摘要:Objective: To investigate the effect of electroacupuncture(EA) on dysfunction of hypothalamus-pituitary-ovary axis(HPOA) in ovariectomized(OVX) rats so as to explore the neuroendocrine mechanism of acupuncture in the treatment of woman perimenopausal syndrome.Methods: Forty-eight female SD rats(180-(200 g)) were randomized into OVX, OVX+EA,normal control(NC) and EA groups with 12 cases(6 cases used for immunohistochemical study and the other 6 for real-time PCR) in each group.EA((3 Hz),(2 mA)) was applied to "Guanyuan"(RN 4),"Zhongji"(RN 3),"Sanyinjiao"(SP 6) and bilateral "Zigong"(EX-CA 1) for(30 min),once daily and continuously for 3 days.Radioimmunoassay(RIA) was used to detect blood luteinizing hormone(LH) and estrin(E)2 contents,and real-time reversed transcription polymerase chain reaction(RT-PCR) and immunohistochemistry were utilized for assaying expression of prolactin-releasing peptide(PrRP) and PrRP mRNA in the medulla oblongata and hypothalamus.Results: Comparison of NC,EA, OVX and OVX+EA groups showed that serum E_2 contents of OVX and OVX+EA groups were significantly lower than that of NC group,while serum LH contents of OVX and OVX+EA groups significantly higher than that of NC group(P<0.01,0.05),and no significant differences were found between NC and EA groups in these two indexes(P>0.05).Compared with OVX group,serum LH level was markedly lower and serum E_2 level markedly higher in OVX+EA group(P<0.05).In comparison with NC group,the number of PrRP immunoreaction(PrRP-ir) positive neurons in nucleus tractus solitarii(NTS) and ventral lateral reticular nucleus(VLRN) and the relative optical density(OD) of bed nucleus of the stria terminalis(BST) in OVX group,and that in NTS in OVX+EA group were markedly fewer(P<0.01),while compared with OVX group,PrRP-ir positive neurons of NTS and VLRN were significantly more and the OD value of PrRP-ir positive fibers of BST was significantly higher in OVX+EA group(P<0.01,0.05).The relative contents of PrRP mRNA expression(PrRP β-actin) of hypothalamus and medulla oblongata in OVX and OVX+EA groups were significantly lower than those of NC group(P<0.01);comparison between OVX and OVX+EA showed that the relative content of PrRP mRNA expression in the medulla oblongata in the later group was markedly higher than that of the former(P<0.01).No significant differences were found between NC and EA groups in the number of PrRP-ir positive neurons of NTS and VLRN,OD values of the positive fibers of BST and the relative content of PrRP mRNA expression in the hypothalamus and medulla oblongata,and between OVX and OVX+EA groups in the relative content of PrRP mRNA expression in the hypothalamus(P>0.05).Results indicated that in OVX rats,serum E_2 level,PrRP contents in NTS, VLRN and BST,and hypothalamic PrRP expression decreased remarkably and serum LH content increased significantly,while EA could reverse most of the aforementioned changes.Conclusion: PrRP,a new neuropeptide or neurohormone,participates in the effect of EA in normalizing the subnormal function of HPOA in OVX rats,which may be one of the neuroendocrine mechanisms of EA in relieving woman perimenopausal syndrome.

摘要:Objective: To explore the mechanism of acupuncture in the treatment of prostate hyperplasia in rats.Methods: Thirty SD male rats were randomized into control(sham-operation),model and acupuncture groups,with 10 cases in each group.Prostate hyperplasia model was established by castration under anesthesia(25% Pentobarbital Sodium,(2 mL/kg),i.p.),and subcutaneous injection of Testosterone Propionate((4 mg/kg/d)) for 30 days.The rats in control group were treated with surgical isolation and subcutaneous injection of normal saline."Guanyuan"(CV 4),bilateral "Qihai"(CV 6) and bilateral "Sanyinjiao"(SP 6) were punctured and stimulated by manipulating the needle with twirling reinforcing method.The treatment was conducted once daily,21 days altogether.The prostate weight index [(prostate damp weight/body weight)×100%],serum testosterone(Ts),estradiol(E_2) contents were detected with radioimmunoassay,and the anatomical structure of the prostate was observed under light microscope and electronic microscope.Results: Compared with control group,the prostate damp weight,prostate weight index,serum E_2 content and E_2/Ts of model group increased significantly(P<0.05),but the body weight of model group decreased considerably(P<0.05);while compared with model group,the prostate damp weight,prostate weight index,serum E_2 content and E_2/Ts of acupuncture group reduced apparently(P<0.05).No significant differences were found between model and acupuncture groups in body weight,and among the 3 groups in serum Ts level(P>0.05).In model group,the glandular cells arranged in disorder,the lumen of gland enlarged,and the epithelial cells increased in number,while the state of acupuncture group was much better.Conclusion: Acupuncture can significantly restrain prostate hyperplasia,lower serum E_2 level and prostate index in prostate hyperplasia rats.
摘要:Objective: To observe the effect of electroacupuncture(EA) on the expression of brain derived neurotrophic factor(BDNF) mRNA in nucleus of facial nerve of rabbits with facial nerve injury so as to reveal the mechanism of acupuncture in improving facial nerve injury.Methods: One hundred and twenty rabbits(Japanese big-earwhite) were randomly divided into control(sham-operation),model,medication,hand-acupuncture and EA groups,with 24 cases in each group which was further divided into(7 d),(14 d),(21 d) and(28 d) phases.Facial nerve injury model was established by cramping the facial nerve(0.5 cm) to the stylomastoid foramen with a pair of forceps."Yifeng"(TE 17),"Quanliao"(SI 18),"Dicang"(ST 4),"Jiache"(ST 6),etc were punctured with filiform needles and stimulated with hand manipulation in hand acupuncture group,and stimulated electrically with the stimulation parameters being 1-(100 Hz) in frequency,(2 V) in strength and duration of(30 min).In medication group,muscular injection of the mixture solution of Vitamin B_1((10 mg)),B_(12)((50 μg)) and dexamethasone(DXM,(2 mg/d) in the first 7 days,(1 mg/d) later) was performed once daily.The treatment was given 1 week,2 weeks,3 weeks and 4 weeks respectively in different subgroups of every group.BDNF mRNA expression of the nucleus of facial nerve was detected by using in situ hybridization(ISH) technique.Results: Compared with control group,the number of BDNF mRNA expression-positive neurons in(7 d),(14 d),(21 d) and(28 d) phases of model group increased significantly(P<0.05),while in comparison with model group,the number of BDNF mRNA expression-positive neurons in the 4 phases of medication,hand-acupuncture and EA groups increased further and pronouncedly(P<0.01),and the effect of EA group was superior to those of medication and hand-acupuncture groups(P<0.01).Self-comparison indicated that compared with phase(7 d),the number of BDNF mRNA expression-positive neurons in phase(28 d) of each treatment group was markedly more(P<0.01).No significant differences were found between medication and handacupuncture groups in the number of BDNF mRNA expression-positive neurons in the 4 phases(P>0.05).Results of hematoxylin-eosin(HE) staining showed a more significant improvement of neuronal degeneration in the nucleus of facial nerve in EA group.Conclusion: EA can promote the recovery of the injured facial nerve and upregulate BDNF mRNA expression in the nucleus of facial nerve in rabbits with facial nerve injury.
摘要:Objective: To observe the effect of electrothermo-needle(ETN) treatment on Cyclooxygenase(COX)-2 mRNA expression osteosynovial cells in collagen-induced arthritis(CIA) rats.Methods: A total of 50 Wistar male rats(180-(200 g)) were randomized into normal control,model,higher electric current((40 mA)) ETN,low-current((20 mA)) ETN,no-current((0 mA)) ETN groups,with 10 cases in each group.Arthritis model was established by injecting type-Ⅱ collagen((100 μg)) into the rats' subcutaneous tissues of the bilateral hind-paws.ETN was applied to bilateral "Yanglingquan"(GB 34) once every other day with(20 min) each time and 20 times altogether.The rats' ankle-joint swelling degree(intensity of pressure) was detected by using joint volume detector.COX-2 mRNA expression of osteosynovial cells of the left ankle joint was assayed by using in situ hybridization technique.Results: In model group,the intensity of pressure of the rats' bilateral ankle joints increased significantly in comparison with that of rats in normal control group,while the intensity values of the ankle joints in high-current ETN,low-current ETN and no-current ETN groups were remarkably lower than that of model group(P<0.01);no significant differences were found among high-current ETN,low-current ETN and no-current ETN groups in the swelling degree(P>0.05),suggesting that after ETN,the joint swell decreased significantly.Compared with normal group,there was a significant increase in COX-2 mRNA expression in model group(P<0.05),while in comparison with model group,COX-2 mRNA expression was downregulated significantly in joint synovitis in high-current ETN group(P<0.05).There were no significant differences between low-electric current and model groups,and between model and no-electric current groups in COX-2 mRNA expression(P>0.05).Conclusion: Electrothermo-needle has a favorable effect in anti-arthritis,and high intensity ETN can downregulate the expression of COX-2 mRNA in joint synovitis in arthritis rats.

摘要:Objective: To observe the effect of electroacupuncture(EA) on protein Bcl-2 and Bax expression in hippocampus in hyperlipemia rats with concomitant cerebral ischemia(CI).Methods: Forty male SD rats were randomized into control,CI,hyperlipemia+CI,and hyperlipemia+CI+EA groups,with 10 cases in each group.Hyperlipemia model was established by feeding the animals with high fat forage for 6 weeks and CI model established by occlusion of the unilateral middle cerebral artery.EA(1-(3 mA),(15 Hz)) was applied to bilateral "Sanyinjiao"(SP 6) and "Fenglong"(ST 40) for (20 min) every time;and "Baihui"(GV 20) and "Shuigou"(GV 26) were punctured and stimulated by twirled the acupuncture needle with hand continuously for(1 min).The treatment was conducted once daily and continuously for 17 days. The expression of protein Bcl-2 and Bax in the hippocampus was displayed by using immunohistochemical method.Results: In comparison with normal control group,Bcl-2 immuno-reaction(IR) positive cells in the hippocampus in CI and hyperlipemia+CI groups decreased significantly(P<0.01),Bax IR positive cells in the same two groups increased significantly(P<0.01),while Bcl-2/Bax also reduced remarkably(P<0.01),meaning that after ischemia the expression of Bcl-2 was downregulated and Bax upregulated markedly.In comparison with hyperlipemia+CI group,Bcl-2 IR activity increased,Bax IR activity reduced and Bcl-2/Bax increased considerably in hyperlipemia+CI+EA group(P<0.01,0.05),suggesting that EA could reverse the effects of hyperlipemia and CI.Conclusion: EA can eliminate hyperlipemia-and cerebral ischemia-induced abnormal expression of Bcl-2 and Bax in hyperlipemia rats with concomitant cerebral ischemia.
摘要:Objective: To explore the mechanism of electroacupuncture(EA) of "Neiguan"(PC 6) in relieving tachyarrhythmia in rats.Methods: Forty Wistar rats were randomly divided into normal control,model,PC-6,and non-acupoint groups,with 10 cases in each group.Tachyarrhythmia model was established by sublingual venous injection of 0.002% Aconitine((1 mL/kg)).EA(dense-sparse waves,(2/20 Hz),2-(3 mA)) was applied to bilateral "Neiguan"(PC 6) or non-acupoints(the prominence sites of the bilateral shoulder deltoid muscles) for(10 min) after successful establishment of tachyarrhythmia.The heart rates were recorded,and myocardial stimulatory G-protein(Gs) mRNA expression was detected by using reverse transcription-polymerase chain reaction(RT-PCR) method.Results: After injection of Aconitine,the heart rate of model group increased significantly compared with normal control group(P<0.01),and ventricular tachyarrhythmia and ventricular premature beat appeared;after acupuncture of "Neiguan"(PC 6),the rat's heart recovered its normal sinus rhythm in PC-6 group,while the arrhythmia and heart rate of non-acupoint group had no marked recovery.Compared with normal control group,the heart rate of non-acupoint group increased apparently(P<0.01).In comparison with normal control group,the optical density(OD) values of Gs mRNA expression of model group and non-acupoint group increased significantly(P<0.01);while compared with model group,the OD value of PC-6 group decreased considerably(P<0.01),and no significant difference was found between model and non-acupoint group in OD values(P>0.05).It suggested that EA could downregulate the expression of Gs mRNA.Conclusion: EA of "Neiguan"(PC 6) can correct Aconitine induced increase of heart rate and tachyarrhythmia which is related to its effect in reducing the upregulated expression of Gs mRNA in rats with tachyarrhythmia.

摘要:Objective: To observe the effect of electroacupuncture(EA) of acupoints on experimental cutaneous pruritus induced by intradermal injection of hydroxytryptamine(5-HT) in rats.Methods: For establishing cutaneous pruritus model,14 male Spraque-Dawley rats were randomly divided into normal saline(NS) group(n=6) and model group(n=8).Pruritus model was established by intradermal injection of(10 μL) of NS or 2% 5-HT at the nape of the neck.After injection,the rats were quickly put into an observation chamber and videotaped for(60 min).The scratching response toward the injected site was counted by playing back the video.Thereafter,other 48 rats were randomly allocated to post-anesthetization model(PAM) group(n=12),EA-I group(n=12),EA-II group(n=12) and sham-EA group(n=12).For rats of PAM group,pentobarbital sodium((40 mg/kg)) was injected intraperitoneally (2.5 h) prior to the injection of 5-HT.EA(4-(16 Hz),1-(3 V)) was applied to unilateral "Quchi"(LI 11) and "Hegu"(LI 4) for EA-I group,"Zusanli"(ST 36) and "Sanyinjiao"(SP 6) for EA-II group,and "nonacupoints" nearby the above-mentioned 4 acupoints for sham-EA group after the rats were anesthetized.When the rats in EA and sham-EA groups regained consciousness from anesthesia,5-HT was injected intradermally.Results: Compared with NS group,the scratching times in model group increased significantly after injection of 5-HT(P<0.001).There was no significant difference between model group and PAM group in the scratching times(P>0.05).EA of LI11 and LI-4 or ST-36 and SP-6 could significantly reduce the rats' scratching times(P<0.01,0.05) in comparison with PAM group,and no statistical difference was found between two EA groups(P>0.05).Compared with PAM group,the scratching times of sham-EA had no marked changes,suggesting no inhibitory role of sham-EA on 5-HT induced itching reaction(P>0.05).Conclusion: Intradermal injection of 5-HT induces itching response,and EA can relieve the itching reaction and EA of acupoints has a special effect.
摘要:Objective:To study the effect of electroacupuncture(EA) at "Hegu"(LI 4) with different stimulation duration in combination with needle retention in "Sanyinjiao"(SP 6) for different duration on uterine contraction in late-stage pregnant rats.Methods: A total of 50 late-stage pregnant(19(th) day after pregnancy) Wistar rats were randomized into control group,LI 4-EA-(20 min)+SP 6-needling(N)-(5 min)(i.e.,EA stimulation of LI 4 for(20 min) first and then SP 6 was punctured with the needle retained for(5 min) without electrical stimulation,similar expression in the following groups),LI 4-EA-(20 min)+SP 6-N-(20 min),LI 4-EA-(40 min)+SP 6-N-(5 min),LI 4-EA-(40 min)+SP 6-N-(20 min) groups with 10 cases in each group according to the requirements of 2×2 factorial experiment design.In addition,other 10 non-pregnancy Wistar rats were used as normal control group.EA((20 Hz),1-(3.5 V)) was applied to LI 4 for(20/40 min) in the abovementioned 4 EA groups respectively.A muscular tension transducer was fixed to the mid-segment of the uterus for detecting the amplitude,frequency and contractility(amplitude X frequency) of the uterine contraction. Results: In comparison with normal control group,the amplitude,frequency,and contractility of the uterine contraction in control group increased significantly(P<0.05).Comparison between factor A(LI 4: EA-20/40 min) and factor B(SP 6: needle kept in place for 5/20 min) among the 4 EA groups showed that 5-(10 min) and 15-(20 min) after removal of the needle,both factor A and factor B had no significant influence on the amplitude of the uterine contraction;factor B could significant raise the frequency and contractility of the uterus contraction(P<0.05,0.01);and the effects of SP 6-N-(5 min) in increasing the frequency and contractility were markedly superior to those of SP 6-N-20 min(P<0.05,0.01);while factor A had no marked influence on the frequency and contractility of the uterus.Comparison of the 4 EA groups showed that 5-(10 min) after removal of the acupuncture needle,the amplitude of the uterine contraction in LI 4-EA-20 min+SP 6-N-(5 min) group was markedly higher than that of LI 4-EA-(20 min)+SP 6-N-(20 min) group;(5 min) after needling SP 6 and 5-(10 min) after removal of needle,the frequency of uterine contraction in LI 4EA-(40 min)+SP 6-N-(5 min) group was significantly higher than that of LI 4-EA-(40 min)+SP 6-N-(20 min) group(P<0.05).Conclusion: EA of LI 4 plus needling SP 6 can promote the uterine contraction activity in pregnancy rats,and the best combination is EA LI 4(20 min) first plus needling SP 6(5 min) later.

摘要:Objective: To investigate the effect of acupuncture on visceral sensitivity in patients with functional dyspepsia(FD) who were undergoing mechanical gastric distention.Methods: Thirty volunteer FD patients were randomly divided into control group(n=11) and acupuncture group(n=19) according to the designed random table.For patients of acupuncture group,Zusanli(ST 36) and Neiguan(PC 6) were punctured and stimulated with an electric pulse generator by setting the parameters being(10 mA) in strength,(500 μs) in the duration of pulses,(40 Hz) in frequency,12 cpm of pulse trains,and(15 min) in the duration of stimulation.Tianshu(ST 25) was punctured with the needle retained.Isovolumetric gastric distention was performed in all the subjects through a balloon which was swallowed before hand and filled with(300 mL) of air and then connected to SVS/Barostat(Synectice Visceral Stimulator).Heart rate variability(HRV) was recorded and analyzed by using HS9616 Dynamic Electrocardiography Recorder.The maximal tolerable volume and pressure of the stomach were recorded,the epigastric pain,fullness and nausea were scored simultaneously.The automatic nervous function was assessed by the power spectral analysis of HRV.Results: Self-comparison showed that in acupuncture group,the scores of gastralgia,fullness and nausea,and low frequency/high frequency(LF/HF) and ultimate LF of HRV decreased remarkably(P<0.05),and the maximal tolerable volume and pressure,LF and HF of HRV increased significantly after acupuncture stimulation(P<0.05,0.01),suggesting that acupuncture stimulation could lower gastric distension induced increase of the visceral sensitivity(noxious reactions),lower the activity of the sympathetic nerve and potentiate vagal tension in FD patients.No significant changes were found in control group(P>0.05).Conclusion: Acupuncture can reduce the increased visceral sensitivity in FD patients,which may be realized by favorable regulation of activities of the automatic nerve.
摘要:Objective: To observe the effect of electroacupuncture(EA) on the learning and memory ability in diabetic cognitive dysfunction patients.Methods: Forty-five cases of diabetic mellitus(DM) patients with cognitive dysfunction were randomly divided into EA group(n=25) and medication group(n=20).For patients of EA group,main acupoints as Sishenchong(EX-HN 1),Baihui(GV 20),bilateral Fengchi(GB 20), Benshen(GB 13) and Neiguan(PC 6) were punctured with filiform needles and stimulated electrically by setting the stimulating parameters being dense-sparse waves,frequency of 14-26 pulses/min,strength of 3-(5 mA) and duration of (30 min).The treatment was conducted once daily,5 times a week and continuously for 6 weeks.Patients of medication group were ordered to take Nimotop((30 mg)/time),3 times everyday and continuously for 6 weeks.Modified Hastgawa Dementia Scale(HDS) and Wilson's Memory Scale(WMS) were used to evaluate the patients' cognitive function,memory ability and general conditions before and after the treatment.Results: After the treatment,according to HDS, of the 20 and 25 cases in medication and EA groups,1(5%) and 4(16%) experienced marked improvement,5(25%) and 15(60%) had moderate improvement,10(50%)and 5(20%) had mild improvement,and 4(20%) and 1(4%) had no improvement, with the total effective rates being 80% and 96% respectively.Rank test showed that the therapeutic effect of EA group was significantly superior to that of medication group(P<0.05) in improving patients' cognitive function.Self-comparison indicated that WMS of both groups increased significantly after the treatment(P<0.001,0.05),including adding test of long-term memory and touching test of short-term memory in medication group,counting-down test of long-term memory,association test and touching test of short-term memory in EA group;and the effects of EA were apparently superior to those of medication in raising WMS(MQ),counting-down test and association test(P<0.05,0.01).Conclusion: The therapeutic effect of EA is significantly superior to that of Nimotop in improving DM patients' cognitive function,learning and memory ability.

摘要:Objective: To investigate the regularities and characters of the ventrimeson pigmentation phenomenon in pregnant women and puberal college students so as to explore the correlation between the ventrimeson pigmentation and the Conception Vessel(CV).Methods: The investigation was conducted in 68 predelivery women including 64 primiparae and 4 pluriparae ranging in age from 21 to 39 years(mean 28.03±3.62 years) and 148 puberal college students including 99 males and 49 females ranging in age from 17 to 24 years(mean 21.24±1.19 years).In these volunteer subjects,the pigmentation trace was photographed and recorded by a 3 people-composed group of the present paper.Results: The ventrimeson pigmentation appeared in all the 68(100%) predelivery women being dark-brown in color,starting from clitoris and ascending along the running course of the CV,ranging in length from(27 cm)(reaching the inferior part of the xiphoid) to(45 cm)(over the xiphoid,mean 34.91±(3.58 cm)) and ranging in width from (3-16 mm)(mean 8.07±(2.57 mm));while in 148 college students, the pigmentation band appeared in 41(83.67%) girls and 7(7.07%) boys,starting from the clitoris or the root of penis to ascend along the ventrimeson and ending below the umbilicus in most girls and in the 7 boys,ranging in length from the upper border of the pubic symphysis(artificial measure point) to the ending point being 13-(28 cm)(mean 15.04±(2.40 cm)) in 41 girls and 12-(36 cm)(mean 19.14±(6.94 cm)) in 7 boys,ranging in width from(2-13 mm)(3.01±(0.58 mm)) in these girls and from 2-(3 mm)(2.75±(0.50 mm)) in these boys.The appearance rate,average length and width of the pigmentation band of pregnant women were significantly bigger than those of puberal college students(P<0.01),and the color of the pigmentation in pregnant was also apparently heavier.Conclusion: The ventrimeson pigmentation,coincident to the running course of CV,is one of the visible meridian phenomena,which is important to learn the essence of meridian by making clear of its regularities,characters and mechanisms.
摘要:In this article,the authors discuss the depth of the acupoint and the depth of the acupuncture needle insertion from the following 5 aspects: 1) the depth of the acupoint depends on the location of the meridian,2) the depth of needle insertion depends on the depth of the acupoint,3) both the depth of the acupoint and needle insertion depend on the needling sensations,(4) the) depth of the acupoint doesn't equal to the safe depth of needle insertion,and 5) the depth of needle-insertion is judged by the therapeutic effect.The depth of the acupoint,the depth of needle insertion and the safe depth of needle insertion are closely related to each other but their concepts are different.The needle insertion should be within the safe depth so as to avoid unnecessary injury or accidents.In particular,when an acupuncture needle inserted,the local anatomical structure,the patient's constitution,the state of the disease,the season,the direction and angles of the needle-tip,different layers(dermal part,aponeurotic system,meridian or the collateral of the meridian,etc) of the body should be considered together in clinical practice.
摘要:Currently,the application of acupuncture therapy for acute facial palsy still has some arguments.In the present paper,the authors make some comments on the applicability of electroacupuncture(EA) therapy for early treatment of idiopathic facial palsy during acute stage from 1) results of clinical random controlled studies,2) results of animal studies,and 3) the basic theory of traditional Chinese medicine.The authors conclude that electroacupuncture therapy is adoptable for idiopathic facial palsy during acute stage under the premise of proper choice of stimulating parameters.
摘要:In the treatment of diseases,the essence of moxibustion therapy of traditional Chinese medicine is the application of thermal conduction and thermal radiation when the local acupoints are stimulated by the ignited moxa.The inspection and analysis techniques of the infrared radiation spectrum of human acupoints and meridians are applied to exploring the objective regularities of meridians by using the non-touch and non-injurious characters of the infrared radiation.In the present paper,the authors review recent situations of the application of infrared radiation spectrum in the studies on acupuncture,moxibustion and meridians from 1) the infrared radiation spectrum of moxibustion,2) the infrared radiation spectrum of acupoints,3) diagnosis of some diseases and observation on the therapeutic effect of acupuncture,and 4) tracks of infrared radiation along the running course of the meridian in the human body and the affected factors.
