      摘要:Objective To observe the inhibitory effects of acupuncture pretreatment on arrhythmia and intracellular calcium(i) oscillation in single cardiocyte in rats subjected to myocardial ischemia-reperfusion(MIR).Methods Male SD rats were evenly randomized into normal control(NC),MIR and electroacupuncture(EA) groups.MIR was performed by ligation(30 min) and reperfusion(10 min) of the left anterior descending coronary artery in vivo in rats or in the isolated and perfused rat heart subjected to a 40 min global ischemia followed by a 10 min reperfusion.EA pretreatment was applied to bilateral "Neiguan"(PC 6) for 30 min once a day in 3 consecutive days.Arrhythmic score was calculated according to Curtis's and Walker's method,and the count of i oscillation was measured by using InCytPm 2 intracellular ion detecting system.Results Both the arrhythmic score and the count of i oscillation in MIR group were considerably higher than those in NC group(P<0.05).While compared with MIR group,the arrhythmia score and the count of i oscillation in EA group were decreased significantly(P<0.05).Conclusion Pretreatment of EA can effectively resist MIR-induced arrhythmia and intracellular calcium oscillation in the rat.

      摘要:Objective To observe the accumulative analgesic effect of electroacupuncture(EA)and the simultaneous changes of protein kinase A(PKA) expression in hypothalamus and hippocampus in rats with chronic constrictive injury(CCI) or/and learning-memory impairment.Methods Sixty-eight female Wistar rats were randomized into normal control,CCI,CCI+EA 2 d,CCI+EA 2 w,OVX+CCI,OVX+CCI+EA 2 d and OVX+CCI+EA 2 w groups,with 10 cases in each except OVX+CCI group(n=8).CCI pain model was established by ligature of the right sciatic nerve with surgical suture.EA(2/15 Hz,1-2 mA,30 min) was daily applied to bilateral "Zusanli"(ST 36) and "Yanglingquan"(GB 34) for 2 days(2 d) and 2 weeks(2 w) respectively.Neuro-memory impairment was induced by OVX plus subcutaneous injection of D-galactose.Hyperalgesic scores(HS) were detected with radiation-heat irradiation and PKA activity was detected by immunohistochemical method.Results After CCI,HS of all groups increased significantly in comparison with normal control group(P<0.05).In simple CCI rats,compared with CCI group,HS of both CCI+EA 2 d and CCI+EA 2 w groups decreased markedly(P<0.05),and that of CCI+EA 2 w group was evidently lower than that of CCI+EA 2 d group on the 18th day after CCI(P<0.05).In memory impairment rats,HS of both OVX+CCI+EA 2 d and OVX+CCI+EA 2 w groups was significantly lower than that of OVX+CCI group(P<0.05);and no significant difference was found between OVX+CCI+EA 2 w and OVX+CCI+EA 2 d groups in HS(P>0.05).Results of immunohistochemistry showed that the integral grey values(IGV) of PKA of paraventricular nucleus(PVN),arcuate nucleus(ARC) and supraoptic nucleus(SON) of hypothalamus in CCI+EA 2 w group were significantly lower than those in normal control and CCI+EA 2 d groups(P<0.05).IGV of PVN and SON in OVX+CCI+EA 2 w group were also markedly lower than those in OVX+CCI group(P<0.05),displaying upregulation and cumulative increase of PKA expression by EA 2 w.IGV of PKA,ARC and SON in OVX+CCI+EA 2 w group were markedly higher than those in CCI+EA 2 w group(P<0.05),suggesting an apparent attenuation of the cumulative upregulation effect of PKA expression after learning-memory injury.The changing tendency of PKA expression in hippocampal CA1 region was similar to that of SON after repeated EA of ST36 and GB34.Conclusion Repeated EA has a cumulative analgesic effect in CCI rats,which is probably related to its resultant upregulation of PKA expression in neurons of hypothalamus and hippocampus and the animals' neuro-memory.

      摘要:Objective To observe the effects of electroacupuncture(EA) of "Neiguan"(PC 6),"Shenmen"(HT 7),"Taiyuan"(LU 9),"Zhizheng"(SI 7) and "Sanyinjiao"(SP 6) on cardiac function in acute myocardial ischemia(AMI) rabbits.Methods A total of 60 rabbits were randomly divided into normal control,model,EA-PC6,EA-HT7,EA-LU9,EA-SI7 and EA-SP6 groups.AMI model was established by intravenous administration of pituitrin.ECG,maximal ascending rate(+dp/dt max) and maximal descending rate(-dp/dt max) of the left ventricular pressure,and left ventricular systolic pressure(LVSP) were recorded.Results The heart rate(HR),+dp/dt max,-dp/dt max and LVSP decreased significantly after AMI(P<0.01).Compared with model group,the 4 indexes of EA-PC6,EA-HT7 and EA-SI7 groups increased considerably(P<0.05,0.01) at most time-points after AMI,and the effects of EA-PC6 and EA-HT7 were markedly better than those of EA-SI7,EA-LU9 and EA-SP6(P<0.05,0.01).No significant differences were found among model group,EA-LU9 and EA-SP6 groups in HR,+dp/dt max,-dp/dt max and LVSP at most time-points after AMI.Conclusion EA of PC6,HT7 and SI7 has a good effect in improving ischemic cardiac performance,and the effect of PC6 and HT7 is markedly better than that of SI7.The effect of EA of SP6 and LU9 is poorer on ischemic cardiac function.

      摘要:Objective To observe the effect of electroacupuncture(EA) on mitochondrial SDH and Na+-K+-ATPase activity,and lactate acid(LA) content in the peri-hemotoma tissue in rats with intracerebral hemorrhage(ICH) so as to explore its mechanisms in resisting cerebral ischemia.Methods A total of 80 Wistar rats were randomized into normal group,sham group,model group,EA group.The last three groups were further divided into 24 h,72 h and 168 h subgroups respectively,with 8 cases in each group/subgroups.ICH model was established by caudatum-injection(right) of Collagense Ⅶ/heparini solution(2 μl).EA(2 Hz,1 mA) was applied to "Baihui"(GV 20)-"Taiyang"(EX-HN 5) for 30 min.Chondriosome suspension of the cerebral peri-hemotoma tissue was got by gradient centrifugation for detecting SDH and Na+-K+-ATPase activity,and LA content with chromometry.Results In comparison with sham-operation group and control group,SDH and mitochondrial Na+-K+-ATPase activity in cerebral peri-hemotoma tissue declined significantly from 72 h and 24 h on respectively(P<0.05),while LA level increased markedly from 24 h on(P<0.05) in model group.Compared with sham-operation group,SDH activity(168 h) and Na+-K+-ATPase activity(24 h,72 h and 168 h) in EA group also decreased significantly(P<0.05);LA level(24 h,72 h) in EA group increased considerably(P<0.05).Whereas compared with model group,SDH and Na+-K+-ATPase activity increased significantly at most time courses(P<0.05),and LA level decreased remarkably(P<0.05) in EA group.Conclusion EA can improve the energy metabolism,upregulate the activity of chondriosome,and reduce the accumulation of LA in the cerebral peri-hemotoma tissue of ICH rats.

      摘要:Objective To observe the effect of electroacupuncture(EA) on learning-memory ability of vascular demen-tia(VD) rats,and the simultaneous changes of plasma and cerebral somatostatin(SS) and β-endorphin(EP) contents.Methods Forty-one SD rats were randomly divided into control(n=8),model A(n=8,no treatment) and B(n=8,intragastric perfusion of 15% saline),EA(n=9) and medication(n=8,intragastric perfusion of Nimoldipine,12 mg/kg) groups.VD model was established by using modified 4-vessels occlusion method.EA(150 Hz,1-2 mA) was applied to "Baihui"(GV 20),"Dazhui"(DU 14),"Pishu"(BL 20) and "Shenshu"(BL 23) for 20 min,once daily for 15 days.Morris water maze tests were conducted for evaluating the rats' learning-memory ability.The contents of SS and β-EP in plasma and brain tissue were measured by radioimmunoassay(RIA).Results In comparison with sham-operation group,the escape latency(EL) prolonged significantly and the target-platform crossing times decreased remarkably(P<0.01) in model group B.In comparison with model group B,EL shortened and target-platform crossing times increased both significantly in EA and medication groups(P<0.01).Plasma and cerebral SS,and cerebral β-EP contents of model groups A and B were significantly lower than those of sham-operation group(P<0.01),while plasma β-EP level had no obvious change(P>0.05).Plasma and cerebral SS,and cerebral β-EP contents in both EA and medication groups were considerably higher than those in model groups A and B(P<0.01).No significant differences were found between EA and medication groups in EL,target-platform crossing times,plasma and cerebral SS and β-EP levels,and between model group A and model group B in plasma and cerebral SS and β-EP levels(P>0.05).Conclusion EA can raise plasma and cerebral SS and cerebral β-EP levels,and improve the learning-memory ability in VD rats.

      摘要:Objective To observe the effect of electroacupuncture(EA) pretreatment on hippocampal glutamate(Glu) content and N-methyl-D-aspartate receptor(NMDAR)1 mRNA expression in rats with vascular dementia(VD) so as to explore its underlying mechanism in improving VD.Methods A total of 72 SD rats were randomized into control(n=16),sham-operation(n=16),model(n=20) and EA pretreatment(n=20) groups.VD model was established by modified middle cerebral artery occlusion.EA(1 mA,1.7 Hz) was applied to "Baihui"(GV 20),"Shenshu"(BL 23) and "Zusanli"(ST 36),once daily for 10 days.Glu content of right hippocampus tissue was detected with chromatometry.NMDAR1mRNA expression of hippocampus was detected by in situ hybridization histochemistry.Results In comparison with sham operation group,Glu content and NMDAR1 mRNA expression of hippocampus in model group increased significantly(P<0.01).Compared with model group,Glu content and NMDAR1mRNA expression of hippocampus in EA pretreatment group decreased significantly(P<0.01,0.05).Conclusion EA pretreatment can suppress the increase of Glu content,down-regulate NMDAR1mRNA expression in VD rats,which may contribute to its effect in relieving VD via reducing apoptosis and protecting cerebral neurons.

      摘要:Objective To investigate the effect of electroacupuncture(EA) on mitochondrial membrane potential and neuronal apoptosis in the cerebral cortex of rats with focal cerebral ischemia/reperfusion(CI/R) injury.Methods Thirty-six male SD rats were uniformly randomized into sham operation(sham),CI/R and CI/R+ EA groups.CI/R model was established by middle cerebral artery occlusion(MCAO) and reperfusion.EA(2/15 Hz,1 mA) was applied to "Shuigou"(GV 26) and "Baihui"(GV 20) for 30 min after CI/R.The mitochondrial membrane potential and neuronal apoptosis in the cerebral cortex(fronto-parietal lobes) in each group were detected by flow cytometer(FCM).Results In comparison with sham group,the mitochondrial membrane potential of CI/R group decreased significantly(P<0.01),while compared with CI/R group,it increased remarkably in CI/R+EA group(P<0.01).The percentage of neuronal apoptosis in CI/R group was significantly higher than that in sham group(P<0.01),and that of CI/R+EA group was significantly lower than that of CI/R group(P<0.01).Conclusion EA can alleviate CI/R injury partially by up-regulating the mitochondrial membrane potential,which may contribute to its effect in reducing cerebral neuronal apoptosis.

      摘要:Objective To investigate the mechanism of electroacupuncture(EA) in treating nonalcoholic fatty liver disease(NAFLD) in rats.Methods Thirty male SD rats were randomized into normal control,NAFLD model and EA groups,with 10 cases in each.NAFLD model was established by feeding the rats with high-fat food for 8 weeks.EA(2 Hz,4 mA) was applied to "Zusanli"(ST 36),"Fenglong"(ST 40),"Sanyinjiao"(SP 6),and "Taichong"(LR 3) for 15 min,once daily for 4 weeks.Fasting blood sugar(FBG) and serum free fatty acids(FFA) were detected with chromatometry;serum insulin(INS) was determined with radioimmunoassay;homeostasis model assessment insulin resistance(HOMA-IR) and liver index(LI) were calculated.Liver tissue was cut into sections for detecting pathological changes.Results Compared with normal control group,FBG,INS,FFA,HOMA-IR and LI of model group increased significantly(P<0.05),and hepatic diffuse steatosis with cellular necrosis and inflammatory cell infiltration was obvious.In comparison with model group,all the 5 indexes in EA group decreased significantly(P<0.05),and pathological changes as hepatic steatosis,etc.were significantly reduced.Conclusion EA may be used to effectively improve NAFLD by lowering insulin resistance.

      摘要:Objective To observe the effect of electroacupuncture(EA)on synovia superoxide dismutase(SOD) activity,malondialdehyde(MDA) and nitric oxide(NO) contents in rabbits with knee osteoarthritis(KOA).Methods A total of 40 New Zealand rabbits were randomly divided into control,model,massage and EA groups,with 10 cases in each.KOA model was established by gypusum fixing method.EA(1.6-2 Hz,1-2 mA) was applied to left "Yanglingquan"(GB 34),"Xuehai"(SP 10),"Zusanli"(ST 36) and "Liangqiu"(ST 34) for 25 min,once daily for 21 days.For massage group,the affected knee joint was pressed,kneaded,stretched and rotated repeatedly for 15 min every time,followed by forced running about 100 m.The intra-joint synovia was collected(0.4-0.6 ml) for detecting contents of SOD with xanthine oxidase method,MDA with thiobarbituric acid method and NO with nitrate reductase method.Results Compared with control group,synovia SOD activity in model group decreased considerably(P<0.05),while MDA and NO contents increased significantly(P<0.05).After 3 weeks' treatment,compared with pre-treatment and model group,synovia SOD activity increased markedly(P<0.05),and MDA and NO contents lowered remarkably in both EA and massage groups(P<0.05).No significant differences were found between EA and massage groups in these 3 indexes(P>0.05).Conclusion Both EA and massage can raise synovia SOD activity and lower MDA and NO content,which may contribute to their effect in relieving knee osteoarthritis in the rabbit.

      摘要:Objective To observe the effect of transcutaneous electric acupoint stimulation(TEAS) on plasma superoxide dismutase(SOD) and malondialdehyde(MDA) in rats with sports fatigue so as to explore its mechanisms in resisting exercise-induced fatigue.Methods Twenty-seven male adult SD rats were randomly divided into control group(n=9),model group(n=9) and TEAS group(n=9).Sports fatigue model was established by using treadmill method,i.e.forcing the rat to run 10 min at a speed of 10 m/min,15 m/min,20 m/min,24 m/min and 28 m/min,once daily for 6 days.TEAS(continuous waves,2 Hz,5 mA) was applied to unilateral "Zusanli"(ST 36) for 30 min,once per day for 7 days.The exhausted exercise time from starting running to exhaustion was recorded on the 7th day and the lactate levels,SOD activity and MDA contents in plasma were measured by lactate oxidase method,xanthine oxidase method and thiobarbituric acid method respectively.Results The duration of exhausted exercise in model group and TEAS group were(56.00±12.27) min and(70.88±13.74) min respectively,displaying significant increase in exercise tolerance after TEAS(P<0.05).The lactate level and MDA content in model group were markedly higher than those in control group(P<0.05);while compared with model group,lactate and MDA contents in TEAS group were significantly lower(P<0.05),and plasma SOD activity in TEAS group was significantly higher(P<0.01).No significant differences were found in plasma lactate content between control and TEAS groups and in SOD activity between model group and control group(P>0.05).SOD/MDA of model group was significantly lower than those of control group and TEAS group(P<0.05,0.01).Conclusion TEAS at "Zusanli"(ST 36) can effectively postpone exercise-induced fatigue by reducing accumulation of blood lactate,improving anti-oxidative ability and relieving lipid peroxidation in the rat.

      摘要:Objective To observe the effects of weak and strong electroacupuncture(EA) on endotoxin(ET) thermolysis-induced changes of discharges of neurons in the preoptic region and anterior hypothalamus(PO-AH) so as to explore its underlying mechanism in antipyretic and thermolytic actions and its relation to the receptive system of acupoints.Methods Eighteen New Zealand rabbits were randomly divided into ET group,ET+weak-EA group and ET+strong-EA group.Extracellular discharges of the PO-AH neurons were recorded by using tungsten microelectrodes.A "U"-shape stainless steel tube was implanted in the region(P0.4-A4.4,L0.5-1.7) crossing the hypothalamus for changing local temperature by perfusion of cool(25 ℃) or warm(41 ℃) solution in order to distinguish the heat sensitive neurons(HSN),cold sensitive neurons(CSN) and insensitve neurons to temperature changes.Intravenous injection of endotoxin(25 EU/rabbit) was given to rabbits to induce increase of tempe rature.EA(8 Hz,wave width 0.1 ms,weak stimulation: 4.5 V,strong stimulation: 25 V) was applied to bilateral "Yongquan"(KI 1) for observing changes of firing rates of HSN in PO-AH.Results Compared with the basal values of firing rates of PO-AH neurons in each group,the average changing ratios of both ET and ET+weak-EA groups decreased significantly from 55-60 min on in ET group and from 40-45 min on in ET+weak-EA group after intravenous injection of ET(P<0.05),suggesting no marked effect of weak EA for preventing discharges of PO-AH neurons from decrease.While in ET+strong-EA group,the firing rates of HSN of PO-AH kept stable after injection of ET during EA and after cease of EA(P>0.05 vs basal value),suggesting that strong EA could antagonize ET thermolysis-induced decrease of firing rates of PO-AH neurons.Conclusion Stronger EA stimulation of KI1 can antagonize ET thermolysis-induced effect on electrical activities of PO-AH HSN,which may be initiated by the activation of the high-threshold thin nerve fibers in the acupoint region.

      摘要:Objective To observe the effect of electroacupuncture(EA) in the treatment of vascular dementia(VD) so as to explore an effective therapy for it.Methods Sixty cases of VD were randomly divided into acupuncture group and medication(Duxil,40 mg/time,2 times/d for 10 weeks) group,with 30 cases in each group."Xiusanzhen" means that two acupuncture needles were penetrated subcutaneously from bilateral Yingxiang(LI 20) to Shangyingxiang(EX-HN 8) respectively,and the 3rd needle was penetrated from the midpoint between two Yangbai(GB 14) through Yintang(EX-HN 3) to the nasal root,followed by applying EA stimulation to the acupoints.Hasegawa's Dementia Scale(HDS),Mini-mental state examination(MMSE),and Functional Activities Questionnaire(FAQ) were used for assessment of the VD patient's state before and after the treatment.Results After the treatment,the total effective rates of acupuncture and medication groups were 80.00% and 73.33% respectively.The effective rate of acupuncture group was significantly superior to that of medication group(P<0.05).Before treatment,scores of HDS,MMSE and FAQ in both groups had no significant differences(P>0.05);after the treatment,the scores of HDS and MMSE increased significantly,and that of FAQ decreased remarkably in both groups(P<0.01),and the difference of scores(or absolute value of FAQ) between post-treatment and pre-treatment in EA group was obviously higher than that in medication group(P<0.05).Conclusion Acupuncture of "Xiusanzhen" is effective in the treatment of VD patients.

      摘要:In the present paper,the authors introduce the development of researches on the specificity of acupoints in morphological structure(nerves,blood vessels,musculotendons,connective tissues,etc),biophysical characteristics(electricity,sonics,photology,thermology,electromagnetism),pathological response,and effectiveness.However,the aforementioned researches are basically at the stage of phenomenon-observation,some key factors influencing acupoint specificity and the underlying mechanisms have no definite or lack convincing results.Further studies should aim at those acupoints for some effective diseases(or symptoms) of acupuncture by using multiple modern scientific techniques.Some key scientific issues about the biophysical characteristics,pathological responses,efficacy,and combination regularity of acupoints should be solved reasonably.Moreover,theoretical models about acupoint specificity should be established for developing the theory of acupoint learning.

      摘要:The curative effect of acupuncture depends much on accurate location of acupoints.But,in the developmental course of Chinese acu-moxibustion medicine,some divarications about the location of acupoints have occurred for ages.In the process of formulating international standard of acupoint location,experts from China,Japan and Korea had different viewpoints about the location of 92 acupoints.After reviewing Chinese ancient and modern literatures,authors of the present paper sum up five basic factors influencing the location of acupoints: ① different understandings about the bone-length;② changes of bone-length;③ faults during copying literature;④ different understandings on the location in the text of books;⑤ evolvement of concepts.These viewpoints may be helpful to solve the divarication of acupoints' location.

    • Discussion on the Qi-passage of Meridian-collateral System

      摘要:The author of the present paper holds that "Qi-passage" is in fact the essence of meridian-collateral system in the human body.In my opinion,the so called "Jing" of the fourteen meridians is referred to the tissue structure of the longitudinal fascia space;the so called "Luo" of the fifteen collaterals referred to the tissue structure of the transversal fascia space,and the "Mai" of meridians and collaterals referred to tissue liquid-Qi of both longitudinal and transversal fascia space.The "Dao"(Qi-passage) of meridians and collaterals is the tissue structure of both longitudinal and transversal fascia space.The tissue liquid-Qi and tissue structure depend on each other.The doctrine of "Qi-passage" of meridian-collateral system has its original evidence,scientific and substantive basis.The meridian-collateral system is a regulative and control system of the unknown synthetic functions of multiple substances including loose connective tissue,tissue liquid-Qi,energy substances,nerves,blood vessels and lymph,etc..
