目的:探讨壮医药线点灸联合针挑疗法对血管氧化应激损伤的影响。方法:Wistar大鼠随机分为对照组、假手术组、模型组、联合组
每组20只。采用坐骨神经慢性压迫损伤法复制血管氧化损伤模型。联合组取双侧"足三里"进行药线点灸
取左侧"阳陵泉""环跳"进行针挑
点灸和针挑均7d为1个疗程
共进行3个疗程的治疗。采用放射免疫分析法检测血浆6-酮-前列环素F 1α(6-keto-PGF 1α)、血栓素B 2(TXB 2)、一氧化氮(NO)、内皮素(ET)的水平
免疫组化法观察股动脉环氧合酶2(COX-2)表达及血管形态学改变。结果:和对照组比较
模型组6-keto-PGF 1α、NO水平下降(P<0.05)
而TXB 2、ET水平显著上升(P<0.01)。和模型组比较
联合组6-keto-PGF 1α、NO水平上升(P<0.05)
而TXB 2、ET水平显著下降(P<0.01)。模型组COX-2表达较对照组增多
联合组COX-2表达较模型组显著减少(P<0.01)。模型组血管内膜增生
管壁增厚;联合组血管内膜增生
管壁增厚现象减轻。结论:壮医药线点灸联合针挑疗法可以下调COX-2表达
进而调节前列环素2/血栓素A 2及NO/ET平衡
抑制血管内膜增生和血管重构
可能是其抗血管氧化损伤的作用机制之一。
Objective To observe the effect of Zhuang-medical thread moxibustion combined with needle-pricking on vascular oxidative stress injury in oxidative stress injury rats.Methods Eighty Wistar rats were randomly allocated to normal control
sham operation(sham)
model
and combined treatment groups(n=20in each group).The oxidative stress injury model was established by ligation of the left sciatic nerve to induce chronic constriction injury(CCI)pain stress stimulation.Zhuangmedical thread moxibustion was applied to bilateral"Zusanli"(ST 36)
once a day for 3weeks.Needle-pricking was applied to left"Yanglingquan"(GB 34)and left"Huantiao"(GB 30)
once a day for 3weeks except Sundays.Plasma 6-keto-PGF 1α
thromboxane B 2(TXB 2)
NO and ET contents were assayed by radioimmunoassay.COX-2immunoactivity of the femoral artery was determined by immunohistochemistry
and pathological changes of the femoral artery were detected by H.E.staining.Results Compared with the control group
the levels of plasma 6-keto-PGF 1αand NO in the model group were significantly reduced(P<0.05)
while those of plasma TXB 2and ET and COX-2expression in the femoral artery were obviously increased in the model group(P<0.01).After moxibustion plus needle-pricking treatment
CCI-induced decrease of plasma 6-keto-PGF 1αand NO contents
and increase of plasma TXB 2and ET and COX-2expression levels were obviously reversed(P<0.05
P<0.01).The tubal wall of the femoral artery in rats of the model group got thicker
while that of the combined treatment group was relatively thinner
suggesting an inhibition of vascular intimal hyperplasia after the treatment.Conclusion Zhuang-medical thread moxibustion combined with needle-pricking of ST 36
GB 34 and GB 30 can reduce the expression of femoral artery COX-2and regulate the balance of both plasma PGI 2/TXA 2and plasma NO/ET in CCI-induced oxidative stress rats
which may contribute to its effect in suppressing oxidative stress-induced vascular intimal hyperplasia.
