目的:观察电针督脉"至阳""脊中"对脊髓损伤(SCI)大鼠脊髓损伤区降钙素基因相关肽(CGRP)和Nod样受体蛋白3(NLRP3)炎性小体表达的影响
探讨电针对脊髓损伤的保护作用机制。方法:将36只雄性SD大鼠随机分为假手术组、模型组和电针组
每组再随机分为7 d组和14 d组两个亚组
每组6只。采用钳夹法复制SCI大鼠模型。电针组予电针刺激"至阳""脊中"
每次30 min
分别连续治疗7 d或14 d。采用BBB行为学评分法观察术后1、3、7、14 d大鼠运动功能的变化
采用蛋白免疫印记法检测术后7 d和14 d脊髓损伤处CGRP、NLRP3、凋亡相关的斑点样蛋白(ASC)和半胱氨酸天冬氨酸蛋白酶-1(caspase-1)的蛋白表达水平。结果:与同时点假手术组比较
模型组的BBB评分降低(P<0.05);与同时点模型组比较
电针组术后7、14 d的BBB评分升高(P<0.05)。与同时点假手术组比较
模型组术后7、14 d脊髓损伤处NLRP3、 ASC和caspase-1的蛋白表达均升高(P<0.05
P<0.01);与同时点模型组比较
电针组术后7、14 d脊髓损伤处NLRP3、 ASC和caspase-1的蛋白表达均降低(P<0.01)
CGRP蛋白表达均升高(P<0.01)。结论:电针督脉可明显改善SCI大鼠的运动功能
其机制可能与上调CGRP的表达
抑制NLRP3炎性小体的活化有关
CGRP可能是电针抑制NLRP3启动的上游因子。
Objective To observe the effect of electroacupuncture(EA) at “Zhiyang”(GV9) and “Jizhong”(GV6) of the Governor Vessel on the expression of calcitonin gene related peptide(CGRP)and NOD-like receptor pyrin containing 3(NLRP3) in the injured anterior horn(AH) of rats with spinal cord injury(SCI)
so as to explore its mechanisms underlying improvement of SCI. Methods Thirty-six male SD rats were randomly divided into sham operation(sham) group
model group and EA group which were further randomly allocated to 7 day(d) and 14 d subgroups(n=6 per subgroup). The SCI model was established by clamping the exposed spinal cord with an aneurysm clip. Rats of the EA group received EA of GV6 and GV9 for 30 min
once daily for 7 and 14 days
