目的:观察针刺对慢性束缚应激(CRS)大鼠海马、前额叶皮层星形胶质细胞(AST)中胶质纤维酸性蛋白(GFAP)及血清白介素-10(IL-10)表达的影响
探讨针刺抗抑郁效应机制。方法:SD大鼠随机分为空白组、模型组、针刺组、氟西汀组
每组8只。采用28dCRS结合孤养方法建立抑郁大鼠模型。针刺组针刺"百会""印堂"和双侧"三阴交"穴20min
氟西汀组予以氟西汀灌胃治疗
均每日治疗1次
共治疗28d。采用糖水消耗实验和旷场实验进行行为学评价
用蛋白免疫印迹法、酶联免疫吸附法分别检测海马、前额叶皮层GFAP及血清IL-10含量。结果:模型组大鼠糖水消耗量、水平穿越格数和垂直竖立次数较空白组均显著降低(P<0.01);针刺组、氟西汀组糖水消耗量、水平穿越格数较模型组均增加(P<0.05)。模型组大鼠海马GFAP含量较空白组明显降低(P<0.01);针刺组、氟西汀组海马中GFAP含量较模型组均明显升高(P<0.01)。模型组大鼠前额叶皮层GFAP含量较空白组明显升高(P<0.01);针刺组、氟西汀组前额叶皮层GFAP含量较模型组均明显下降(P<0.01)。模型组血清IL-10含量明显低于空白组(P<0.01);针刺组、氟西汀组均明显高于模型组(P<0.01);氟西汀组明显低于针刺组(P<0.01)。结论:针刺可能是通过调节CRS抑郁大鼠海马、前额叶皮层AST中GFAP的表达
改善海马、前额叶皮层AST的功能
并增加血清抗炎性细胞因子IL-10的含量
从而发挥抗抑郁效应。
Objective To observe the effect of acupuncture on the expression of glial fibrillary acidic protein(GFAP)in hippocampus and prefrontal cortex and the level of serum interleukin 10(IL-10)in rats with depression
so as to explore its mechanism underlying improvement of depression.Methods Thirty-two male SD rats were randomly divided into four groups:control
model
acupuncture
and medication(Fluoxetine
Flu)(n=8 rats in each).The depression model was established by using chronic restraint stress(constraint
fasting
water deprivation
etc.)combined with solitary raising for 28 days.Acupuncture was applied to"Baihui"(GV 20)and"Yintang"(GV 29)
and bilateral"Sanyinjiao"(SP 6)for 20 min
once daily for 28 days.Fluoxetine(1.8 mg/kg)was given to rats of the medication group by gavage once every day for 28 days.Sucrose consumption test and open field test(crossing and rearing locomotion)were carried out to evaluate the behavioral changes.Western blot was used to detect the expression of GFAP in the hippocampus and prefrontal cortex
and the content of serum IL-10 was detected by enzyme linked immunosorbent assay(ELISA).Results After modeling

hippocampal GFAP protein expression and serum IL-10 content were significantly decreased and prefrontal GFAP protein expression was up-regulated markedly in the model group relevant to the control group(P<0.01).After the treatment
the decreased levels of sucrose consumption and crossing numbers
hippocampal GFAP protein expression and serum IL-10 content and the increased prefrontal GFAP protein expression were considerably suppressed in both medication and acupuncture groups compared with the model group(P<0.05
P<0.01).No significant differences were found between the acupuncture and medication groups in increasing the rats' locomotion
sucrose consumption
hippocampal GFAP protein expression(P>0.05)and in downregulating prefrontal GFAP protein expression(P>0.05)except up-regulation of IL-10 level.Conclusion Acupuncture intervention plays a positive role in anti-depression in rats
which may be related to its effects in regulating the expression of GFAP in the hippocampal and prefrontal astrocytes
and in increasing the content of serum anti-inflammatory cytokine IL-10.
