目的:观察电针对模拟失重大鼠心肌组织形态学及能量代谢的调节效应
探讨不同腧穴改善失重大鼠心肌形态的可能机制。方法:40只雄性Wistar大鼠随机分为正常对照组、模拟失重组、内关组、三阴交组
每组10只。采用尾部悬吊法复制失重大鼠模型。内关组隔日电针"内关"穴30min
三阴交组隔日电针"三阴交"穴30min
共治疗4周。光镜下观察心肌组织形态学改变
化学比色法检测三磷酸腺苷(ATP)酶、琥珀酸脱氢酶(SDH)、乳酸脱氢酶(LDH)活性的变化。结果:模拟失重组大鼠心肌纤维排列稀松
可见大量肌纤维断裂
并可见局部溶解性坏死。内关组、三阴交组大鼠心肌细胞较完整
细胞核呈卵圆形居中
肌纤维排列整齐
无明显坏死、溶解
细胞间质轻度水肿。模拟失重组较正常对照组心肌ATP酶活性显著降低(P<0.05)
LDH活性增高(P<0.05);电针"内关"穴能显著降低LDH活性(P<0.05);电针"三阴交"穴能够显著提高ATP酶的活性(P<0.05)
显著降低LDH活性(P<0.05)。结论:电针能通过影响三羧酸循环
提高心肌线粒体功能
增加心肌组织氧气供应
提高ATP酶活性
进而改善心肌能量供应
并能够促进心肌病理形态学结构的修复
从而起到防护模拟失重大鼠引发心肌组织损伤的作用。
Objective To observe the effect of electroacupuncture(EA)stimulation of"Neiguan"(PC 6)and"Sanyinjiao"(SP 6)on morphological changes
myocardial energy metabolism in rats undergoing simulated weightlessness.MethodsA total of 40 male Wistar rats were randomly divided into normal control
weightlessness model
Neiguan(PC 6)and Sanyinjiao(SP 6)groups(10rats/group).EA was applied to PC 6and SP 6acupoints for 30 min
once every other day.The morphological changes were observed by light microscopy after H.E.staining.Lactate dehydrogenase(LDH)
succinate dehydrogenase(SDH)and ATPase activities in the left ventricular myocardium were measured by colorimetry.Results In weightlessness rats
myocardial pathological changes as loose arrangement
breakage and lytic necrosis of myocardial fibers were found
these changes were obviously improved in rats of both PC 6and SP 6groups.Compared with the control group
the ATPase activity was significantly decreased
while the LDH activity was significantly increased in the model group(P<0.05).After EA intervention
the ATPase activity was obviously increased in the SP 6group(P<0.05)
and LDH activity was apparently reduced in both PC 6and SP 6groups(P<0.05).No significant changes were found in myocardial SDH activity after modeling and EA of the two acupoints(P>0.05).Conclusion EA intervention has functions in up-regulating ATP activity and suppressing the myocardial LDH activity in weightlessness rats
which may be related to its effect in improving myocardial pathological changes.
