 3,7,14 and 28 in the EA+PDTC group,IRAK mRNA on day 3 in the EA and EA+PDTC group were significantly down-regulated(P<0.05),but those of IRAK mRNA on day 14 and 28 in the EA group were significantly up-regulated in comparison with those of the model group(P<0.05).The effect of the EA+PDTC was obviously superior to that of simple EA in down-regulating the expression of TLR2(on day 28),and IRAK(on day 3,14,28),and NF-κB(on day 3,7 and14)(all P<0.05).Conclusion EA stimulation can improve the symptoms of neurological deficits in CIRI rats,which may be related to its effect in suppressing the expression of TLR2,NF-κB and IRAK mRNAs of the ischemic cerebral tissue,i.e.,down-regulating the activity of TLR2/NF-κB signaling.
摘要:Objective To investigate the effect of"Jiaji"(EX-B2)-electroacupuncture(EA)preconditioning on structural changes of myocardium and expression of cytochrome P450(CYP450)enzymes in acute myocardial ischemia-reperfusion injury(MIRI)rats.Methods Sixty male Wistar rats were randomly divided into sham operation,model,EX-B2,Neiguan(PC6),Yanglingquan(GB34)and Quchi(LI11)groups(n=10 in each group).The MIRI model was established by occlusion of the descending anterior branch of the left coronary artery for 30 min,followed by reperfusion for 30 min.EA preconditioning(2 Hz/100 Hz,1 mA)was respectively applied to EX-B2,PC6,GB34,and LI11 for 30 min,once daily for 7 days.Morphological changes of myocardium were observed by H.E.staining.The ultrastructure of myocardial cells was observed by transmission electron microscope(TEM).The protein expression levels of myocardial CYP450(CYP1 A1,CYP2 B2,CYP2 C11 and CYP4 A2)were detected by Western blot.Results In MIRI rats,myocardial pathological changes as swollen,disorganized arrangement,and partially ruptured myocardial fibers with unclear limit and necrosis and infiltration of lots of inflammatory cells under microscope,and focal myofilament degeneration,dissolution and necrosis with interstitial edema,vague mitochondria with swelling and vacuolation,ruptured and disorganized arrangement of crests under TEM were found in the ischemic area,which was obviously milder in rats of both EX-B2 and PC6 groups.After modeling,the expression of myocardial CYP1 A1 and CYP2 B2 protein were significantly up-regulated in the model group relevant to the sham operation group(P<0.05).In EA preconditioning groups,the expression levels of CYP1 A1 and CYP4 A2 proteins in the EX-B2,PC6 and GB34 groups were obviously down-regulated(P<0.01),and those of CYP2 B2 and CYP2 C11 proteins in both EX-B2 and PC6 groups were markedly up-regulated relevant to the model group(P<0.01).Compared with the EX-B2 group,the expression levels of CYP1 A1,CYP4 A2 proteins in the LI11 group were significantly up-regulated(P < 0.05),and CYP2 B2,CYP2 C11 proteins were significantly down-regulated(P<0.05).No significant differences were found between the model and sham operation groups in the expression levels of myocardial CYP2 C11 and CYP4 A2 proteins,between the LI11 and model groups in the expression of CYP1 A1,CYP2 B2,CYP2 C11 and CYP4 A2,between the GB34 and model groups in the expression of CYP2 B2,CYP2 C11(all P>0.05).Conclusion EA preconditioning at EX-B2 and PC6 can suppress MIRI induced up-regulation of myocardial CYP450ω-hydroxylase CYP1 A1 and CYP4 A2 expression and further up-regulate the expression of myocardial epoxygenase CYP2 B2 and CYP2 C11 proteins in acute MIRI rats,thus playing a role in myocardial protection.
摘要:Objective To observe the effect of electroacupuncture(EA)on morphological changes of denervated gastrocnemius(GS)and the expression of fork-head protein(FOXO3 A),muscle atrophy F-box(MAFbx)and myogenic differentiation antigen(Myod1)in sciatic nerve injury rats,so as to reveal its mechanism underlying improvement of myoatrophy.Methods Eighteen male Sprague-Dawley rats were randomly divided into sham operation,model and EA groups(n=6 per group).The model of gastrocnemius atrophy was established by crushing the right sciatic nerve.Then,EA(2 Hz)was applied to the right"Zusanli"(ST36)and"Huantiao"(GB30)for 10 min,once a day for 14 successive days.The wet weight of the GS on both sides was weighted to calculate the wet weight ratio(the injured side/the healthy side),and the cross-sectional area(CSA)and diameter of GS fibers were measured after H.E.staining.The expressions of FOXO3 A,MAFbx and Myod1 protein and mRNA in the GS tissue were tested using Western blot and fluorescence quantitative PCR,separately.Results Following modeling,the GS wet weight ratio,CSA and fiber diameter were smaller in the model group than those in the sham group(P<0.01),and were significantly higher in the EA group than in the model group(P<0.01).H.E.staining showed that the GS fibers became smaller and the myocyte got round in the model group,while the GS fibers were bigger and the myocyte was relatively regular in morphology in the EA group.After modeling,the expression levels of FOXO3 A,MAFbx and Myod1 mRNA and protein were evidently higher in the model group(P<0.01);Moreover,after EA treatment,modeling-induced increasing of expression levels of FOXO3 Aand MAFbx mRNA and protein were revised(P<0.01),while the increased expression level of Myod1 was further up-regulated relavant to that in the model group(P<0.01).Conclusion EA of ST36 and GB30 can suppress the up-regulated expression of FOXO3 Aand MAFbx mRNA and protein and further promote the expression of Myod1 mRNA and protein in the GS tissue in rats with denervated GS atrophy,which may contribute to its function in relieving the myoatrophy,promoting the skeletal muscle protein hydrolysis and differentiation of satellite cells.
摘要:Objective To observe the effect of electroacupuncture(EA)stimulation on the levels of a disintegrin and metalloproteinase with thrombospondin motifs-4(ADAMTS-4)protein in the lumbar intervertebral disc tissue and serum in prolapsed lumbar intervertebral disc degeneration(PLIDD)rats,so as to explore its mechanism underlying improvement of intervertebral disc degeneration(IDD).Methods A total of 48 Sprague-Dawley rats were divided into sham operation(n=12),model(n=18)and EA(n=18)groups.The PLIDD model was established by puncturing the lumbar discs(L4—L5,L5—L6)with a gauge-22 syringe needle.After modeling,EA stimulation was applied to"Pangguangshu"(BL28),"Zusanli"(ST36)and"Zhijian"for 20 min,6 times per week for 4 weeks.The lumbar intervertebral disc tissue and blood samples were collected at the 4 th,6 th and 8 th week after modeling,respectively.The expression of ADAMTS-4 in the lumbar intervertebral disc tissue was detected by immunohistochemistry and Western blot(WB),separately.The content of ADAMTS-4 in the serum was detected by ELISA.Results Both immunohistochemical stain and WB showed that the expression levels of lumbar ADAMTS-4 at the 4 th,6 th and 8 th week were significantly up-regulated in the model group relevant to the sham operation group(P<0.01).Following EA treatment,the expression levels of ADAMTS-4 on day 14 and 28 were notably lower in the EA group than in the model group(P<0.05).The serum ADAMTS-4 contents were significantly increased in the model group than in the sham operation group at the 3 time-points(P<0.05,P<0.01),and considerably decreased in the EA group than in the model group on day 28 after EA intervention(P<0.05).Conclusion EA can down-regulate the ADAMTS-4 expression of lumbar intervertebral disc tissue in PLIDD rats,which may contribute to its effect in relieving lumbar intervertebral disc degeneration.

摘要:Objective To observe and compare the effect of electroacupuncture(EA)of"Tianshu"(ST25)and"Dachangshu"(BL25)on intestinal sensitivity and the expression of M3 muscarinic acetylcholine receptor(M_(3R))and5-hydroxytryptamine 3 Areceptor(5-HT_(3A)R)in the colon tissue in irritable bowel syndrome(IBS)rats,so as to explore its mechanism underlying improvement of IBS.Methods Forty newborn Wistar rats were randomly divided into blank control,model,ST25 and BL25 groups(n=10 rats per group).The IBS model was established by joint application of maternal separation,acetic acid enema plus colorectal distension from day 8 to 21 after birth.At the age of 9 weeks,EA(2 Hz/100 Hz,0.1-0.3 mA)was applied to ST25 and BL25 for 20 min,once every other day for 5 times.The modified method was used to record the abdominal withdrawal reflex(AWR),so as to evaluate the visceral sensitivity by referring to the modified Al-Chaer's and colleagues' methods.The threshold(initial wave),the number and peak-peak value of contraction waves(AWR)within 90 swere recorded.The immunoactivity of M_(3R) and 5-HT_(3A)R in the colon tissue was detected by immunohistochemistry.Results After modeling,the latency was obviously shortened,the number and peak-peak values of contraction waves of intestine were significantly increased,and the immunoactivity of M_(3R) and 5-HT_(3A)R was notably up-regulated in the model group compared with the blank control group(P<0.01).After the EA intervention,the latency was obviously prolonged,the peak-peak value in both ST25 and BL25 groups,and the number of contraction waves in the ST25 group were significantly decreased(P<0.01),and the expression levels of M3 and 5-HT_(3A)R were considerably down-regulated in both ST25 and BL25 groups relevant to the model group(P<0.01).The therapeutic effects were significantly better in the ST25 group than in the BL25 group in prolonging the latency and reducing the contraction wave number,and in down-regulating the expression of colonic M_(3R) and 5-HT_(3A)R(P<0.05,P<0.01).Conclusion EA at ST25 and BL25 can reduce the intestinal sensitivity in IBS rats,which is probably associated with its effects in down-regulating the expression of M_(3R) and 5-HT_(3A)R in the colon.The therapeutic effects of ST25 were obviously better than those of BL25 in suppressing intestinal sensitivity and expression of colonic M_(3R) and 5-HT_(3A)R.
摘要:Objective To observe the effect of electroacupuncture(EA)on expression of hypothalamic sirtuin 1(SIRT1)and proopiomelanocortin(POMC),and body weight,food-intake,blood glucose,and blood lipid levels in obese rats,so as to explore its mechanisms underlying improvement of obesity.Methods Forty male Wistar rats were randomly divided into normal,model,EA,and sham EA groups(n=10 rats in each group).The obesity model was established by feeding the rats with high fat diet.EA(2 Hz,1 mA)was applied to "Zusanli"(ST36),"Zhongwan"(CV12),"Guanyuan"(CV4)and "Fenglong"(ST40)or sham acupoints(about 5 mm beside each acupoint,shallow needling)for 20 min,once every other day for 8 weeks.The rats' body weight and food-intake were recorded.The blood glucose(fasting plasma glucose:FPG,postprandial plasma glucose:PPG)and blood lipids(triglyceride:TG,total cholesterol:TC,non esterified fatty acid:NEFA)were assayed by using an automatic biochemical analyzer.The protein and mRNA expression levels of SIRT1 and POMC in the hypothalamus were detected by Western blot and quantitative real-time PCR,respectively.Results In comparison with the normal group,the body weight,foodintake,blood lipids,and PPG levels were significantly increased(P<0.05,P<0.01),and the expression levels of SIRT1 protein and mRNA in the hypothalamus were significantly doun-regulated in the model group(P<0.05).Following EA,the body weight,food-intake,blood lipids,and PPG levels were considerably down-regulated(P<0.01,P<0.05),and the expression levels of SIRT1 and POMC protein and mRNA in the hypothalamus were significantly up-regulated in the EA group rather than those in the sham EA and the model groups(P<0.05).Conclusion EA can reduce the obese rats' body weight,food-intake,blood lipids and blood glucose,which may be associated with its effect in up-regulating the SIRT1 and POMC expression of hypothalamus.
摘要:Objective To observe the effect of acupoint injection on expression of fork head/winged helix protein 3(Foxp3),retinoic acid-related orphan receptorγt(RORγt)in nasal mucosa and serum interleukin-17(IL-17)level in allergic rhinitis(AR)rats,so as to explore its mechanism underlying improvement of AR in terms of balancing Th17/Treg.Methods Thirty-two SD rats(half male and half female)were randomized into normal control,AR model,acupoint injection and non-acupoint injection groups(n=8 in each group).The AR model was established by ovalbumin sensitization.In the acupoint injection group,"Yintang"(EX-HN3)and bilateral"Yingxiang"(LI20)were selected for injection of mixture solution of dexamethasone(DEX)and transfer factor and lidocaine(0.1 mL/acupoint),once every 3 days for a total of 4 times.The non-acupoints,located at the midpoint between the"Houhai"(GV1)and"Huantiao"(GB30)on the bilateral hips and the sites 5 cm inferior to the axillary were injected with the same dose of mixture solution as that in the acupoint injection.The AR severity was assessed by cumulative quantification scoring methods(including the numbers of nose-catching and sneezes,and the amount of nasal secretions in 30 min).The expressions of Foxp3 and RORγt in the nasal mucosa were detected by immunohistochemistry.The serum IL-17 content was detected by enzyme linked immunosorbent assay(ELISA).Results The AR symptom score and serum IL-17 content were significantly higher in the AR model group than those in the normal control group(P<0.05),and significantly down-regulated in the acupoint injection group(not in the non-acupoint group)relevant to the AR model group(P<0.05).Following modeling,the expression levels of nasal Foxp3 protein was significantly down-regulated while that of RORγt protein markedly up-regulated in the AR model group relevant to the normal control group(P<0.05),indicating an imbalance between Foxp3 and RORγt activity(P<0.05).After EA intervention,the increased expression of Foxp3 and the down-regulated expression of RORγt were revised in the acupoint injection group(P<0.05)but not in the non-acupoint group(P>0.05).The percentage of the Foxp3 positive cells and the ratio of Foxp3/RORγt were negatively correlated with the AR symptom score(P<0.05),the expression of RORγt and the content of IL-17 were positively correlated with the symptom score(P<0.05).Conclusion Acupoint injection is able to improve symptoms of RA rats,which may be related with its function in up-regulating the level of nasal mucosal Foxp3 and suppressing the levels of nasal RORγt and serum IL-17 to correct the imbalance of Th17/Treg.
摘要:Objective To observe the effect of acupuncture of intraorbital and extraorbital acupoints on apoptosis and expression of Bax,Bcl-2 and Caspase-3 proteins of retinal ganglion tissue in rabbits with nonarteritis anterior ischemic optic neuropathy(NAION),so as to reveal its mechanism underlying improvement of NAION.Methods Female New Zealand rabbits were randomly divided into four groups:model,intraocular needling(ION),extraocular needling(EON),ION+EON groups(n=5 per group),and the 5 healthy eyes of those rabbits in the model group were selected to be used as the control group.The NAION model of the right eye was established by photodynamic stroke method.For ION group,acupoints "Jingming"(BL1),"Chengqi"(ST1)and "Qiuhou"(EX-HN7)were needled,and for EON group,"Cuanzhu"(BL2),"Yuyao"(EX-HN4)and"Qiaoming"(EX)were needled with filiform needles,followed by retaining the needles for 30 min.For ION+EON group,the 6 acupoints were needled simutaneously.The treatment was conducted once daily for 3 days.The apoptosis of retinal ganglion tissue was detected by using TUNEL fluorescence labeling,and the expression of Bax,Bcl-2 and active Caspase-3 in the retinal ganglion were detected by immunohistochemistry.Results Following modeling and compared with the control group,the number of the apoptotic cells,and the expression levels of Bax and Caspase-3 proteins,as well as the ratio of Bax/Bcl-2 were significantly increased in the model group(P<0.01),while the expression of Bcl-2 was significantly decreased(P<0.01).After intervention,the number of the apoptotic cells in the ION,EON and ION+EON groups,and the ratio of Bax/Bcl-2 in EON and ION+EON groups,and the expression of Caspase-3 proteins in the ION and ION+EON groups were significantly down-regulated(P <0.01,P<0.05),and the expression levels of Bcl-2 in both EON and ION+EON groups were significantly up-regulated in comparison with the model group(P<0.01).No significant changes were found in the expression levels of Bax in the 3 needling groups relevant to the model group(P>0.05).Conclusion Acupuncture of intraorbital and extraorbital acupoints can reduce apoptosis of retinal ganglion in NAION rabbits via inhibiting the activation of Caspase-3 protein and ratio of Bax/Bcl-2,and up-regulating the expression of Bcl-2 protein.
摘要:Objective To explore the effects of moxibustion intervention on wound healing in rats with full-thickness cutaneous wounds.Methods A total of 28 adult SD male rats were randomly assigned to model group and moxibustion group(n=14 in each one).The skin wound model was established by removal of a piece of full-thickness skin from the median line of the rats'back(about 2 cm below the shoulder blade).Moxibustion intervention was applied to the surrounding area of the focus for 25 min,once daily for 6 days.The wound healing was observed and photographed after each moxibustion intervention.The wound tissues were harvested after transcardiac perfusion with 4% paraformaldehyde solution on the 2 nd and 7 thday after modeling,and stained with H.E.and Masson methods for displaying histopathological changes and collagen fiber growth status,respectively.Results After modeling,the cutaneous wound was healed gradually in both groups,and the wound area was significantly smaller from the 2 nd day to the 5 th day in the moxibustion group than in the model group(P<0.01,P<0.05).Correspondingly,the wound closure area was significantly bigger from the 2 nd to the 5 th day in the moxibustion group than in the model group(P<0.01).H.E.and Masson staining showed that the number of the inflammatory cells(monocytes,macrophages,neutrophile,etc.)and the collagen fiber area in the local wound tissue of the moxibustion group were significantly bigger than those of the model group on the 2 nd day after intervention(P<0.01).After 6 sessions of moxibustion intervention,the number of fibroblasts and collagen fibers in the moxibustion group were obviously increased than that in the model group,characterized by closer arrangement of fibroblasts and collagen fibers,more and larger of the new-born blood vessels,and thicker of the scab in the wound area.Conclusion Moxibustion can promote the wound healing by enhancing the growth of collagen fibers and cell proliferation in the traumatic cutaneous area in full-thickness cutaneous wound rats.
摘要:Objective To investigate the clinical effect of shallow acupuncture combined with ear-acupoint pellet-pressing in the treatment of primary insomnia in patients with qi-stagnation constitution.Methods A total of 60 primary insomnia outpatients with qi-stagnation constitution were randomly divided into treatment group and control group,with 30 patients in each group.The patients in the control group were given oral Alprazolam tablets once a day,and those in the treatment group given shallow acupuncture of Yintang(EX-HN3),Shangen(inferior to EX-HN3),Anmian(EX-HN16)and bilateral Xingjian(LR2)and bilateral Taichong(LR3),once a day,and combined with pellet-pressing of ear acupoints"Shenmen""Sympathy""Subcortex""Heart"and"Liver"once every other day.Each course of treatment was 10 consecutive days,and both groups were treated for three courses.The sleep quality was assessed using Pittsburgh Sleep Quality Index(PSQI),the emotional status assessed using Self-rating Depression Scale(SDS),and the qi-stagnation state evaluated according to"the Criteria for Classification and Judgement of Constitution of Traditional Chinese Medicine(2009)".The therapeutic effect was evaluated according to"the Criteria for Diagnosis and Evaluation of Therapeutic Effect of Diseases of Traditional Chinese Medicine".Results Of the two 30 cases in the control and treatment groups,2(6.7%)and 6(20.0%)were cured,6(20.0%)and 14(46.7%)experienced marked improvement in their symptoms,21(70.0%)and 6(20.0%)were effective,and 1(3.3%)and 4(13.3%)ineffective,with the effective rate being 86.7%and96.7%,respectively.No significant difference was found between the two groups in the short-term effect(P>0.05).One month's follow-up showed that,of the two 30 cases in the control and treatment groups,2(6.7%)and 5(16.7%)were cured,4(13.3%)and 14(46.6%)experienced marked improvement,10(33.3%)and 6(20.0%)were effective,and 14(46.7%)and 5(16.7%)ineffective,with the effective rate being 53.3%and 83.3%,respectively.The long-term therapeutic effect of the treatment group was significantly superior to that of the control group(P<0.05).After the treatment,both PSQI and SDS scores in the two groups,and qi-stagnation score in the treatment group showed a significant reduction in comparison with their own pretreatment(P<0.05),and one-month's follow-up(not the short-term outcome)displayed that the PSQI,SDS and qi-stagnation scores of the treatment group were significantly lower than those of the control group(P<0.05).Conclusion Shallow acupuncture combined with ear-acupoint pellet-pressing can significantly improve sleep quality,depression symptoms,and pathological constitution in primary insomnia patients with qi-stagnation constitution,possessing a stable long-term clinical effect.
摘要:Objective To compare the clinical therapeutic effect of intensive moxibustion plus acupuncture and simple acupuncture therapy in the treatment of frozen shoulder so as to provide a reference for clinical practice.Methods A total of60 patients with frozen shoulder were randomly divided into intensive moxibustion plus acupuncture group and simple acupuncture group(n=30 per group).Acupoints Jianyu(LI15),Jianzhen(SI9),Jianliao(TE14),Tiaokou(ST38),Chengshan(BL57)and Ashi points(Extra)were punctured with filiform needles which were manipulated till the patients experienced feelings of soreness and distension in the local region,followed by retaining the needles for 40 min.For patients of the moxibustion plus acupuncture group,after acupuncture,an ignited moxa-stick segment was applied to the affected shoulder for 40-50 min.After the treatment,all patients were ordered to make a shoulder exercise(anteflexion,rear protraction,abduction and upthrow movements,etc.).The visual analogue scale(VAS)was used to assess the pain severity,and the Constant-Murley shoulder assessment scale(the total score is 100 points,including 15 points in pain severity and 20 points in daily living activities,40 points in joint motion range,and25 points in myodynamia)used to assess the functional state of the shoulder,and the"Standards for Diagnosis and Therapeutic Effect Evaluation of Diseases of Traditional Chinese Medicine"(1994)were employed to evaluate the therapeutic effect.Results Following the treatment,intra-group comparison showed that the VAS score was significantly reduced in both groups in comparison with their own pre-treatment(P<0.05),and the total scores of Constant-Murley scale,the scores of activities of daily living and active motion range,as well as the scores of pain integration and myodynamia,were all considerably increased in the two groups in comparison with their own pre-treatment(all P<0.05).The therapeutic effect of the moxibustion plus acupuncture was significantly superior to that of the simple acupuncture in reducing VAS score and increasing total score of Constant-Murley scale,and scores of activities of daily living and active motion range,as well as the scores of pain integration(P<0.05).Of the two 29 cases in the moxibustion plus acupuncture and simple acupuncture groups,19(65.52%)and 10(34.48%)were cured,8(27.58%)and 12(41.38%)experienced improvement,2(6.90%)and 7(24.14%)were failed,with the effective rate being 93.10%and 75.86%,respectively.The therapeutic effective rate of the moxibustion plus acupuncture was evidently higher than that of the simple acupuncture(P<0.05).Conclusion The therapeutic effect of intensive moxibustion plus acupuncture is superior to that of simple acupuncture in improving symptoms of frozen shoulder in patients.

摘要:The inflammatory reaction induced by ischemic myocardial injury(IMI)is divided into three phases,i.e.the inflammatory phase,the fibrous proliferative phase and the stable phase.The appropriate inflammatory reaction effectively removes the fragments of myocardial cells,which is the essential phase in the pathological progression of myocardial ischemia(MI).However,the excessive inflammatory reaction may aggravate the myocardial injury.For this reason,the immediate control of the post-injury inflammatory reaction is the principal therapeutic measure and the research hotspot at the present.Acupuncture intervention has been demonstrated to have positive roles in relieving MI and inflammatory reaction by suppressing myocardial inflammatory cytokines(suppressing IL-1β,TNF-α,IL-8,etc.),adjusting inflammatory reaction pathway(NF-κB signaling,TGF-β,etc.)and activating cholinergic anti-inflammatory pathway.Therefore,it is feasible to explore the underlying mechanism of acupuncture therapy in protecting ischemic myocardium based on anti-inflammatory efficacy.
摘要:There are some records about the needling and moxibustion sensations in Chinese ancient medical literature,in which,the concept of Deqi(qi arrival)and its relationship with the production of therapeutic effects were put forward.The generation of needling or moxibustion sensations is one of the symbol for qi reaching the affected site during the manipulation and is also related to the treatment outcomes.In the present paper,we reviewed current situations about the mechanisms of acupuncture and moxibustion stimulation induced sensations and their relation to the propagated sensation(PS)published in recent 10 years from the following four aspects:1)the patterns of manifestation of needling sensations(including general sensations,and specific sensations,as well as the practitioner's hand felt heavy,tight,hard-going senses,etc.)and moxibustion sensations(local superficial and deep warmth);2)modern anatomical and physiological basis(skin,loose and dense connective tissue,free nerve ending,various receptors);3)neurohumoral mechanism of acupuncture stimulation induced PS phenomenon(mechanical receptors,afferent and efferent nerve fibers,neural center,various neurotransmitters/modulators,chemical substances,etc.);4)neurohumoral mechanism of moxibustion stimulation induced channel transmission(transient receptor potential vanilloid receptors-heat sensitive receptors,heat-shock protein,immunocytes,etc.).Additionally,the authors also put forward some suggestions about future researches on acupuncture-moxibustion sensations.
