目的:通过观察悬灸"大椎"穴不同灸量对哮喘大鼠支气管肺泡灌洗液(BALF)中神经生长因子(NGF)、P物质(SP)、降钙素基因相关肽(CGRP)和神经激肽A(NKA)、神经激肽B(NKB)、磷酸化细胞外信号调节激酶(pERK)含量的影响
探讨艾灸治疗哮喘的神经源性炎性反应机制以及最佳灸量。方法:SD雄性大鼠60只
随机均分为空白组、模型组、悬灸15min组、悬灸30min组、悬灸60min组、悬灸90min组。于第1天、第8天腹腔注射致敏混悬剂1mL
第15天开始用1%卵白蛋白生理盐水雾化吸入激发哮喘
每日1次
每次20min
共14d。21d后
温和灸"大椎"穴
各组悬灸不同的时间
共治疗7d。运用ELISA双抗体夹心法测定各组大鼠BALF中的NGF、SP、CGRP、NKA、NKB、pERK等神经源性炎性反应相关因子的含量。结果:模型组BALF中NGF、SP、CGRP、NKA、NKB、pERK含量明显高于空白组(P<0.01);4个治疗组BALF中NGF、SP、CGRP、NKA、NKB、pERK含量较模型组均有显著降低(P<0.01);悬灸15、30、60min组之间各项指标比较差异有统计学意义(P<0.01)
随悬灸时间的延长
各指标变化更加明显;悬灸60min组与悬灸90min组之间各项指标比较差异无统计学意义(P>0.05)。结论:悬灸"大椎"穴通过降低大鼠BALF中NGF、SP、CGRP、NKA、NKB的含量来减轻哮喘的神经源性炎性反应
其信号转导可能通过ERK细胞外信号转导途径;随着灸量的增加
对哮喘大鼠的神经源性炎性反应的改善作用也在增加
艾灸60min是饱和灸量
而90min的治疗时间并不能进一步提高疗效。
Objective To observe the effect of suspending-moxibustion stimulation of"Dazhui"(GV 14)with different quantities on the levels of nerve growth factor(NGF)
substance P(SP)
calcitonin gene-related peptide(CGRP)
neurokinin A(NKA)
neurokinin B(NKB)and phosphorylated extracellular signal-regulated kinases(pERK)in the bronchoalveolar lavage fluid(BALF)of asthma rats
so as to analyze its mechanisms underlying improving asthma.Methods Sixty male SD rats were randomly divided into six groups:blank control
model
15min-moxibustion(15min-moxi)
30min-moxi
60min-moxi and 90minmoxi(n=10rats in each group).The asthma model was established by intraperitoneal injection of suspension of egg protein
magaldrate
and inactivated Bacillus pertussis(on day 1and 8)
and inhaling the atomized ovalbumin saline(from day 15 on for 14days).Mild moxibustion was conducted at"Dazhui"(GV 14)for 15 min
30min
60 min and 90 min
respectively
once daily for7 days.The levels of NGF
SP
CGRP
NKA
NKB
and pERK in the BALF were detected by ELISA(enzyme-linked immuno sorbent assay).Results The contents of NGF
SP
CGRP
NKA
NKB and pERK in the BALF in the model group were obviously higher than those in the blank control group(P<0.01)
suggesting an apparent inflammatory reaction in rats after modeling.Following moxibustion
the levels of NGF
SP
CGRP
NKA
NKB and pERK of the four treatment groups were significantly downregulated compared with the model group(P<0.01).The effect of 30min-moxi group was obviously superior to that of 15 minmoxi group(P<0.01)
and those of 60min-moxi and 90min-moxi groups were markedly superior to those of 15min-moxi and 30min-moxi groups(P<0.01)in down-regulating NGF
SP
CGRP
NKA
NKB
and pERK levels in the BALF.No significant differences were found between the 60min-moxi and 90min-moxi groups in down-regulating NGF
SP
CGRP
NKA
NKB
and pERK levels(P>0.05).Conclusion Suspending-moxibustion stimulation of GV 14 can down-regulate the contents of NGF
SP
CGRP
NKA
NKB
and pERK levels in the BALF in asthma rats
suggesting a relief of neurogenic inflammation reaction after moxibustion.The effect of moxibustion presents a time-dependant manner and peaks at 60 min.
